발효 쌍별귀뚜라미 추출물의 생리활성 연구 (Biological Activity of Fermented Gryllus bimaculatus extracts)

  • Gryllus bimaculatus extract (GbE) have reported that anti-inflammatory activity by suppression of pro-inflammatory cytokines. However, the effects of fermented Gryllus bimaculatus extract (FGbE) have not yet been investigated. In this study, we evaluated the anti-inflammatory and anti-wrinkle effect of the fermented Gryllus bimaculatus extracts using Bacillus subtilis (JB PMB-18) in RAW264.7 cells. Both GbE and FGbE exerted no cytotoxic effects until 1000 ㎍/mL concentration. FGbE decreased NO production and decreased iNOS and COX-2 mRNA levels in a concentration-dependent manner. In addition, the protein production of inflammatory cytokines TNF-α, IL-1β and IL-6 was effectively reduced compared to the GbE. Inhibitory activities of elastase and collagenase associated with skin wrinkle improvement were measured to be 45% and 69%, respectively, at a concentration of 500 ㎍/mL in FGbE. From these results, FGbE can be used as a health functional food and skin functional cosmetic materials for preventing inflammatory diseases because it has excellent anti-inflammatory and anti-wrinkle effects.

Analytical Method Validation of (-)Epicatechin gallate in Penthorum chinense Pursh Extract using HPLC

  • This study attempted to establish a High Performance Liquid Chromatography (HPLC) analysis method for the determination of (-)-epicatechin gallate as a part of the quality control for the development of functional cosmetic materials from Penthorum chinense Pursh extracts. HPLC was performed on a Unison US-C18 column ($4.6{\times}250mm$, $5{\mu}m$) with a gradient elution of 0.05% (v/v) trifluoroacetic acid (TFA) and methyl alcohol at a flow rate of 1.0 mL/min at $30^{\circ}C$. The analyte was detected at 280 nm. The HPLC method was performed in accordance with the International Conference on Harmonization (ICH) guideline (version 4, 2005) of analytical procedures with respect to specificity, precision, accuracy, and linearity. The limits of detection and quantitation were 0.11 and 0.33 mg/mL, respectively. Calibration curves showed good linearity (r2 > 0.9999), and the precision of analysis was satisfied (less than 0.6%). Recoveries of quantified compounds ranged from 99.51 to 101.92%. This result indicates that the established HPLC method is very useful for the determination of marker compound in P. chinense Pursh extracts.

마이크로웨이브 추출방법을 이용한 작약의 유효성분 추출 및 생리활성 측정 (Development of Microwave Extraction Method for the Active Ingredients and Functional Constituents of Paeonia Root)

  • The heat-mediated reflux apparatus extraction of Paeonia lactiflora Pall. has been widely used as a traditional extraction method. In this paper, the microwave apparatus extraction method of Paeonia Radix was performed and the active ingredients and functional constituents were compared with the reference extraction method. The most effective extraction condition of albiflorin using the microwave was 120℃ in 50% methanol, and paeoniflorin was maximally extracted at 60℃. The reduced level of paeoniflorin molecule at high-pressure and high-temperature extraction condition was caused by the molecular instability. Additionally, the microwave extraction of 50% methanol extracts at 150℃ showed the highest functional constituents determined by in vitro DPPH radical scavenging activity, polyphenol concentration, and tyrosinase inhibition assay. The microwave apparatus was adapted as a rapid, low-cost, and environmentally friendly method to extract active ingredients and the practical extraction conditions of Paeonia Radix can be used in industrial applications.

구실잣밤나무 열매의 성분분석 및 항산화활성 평가 (Component Analysis and Antioxidative Activity of Castanopsis cuspidata var. sieboldii Nut)

  • 난대성 상록활엽수종의 자원화 및 식품 소재화 탐색을 목적으로 구실잣밤나무(Castanopsis cuspidata var. sieboldii $N_{AKAI}$) 열매(구실잣밤) 추출물의 수율, pH, 당도, 색도, 구성당, 유기산 함량, 페놀 함량 및 항산화활성을 검정하였다. 각 용매에 대한 추출물의 수율은 물 추출물에서 3.6%로 가장 높았고, pH는 6.43의 중성값을 보였으며, 당도는 3.17 $^{\circ}Brix$를 나타내었다. 색도 중 L값은 83.07, a값은 1.49 그리고 b값은 10.48을 나타내었다. 구성당의 조성 및 함량은 fructose 27.11 mg%, glucose 26.29 mg%, galactose 0.86 mg%로 나타났다. 유기산은 총 5종이 동정되었으며, oxalic acid(59.44%), formic acid (23.99%), malic acid (11.24%), citric acid (3.34%), succinic acid (1.99%) 순으로 나타났다. 총 페놀 함량은 물 추출물에서 27.69 mg% 수준으로 가장 높았고, DPPH radical-scavenging 활성은 대조구인 $\alpha$-tocopherol ($SC_{50}\;12.47{\mu}g/mL$)에 비해 낮은 수준이었으나 EtOAc 추출물($SC_{50}\;74.88{\mu}g/mL$)에서 가장 강한 활성을 보였다. 이와 같은 결과를 바탕으로 구실잣밤은 다양한 식품소재 및 기능성 소재로서의 활용성이 기대되었으며, 또 도토리의 대용 및 병용이 가능한 소재로서 실용화 가능성이 확인되었다.

