유청을 이용한 Bacillus subtilis와 Lactobacillus plantarum의 혼합발효를 통한 γ-aminobutyric acid와 생리활성물질 강화 (Fortification of γ-aminobutyric acid and bioactive compounds in whey by co-fermentation using Bacillus subtilis and Lactobacillus plantarum)

  • 본 연구는 모짜렐라 치즈 제조과정에서 분리된 유청을 B. subtilis HA와 L. plantarum EJ2014의 혼합 발효를 통해 ${\gamma}$-PGA, GABA 등의 기능성 물질이 강화된 발효물을 생산하고자 하였다. 유청 B. subtilis 발효 1일차의 시료 분석결과, pH는 6.51, 산도는 0.32%, 생균수는 8.39 log CFU/mL을 나타냈으며, 점질물과 점조도는 각각 6.06%와 $4.09Pas^n$로 발효 전보다 유의적으로 증가하였다. 2차 L. plantarum 발효를 통해 유청 혼합발효물의 최종 pH는 4.57까지 감소하였고 산도는 L. plantarum 발효 1일째 1.39%로 증가하여 최종 산도는 1.73%를 나타내었다. B. subtilis 생균수는 2차 L. plantarum 발효가 진행됨에 따라 5.83 log CFU/mL까지 감소하였으나 L. plantarum 생균수는 5.73 log CFU/mL에서 L. plantarum 발효 1일째 9.08 log CFU/mL로 급격히 증가한 후 L. plantarum 발효 7일까지 유지하였다. TLC 정성분석한 결과 L. plantarum 발효 5일 이후 MSG가 모두 소진되어 GABA로 전환되는 것을 확인할 수 있었다. HPLC 정량분석으로 MSG는 L. plantarum 발효 초기 3.40%에서 L. plantarum 발효 7일째 거의 소진 되면서 혼합발효물의 GABA 함량은 2.21%를 보였다. 환원당과 젖당 함량은 각각 발효 전 11.07과 6.73%에서 L. plantarum 발효 7일째 각각 4.97과 3.68%로 크게 감소하는 것을 보였다. 타이로신 함량은 발효가 진행되는 동안 증가되어 L. plantarum 발효 7일째 38.24 mg%를 나타내었다. 단백질 가수분해 정도를 확인하기 위해 SDS-PAGE를 통해 발효 후 유청 단백질들이 대부분 가수분해되어 저분자화되는 것을 확인하였다. 유청의 발효 전후에 따른 항산화 활성을 측정한 결과 ABTS $RC_{50}$값은 26.81 mg/g에서 발효 후 8.78 mg/g, DPPH $RC_{50}$값 또한 발효 전 17.58 mg/g에서 발효 후 10.38 mg/g으로 감소하면서 혼합발효를 통해 전자 공여능이 향상되는 것으로 확인되었다.

  • 반추가축의 반추위내 미생물단백질은 단백질의 공급원중 일부이며 글루타메이트 생산을 위한 공급원이기도 하다. 글루탐산은 신체의 대사반응, 근육 및 기타 세포구성에 필요한 단백질 합성물질로 이용되며 면역기능항진에도 매우 필수적으로 이용된다. 또한 계면활성제, 완충제, 킬레이트제제, 향미 증강제, 배양배지 및 농업 분야에서 성장촉진제로 이용된다. 글루탐산은 감마-아미노부티르산(GABA)생산을 위한 기질로서 본 연구는 글루탐산의 기능과 글루탐산 탈 탄산효소 유전자를 포함하는 미생물에 대한 정보를 제공하는데 있다. GABA는 체온 조절, 건물섭취량, 유생산량 및 유성분을 개선시키는 것으로 알려져 있다. 대부분의 글루탐산과 GABA 생성 미생물은 대부분 Lactococcus, Lactobacillus, Enterococcus 및 Streptococcus 종과 같은 젖산생성 미생물로 이루어져 있다. 반추위내 대사기전을 보면 GABA 합성을 통해 succinate 생산과정을 거치고, succinate는 탈수소효소반응을 통해 프로피온산과 기타 대사산물을 생산할 수 있다. 또한 Clostridium tetanomorphum과 혐기성 Micrococci는 글루타메이트 발효과정에서 아세트산과 낙산을 생성한다. 프로피온산과 기타 대사산물은 간에서의 혈당으로 전변되어 반추가축의 유선세포에서 유당 및 체중증가를 위한 에너지를 제공한다. 이를 통해 반추가축의 건강상태 개선 및 성장촉진을 위한 중요한 미생물로 이용가능하다.

