Purpose: Foot and ankle disease (FAD) is a frequent cause of morbidity among the homeless population. Various conditions, exacerbated by malnutrition, poor lifestyle habits, psychiatric disorders, physical injuries, poor hygiene, and limited access to healthcare, have been described in this population. The purpose of this study was to investigate the cognition and management status of FAD in shelter based homeless people. Materials and Methods: Fifty-two male and twenty-five female volunteer homeless individuals were recruited from two homeless shelters. Each person completed a questionnaire assessing any presence of pain, pain management, as well as foot and ankle care status. A foot and ankle surgeon examined the physical status of the individual's foot and ankle, including tenderness and instability. A radiologic evaluation was done for 18 male and 11 female homeless people who agreed to participate in this test for the existence osteophyte or joint space narrowing representing osteoarthritis and some reference angles for hallux valgus, flatfoot and cavus foot. Results: Homeless people had higher prevalence of body mass index, diabetes, and smoking than the general population. The most prevalent infectious disease was fungal infection (male 78%, female 68%), with a low compliance of management for FAD. Conclusion: Although most of shelter-based homeless people showed an acceptable foot and ankle management status, the potential risk for FAD development and exacerbation of mild FAD was high.

The primary cilium is a non-motile microtubule-based organelle that protrudes from the surface of most human cells and works as a cellular antenna to accept extracellular signals. Primary cilia assemble from the basal body during the resting stage ($G_0$ phase) and simultaneously disassemble with cell cycle re-entry. Defective control of assembly or disassembly causes diverse human diseases including ciliopathy and cancer. To identify the effective compounds for studying primary cilium disassembly, we have screened 297 natural compounds and identified 18 and 17 primary cilium assembly and disassembly inhibitors, respectively. Among them, the application of KY-0120, identified as Brefeldin A, disturbed Dvl2-Plk1-mediated cilium disassembly via repression of the interaction of $CK1{\varepsilon}-Dvl2$ and the expression of Plk1 mRNA. Therefore, our study may suggest useful compounds for studying the cellular mechanism of primary cilium disassembly to prevent ciliopathy and cancer.
Parasitic leeches could directly (through causing poor growth, anemia and wound in the fish) and indirectly (by predisposition of the fish to secondary bacterial and fungal infections) affects their hosts. In the present study, fishes that were attacked by leeches in natural and experimental environment were studied. Pathologic samples were obtained from damages at the site of leech bite, as well as kidney and liver of the fish. Histopathological examination revealed numerous lesions at the site of leech bite including tissue demolition, detachment at the site of leech bite in the epidermis of epithelial tissue in the skin, destructed nucleus in epithelial cells of the skin plus necrosis in the damaged skin and weak inflammatory penetration to acute necrotic damages along with piercing dermis layer. Pathologic lesions in the kidney included some changes such as proliferation by increasing glomerular cells and membrane cells in capillary vein of the kidney, blood cell necrosis in kidney with infiltration of white blood cells mainly mononuclear and less polymorphonuclear which are the symptoms of anemia due to blood feeding and sucking by leeches. There was also a chronic kidney infection probably originated from another part of body such as skin. Moreover, leeches caused hemorrhagic anemia due to blood consumption of the hosts, which led to observation of immature red blood cells. Also results showed that diseases induced by leeched in fish could be acute or chronic, which depends on size of fish, species of leech and severity of infection.

Chitinase-producing Paenibacillus elgii strain HOA73 has been used to control plant diseases. However, the antimicrobial activity of its extracellular chitinase has not been fully elucidated. The major extracellular chitinase gene (PeChi68) from strain HOA73 was cloned and expressed in Escherichia coli in this study. This gene had an open reading frame of 2,028 bp, encoding a protein of 675 amino acid residues containing a secretion signal peptide, a chitin-binding domain, two fibronectin type III domains, and a catalytic hydrolase domain. The chitinase (PeChi68) purified from recombinant E. coli exhibited a molecular mass of approximately 68 kDa on SDS-PAGE. Biochemical analysis indicated that optimum temperature for the actitvity of purified chitinase was $50^{\circ}C$. However, it was inactivated with time when it was incubated at $40^{\circ}C$ and $50^{\circ}C$. Its optimum activity was found at pH 7, although its activity was stable when incubated between pH 3 and pH 11. Heavy metals inhibited this chitinase. This purified chitinase completely inhibited spore germination of two Cladosporium isolates and partially inhibited germination of Botrytis cinerea spores. However, it had no effect on the spores of a Colletotricum isolate. These results indicate that the extracellular chitinase produced by P. elgii HOA73 might have function in limiting spore germination of certain fungal pathogens.
