Enriching CCL3 in the Tumor Microenvironment Facilitates T cell Responses and Improves the Efficacy of Anti-PD-1 Therapy

  • Chemokines are key factors that influence the migration and maintenance of relevant immune cells into an infected tissue or a tumor microenvironment. Therefore, it is believed that the controlled administration of chemokines in the tumor microenvironment may be an effective immunotherapy against cancer. Previous studies have shown that CCL3, also known as macrophage inflammatory protein 1-alpha, facilitates the recruitment of dendritic cells (DCs) for the presentation of tumor Ags and promotes T cell activation. Here, we investigated the role of CCL3 in regulating the tumor microenvironment using a syngeneic mouse tumor model. We observed that MC38 tumors overexpressing CCL3 (CCL3-OE) showed rapid regression compared with the wild type MC38 tumors. Additionally, these CCL3-OE tumors showed an increase in the proliferative and functional tumor-infiltrating T cells. Furthermore, PD-1 immune checkpoint blockade accelerated tumor regression in the CCL3-OE tumor microenvironment. Next, we generated a modified CCL3 protein for pre-clinical use by fusing recombinant CCL3 (rCCL3) with a non-cytolytic hybrid Fc (HyFc). Administering a controlled dose of rCCL3-HyFc via subcutaneous injections near tumors was effective in tumor regression and improved survival along with activated myeloid cells and augmented T cell responses. Furthermore, combination therapy of rCCL3-HyFc with PD-1 blockade exhibited prominent effect to tumor regression. Collectively, our findings demonstrate that appropriate concentrations of CCL3 in the tumor microenvironment would be an effective adjuvant to promote anti-tumor immune responses, and suggest that administering a long-lasting form of CCL3 in combination with PD-1 blockers can have clinical applications in cancer immunotherapy.

  • FPGA(Field Programmable Gate Array) devices are widely used due to their merits in circuit development time, and development cost. Among various FPGA technologies, SRAM-based FPGAs have large cell capacity so that they are attractive for complex circuit design and their reconfigurability. However, they are weak in space environment where radiation energy particles cause Single Event Upset(SEU). In this paper, we designed a controller supervising SRAM-based FPGA to protect configuration memory inside. The controller is implemented on an Anti-Fusing FPGA. Radiation test was performed on the implemented computer board and the result show that our controller provides better SEU-resilience than TMR-only system.

  • The topic of 21st century's culture is the appearance of digital media. It made changes as big as the industrial revolution, and our society is now ruled by the digital media. The main objective of this study is to forecast the direction of current visual design education by researching and analyzing how the introduction of digital media is influencing the evolution of visual design's identity, which is an ever changing and developing science. Also, since the rain target of digital media is the young generation, the change in the method of expressing visual language is inevitable In fact, there have been a lot of changes in the methods of creating and distributing visual communication due to the introduction of digital media. In the past, most educational institutions of design had similar objectives, curriculums and teaching methods to provide education that prepares students for practical business. However, in this digital media era, the application and utilization of visual design are uncomparably diversified, and it is generally classified as interaction. The purpose of this study is to find a wat to train visual design professionals in this digital era. For this purpose, this study will identify a new educational system that fulfills the demands of this society by fusing the traditional education and the new digital education, and will suggest what an design education institute that is ahead of the demands of society should be like.

Efficient Fusion Method to Recognize Targets Flying in Formation (편대비행 표적식별을 위한 효과적인 ISAR 영상 합성 방법)

  • This paper proposes a novel method for the recognition of the inverse synthetic aperture radar(ISAR) image of multiple targets flying in formation. Rather than separating the ISAR image of each target, the proposed method combines an ISAR image obtained by fusing the ISAR images in the training database. Fusion is conducted by optimizing the non-linear problem whose parameters are the aspect angle and the target location. Assuming that the aspect angle is properly estimated, the proposed method estimates the number of the targets and their locations by optimizing the template matching using PSO. In simulations using the F-16 scale model, the efficiency of the proposed method was demonstrated by yielding the ISAR image identical to that of targets in formation.

  • STATs activated by various cytokine and growth factors trigger a quick response in the nucleus and induce changes in gene expression. We have found the sequences homologous to STAT binding site in the 5'-flanking region of the D-raf gene. In this study, we examined role of the STAT binding site in D-raf gene promoter activity in vivo by using transgenic flies. The reporter plasmid pDraf-STATmut-lacZ was constructed by fusing D-raf promoter fragment having the base-substituted STAT binding site with the lacZ gene in a P-element vector. Transgenic flies bearing the Draf-STATmut-lacZ fusion genes were established by P-element mediated transformation. The expression of lacZ in transgenic flies bearing Draf-STATmut-lacZ fusion genes carrying base substitution in STAT site throughout various developmental stages was extensively reduced in comparison with that in transgenic flies bearing wild type Draf-lacZ fusion gene. These results show that the STAT binding site plays an important role in regulation of the D-raf gene.

  • Galacto-oligosaccharides (GOS) are prebiotics that have a beneficial effect on human health by promoting the growth of probiotic bacteria in the gut, in addition to having various applications in the food industry. GOS are generally produced from lactose in a reaction catalyzed by β-galactosidase. Synthesis of GOS from whey permeate (WP) (ultrafiltration of whey, concentrated then spray dried) using surface engineered β-galactosidase in Pichia pastoris (P. pastoris) is a novel method to convert waste into a valuable product. Cell-surface display is the expression of peptides and proteins on the surface of living cells by fusing them to functional components of cells. Surface engineered cells have many potential uses. The Flo1p flocculation functional domain, thought to be located near the N terminus, recognizes and adheres non-covalently to cell-wall components such as α-mannan carbohydrates, causing reversible aggregation of cells into flocs.