Isolation and Structural Elucidation of Antimicrobial Compounds from Buckwheat Hull

  • Three new compounds were isolated from the methanol extracts of buckwheat (Fagopyrum esculentum) hull, and their structures were determined to be 6,7-dihydroxy-3,7-dimethyl-octa-2(Z),4(E)-dienoic acid (1), 6,7-dihydroxy-3,7-dimethyl-octa-2(E),4(E)-dienoic acid (2), and 4,7-dihydroxy-3,7-dimethyl-octa-2(E),5(E)-dienoic acid (3) by NMR and MS spectroscopic analyses. These compounds at $500{\mu}g$ concentration showed antimicrobial activity against Staphylococcus aureus, demonstrated by the paper disc method.

추출용매에 따른 진피 추출물의 항산화 활성 (Antioxidant Activities of Citrus unshiu Extracts obtained from Different Solvents)

  • 본 연구에서는 진피의 활용도를 높이기 위한 연구의 일환으로 유기용매별에 따른 생리활성물질의 용출량을 측정하기 위해, 진피와 에탄올 진피 추출물을 대상으로 유기용매인 에틸 아세테이트, 아세톤, 염화 메틸렌, 메탄올을 이용하여 추출한 시료를 대상으로 총 폴리페놀 함량, 전자공여능, glutathione S-transferase(GST)의 활성 저해력을 측정한 결과는 다음과 같다. 1. 총 폴리페놀 함량은 ethyl acetate인 경우 진피는 $928.48{\pm}1.19{\mu}g\;GAE/mL$, 에탄올 추출 진피는 $664.64{\pm}0.74{\mu}g\;GAE/mL$로, acetone인 경우 진피는 $886.03{\pm}0.44{\mu}g\;GAE/mL$, 에탄올 추출 진피는 $702.67{\pm}0.85{\mu}g\;GAE/mL$로, methylene chloride인 경우 진피는 $413.08{\pm}1.39{\mu}g\;GAE/mL$, 에탄올 추출 진피는 $429.64{\pm}0.61{\mu}g\;GAE/mL$로, methanol인 경우 진피는 $12,648.60{\pm}0.56{\mu}g\;GAE/mL$, 에탄올 추출 진피는 $16,108.20{\pm}0.73{\mu}g\;GAE/mL$로 나타나, 진피나 에탄올 추출 진피는 모두 methanol로 추출한 것이 상대적으로 높게 나타났으며, 총 폴리페놀의 함량 차이는 유기용매별 통계적으로 유의한 차이가 나타났다(p<0.05). 2. 전자공여능은 ethyl acetate인 경우 진피는 62.80%, 에탄올 추출 진피는 51.49%로 나타났으며, acetone인 경우 진피는 97.43%, 에탄올 추출 진피는 63.17%로 나타났으며, methylene chloride인 경우 진피는 52.20%, 에탄올 추출 진피는 67.68%로 나타났으며, methanol인 경우 진피는 97.63%, 에탄올 추출 진피는 96.18%로 나타났다. Electron donating ability(EDA)는 유기용매 중 메탄올로 추출하였을 때가 상대적으로 가장 높게 나타났으며, 유기용매별로 통계적으로 유의한 차이가 나타났다(p<0.05). 3. Glutathione S-transferase(GST)에 대한 활성 저해능은 ethyl acetate인 경우 진피는 76.22%, 에탄올 추출 진피는 75.54%로 나타났으며, methylene chloride인 경우 진피는 31.73%, 에탄올 추출 진피는 73.53%로 나타났으며, methanol인 경우 진피는 97.48%, 에탄올 추출 진피는 48.70%로 나타났다. Glutathione S-transferase(GST)에 대한 활성 저해능은 진피인 경우 유기용매 중 메탄올로 추출하였을 때가 가장 높게 나타났고, 에탄올 추출 진피인 경우는 에탄올과 methylene chloride로 추출할 때가 높게 나타났으며, 유기용매별로 통계적으로 유의한 것으로 나타났다(p<0.05).

  • This experiment was planned to develop a functional supplements by food resources to prevent and lessen the bad effects caused by the environmental pollution such as bad food, air, water and heavy metals exposed to people these days. As for Its primary stage, the nutrient profile and sensory characteristics of the duck-extract were evaluated in this study. The duck-extract was formulated by the mixture of bone and internal organs of ducks and 6 medicinal herbs which were able to be used as foods and known to help the excretion and the repression of the poison inside the body for a long time. As we compared the six medicinal herbs mentioned above to the vegetable herbs, the nutrient profile of the medicinal herbs were superior to the vegetable herbs. The duck-extract was composed of protein 49.92%, carbohydrate 37.02%, dietary fiber 20.99%, lipid 7.60% and ash 5.46%. The ratio of Ca to P was 1 : 1.4, which was a suitable ratio for absorption, the contents of Na and K were low and those of micro-elements such as Fe, Mn, Zn and Cu high compared to other meat extracts. The essential amino acids accounted for 30.91% of total amino acids. The result of sensory evaluation was better in overall preferences than the ones which are already in the market. From these results, the nutrient profile of the duck-extract was estimated to be able to supply enough nutrients to the people whose nutritional balance was destroyed these days. This study also showed the effective way of using duck-extract and its application to the oriental medicine.