유산균 발효 다시마(Saccharina japonica) 분말이 첨가된 조미간장의 품질 특성 (Characteristics of Seasoning Soy Sauce with Added Saccharina japonica Powder Fermented by Lactic Acid Bacteria)

  • This study investigated changes in the quality and antioxidant activity of soy sauce with added Saccharina japonica powder (FSP). Soy sauce was prepared with acid-hydrolyzed soy sauce, brewed soy sauce, honey, ginger, garlic, dried red pepper, ${\gamma}$-aminobutyric acid (GABA)-enriched sea tangle fermented in lactic acid, and 1% or 3% S. japonica powder by soy sauce volume. The color, pH, microbiology, amino nitrogen, free amino acids, antioxidant activity, and sensory qualities of the soy sauce were evaluated. There were no significant differences in the general characteristics or antioxidant activity. However, the addition of FSP to the soy sauce enhanced the delicate flavor. Furthermore, soy sauce containing FSP had higher levels of GABA (50 mg/100 mL), a biofunctional ingredient. Therefore, the development of seasoning soy sauce with enhanced functionality seems possible.

청소년기 동물모델에서 구속 스트레스로 유발된 불안, 우울행동에 미치는 영향 (The Alleviating Effects of Sweet Drinks on Restraint Stress-Induced Anxiety and Depressive Behavior in Adolescent Rats)

  • Purpose: Some of the adolescent drinks more sugar-sweetened beverages. However, there is little evidence on the effect of eating behavior on emotional state and neurochemical changes under stress, especially on the levels of typical inhibitory neurotransmitters and gamma-aminobutyric acid. This article demonstrates that sucrose or saccharin drink reduces stress-related behavior responses and GABAergic deficits in adolescent rats. Methods: We randomly assigned 7-weeks-old Sprague-Dawley male rats to three groups: control group (Control), restraint stress only group (Stress), and restraint stress with unrestricted access to saccharin solution (Saccharin) and sucrose solution (Sucrose) as a positive control. We evaluated both anxious and depressive moods using an open field test and forced swim test, respectively. Using western blot analyses, the expression of a GABA-synthesizing enzyme, glutamate decarboxylase-67 (GAD67) and GABAergic markers, including calbindin and parvalbumin was assessed in the prefrontal cortex and the amygdala. Results: We found that both the drinks alleviated anxiety and depressive moods, induced significant attenuation in GAD67 level, and reduced calbindin level under stress in the prefrontal cortex and the amygdala. Conclusion: The results provide an understanding of the effect of sucrose or saccharin drink on stress-related responses. We propose the consumption of sweet drinks as a plausible strategy to alleviate stress-related alterations in adolescents.

Effects of ${\gamma}-Aminobutyric$ Acid on Pancreatic Amylase Secretion Evoked by Sodium Oleate in Anesthetized Rats

  • ${\gamma}-Aminobutyric$ Acid (GABA) is contained in pancreatic islet ${\beta}-cells$ although its physiological role in pancreatic exocrine function is completely unknown at the present time. Recently, we have reported that exogenous GABA enhances secretagogue-evoked exocrine secretion in the isolated, perfused rat pancreas. This study was aimed to investigate an effect of exogenous GABA on pancreatic exocrine secretion in vivo evoked by intestinal stimulation. Rats were anesthetized with urethane (1.4 g/kg) after 24-h fast with free access to water. GABA $(10,\;30\;and\;100\;{\mu}mol/kg/h),$ given intravenously, did not change spontaneous pancreatic amylase secretion but dose-dependently elevated the amylase secretion evoked by intraduodenal sodium oleate (0.05 mmol/h). GABA $(30\;{\mu}mol/kg/h)$ also further increased the amylase secretion stimulated by CCK (30 pmol/kg/h) plus secretin (20 pmol/kg/h) but failed to modify the amylase secretion induced by secretin alone. GABA $(10,\;30\;and\;100\;{\mu}mol/kg/h)$ also dose-dependently elevated pancreatic amylase secretion evoked by CCK alone. Bicuculline $(100\;{\mu}mol/kg/h),$ a $GABA_A-receptor$ antagonist, markedly reduced the GABA-enhanced pancreatic responses to sodium oleate, CCK plus secretin or CCK alone. The results indicate that GABA enhances the sodium oleate-evoked pancreatic amylase secretion via $GABA_A-receptor$ in anesthetized rats, which may account for elevating the action of CCK released by sodium oleate.