Biosynthesis of a Biological Active Single Chain Equine Chorionic Gonado-tropin

  • The equine chorionic gonadotropin (eCG) subunits $\alpha$ and ${\beta}$ are transcribed from different genes and associate noncovalently to form the bioactive eCG heterodimer. Dimerization is rate limiting for eCG secretion, and dissociation leads to hormone inactivation. The correct conformation of the heterodimer is alto important for efficient secretion, hormone-specific post-translational modifications, receptor binding and signal transduction. To determine whether ${\alpha}$ and ${\beta}$ subunits can be synthesized as a single polypeptide chain (tethered-eCG) and also display biological activity, the tethered-eCG molecule by fusing the carboxyl terminus of the eCG ${\beta}$-subunit to the amino terminus of the af-subunit was construe-ted and transfected into chinese hamster ovary (CHO-Kl) cells. LH- and FSH-like activities were assayed in terms of testosterone production and aromatase activity in primary cultured rat Leydig cells and granulosa cells, respectively. The tethered-eCG was efficiently secreted and showed similar LH-like activity to the dimeric eCG ${\alpha}$/${\beta}$ and native eCG. FSH-like activity of the tethered-eCG was also shown similarly in comparison with the native and wild type eCG ${\alpha}$/${\beta}$. Our data for the first time suggest that the tethered-eCG can be expressed efficiently and the produced product by the CHO-K1 cells is fully LH- and FSH-like activities in rat in vitro bioassay system. Our results also suggest that this molecular can imply particular models of FSH-like activity not LH-like activity in the eCG. Taken together, these data indicate that the constructs of tethered molecule will be useful in the study of mutants that affect subunit association and/or secretion.

A Statistical Analysis of JERS L-band SAR Backscatter and Coherence Data for Forest Type Discrimination

  • Synthetic aperture radar (SAR) from satellites provides the opportunity to regularly incorporate microwave information into forest classification. Radar backscatter can improve classification accuracy, and SAR interferometry could provide improved thematic information through the use of coherence. This research examined the potential of using multi-temporal JERS-l SAR (L band) backscatter information and interferometry in distinguishing forest classes of mountainous areas in the Northeastern U.S. for future forest mapping and monitoring. Raw image data from a pair of images were processed to produce coherence and backscatter data. To improve the geometric characteristics of both the coherence and the backscatter images, this study used the interferometric techniques. It was necessary to radiometrically correct radar backscatter to account for the effect of topography. This study developed a simplified method of radiometric correction for SAR imagery over the hilly terrain, and compared the forest-type discriminatory powers of the radar backscatter, the multi-temporal backscatter, the coherence, and the backscatter combined with the coherence. Statistical analysis showed that the method of radiometric correction has a substantial potential in separating forest types, and the coherence produced from an interferometric pair of images also showed a potential for distinguishing forest classes even though heavily forested conditions and long time separation of the images had limitations in the ability to get a high quality coherence. The method of combining the backscatter images from two different dates and the coherence in a multivariate approach in identifying forest types showed some potential. However, multi-temporal analysis of the backscatter was inconclusive because leaves were not the primary scatterers of a forest canopy at the L-band wavelengths. Further research in forest classification is suggested using diverse band width SAR imagery and fusing with other imagery source.

A Study for Cyber Situation Awareness System Development with Threat Hunting (위협 헌팅을 적용한 사이버 상황인식 시스템 개발에 관한 연구)

  • Threat hunting is defined as a process of proactively and iteratively searching through networks to detect and isolate advanced threats that evade existing security solutions. The main concept of threat hunting is to find out weak points and remedy them before actual cyber threat has occurred. And HMM(Hunting Maturity Matrix) is suggested to evolve hunting processes with five levels, therefore, CSOC(Cyber Security Operations Center) can refer HMM how to make them safer from complicated and organized cyber attacks. We are developing a system for cyber situation awareness system with pro-active threat hunting process called unMazeTM. With this unMaze, it can be upgraded CSOC's HMM level from initial level to basic level. CSOC with unMaze do threat hunting process not only detecting existing cyber equipment post-actively, but also proactively detecting cyber threat by fusing and analyzing cyber asset data and threat intelligence.

3D Reconstruction of Structure Fusion-Based on UAS and Terrestrial LiDAR (UAS 및 지상 LiDAR 융합기반 건축물의 3D 재현)

  • Digital Twin is a technology that creates a photocopy of real-world objects on a computer and analyzes the past and present operational status by fusing the structure, context, and operation of various physical systems with property information, and predicts the future society's countermeasures. In particular, 3D rendering technology (UAS, LiDAR, GNSS, etc.) is a core technology in digital twin. so, the research and application are actively performed in the industry in recent years. However, UAS (Unmanned Aerial System) and LiDAR (Light Detection And Ranging) have to be solved by compensating blind spot which is not reconstructed according to the object shape. In addition, the terrestrial LiDAR can acquire the point cloud of the object more precisely and quickly at a short distance, but a blind spot is generated at the upper part of the object, thereby imposing restrictions on the forward digital twin modeling. The UAS is capable of modeling a specific range of objects with high accuracy by using high resolution images at low altitudes, and has the advantage of generating a high density point group based on SfM (Structure-from-Motion) image analysis technology. However, It is relatively far from the target LiDAR than the terrestrial LiDAR, and it takes time to analyze the image. In particular, it is necessary to reduce the accuracy of the side part and compensate the blind spot. By re-optimizing it after fusion with UAS and Terrestrial LiDAR, the residual error of each modeling method was compensated and the mutual correction result was obtained. The accuracy of fusion-based 3D model is less than 1cm and it is expected to be useful for digital twin construction.
