Aflatoxins are classified as Group 1 (carcinogenic to humans) by the International Agency for Research on Cancer. In this study, a total of 134 fungal strains were isolated from 65 meju samples, and two fungal isolates were selected as potential aflatoxin $B_1$ ($AFB_1$)-biodetoxification fungi. These fungi were identified as Aspergillus oryzae MAO103 and A. oryzae MAO104 by sequencing the beta-tubulin gene. The two A. oryzae strains were able to degrade more than 90% of $AFB_1$ (initial concentration: $40{\mu}g/l$) in a culture broth in 14 days. The mutagenic effects of $AFB_1$ treated with A. oryzae MAO103 and MAO104 significantly decreased to 5.7% and 6.4%, respectively, in the frame-shift mutation of Ames tests using Salmonella typhimurium TA98. The base-substituting mutagenicity of $AFB_1$ was also decreased by the two fungi. Moreover, $AFB_1$ production by Aspergillus flavus was significantly decreased by the two A. oryzae strains on soybean-based agar plates. Our data suggest that the two $AFB_1$-detoxifying A. oryzae strains have potential application to control $AFB_1$ in foods and feeds.

Yeasts that are present in marine environments have evolved to survive hostile environments that are characterized by high exogenous salt content, high concentrations of inhibitory compounds, and low soluble carbon and nitrogen levels. Therefore, yeasts isolated from marine environments could have interesting characteristics for industrial applications. However, the application of marine yeast in research or industry is currently very limited owing to the lack of a suitable isolation method. Current methods for isolation suffer from fungal interference and/or low number of yeast isolates. In this paper, an efficient and non-laborious isolation method has been developed and successfully isolated large numbers of yeasts without bacterial or fungal growth. The new method includes a three-cycle enrichment step followed by an isolation step and a confirmation step. Using this method, 116 marine yeast strains were isolated from 14 marine samples collected in the UK, Egypt, and the USA. These strains were further evaluated for the utilization of fermentable sugars (glucose, xylose, mannitol, and galactose) using a phenotypic microarray assay. Seventeen strains with higher sugar utilization capacity than the reference terrestrial yeast Saccharomyces cerevisiae NCYC 2592 were selected for identification by sequencing of the ITS and D1/D2 domains. These strains belonged to six species: S. cerevisiae, Candida tropicalis, Candida viswanathii, Wickerhamomyces anomalus, Candida glabrata, and Pichia kudriavzevii. The ability of these strains for improved sugar utilization using seawater-based media was confirmed and, therefore, they could potentially be utilized in fermentations using marine biomass in seawater media, particularly for the production of bioethanol and other biochemical products.
Objective: As the climate changes, it influences ruminant's feed intake, nutrient digestibility, rumen methane production and emission. This experiment aimed to evaluate the effect of feeding Sweet grass (Pennisetum purpureum cv. Mahasarakham; SG) as a new source of good quality forage to improve feed utilization efficiency and to mitigate rumen methane production and emission. Methods: Four, growing crossbred of Holstein Friesian heifers, 14 months old, were arranged in a $4{\times}4$ Latin square design to receive four dietary treatments. Treatment 1 (T1) was rice straw (RS) fed on ad libitum with 1.0% body weight (BW) of concentrate (C) supplementation (RS/1.0C). Treatment 2 (T2) and treatment 3 (T3) were SG, fed on ad libitum with 1.0% and 0.5% BW of concentrate supplementation, respectively (SG/1.0C and SG/0.5C, respectively). Treatment 4 (T4) was total Sweet grass fed on ad libitum basis with non-concentrate supplementation (TSG). Results: The results revealed that roughage and total feed intake were increased with SG when compared to RS (p<0.01) while TSG was like RS/1.0C treatment. Digestibility of nutrients, nutrients intake, total volatile fatty acids (VFAs), rumen microorganisms were the highest and CH4 was the lowest in the heifers that received SG/1.0C (p<0.01). Total dry matter (DM) feed intake, digestibility and intake of nutrients, total VFAs, $NH_3-N$, bacterial and fungal population of animals receiving SG/0.5C were higher than those fed on RS/1.0C. Reducing of concentrate supplementation with SG as a roughage source increased $NH_3-N$, acetic acid, and fungal populations, but it decreased propionic acid and protozoal populations (p<0.05). However, ruminal pH and blood urea nitrogen were not affected by the dietary treatments (p>0.05). Conclusion: As the results, SG could be a good forage to improve rumen fermentation, decrease methane production and reduced the level of concentrate supplementation for growing ruminants in the tropics especially under global climate change.