비타민나무 추출물 및 착즙 분말의 항산화 활성 및 Nitric Oxide 생성과 Elastase에 대한 저해활성 (Antioxidative and Inhibitory Activities of Extract and Juice Powder from Hippophae rhamnoides L. against Nitric Oxide and Elastase Production)

  • Background: The flowering plant Hippophae rhamnoides L. has been used for many studies on fruit or leaf extracts. This study was conducted to investigate the development of a new cosmetic material from H. rhamnoides fruits and leaves that have by antioxidant, anti-inflammatory and wrinkle improvement activities. Methods and Results: The antioxidant abilities of H. rhamnoides extracts, including of a water-soluble fruit powder (FW), a fatsoluble fruit powder (FF), a supercritical extract of fruit by-product (BS), and a mixture of leaf and fruit (MIX), were investigated in vitro. A DPPH radical assay for antioxidant activity was performed for these fractions alongside assay to evaluate the total phenolic and flavonoid content (TPC and TFC). As expected, the MIX had the highest DPPH radical scavenging activity ($RC_{50}=10.27{\mu}g/m{\ell}$), and the TPC and TFC also were highest in MIX ($225.7mg{\cdot}GAE/g$, and $25.18mg{\cdot}QE/g$, respectively). Nitric oxide (NO) production in LPS-induced RAW264.7 cells was estimated and the results indicated an over 75% decrease of NO production in FF and MIX. In other assays, the highest elastase inhibitory activity was found in FW. Conclusions: These results revealed that H. rhamnoides extracts have a high potential for antioxidant, anti-inflammatory and antiwrinkle activities. H. rhamnoides products are suggested to be applied as the functional materials of cosmetic ingredients.

연근 추출물에서 주름개선 소재의 개발 (Development of Anti-Wrinkle Agent from Nelumbo nucifera Root Extract)

  • 본 연구에서는 연근의 추출물에서 기능성화장품 소재를 개발하고자 하였다. 용매는 70~100%의 에탄올을 이용하였으며, 연근의 유효성분으로 nuciferine을 확인하였다. 연근 추출물의 화장품소재로서의 응용가능성을 보기 위해 안전성시험(MTT assay), 주름개선 효과시험(Elastase 활성저해 시험) 및 항산화효과시험(DPPH free radical scavenging assay)을 실시하였다. 100% 에탄올로 연근을 추출할 경우 $100{\mu}g/ml$의 농도에서 세포생존율이 80% 이상이어서 연근 추출물은 화장품소재로서 비교적 안전함을 알 수 있었다. 또한 연근 추출물은 $100{\mu}g/ml$의 농도에서 elastase 활성억제가 40~50%를 보여주어서 비교적 우수한 주름개선 효과를 나타내었다. 50% 항산화 저해능을 보이는 농도($FSC_{50}$)는 $5.0{\sim}38{\mu}g/ml$로 연근 추출물의 항산화효과가 매우 우수하였다. 따라서 연근 에탄올추출물은 주름개선 기능성화장품 소재로서 그 가능성이 높음을 알 수 있었다.

Comparison of Functional Constituents and Biological Activity of the Seed Extracts from Two Mulberry Fruits

  • The seeds from two mulberry fruits [Morus alba (MA) and Cudrania tricuspidata (CT)] were examined for their oil content, and fatty acid, phytosterol and tocopherol compositions and contents. Moreover, polyphenolic compounds and biological activity of the two defatted seed residue extracts were also evaluated. Oil contents of MA and CT seeds were 29.36% and 16.69%, respectively, while MeOH extracts of the defatted MA and CT seed residues were 5.10% and 6.22%, respectively. The two seed oils were composed of 81.4 and 74.37% linoleic, 5.75 and 11.39% oleic, 8.40 and 10.18% palmitic acid, and 3.52 and 3.0% stearic acids, and two other minor fatty acids, such as linolenic and arachidic acids. MA seed had higher contents of phytosterols (507.59 mg/100 g of oil), tocopherols (99.64 mg/100 g of oil), and total flavonoid (106.50 mg/100 g of seed) than CT seed, whereas CT seed had higher levels of total polyphenol than MA seed. The MeOH extract of MA seed residue showed higher antioxidant, anti-diabetic, and anti-melanogenic activity than that of CT seed residue. trans-Resveratrol (9.62 mg/100 g), quercetin (54.83 mg/100 g), and 4-prenylmoracin (48.70 mg/100 g), were found to be the main polyphenolic components in the MeOH extract of MA seed residue. These results indicate that MA seeds are good sources of essential dietary phytochemicals with antioxidant, anti-diabetic and anti-melanogenic activity.
