The purpose of this study was to determine the optimum ethanol extraction conditions for maximum extraction of functional components such as ferulic acid, oryzanol, and toopherol from black rice bran using Response Surface Methodology (RSM). A central composite design was applied to investigate the effects of the independent variables of solvent ratio ($X_{1}$), extraction temperature ($X_{2}$) and extraction time ($X_{3}$) on the dependent variables such as total phenol components ($Y_{1}$), total flavonoids compounds ($Y_{2}$), electron donating ability ($Y_{3}$), $\gamma$-oryzanol ($Y_{4}$), ferulic acid ($Y_{5}$) and $\alpha$-toopherol components ($Y_{6}$). ANOVA results showed that coefficients of determination (R-square) of estimated models for dependent variables ranged from 0.8939 to 0.9470. It was found that solvent ratio and extraction temperature were the main effective factors in this extraction proess. Particularly, the extraction efficiency of ferulic acid, $\gamma$-oryzanol and $\alpha$-toopherol components were significantly affected by extraction temperature. As a result, optimum extraction conditions were 20.35 mL/g of solvent ratio, 79.4$^{\circ}C$ of extraction temperature and 2.88 hr of extraction time. Predicted values at the optimized conditions were acceptable when compared with experimental values.
The purpose of this study was to analyze the growth and functional differences between C. rotundus and G. littoralis according to different electrical conductivity (EC) conditions in reclaimed soil conditions. C. rotundus and G. littoralis seeds were sown in a tray and managed for seedlings stage for eight weeks. They were transplanted in the pots containing reclaimed soils sampled in the Saemangum region. The plants were grown in the reclaimed land soil for 12 weeks under the control, 1, 2, 4, and $8dS{\cdot}m^{-1}$ conditions and in horticultural soils with EC $1.0dS{\cdot}m^{-1}$. Plant height, leaf length and width of C. rotundus were the highest in EC $1dS{\cdot}m^{-1}$. Leaf, flower and tuber numbers of C. rotundus were the highest in EC $2dS{\cdot}m^{-1}$ and the lowest in EC $8dS{\cdot}m^{-1}$, and SPAD was the highest in EC 2 and $4dS{\cdot}m^{-1}$ and the lowest in EC $8dS{\cdot}m^{-1}$. The fresh weights of shoot and root of C. rotundus grown under EC $2dS{\cdot}m^{-1}$ increased and then decreased as the concentration increased. When compared plant growth between reclaimed soil and horticulture soil with EC $1dS{\cdot}m^{-1}$, the fresh weights of shoot and root, SPAD, leaf number, flower number, and tuber number were higher in horticultural soils. Although G. littoralis grown under EC $8dS{\cdot}m^{-1}$ was the lowest in all growth parameters, there were no significant differences among other EC treatments. C. rotundus had the highest p-coumaric acid content in EC $1dS{\cdot}m^{-1}$. And the catechin content in shoot of G. littoralis was the highest in the control, and root of Glehnia littoralis had the highest benzoic acid contents in EC $1dS{\cdot}m^{-1}$. If the soil EC is well managed within $4.0dS{\cdot}m^{-1}$, two plants would be cultivated in reclaimed land.

This study was conducted to investigate the possibility of peanut shell, a by-product of peanut, as a functional cosmetic ingredient. Peanut shell extract showed high antioxidant activity with IC50 values of 75.00, 46.33, and 472.83 ㎍/mL for DPPH and ABTS radical scavenging and SOD-like activity, respectively. Furthermore, peanut shell extract was efficiently decreased the MMP-1 and MMP-3 protein level in the UVB treated-HaCaT cell and maintained procollagen protein level similar to normal control. Similar to anti-wrinkle related protein expression assay, the IC50 value of elastase and collagnease inhibition in peanut shell extract was lower as 0.30 and 0.09 mg/mL, respectively, than that of the positive control. Additionally, eriodictyol and luteolin, which are isolated from peanut shell extract, showed 53.8 and 98.0% elastase inhibition rate, respectively, and 60.1 and 72.5% collagenase inhibition rate, respectively, at a concentration of 0.1 mg/mL. Thus, luteolin was assumed to be the effective ingredient for wrinkle inhibition in peanut shell extract. As a result of stability evaluation of lotion and cream formulations containing peanut shell extract, it was confirmed to be a stable formulation with no significant changes. Therefore, it is considered that peanut shell extract can be applied as a cosmetic ingredient for wrinkle inhibition.
The transitivity of Chemical constituents by Pleurotus ostreatus cultivated in different raw sawdusts, which are Juglans mandchurica, Cudrania tricuspidata and Lindera glauca, was investigated. The HPLC chromatography patterns on the chemical constituents of P. ostreatus showed the similar chromatography patterns in all different raw sawdusts and control sawdust. The unknown chemical constituents of P. ostreatus cultivated in the 10%, 20% mixed medium added 10 %, 20% different raw sawdusts, respectively, were increased. But the significance results in the mixed medium added 50% different raw sawdusts were not showed. The chromatography patterns of mycelia grown in media added the 80% MeOH extracts of three tree species showed the similar patterns in comparison with control mycelia. In the results, the secondary metabolites of functional media were not degrade and changed to other derivatives compounds by P. ostreatus.

