The Effect of Tai-Chi Exercise on the Function of Shoulder Joint, Heart Rate in Breast Cancer Patients (타이치(Tai-Chi) 운동이 유방암 수술환자의 어깨관절 가동범위와 심박동수 변화에 미치는 영향)

  • Good health-related quality of life is regarded as the goal of elderly residential care. Treatment for breast cancer produces side effects that diminish functional capacity and quality of life among survivors. Tai-Chi exercise offers the benefits of fitness and health. Tai-chi has been claimed to generate beneficial effects with respect to a wide range of diseases. The object of this research is to evaluate the effects of tai chi exercise on shoulder joint movement and cardiovascular function in elderly breast cancer patients. The subjects were 40 patients residing in B metropolitan city, who had undergone surgery for breast cancer within 2 months. They underwent post-operative radiotherapy and an anti-cancer chemical treatment. They practiced tai-chi exercise for 60 min a day, four times a week for 12 weeks under the direction of a special tai-chi instructor. Shoulder joint movement and heart rate, were measured before and after the experiment. The results were as follows; Shoulder joint movement, including flexion, extention, adduction and abduction, significantly increased (p<0.05) after treatment, and heart rates significantly decreased after treatment (p<0.05). From the results, Tai-Chi exercise was shown to be extremely effective in improving shoulder joint movement and heart rate in elderly breast cancer patients. Further study is needed in this area.

Antioxidant Effect of Oil Containing Cellulase-Treated Red Ginseng. (효소 처리 홍삼을 함유한 오일의 항산화 효과)

  • In this study we evaluated the method to develop red ginseng oil containing high content of phytochemicals by enzymes treatment. To select the optimum extraction process of red ginseng with oils, the antioxidant activities of red ginseng using various enzymes were measured. Red ginseng after 0.5% cellulase treatment for 1 hr at $50^{\circ}C$ had higher antioxidant activity than the other conditions. We found that red ginseng/soybean oil extracted for 15 days at $40^{\circ}C$ after 0.5% cellulase treatment increased DPPH radical scavenger activity and decreased the TBA and POV values. However, red ginseng/olive oil had little functional activities compare to the red ginseng/soybean 0il. We also analyzed vitamin A and E by HPLC and found that vitamin E was increased by 0.5% cellulase treatment in the oil. This is the first report that red ginseng oil extracted by enzyme treatment has various beneficial effects.

Anti-proliferative Effects of Acid Extract of Gracilaria Verrucosa on Primary Human Prostate Cancer Cells (꼬시래기 산추출물의 primary 인체 전립선 암세포 증식억제 효과)

  • The purpose of this study was to investigate the anti-proliferative and apoptotic effects of acid extract of Gracilaria verrucosa (AEG) on RC-58T/h/SA#4 primary human prostate cancer cells. AEG significantly decreased the cell viability of prostate cancer cells in a dose-dependent manner. AEG also showed relatively low cytotoxicity on normal cell (RWPE-1). The morphology of prostate cancer cells treated with AEG was distorted to shrunken cell masses. In addition, it was revealed that AEG induced cell death as evidenced by increased formation of apoptotic body and nuclear condensation. Furthermore, AEG clearly modulated the down regulation of Bcl-2 (anti-apoptotic)/Bax (pro-apoptotic) family and activated caspase-3 as an effector caspase in a dose-dependent manner. AEG inhibited cell proliferation induced by environmental hormones as a bisphenol A in a dose-dependent manner. These results indicate that AEG act as anti-proliferative effects as a potential therapeutic agent on primary human prostate cancer cells.

Physicochemical Characteristics and Antioxidant Activities of Freezing Pretreated Black Garlic (동결 전처리한 숙성 흑마늘의 이화학적 특성 및 항산화 활성)

  • Freezing pretreatment can destroy the cell membrane of garlic and may improve some food-quality of garlic. Therefore we investigated the effect of freezing pretreatment at $-20^{\circ}C$ and $-70^{\circ}C$ on quality of aged black garlic, compared with traditional processing methods. Our results showed that freezing pretreatment at $-70^{\circ}C$ had the greatest impact on qualities and antioxidant activities of black garlic. Browning degree and pH of black garlic after both the freezing pretreatment and aging process were 3.14 and 3.55, respectively. Furthermore, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity and reducing power of aged black garlic can be enhanced by pre-freezing processing. Reducing sugar and 5-hydroxymethyl-2-furaldehyde (5-HMF) contents of freezing pretreated and aged black garlic were increased by 1.69 and 1.14 fold, respectively, compared with the control samples. The results indicated that freezing pretreatment had improved the overall qualities (such as browning degree, pH, reducing sugar) and functional materials of black garlic.