Implantation of allografts has increased widely with not only the availability of many allogenic bone but also allogenic soft tissues. The aim of tissue banking is to provide surgeons with safe tissues compatible with their intended clinical application. The incidence of tissue transplant-transmitted infection is unknown and can only be inferred from prospective studies. The possibility of donor-to-recipient disease transmission through soft tissue transplantation can be considered by reviewing the risk associated with other transplanted hard tissues. Viral, bacterial, and fungal infections have been transmitted via transplantation of soft tissue allografts such as skin, cornea, dura, pericardium. fascia lata, and heart valves. Corneas have transmitted rabies, Creutzfeldt-Jakob disease (CJD), hepatitis B (HBV), cytomegalovirus (CMV), herpes simplex virus (HSV), bacteria, and fungi. Heart valves have been implicated in transmitting tuberculosis, hepatitis B. HIV-1 and CMV. CJD has been transmitted by dura and pericardium transplants. Skin has transmitted CMV, bacteria, and fungi. Cadaveric skin, pericardium, dura, and fascia lata have been used in dental patients with intra-oral soft tissue injuries and GBR. This study is review of the considering transmission of infectious disease in allogenic soft tissues and guidelines of reducing the risk. Prior to use, many tissues are exposed to antibiotics, disinfectants, and sterilants, which further reduce or remove the risk of transmitted disease. Because some soft tissue grafts cannot be subjected to sterilization steps, the risk of infectious disease transmission remains and thorough donor screening and testing is especially important.
Amphotericin B (AmB) is a drug of choice for the treatment of systemic fungal diseases, but its use is considerably limited due to a high incidence of toxicity, particularly nephrotoxicity. It has been demonstrated that the toxicity of AmB is caused by self-aggregated species of the drug and that unaggregated (monomeric) drug is nontoxic but still expresses antifungal activity. Poly (ethylene oxide) (PEO) is a water-soluble polymer, which may impact the aggregation state of AmB. We have studied the aggregation state of AmB as a function of PEO molecular weight and concentration. At 3,000 and 8,000 g/mole, there was minimal or no change of critical aggregation concentration (CAC) of AmB regardless of the concentration of polymer. By contrast at 20,000 g/mole, the CAC of AmB strikingly increased to 24.3 and $37.5\;{\mu}M$ at 5.0% and 10 % w/v of polymer, respectively. The critical overlap concentration (COC) of PEO 20,000 g/mole was 5.5%. It appears that an interaction between monomeric AmB and polymer coil increases above the COC, competing with self-aggregation of the drug. Accordingly, the degree of aggregation of AmB stayed low and the toxicity became less. There was no such effect at 3,000 and 8,000 g/mole of PEO, owing perhaps to small dimensions in comparison to AmB. Based upon these findings, less toxic AmB formulation may be developed by a pharmaceutical technique such as solid dispersion system containing both AmB and PEO 20,000 g/mole.
Malassezia globosa is a common pathogenic fungus that causes skin diseases including dandruff and seborrheic dermatitis in humans. Analysis of its genome identified a gene (MGL_1677) coding for a putative NADPH-P450 reductase (NPR) to support the fungal cytochrome P450 enzymes. The heterologously expressed recombinant M. globosa NPR protein was purified, and its functional features were characterized. The purified protein generated a single band on SDS-PAGE at 80.74 kDa and had an absorption maximum at 452 nm, indicating its possible function as an oxidized flavin cofactor. It evidenced NADPH-dependent reducing activity for cytochrome c or nitroblue tetrazolium. Human P450 1A2 and 2A6 were able to successfully catalyze the O-deethylation of 7-ethoxyresorufin and the 7-hydroxylation of coumarin, respectively, with the support of the purified NPR. These results demonstrate that purified NPR is an orthologous reductase protein that supports cytochrome P450 enzymes in M. globosa.

