사람 치수 세포와 치주 인대 세포의 유전자 발현에 관한 비교 연구 (THE COMPARISON OF GENE EXPRESSION FROM HUMAN DENTAL PULP CELLS AND PERIODONTAL LIGAMENT CELLS)

  • 본 연구는 사람 치수세포 및 치주인대세포의 차이를 알아보고자 배양한 각각의 세포를 CDNA microarray assay를 통하여 유전자의 발현정도의 차이를 비교하였다. 그 결과를 바탕으로 각각의 세포에서 2배 이상의 유전자 발현의 차이를 보이는 유전자중 특징적인 3가지 유전자를 선택하여 RT-PCR로 검증한 결과 다음과 같은 결론을 얻었다; 1. Microarray assay 결과, 치주인대 세포에 비해 치수 세포에서 2배 이상 발현한 유전자 수는 총 51개가 나타났다. 2. RT-PCR의 결과, 치주인대세포에 비해 치수 세포에서 ITGA4, TGF-${\beta}2$ 등이 높게 나타났다. 3. Microarray assay결과, 치수 세포에서 비해 치주인대 세포에서 2배 이상 발현한 유전자 수는 총 19개가 나타났다. 4. RT-PCR의 결과, 치수 세포에 비해 치주인대세포에서 LUM, WISP1, MMP1 등이 높게 나타났다. 본 연구 결과로 치수세포에는 상아질 형성에 관여하는 특징적인 유전자가 치주인대세포에 비해 높게 발현되었으며, 치주인대세포에는 교원질 합성에 관여하는 특징적인 유전자가 치수세포에 비해 높게 발현되어, 치수세포와 치주인데 세포는 유전자 발현의 차이가 나타남을 알 수 있었다.

SPECT 영상을 사용한 기능적 심근형태의 자동 계측법 개발 (Automated Functional Morphology Measurement Using Cardiac SPECT Images)

  • 핵의학 검사에 있어서 심근 관류스캔은 관상동맥질환의 혈역학적 중요성을 평가하는 좋은 방법이다. 그러나 판독효율을 높이기 위해서 자동화된 정량적 계측 방법이 추가적으로 제시되어야한다. 본 연구에서는 판독에 필요한 심근의 3차원 기능모델과 심근 두께 계산 모델을 제시한다. 개발을 위해서 SPECT로 부터 심장의 단축단면상을 얻었고 전처리를 방정식을 적용하여 좌심근 두께의 모델링을 구현하였다. 실험결과 슬라이스 단축방향 영상으로부터 내벽과 외벽을 계측하는데 성공하였고, 계산된 좌표를 이용해서 3차원 모델링을 구현하였다. 다음 라플라스 식을 사용하여 심벽 두께의 3차원 모델을 완성하였다. 3차원 모델을 통해서 결절 부위가 쉽게 관찰할 수 있고, 3차원 모델의 회전을 통해서 병변의 위치를 빨리 파악할 수 있는 특징을 가진다. 판독 보조지표로서의 개발된 제안된 모델은 보조적 판독정보를 제공하고 오진의 확률을 낮추는데 기여할 것으로 예상한다. 허혈성 심장질환 환자의 조기 진단에도 큰 역할을 할 것이다.

N-Terminal Pro-B-type Natriuretic Peptide Is Useful to Predict Cardiac Complications Following Lung Resection Surgery

  • Background: Cardiovascular complications are major causes of morbidity and mortality following non-cardiac thoracic operations. Recent studies have demonstrated that elevation of N-Terminal Pro-B-type natriuretic peptide (NT-proBNP) levels can predict cardiac complications following non-cardiac major surgery as well as cardiac surgery. However, there is little information on the correlation between lung resection surgery and NT-proBNP levels. We evaluated the role of NT-proBNP as a potential marker for the risk stratification of cardiac complications following lung resection surgery. Material and Methods: Prospectively collected data of 98 patients, who underwent elective lung resection from August 2007 to February 2008, were analyzed. Postoperative adverse cardiac events were categorized as myocardial injury, ECG evidence of ischemia or arrhythmia, heart failure, or cardiac death. Results: Postoperative cardiac complications were documented in 9 patients (9/98, 9.2%): Atrial fibrillation in 3, ECG-evidenced ischemia in 2 and heart failure in 4. Preoperative median NT-proBNP levels was significantly higher in patients who developed postoperative cardiac complications than in the rest (200.2 ng/L versus 45.0 ng/L, p=0.009). NT-proBNP levels predicted adverse cardiac events with an area under the receiver operating characteristic curve of 0.76 [95% confidence interval (CI) 0.545~0.988, p=0.01]. A preoperative NT-proBNP value of 160 ng/L was found to be the best cut-off value for detecting postoperative cardiac complication with a positive predictive value of 0.857 and a negative predictive value of 0.978. Other factors related to cardiac complications by univariate analysis were a higher American Society of Anesthesiologists grade, a higher NYHA functional class and a history of hypertension. In multivariate analysis, however, high preoperative NT-proBNP level (>160 ng/L) only remained significant. Conclusion: An elevated preoperative NT-proBNP level is identified as an independent predictor of cardiac complications following lung resection surgery.