Anthocyanins belong to a group of flavonoid compounds and are well known for their various health beneficial effects, which include antioxidative activities. Among them, the major anthocyanins isolated from seed coat of black soybean (Glycine max L.) were previously characterized as glycosides containing glucopyranose. Asthma is an allergic disease that is strongly associated with various immune cells, including basophils and mast cells. Eosinophils, basophils, and mast cells play important roles in allergic asthma through the release of inflammatory mediators such as asthma-specific T-helper 2 (Th2) cytokines and subsequent amplification of asthma symptoms via degranulation. Rat basophilic leukemia RBL-2H3 cells are the most common in vitro models for evaluating allergic reactions. In this study, we examined the effects of anthocyanin from seed coat of black soybean on antigen-stimulated degranulation and Th2 cytokine production in RBL-2H3 cells. Cell degranulation was evaluated by measuring the release of ${\beta}$-hexosaminidase. ${\beta}$-Hexosaminidase release and Th2 cytokine production in RBL-2H3 cells was much higher upon stimulation with IgE-antigen complex than those in untreated control cells. Anthocyanins significantly suppressed IgE-antigen complex-induced degranulation of RBL-2H3 cells and inhibited IgE-antigen complex-mediated interleukin (IL)-4, IL-13, and tumor necrosis factor ${\alpha}$ (TNF-${\alpha}$) production in RBL-2H3 cells. These findings suggest that anthocyanins from seed coat of black soybean effectively inhibit allergic reactions and may have beneficial effects against allergic asthma.
The rice bran, a by-product of rice milling process, is well known for various functional components, such as tocopherol, tocotrienol, ${\gamma}$-oryzanol, carrying antioxidant activities. This study was conducted to investigate the antioxidant components and antioxidant activities in rice bran of different Korean rice cultivars. The 8 isomers of vitamin E, ${\gamma}$-oryzanol, flavonoids, and polyphenolics in rice bran from 16 Korean premium and high quality rice cultivars were quantified. DPPH and ABTS radical scavenging activities and reducing power of the ethanol extracts of rice bran were measured. 'Hopum' showed the highest total vitamin E content, $221.47{\mu}g/g$ among the cultivars, and 'Hanseol' showed the lowest content. The rice bran showed different compositions of ${\alpha}-$, ${\beta}-$, ${\gamma}-$, ${\delta}-$ tocopherol and tocotrienol among rice cultivars. The antioxidant contents were also different by cultivar; the ${\gamma}$-oryzanol contents ranged from 1.99 mg/g (Unkwang) to 4.30 mg/g (Chilbo), the polyphenol contents ranged from 427.22 mg gallic acid eq./100 g (Odaebyeo) to 775.80 mg gallic acid eq./100 g (Hopum). 'Hopum' also had the highest DPPH and ABTS free radical scavenging activities, 9.82% and 187.5 AEAC mg/100 g, respectively. In vitro, the rice bran extracts from 'Hopum' had significantly higher antioxidant activities than that of other cultivars.
Wine extracts of four different berry fruits, such as mulberry, blueberry, strawberry, and raspberry, were investigated for antioxidant, anti-tyrosinase, and ${\alpha}$-glucosidase activities by using in vitro assays. Additionally, quantitative changes of ${\alpha}$-glucosidase inhibitor in mulberry wine were determined by HPLC according to mulberry cultivars and fermentation process. Among four berry wines examined, mulberry wine showed the most potent ${\alpha}$-glucosidase inhibitory activity with 69.37% at 0.23 mg/mL, while blueberry and strawberry wines exhibited the strongest inhibition against DPPH radical and tyrosinase activity, respectively. Four compounds were isolated and purified from mulberry wine by a series of isolation procedures, such as solvent fractionation, and Diaion HP-20, ODS-A, and Sephadex LH-20 column chromatographies. Among them, Comp. 4 exerted the strongest ${\alpha}$-glucosidase inhibitory activity ($I_C_{50}=31.57{\mu}M$), and its chemical structure was identified as quercetin by UV and NMR spectral analysis. Finally, the "Daeseongppong" (16.83 ppm) muberry wine had larger amount of quercetin than the "Iksuppong" (14.85 ppm) and "Cheongilppong" (8.92 ppm) mulberry wines, but their contents of three mulberry wines decreased considerably with aging process. These results suggest that mulberry wine containing quercetin acted as ${\alpha}$-glucosidase inhibitor may be useful as a potential functional wine for improving diabetic disorder.