Background: Atopic dermatitis one of the most common chronic skin diseases, is caused by various environmental and genetic factors. Methods: A total of 2609 healthy newborns who were enrolled in the COCOA study (COCOA) from 2008 to 2015 were surveyed for indoor environmental exposure to fungi during gestation and then diagnosed postnatally for atopic dermatitis. The fungi collected during the gestation of 20 normal subjects and 20 infants that developed atopic dermatitis were identified using Illumina's MiSeq platform and analyzed for their diversity and species. Results: A total of 2,609 respondents were surveyed (52.8% male and 47.2% female) Children, 1, 2, and 3 years old diagnosed with atopic dermatitis comprised 15.2%, 15.7%, and 14.1% of the respondents, respectively. The prevalence of exposure to mold during gestation was 1.46 (95% CI, 1.05-2.04) and 1.52 (95% CI, 0.95-2.43), in the first and third years after birth, respectively. One-year-old children with atopic dermatitis and no fungal markers detected in the bathroom environment during gestation accounted for less than 5% (aOR, 1.51; 95%CI, 0.96-2.38) and in the group less than 5 ~ 30% (aOR, 2.21; 95%CI, 1.00-4.89), 3-year-old children had an increased prevalence of atopic dermatitis of more than 30% (aOR, 9.48, 95%CI 1.42-63.13). Conclusions: Exposure to indoor fungi during gestation and infancy is associated with the development of atopic dermatitis in children. The phyla and genera of the fungi in the indoor house dust differed during gestation. This suggests that exposure to indoor fungi during gestation may be associated with the development of atopic dermatitis in children. Future research will be necessary to establish the underlying mechanisms.
We have developed an efficient synthesis of 2(5H)-furanones using the microwave-promoted Suzuki-Miyaura coupling reaction and evaluated their fungicidal activities against six fungal pathogens in vivo. In addition, 2-cyclopentenones or 2-cyclohexenones possessing one or two additional methylene groups instead of the oxygen in the 2(5H)-furanone skeleton were also prepared by using the similar method. As the results of in vivo fungicidal screening against 6 plant diseases, a few derivatives displayed specific fungicidal activities against rice blast and tomato late blast. Further studies toward the optimization of the chemical structures are necessary for the development of novel fungicides with high potency.
Maize (Zea mays L.; 2N=20) is major staple food crop grown worldwide adapted to several biotic and abiotic stresses. Maydis leaf blight (MLB) and banded leaf and sheath blight (BLSB) are serious foliar fungal diseases may cause up to 40% and 100% grain yield loss, respectively. The present studies were undertaken to work out the efficacy of chemicals, botanicals and bioagents for the management of MLB and BLSB under field condition for two seasons Kharif 2014 and 2015. Five molecules (propiconazole 25 EC, hexaconazole 25 EC, carbendazim 50 WP, mancozeb 75 WP and carbedazim 12 WP + mancozeb 63 WP), two bioagents i.e. Trichoderma harzianum and T. viridae and three botanicals namely azadirachtin, sarpagandha and bel pathar were tested for their efficacy against MLB. Eight newer fungicides viz., difenconazole 250 SC, hexaconazole 5 EC, carbendazim 50WP, validamycin 3 L, tebuconazole 250 EC, trifloxystrobin 50 WG + tebuconazole 50 WG, azoxystrobin 250 EC and pencycuron 250 SC were evaluated against BLSB. Analysis revealed significant effects of propiconazole at 0.1%, carbendazim 12 WP + mancozeb 63 WP at 0.125% and sarpagandha leaves at 10% against MLB pathogen, whereas validamycin at 0.1% and trifloxystrobin 25 WG + tebuconazole 50 WG at 0.05% were found effective against BLSB. The slow rate of disease control virtually by the bioagents might have not shown instant effect on plant response to the yield enhancing components. The identified sources of management can be used further in strengthening the plant protection in maize against MLB and BLSB.