Rice OsACDR1 (Oryza sativa Accelerated Cell Death and Resistance 1) Is a Potential Positive Regulator of Fungal Disease Resistance

  • Rice Oryza sativa accelerated cell death and resistance 1 (OsACDR1) encodes a putative Raf-like mitogen-activated protein kinase kinase kinase (MAPKKK). We had previously reported upregulation of the OsACDR1 transcript by a range of environmental stimuli involved in eliciting defense-related pathways. Here we apply biochemical, gain and loss-of-function approaches to characterize OsACDR1 function in rice. The OsACDR1 protein showed autophosphorylation and possessed kinase activity. Rice plants overexpressing OsACDR1 exhibited spontaneous hypersensitive response (HR)-like lesions on leaves, upregulation of defense-related marker genes and accumulation of phenolic compounds and secondary metabolites (phytoalexins). These transgenic plants also acquired enhanced resistance to a fungal pathogen (Magnaporthe grisea) and showed inhibition of appressorial penetration on the leaf surface. In contrast, loss-of-function and RNA silenced OsACDR1 rice mutant plants showed downregulation of defense-related marker genes expressions and susceptibility to M. grisea. Furthermore, transient expression of an OsACDR1:GFP fusion protein in rice protoplast and onion epidermal cells revealed its localization to the nucleus. These results indicate that OsACDR1 plays an important role in the positive regulation of disease resistance in rice.

보리 잎과 옥수수 수염의 혼합과 유산균 발효를 이용한 γ-aminobutyric acid 생산 증진 (Enhancement of γ-aminobutyric Acid Production by Combination of Barley Leaf and Corn Silk and Its Fermentation with Lactic Acid Bacteria)

  • GABA는 glutamic acid decarboxylase에 의해서 L-glutamic acid가 탈탄산화되어 생합성된 비단백질 아미노산이다. GABA는 식물에서 스트레스에 대항한 대응반응으로 생성된다. 사람의 중추신경계에서는 주요 억제성 신경전달물질 중 하나로 항고혈압, 항당뇨 효능이 있다고 알려져 있다. 본 연구에서 우리는 보리 잎과 옥수수 수염을 유산균과 함께 발효함으로써 GABA 생성을 증진시키고자 하였다. 보리 잎과 옥수수 수염을 다양한 무게 비율로 조합하여 혼합하였고, $30^{\circ}C$에서 48시간 동안 배양기 안에서 L. plantarium과 함께 발효시켰다. 발효된 혼합물을 열수 추출한 후, thin layer chromatography와 GABase assay를 이용하여 GABA의 생산을 분석하였다. 우리는 9:1 혼합발효추출물이 다른 비율의 추출물 보다 GABA 함량이 높은 것을 확인하였는데 이것은 혼합과 발효기술이 보리 잎과 옥수수 수염 내 GABA 양 증진에 효과가 있음을 의미한다. 또한 몇 가지 생리활성을 분석한 결과 혼합발효추출물의 항산화 효능이 비발효 추출물에 비하여 증진되었고 세포독성은 나타나지 않음을 확인하였다. 이러한 결과는 보리 잎과 옥수수 수염의 조합과 이것을 유산균과 함께 발효시키는 방법이 고함량의 GABA와 증진된 생리 활성을 지닌 기능성 식품으로서의 개발 가능성이 있음을 의미한다.