In order to select the raspberry cultivars that have high contents of bioactive constituents and high antioxidant activities, 7 cultivated and 2 wild raspberries which were selected and cultivated in the Bokbunja Institute were evaluated for their physicochemical characteristics, bioactive constituents, and antioxidant activities. The wild raspberry of Asan was the smallest among the sample raspberries but it had the highest sugar and lowest acid contents among the raspberries. Another wild raspberry of Ulleungdo had the highest total phenolic compound and ellagic acid contents, 182.97, $55.25mg{\cdot}100g^{-1}FW$, respectively, although it was small and had low sugar and high acid contents. Among the widely cultivated raspberry cultivars in Kimhae, 'Wangttal' cultivar was a big raspberry with 12.80% sugar content, and another unknown raspberry cultivar was as small as the wild raspberry with 14.60% sugar content. Although 'Wangttal' and the unknown raspberry cultivars cultivated in Kimhae possess lower contents of total phenolic compound (159.62, $165.94mg{\cdot}100g^{-1}$) and ellagic acid (45.7, $52.1mg{\cdot}100g^{-1}$ ) than the wild raspberry of Ulleungdo, the contents of total flavonoids (14.28, $14.90mg{\cdot}100g^{-1}$) and total anthocyanins (28.69, $30.48mg{\cdot}100g^{-1}$) were higher. Also the wild raspberry of Ulleungdo, 'Wangttal', and the unknown raspberry cultivar of Kimhae had higher antioxidant activities measured by FRAP (Ferric reducing antioxidant power), DPPH (2,2-diphenyl-1-picrylhydrazyl), and ABTS (2,2' azinobis-(3-ethylbenzothiazoline-6-sulphonic acid) assays. The present study shows that three raspberry cultivars could be potent resources for raspberry breeding and functional material development.
Raw-red bean (RR) should be boiled in hot water, and only boiled-red bean (BR) has been used in the food industry. In the course of development of functional food using red- bean (Phaseolus radiatus L), hot- water extracts (HWEs) of RR and BR were prepared, respectively and their components and various biological activities were compared. The extraction yield at $100^{\circ}C$ of RR (16.2%) was higher than that of BR (14.8%), and contents of total polyphenols, total flavonoids and reducing sugars of HWE of RR were 2.5-fold, 2.1-fold and 1.5-fold higher than those of HWE of BR. In anti-oxidation activity assay, scavenging activities against DPPH anion and ABTS cation as well as reducing power of RR was higher than those of BR. The results suggest that the anti-oxidant compounds in red bean might be heat-liable or discarded during boiling in hot-water as a cooking drip. Unexpectedly, nitrite scavenging activity was stronger in HWE of BR than RR. In anti-microbial activity assay, HWE of RR ($500{\mu}g/disc$) showed growth inhibition activity against gram-positive bacteria, whereas HWE of BR did not show any activity against any tested bacteria and fungi. Assay of in-vitro anti-diabetes and anti-thrombosis activities, which were previously reported in ethanol extract of red-bean, revealed that HWEs of RR and BR did not show significant activities against ${\alpha}$-amylase, ${\alpha}$-glucosidase, thrombin, prothrombin, or blood coagulation factors. Our results suggest that the anti-oxidation, anti-diabetes and anti-thrombosis activities of HWEs of RR and BR were lower than those of ethanol extracts of red bean, and bioactive substances in RR were destroyed during boiling or discarded after boiling. Further research on suitable boiling and re-use of cooking drip of red bean is necessary.
In the current study, comparisons of Oenothera Biennis seed extracts with water, ethanol, methanol, and 70% ethanol in their total polyphenolics contents, anti-oxidant, anti-neurotoxicity, anti-cancer, and immune-modulatory activities were investigated. Compared with other extracts, those concentrations of total phenolics and flavonoids were the highest in MeOH extract (31.90 mg GAE/g and 20.66 mg QE/g). The radical scavenging and reducing power activities were dose-dependently increased by treatment of O. Biennis seed water, EtOH, MeOH, and 70% EtOH extracts. Furthermore, pretreatment of water, EtOH, and MeOH extracts significantly reduced glutamate-induced cytotoxicity in HT22 hipocampal neuron cells. In the case of cancer cells, MeOH extracts showed lower $IC_{50}$ values in HepG2 ($74.21{\mu}g/ml$), A549 ($188.24{\mu}g/ml$), MCF-7 ($186.42{\mu}g/ml$), and B16 ($101.80{\mu}g/ml$) than other extracts, where those water ($101.96{\mu}g/ml$) and EtOH ($788.39{\mu}g/ml$) extracts showed the lowest $IC_{50}$ activity in HT-29 and PC-3 cells, respectively. O. Biennis seed extracts did not show any cytotoxicity in RAW 264.7 macrophages at the concentration of $1-10{\mu}g/ml$, whereas 70% EtOH extract dose-dependently enhanced nitric oxide (NO) production in RAW 264.7 cells. Overall, we evaluated that various bioactive potentials of O. Biennis seed extracts which would relate with phenolic compounds abundance, thus these can be useful to future developments as functional food ingredients and natural medicines.