In this study, five endophytic fungi belonging to the Aspergillus and Alternaria genera were isolated from Lagopsis supina. The antimicrobial activity of all fungal fermented extracts against Staphylococcus and Fusarium graminearum was tested using the cup-plate method. Among them, Aspergillus ochraceus XZC-1 showed the best activity and was subsequently selected for large-scale fermentation and bioactivity-directed separation of the secondary metabolites. Four compounds, including 2-methoxy-6-methyl-1,4-benzoquinone (1), 3,5-dihydroxytoluene (2), oleic acid (3), and penicillic acid (4) were discovered. Here, compounds 1 and 4 displayed anti-fungal activity against F. graminearum, F. oxysporum, F. moniliforme, F. stratum, Botrytis cinerea, Magnaporthe oryzae, and Verticillium dahlia with diverse MIC values (128-512 ㎍/ml), which were close to that of the positive control antifungal, actidione (64-128 ㎍/ml). Additionally, compounds 1 and 4 also exhibited moderate antibacterial activity against S. aureus, Listeria monocytogenes, Escherichia coli, and Salmonella enterica, with low MIC values (8-64 ㎍/ml). Moreover, compounds 1 and 4 displayed selective cytotoxicity against cancer cell lines as compared with the normal fibroblast cells. Therefore, this study proposes that the endophytic fungi from L. supina can potentially produce bioactive molecules to be used as lead compounds in drugs or agricultural antibiotics.

This study investigates the changes in inorganic composition and the microbial counts during the process of fermentation of mixed domestic organic resources for the development of alternatives for imported oil cake, and examines the characteristics of mixed fermentation organic fertilizer (MFOF). The effect of the MFOF on the lettuce growth is investigated in order to evaluate the possibility of replacing the existing mixed oil cake with the MFOF. Six kinds of domestic by-product resources, which are rice bran, distiller's dried grains, sesame meal, fish meal, and spent mushroom substrate, are mixed by mixing ratio and the composition was analyzed during the fermentation process for 90 days under moisture content 30% and sealed condition. During the 90 days of fermentation, the pH change of the MFOF was little, and the moisture content was maintained at 34-35% until the 60th day of fermentation, and then decreased to 30-31% on the 90th day. Total nitrogen content remained unchanged during the fermentation period, but total carbon content showed a significant difference on the 21st day of fermentation. It was confirmed that the content of fertilizer composition (nitrogen, phosphate, and potash) of the MFOF was 8.7% or more, which is suitable for the minimum amount standard of the main nutrients to be contained in the organic fertilizer. During the fermentation process of organic fertilizer, the density of bacteria and actinomycetes increased until 60 days and 30 days, respectively, and thereafter little changes were shown, and fungal population showed an increasing trend. As a result of lettuce cultivation test in the greenhouse by applying the MFOF, the growth and yield were comparable to that of using the existing mixed oil cake fertilizer when 100% was applied based on crop standard nitrogen fertilizer level. The use of mixed fermentation organic fertilizer made with domestic by-product resources can be used for use in farms in the future and is expected to contribute to the stable production of environment friendly agricultural products.
In order to produce clear and favorable citrus wine from Citrus unshiu produced in Cheju island, chemical and microbiological processes for alcoholic fermentation were investigated. The ratio of pressed juice passed below 100 mesh sieve and peel of mandarin orange were 55.9% and 25.6% respectively. Orange juice for fermentation source contained 8.85% total sugar, 1.43% total acid and 0.056% volatile acid. Pressed juice was adjusted to 24 degree Brix with cane sugar, and was fermented at $20^{\circ}C$ for one month. Starter screened and selected was Saccharomyces cerevisiae IAM 4274. As principal fermentation proceeded for one week, suspended solids began to precipitate slowly after then. After fermentation, clear citrus wine consisted of about 8 degree Brix residual sugar, $13.3{\sim}14.4%$ ethanol, $0.78{\sim}1.11%$ total acid, $0.05{\sim}0.07%$ methanol and $2.25{\sim}3.29%$ extract, was obtained. Color, flavor and taste of citrus wine found good with panel test. Citrus wine which was treated with fungal enzyme derived from Aspergillus niger CCM-4 was cleared much faster, and could be filtered more rapidly than the untreated. The enzyme-producing strain was isolated from field soil of Cheju island and identified.