Cu/MCM-41 메조포러스 촉매 제조 및 NO 제거 특성 (Preparation and Characterization of Cu/MCM-41 Mesoporous Catalysts for NO Removal)

  • 본 연구에서는 제조한 MCM-41에 Cu의 함량에 따른 NO의 전환율을 고찰하였다. MCM-41은 실리카 원으로 colloid silica를 사용하였고, template로 cetyltrimethylammonium chloride (CTMACl)를 사용하여 수열 합성하였으며, Cu/MCM-41은 Cu(II) acetylacetonate를 사용해서 Cu의 농도를 5, 10, 20 그리고 40%로 변화시켜 제조하였다. 표면 특성은 pH, FT-IR로 분석하였고, 육방배열의 1차원 기공 구조는 XRD로 고찰하였다. $N_2/77K$ 등온흡착 특성은 BET식과 t-plot을 이용하여 확인하였으며, NO 제거 효율은 가스크로마토그래프를 이용하여 측정하였다. 실험 결과, Si-OH와 Si-O-Si의 stretching vibration peak가 관찰되었으며, (100), (110), (200) 그리고 (210)의 육방배열의 1차원 구조를 확인하였다. Cu 금속이 도입된 MCM-41은 Cu 도입량이 증가할수록 비표면적과 미세기공부피는 감소한 반면에 NO 제거 효율은 증가하였다. 결과적으로 Cu/MCM-41의 Cu의 함량이 증가함에 따라 전체 촉매작용 반응과 NO 제거율이 증가하였다.

Cyclophosphamide가 마우스의 면역기억에 미치는 영향 (Effects of Cyclophosphamide on Immunological Memory in Mice)

  • The use of alkylating agent cyclophosphamide(CY), a widely used antitumor drug is well known as a potent immunosuppressant and has been used as a probe for investigating the functional capabilities of lymphocyte subsets of both T and B cells that play an important role in the regulation of the immune response. The present study was undertaken in an effort to assess the effects of CY on immunological memory in murine model. CY, given as a single dose of CY(250mg/kg) before sensitization with sheep red blood cells(SRBC) enhanced the primary response of Arthus and delayed-type hypersensitivity(DTH), as measured by footpad swelling reaction, but suppressed their tertiary DTH response. The similar CY pretreatment enhanced both the primary and tertiary hemagglutinin(HA) responses to SRBC, and the tertiary antibody response against polyvinylpyrroridone(PVP), a thymus-independent antigen but not the primary response against PVP. CY, given as a single dose of 250mg/kg 2 days before the primary immunization and two doses of 100mg/kg 2 days before the secondary and tertiary immunization, markedly suppressed the tertiary DTH and HA responses to SRBC. However, CY, given as small multiple daily doses(10mg/kg) over 4 days before sensitization but not after sensitization, enhanced the secondary HA response to SRBC. Contact sensitivity to dinitrofluorobenzene(DNFB) was suppressed by the drug, given either as a single large dose(300mg/kg) or as multiple dose(10mg/kg) administered 2 days before, together with or after DNFB sensitization. This suppression was more pronounced and more significant when CY was given as multiple dose. However, the enhancement of the secondary contact sensitivity to DNFB by CY was not clear-cut. The splenectomy appears to increase the enhancing effect of CY on contact sensitivity. These results suggest that CY selectively influences the immune response depending on the time of the drug administration relative to immunization and that the secondary or tertiary immune response involve memory cells with different susceptibilities to CY. Moreover, these results suggest that multiple low doses may sesectivley inhibit suppressor T cell proliferation involving DTH, HA or contact sensitivity without effecting helper T cells, but high doses presumably inhibit helper T cells and suppressor T cells with effecting B cells.

Stage specific transcriptome analysis of liver tissue from a crossbred Korean Native Pig (KNP × Yorkshire)

  • Korean Native Pig (KNP) has a uniform black coat color, excellent meat quality, white colored fat, solid fat structure and good marbling. However, its growth performance is low, while the western origin Yorkshire pig has high growth performance. To take advantage of the unique performance of the two pig breeds, we raised crossbreeds (KNP ${\times}$ Yorkshire to make use of the heterotic effect. We then analyzed the liver transcriptome as it plays an important role in fat metabolism. We sampled at two stages: 10 weeks and at 26 weeks. The stages were chosen to correspond to the change in feeding system. A total of 16 pigs (8 from each stage) were sampled and RNA sequencing was performed. The reads were mapped to the reference genome and differential expression analysis was performed with edgeR package. A total of 324 genes were found to be significantly differentially expressed (${\left|log2FC\right|}$ > 1 & q < 0.01), out of which 180 genes were up-regulated and 144 genes were down-regulated. Principal Component Analysis (PCA) showed that the samples clustered according to stages. Functional annotation of significant DEGs (differentially expressed genes) showed that GO terms such as DNA replication, cell division, protein phosphorylation, regulation of signal transduction by p53 class mediator, ribosome, focal adhesion, DNA helicase activity, protein kinase activity etc. were enriched. KEGG pathway analysis showed that the DEGs functioned in cell cycle, Ras signaling pathway, p53 signaling pathway, MAPK signaling pathway etc. Twenty-nine transcripts were also part of the DEGs, these were predominantly Cys2His2-like fold group (C2H2) family of zinc fingers. A protein-protein interaction (PPI) network analysis showed that there were three highly interconnected clusters, suggesting an enrichment of genes with similar biological function. This study presents the first report of liver tissue specific gene regulation in a cross-bred Korean pig.