Substitution of Heavy Complementarity Determining Region 3 (CDR-H3) Residues Can Synergistically Enhance Functional Activity of Antibody and Its Binding Affinity to HER2 Antigen

  • To generate a biobetter that has improved therapeutic activity, we constructed scFv libraries via random mutagenesis of several residues of CDR-H3 and -L3 of hu4D5. The scFv clones were isolated from the phage display libraries by stringent panning, and their antiproliferative activity against HER2-positive cancer cells was evaluated as a primary selection criterion. Consequently, we selected AH06 as a biobetter antibody that had a 7.2-fold increase in anti-proliferative activity ($IC_{50}$: 0.81 nM) against the gastric cancer cell line NCI-N87 and a 7.4-fold increase in binding affinity ($K_D$: 60 pM) to HER2 compared to hu4D5. The binding energy calculation and molecular modeling suggest that the substitution of residues of CDR-H3 to W98, F100c, A101 and L102 could stabilize binding of the antibody to HER2 and there could be direct hydrophobic interactions between the aromatic ring of W98 and the aliphatic group of I613 within HER2 domain IV as well as the heavy and light chain hydrophobic interactions by residues F100c, A101 and L102 of CDR-H3. Therefore, we speculate that two such interactions were exerted by the residues W98 and F100c. A101 and L102 may have a synergistic effect on the increase in the binding affinity to HER2. AH06 specifically binds to domain IV of HER2, and it decreased the phosphorylation level of HER2 and AKT. Above all, it highly increased the overall level of p27 compared to hu4D5 in the gastric cancer cell line NCIN82, suggesting that AH06 could potentially be a more efficient therapeutic agent than hu4D5.

The Effect of Activated Ion Calcium for Production of Panax ginseng Seedlings in Paddy Field (논 인삼 우량묘 생산을 위한 활성이온칼슘 처리효과)

  • When ginseng seedlings are cultured in paddy fields, quality degradation and yield reduction are induced by severe plant loss with chlorosis on leaves occurred physiological disorder by excessive salt and poor drainage, rusty-root occurrence, and root rot etc. Accordingly, in order to solve these problems, this study was performed to investigate the treatment method, concentrations and time of activated ion calcium as environment-friendly agricultural materials. Activated ion calcium is an enriched and purified water-soluble mineral calcium component for absorbing quickly into plant as a highly functional calcium and it is an alkaline calcium of 37% (370 $m{\ell}$/1 ${\ell}$) concentration with pH 13. Treatment method was that ginseng seeds were sown after removing water in the shade after seed immersion for 1 minute with active ion calcium of 20-fold diluted solution, and then irrigated $4{\ell}$ per 3.3 $m^2$ with 200-fold, 400-fold, and 600-fold diluted solution before emergence on late March, and supplied 1 time on leaves with 500-fold diluted solution in June and July respectively. The disease rate by treatment of activated ion calcium was that on the treatment of soil irrigated with 200-fold diluted solution compared to non-treated soil, damping-off was 33%, anthracnose was 100% reduced and the occurrence rate of rusty-root was 30% reduced. In addition, when active ion calcium of 200-fold diluted solution were soil irrigated, first and second grade ginseng were respectively 26% and 22% produced more, compared with control.

  • Objectives : Sparganii Rhizoma is frequently used in traditional herbal medicine for treatment of blood stasis, amenorrhea and functional dyspepsia and has been reported to exhibit anti-oxidant, anti-proliferation and anti-angiogenesis peoperties. In this study, we investigated the cytoprotective effect and underlying mechanism of Sparganii Rhizoma water extract (SRE) against oxidative stress-induced mitochondrial dysfunction and apoptosis in hepatocyte. Methods : To determine the effects of SRE on oxidative stress, we induced synergistic cytotoxicity by co-treatment of arachidonic acid (AA) and iron in the HepG2 cell, a human derived hepatocyte cell line. Results : Treatment of SRE increased relative cell viability and altered the expression levels of apoptosis-related proteins such as Bcl-xL, Bcl-2 and procaspase-3. And SRE also inhibited the mitochondrial dysfunction and excessive reactive oxygen species production induced by AA+iron. In addition, SRE activated of AMP-activated protein kinase (AMPK), a potential target for cytoprotection, by increasing the phosphorylation of AMPKα at Thr-172. Morever, SRE increased phosphorylation of acetyl-CoA carboxylase, a direct downstream target of AMPK. Conclusion : These results indicated that SRE has the ability to protect against oxidative stress-induced hepatocyte damage, which may be mediated with AMPK pathway.