Galectin-3-independent Down-regulation of GABABR1 due to Treatment with Korean Herbal Extract HAD-B Reduces Proliferation of Human Colon Cancer Cells

  • Objectives: Many efforts have shown multi-oncologic roles of galectin-3 for cell proliferation, angiogenesis, and apoptosis. However, the mechanisms by which galectin-3 is involved in cell proliferation are not yet fully understood, especially in human colon cancer cells. Methods: To cluster genes showing positively or negatively correlated expression with galectin-3, we employed human colon cancer cell lines, SNU-61, SNU-81, SNU-769B, SNU-C4 and SNU-C5 in high-throughput gene expression profiling. Gene and protein expression levels were determined by using real-time quantitative polymerase chain reaction (PCR) and western blot analysis, respectively. The proliferation rate of human colon cancer cells was measured by using a 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. Results: Expression of ${\gamma}$-aminobutyric acid B receptor 1 (GABABR1) showed a positive correlation with galectin-3 at both the transcriptional and the translational levels. Down-regulation of galectin-3 decreased not only GABABR1 expression but also the proliferation rate of human colon cancer cells. However, Korean herbal extract, HangAmDan-B (HAD-B), decreased expression of GABABR1 without any expressional change of galectin-3, and offset ${\gamma}$-aminobutyric acid (GABA)-enhanced human colon cancer cell proliferation. Conclusions: Our present study confirmed that GABABR1 expression was regulated by galectin-3. HAD-B induced galectin-3-independent down-regulation of GABABR1, which resulted in a decreased proliferation of human colon cancer cells. The therapeutic effect of HAD-B for the treatment of human colon cancer needs to be further validated.

Scopolamine으로 유발한 치매유도 쥐에 대한 저염 오징어 (Todorodes pacificus) 젓갈의 인지 및 기억손상의 개선효과 (Low-salt Todarodes pacificus Jeotgal improves the Learning and Memory Impairments in Scopolamine-induced Dementia Rats)

  • We investigated the effect low salt (2 or 4% salt) concentrations jeotgal made from Todarodes pacificus on the learning and memory impairments in scopolamine-induced (2 mg/kg, i.p.) dementia rats. Rats treated with oral BF-7 (200 mg/kg, p.o.) as a positive control and Todarodes pacificus jeotgal had significantly reduced scopolamine-induced memory deficits in the passive avoidance test. The Morris water maze test or treatment with 2% salt jeotgal made from Todarodes pacificus significantly ameliorated the scopolamine-induced memory deficits in the formation of long- and short-term memory. The acetylcholine content and acetylcholinesterase acitivity paralleled the results of the behavior experiment. There were no significant differences in the brain acetylcholine contents of the experimental groups, while the brain acetylcholine content of the group treated with 2% salt Todarodes pacificus jeotgal was higher than that of the control group. The inhibitory effect of 2% salt jeotgal made from Todarodes pacificus on the acetylcholinesterase activity in the brain was lower than that of the control group. These trends were similar to those of the gamma-aminobutyric acid content. We suggest that Todarodes pacificus jeotgal enhances learning memory and cognitive function by regulating cholinergic enzymes.