Effects of Korean Red Ginseng extract on tissue plasminogen activator and plasminogen activator inhibitor-1 expression in cultured rat primary astrocytes

  • Korean Red Ginseng (KRG) is an oriental herbal preparation obtained from Panax ginseng Meyer (Araliaceae). To expand our understanding of the action of KRG on central nervous system (CNS) function, we examined the effects of KRG on tissue plasminogen activator (tPA)/plasminogen activator inhibitor-1 (PAI-1) expression in rat primary astrocytes. KRG extract was treated in cultured rat primary astrocytes and neuron in a concentration range of 0.1 to 1.0 mg/mL and the expression of functional tPA/PAI-1 was examined by casein zymography, Western blot and reverse transcription-polymerase chain reaction. KRG extracts increased PAI-1 expression in rat primary astrocytes in a concentration dependent manner (0.1 to 1.0 mg/mL) without affecting the expression of tPA itself. Treatment of 1.0 mg/mL KRG increased PAI-1 protein expression in rat primary astrocytes to $319.3{\pm}65.9%$ as compared with control. The increased PAI-1 expression mediated the overall decrease in tPA activity in rat primary astrocytes. Due to the lack of PAI-1 expression in neuron, KRG did not affect tPA activity in neuron. KRG treatment induced a concentration dependent activation of PI3K, p38, ERK1/2, and JNK in rat primary astrocytes and treatment of PI3K or MAPK inhibitors such as LY294002, U0126, SB203580, and SP600125 (10 ${\mu}M$ each), significantly inhibited 1.0 mg/mL KRG-induced expression of PAI-1 and down-regulation of tPA activity in rat primary astrocytes. Furthermore, compound K but not other ginsenosides such as Rb1 and Rg1 induced PAI-1 expression. KRG-induced up-regulation of PAI-1 in astrocytes may play important role in the regulation of overall tPA activity in brain, which might underlie some of the beneficial effects of KRG on CNS such as neuroprotection in ischemia and brain damaging condition as well as prevention or recovery from addiction.

각질형성세포에서 왕불유행 헥산 분획물이 Laminin-332 발현에 미치는 효과 (Hexane Fraction of Melandrium firmum Extract Induces Laminin-332 Expression in Human Keratinocyte)

  • 피부 기저막(basement membrane, BM)이란 표피와 진피 사이에 존재하는 특별한 구조물로 표피와 진피를 단단히 고정시켜 피부 구조를 유지하는 데에 중요한 역할을 수행한다. 노화 및 자외선 노출에 의한 피부 기저막의 구조적 변화와 파괴는 피부 주름 형성과 탄력 저하를 포함하는 피부노화 현상의 요인으로 여겨지고 있다. Laminin-332 (LN-332)는 피부 기저막을 구성하는 주성분으로 피부에서 표피와 진피를 단단히 고정시키는데 중요한 역할을 한다. 본 연구에서는 왕불유행 헥산 분획물(Melandrium firmum hexane fraction, MFHF)이 각질형성세포에서 LN-332 발현에 미치는 효과를 확인하였다. 정량적 real-time PCR (RT-PCR)과 단백질 발현 분석을 통해서 MFHF가 LN-332의 mRNA 발현 및 단백질 발현을 촉진시키는 것을 확인하였다. 또한 MFHF가 어떤 신호전달 경로를 통해 LN-332 발현을 조절하는지 확인하기 위하여 p38 MAPK 억제제인 SB202190과 ERK1/2 억제제인 U0126을 처리한 결과, p38 MAPK 억제제에 의해서 LN-332 발현이 완벽히 억제됨을 확인하였다. 또한, 피부 기저막을 구성하고 있는 콜라겐 타입 VII과 integrin ${\alpha}6$의 mRNA 발현 역시 MFHF에 의해 증가하는 것을 확인하였다. 우리는 본 연구를 통해 MFHF가 각질형성세포에 작용하여 피부 기저막을 구성하는 성분들의 생성을 촉진할 수 있는 소재로 작용할 수 있다는 것을 확인하였다. 이러한 결과는 기저막의 구조적, 기능적 이상에 의해 나타나는 피부노화 현상의 개선을 위해 활용할 수 있을 것이라 제안한다.