An experiment was conducted to study the effect of ionophore enriched cold processed mineral block supplemented with urea molasses on microbial growth and rumen fermentation. Twelve adult male crossbred cattle were divided into four groups on body weight basis. Animals were given wheat straw as a basal diet. The animals of group I and II were supplemented with concentrate mixture and animals of group III and IV were supplemented with cold processed urea molasses mineral block (UMMB). Thirty mg monensin/day/animal were supplemented to the animals of group II and 35 ppm monensin were incorporated in the UMMB supplemented to the animals of group IV. Dry matter (DM) intake did not differ significantly among groups. Mean rumen pH was higher in UMMB fed animals. Total volatile fatty acids (TVFA) concentration (mmole/L strained rumen liquor (SRL) in group III (113.19) was significantly (p<0.05) higher than those of group I (105.83) and II (108.74) but similar to group IV (109.34). TVFA production (mole/day) was similar in all the groups. The molar proportion of acetate was significantly (p<0.01) higher in the group I (59.56) than those of group II (51.73) and IV (55.91) but similar to group III (57.12). The molar proportion of propionate was significantly (p<0.01) higher in the monensin treated groups i.e. group II (38.38) and IV (36.26) than those of group I (27.78) and III (33.06). Butyrate molar percent was significantly (p<0.01) higher in group I (12.65) than those of group II (10.19), group III (9.83) and IV (7.84). The reduction of acetate and butyrate was due to UMMB and monensin resulted in lower A:P ratio. Average bacterial pool and bacterial production rate did not differ significantly among groups. Total N concentration (mg/100 ml SRL) was significantly (p<0.01) higher in the group I (55.30) and III (57.70) as compared to the group II (47.97) and IV (47.59). Ammonia-N concentration (mg/100 ml SRL) of group III (34.99) was significantly (p<0.01) higher than that of the group I (25.76) which was again significantly (p<0.01) higher than that of the group II (20.79) and IV (19.83) indicating slower release of ammonia due to monensin in diet. Total bacterial, cellulolytic, proteolytic bacterial and fungal count at 4 h post feeding did not differ significantly (p<0.05) among treatment groups. However, methanogenic bacterial count was significantly (p<0.01) higher in the group I (11.80) compared to the group II (8.43) which was significantly (p<0.01) higher than that of the group III (4.70) and IV (2.90). Average protozoal population was affected by both treatments. Thus feeding of UMMB and monensin in diet affected the rumen fermentation pattern towards propionate production, slower release of ammonia and reduction in methanogenic bacteria in the rumen.
The fungi play an important role in smell and taste during the fermentation of salami. In this study, the fungi from the salami casing during the fermentation process in Korea were isolated. They were examined by morphological and molecular methods and were identified as Aspergillus cibarius, Penicillium echinulatum, Cladosporium sphaerospermum. Of these, A. cibarius formed white or light green conidial heads on salami casing and it was widely distributed through almost whole casing. P. echinulatum occurred sparsely on the casing of the salami and formed white masses. C. sphaerospermum occurred rarely and formed black spots. It is considered that additional studies on their roles on salami are required.
A systematic investigation was performed on the bacterial, Bacillus, fungal, and yeast communities of the three types of Daqu (mechanically prepared, manually prepared, and mixed prepared) used in Baiyunbian Company by reconditioning PCR-denaturing gradient gel electrophoresis (PCR-DGGE). The DGGE results showed that the microbes in the three types of Daqu were mainly thermotolerant and thermophilic microbes, and the most dominant bacterial species were Bacillus and Virgibacillus, followed by Lactobacillus and Trichococcus. Furthermore, the dominant fungi were found to be molds, such as Rasamsonia, Penicillium, Aspergillus, and Monascus, and the dominant yeasts were Saccharomyces cerevisiae, Saccharomycopsis fibuligera, Pichia anomala, and Debaryomyces hansenii. In general, the three types of Daqu showed slight differences in microbial communities, and the Shannon indexes (H') of the manually prepared and mechanically prepared Daqu were similar. The results suggest that mechanically prepared Daqu can replace manually prepared Daqu in liquor production, and this research provides useful information for liquor production and process improvement.
Two typical microbial communities from Chinese rice wine fermentation collected in Yichang city and Suzhou city in China were investigated. Both communities could ferment glutinous rice to rice wine in 2 days. The sugar and ethanol contents were 198.67 and 14.47 mg/g, respectively, for rice wine from Yichang city, and 292.50 and 12.31 mg/g, respectively, for rice wine from Suzhou city. Acetic acid and lactic acid were the most abundant organic acids. Abundant fungi and bacteria were detected in both communities by high-throughput sequencing. Saccharomycopsis fibuligera and Rhizopus oryzae were the dominant fungi in rice wine from Suzhou city, compared with R. oryzae, Wickerhamomyces anomalus, Saccharomyces cerevisiae, Mucor indicus, and Rhizopus microsporus in rice wine from Yichang city. Bacterial diversity was greater than fungal diversity in both communities. Citrobacter was the most abundant genus. Furthermore, Exiguobacterium, Aeromonas, Acinetobacter, Pseudomonas, Enterobacter, Bacillus, and Lactococcus were highly abundant in both communities.