지황 품종의 부위별 Aucubin, Catalpol, GABA 함량 (Aucubin, Catalpol, and GABA Contents in Different Plant Parts of Rehmannia glutinosa Cultivars)

  • Background: Rehmannia glutinosa is a perennial herb belonging to the family Scrophulariaceae. Its root has been utilized as a traditional medicine but the aerial parts (flower, flower stalk, leaf) are not used. We aimed to determine the content of three compounds [aucubin, catalpol, and ${\gamma}$-aminobutyric acid (GABA)] in the different organs of R. glutinosa cultivars (Dakang, Tokang, and Suwon 9) Methods and Results: The flower, flower stalk, leaf, and root of R. glutinosa were harvested at the end of August. The aucubin and catalpol contents were analyzed by LC/MS, whereas the GABA content was analyzed by GC/MS. The aucubin content was the highest in the leaf, while catalpol and GABA were the highest in the flower. The aucubin contents of leaf in Dakang, Tokang, and Suwon 9 were 1.43, 0.81, and 1.07 mg/g, respectively. The catalpol contents of flower in Dakang, Tokang, and Suwon 9 were 41.06, 28.78, and 37.48 mg/g, respectively, the GABA contents were 0.79, 0.76, and 0.65 mg/g, respectively. Conclusions: The aucubin, catalpol, and GABA contents were higher in the leaf and flower than that in the root. This study show that R. glutinosa leaf and flower can be used as a potential supplement.

큰눈벼와 일품벼의 발아에 의한 생리활성물질 함량 및 암세포 증식억제활성 변화 (Changes in Phytochemical Content and Antiproliferative Activity of Germinated Geunnun and Ilpum Rice Varieties)

  • 본 연구에서는 현미의 발아를 통해 큰눈벼와 일품벼 두 품종에서의 GABA(${\gamma}$-aminobutyric acid) 함량, 항산화성분 함량 및 암세포 증식억제 활성의 변화를 측정하고자 하였다. 연구 결과 두 품종의 현미는 발아 과정을 통해 GABA 함량이 증가하였으며, 특히 일품벼의 미강층에서 발아 후 가장 큰 증가율을 나타내었다. 항산화 성분의 경우, 큰눈벼에서는 발아 후 polyphenol 함량이 증가하였으나, flavonoid 함량의 경우 두 품종 모두 발아 후 감소하는 것으로 나타났다. 암세포 증식억제활성 측정 결과, 폐암 세포주의 경우 두 품종 모두 현미상태에서는 발아 후 암세포 증식 억제활성이 증가하였으며, 위암 세포주에서 발아 후에 암세포 증식 억제활성이 보다 증가하였다. 따라서 현미의 발아 시 항산화 성분이 용출되는 현상을 방지하는 조건이 확립된다면, 향후 발아현미는 건강기능식품 및 건강보조제 등을 위한 기능성 식품 소재로 이용될 수 있을 것으로 사료된다.

Effects of ${\gamma}-Aminobutyric$ Acid on Intrinsic Cholinergic Action in Exocrine Secretion of Isolated, Perfused Rat Pancreas

  • ${\gamma}$-Aminobutyric acid (GABA) has been reported to enhance exocrine secretion evoked not only by secretagogues but also by intrinsic neuronal excitation in the pancreas. The pancreas contains cholinergic neurons abundantly that exert a stimulatory role in exocrine secretion. This study was undertaken to examine effects of GABA on an action of cholinergic neurons in exocrine secretion of the pancreas. Intrinsic neurons were excited by electrical field stimulation (EFS; 15 V, 2 msec, 8 Hz, 45 min) in the isolated, perfused rat pancreas. Tetrodotoxin or atropine was used to block neuronal or cholinergic action. Acetylcholine was infused to mimic cholinergic excitation. GABA $(30{\mu}M)$ and muscimol $(10{\mu}M)$, given intra-arterially, did not change spontaneous secretion but enhanced cholecystokinin (CCK; 10 pM)-induced secretions of fluid and amylase. GABA (3, 10, $30{\mu}M$) further elevated EFS-evoked secretions of fluid and amylase dose-dependently. GABA (10, 30, $100{\mu}M$) also further increased acetylcholine $(5{\mu}M)$-induced secretions of fluid and amylase in a dose-dependent manner. Bicuculline $(10{\mu}M)$ effectively blocked the enhancing effects of GABA $(30{\mu}M)$ on the pancreatic secretions evoked by either EFS or CCK. Both atropine $(2{\mu}M)$ and tetrodotoxin $(1{\mu}M)$ markedly reduced the GABA $(10{\mu}M)$-enhanced EFS- or CCK-induced pancreatic secretions. The results indicate that GABA enhances intrinsic cholinergic neuronal action on exocrine secretion via the $GABA_A$ receptors in the rat pancreas.