Assessments of the VOCs and Smells Compounds Emitted from Properties Exhumed at Sim Seol Tomb (심설(沈偰)묘 출토유물에서 발현되는 VOCs 및 냄새물질의 특성 평가)

  • In this study, the 89 components of the gases emitted from grave goods at Sim Seol(1570-1630) tomb identified and quantitatively analyzed using a thermal desorption system integrated with gas chromatography-mass spectrometer. The concentration of major components show the levels of alpha-pinene 4,113ppbv, beta-pinene 2,510ppbv and limonene 2,424ppbv, that of the others orderly show p-menth-2-ene, acetone, isolongifolene and isoborneol. The functional groups of high concentration level cover 65% of terpenes and 25.8% of alcohols. The expected odor intensity orderly show 35.1% of terpenes, 33.4% of aldehydes and 8.8% of alcohols. Terpenes have generally effect of a perfume, antifungal and antimicrobial activity, and could come out of the pine(Pinus densiflora) coffin, aldehydes and alcohols could be come out of both the Sim Seol mirra and Pinus densiflora for a long time. The analyses of gases emitted from a excavated properties or a tomb are assessed that it can pc001 provide a scientific basis on a counterplan against harmful gases, a method of conservation treatments and a evaluation of effectiveness in conservation of cultural properties in a tomb.

Protective Effect of Phosphatidylcholine on Lipopolysaccharide-Induced Acute Inflammation in Multiple Organ Injury

  • Soybean polyunsaturated phosphatidylcholine (PC) is thought to exert anti-inflammatory activities and has potent effects in attenuating acute renal failure and liver dysfunction. The aim of this study was to investigate the effects of PC in protecting multiple organ injury (MOI) from lipopolysaccharide (LPS). Six groups of rats (N=8) were used in this study. Three groups acted as controls and received only saline, hydrocortisone (HC, 6 mg/kg, i.v.) or PC (600 mg/kg, i.p.) without LPS (15 mg/kg, i.p.) injections. Other 3 groups, as the test groups, were administered saline, HC or PC in the presence of LPS. Six hours after the LPS injection, blood and organs (lung, liver and kidney) were collected from each group to measure inflammatory cytokines and perform histopathology and myeloperoxidase (MPO) assessment. Serum cytokines (TNF-${\alpha}$, IL-6 and IL-10) and MPO activities were significantly increased, and significant histopathological changes in the organs were observed by LPS challenge. These findings were significantly attenuated by PC or HC. The treatment with PC or HC resulted in a significant attenuation on the increase in serum levels of TNF-${\alpha}$ and IL-6, pro-inflammatory cytokines, while neither PC nor HC significantly attenuated serum levels of IL-10, anti-inflammatory cytokine. In the organs, the enhanced infiltration of neutrophils and expression of ED2 positive macrophage were attenuated by PC or HC. Inductions of MPO activity were also significantly attenuated by PC or HC. From the findings, we suggest that PC may be a functional material for its use as an anti-inflammatory agent.

Effect of Conjugated Linoleic Acid (CLA) in Growth of Larval Rockfish (Sebastes schlegeli) (CLA 급이가 우럭치어 성장에 미치는 영향)

  • The multifunctional compound such as conjugated linoleic acid (CLA) was used to producing of functional larval rockfish. Artemia, livefoods for larval fish, enriched with highly unsaturated fatty acids (HUFA) and CLA ($0.5\%$, $1.0\%$, $2.5\%$ and $5.5\%$ group) was fed to larval rockfish. Artemia, enriched with $2.5\~5.0\%$ of CLA, negatively affects the quality of larval rockfish. But $1.0\%$ CLA fed group shown to good specific growth rate, feed efficiency, n-3/n-6 PUFA ratio, DHA/EPA ratio and hematology results. The CLA deposited in muscle lipids of larval rockfish was reached 6.57 to $19.69\%$.
