Cloning and overexpression of lysozyme from Spodoptera litura in prokaryotic system

  • Insect lysozymes are basic, cationic proteins synthesized in fat body and hemocytes in response to bacterial infections and depolymerize the bacterial cell wall. The c-type lysozyme of the insect Spodoptera litura (SLLyz) is a single polypeptide chain of 121 residues with four disulfide bridges and 17 rare codons and is approximately 15 kDa. The full-length SLLyz cDNA is 1039 bp long with a poly(A) tail, and contains an open reading frame of 426 bp long (including the termination codon), flanked by a 54 bp long 5' UTR and a 559 bp long 3' UTR. As a host for the production of high-level recombinant proteins, E. coli is used most commonly because of its low cost and short generation time. However, the soluble expression of heterologous proteins in E. coli is not trivial, especially for disulfide-bonded proteins. In order to prevent inclusion body formation, GST was selected as a fusion partner to enhance the solubility of recombinant protein, and fused to the amplified products encoding mature SLLyz. The expression vector pGEX-4T-1/rSLLyz was then transformed into E. coli BL21(DE3)pLysS for soluble expression of rSLLyz, and the soluble fusion protein was purified successfully. Inhibition zone assay demonstrated that rSLLyz showed antibacterial activity against B. megaterium. These results demonstrate that the GST fusion expression system in E. coli described in this study is efficient and inexpensive in producing a disulfide-bonded rSLLyz in soluble, active form, and suggest that the insect lysozyme is an interesting system for future structural and functional studies.

관성 측정 장치의 융합연구 형태와 방법에 관한 체계적 고찰 (Systematic Review on the Type and Method of Convergence Study of Inertial Measurement Unit)

  • 본 연구의 목적은 관성 측정 장치(Inertial Measurement Unit, IMU)의 융합연구 형태와 방법에 대한 연구를 체계적 문헌고찰 방법으로 분석하여 IMU의 형태와 방법에 대한 경향을 파악하고자 한다. 연구수행은 PRISMA(Preferred Reporting Items for Systematic Reviews and Meta-Analyses)가이드라인을 이용하여 수행되었다. 3개의 데이터베이스에서 검색된 630편 중 최종적으로 선정기준에 부합하는 23편을 선정하였다. 본 연구결과 전 세계적으로 IMU를 사용한 다양한 연구를 진행하고 있음을 알 수 있었고, IMU의 형태는 스트랩, 전신 슈트, 벨트, 손목시계, 신발, 장갑이 있었다. 이 중, 스트랩 형태의 IMU가 11편으로 가장 많았다. IMU의 장점인 간소화와 실시간 데이터 수집, 적용의 쉬움으로 작업 활동, 보행, 관절 가동 범위 등의 측정 방법으로 사용되었다. 본 연구의 결과는 IMU의 연구를 진행하는 의료 및 재활 분야의 전문가들에게 기초 자료로 활용될 수 있으리라 기대된다.

  • It is expected that the typical lifestyle of the future will be transformed into an opulent and comfortable existence as the quality of life improves due to the increase in household income and reduction in working hours. In the meantime, as the standard of living becomes increasingly more comfortable and plentiful, the toll on physical health becomes magnified as a result of obesity and insufficient exercise caused by super nutrition and change in labor conditions. This has instigated a deep awareness in fitness on the part of many people, forcing them to recognize the significance of daily exercise and physical activity. The Elliptical Cross Trainer(ECT), which has drawn wide attention recently, is a non-impact athletic apparatus that not only promotes exercise of the upper body parts in such sports as skiing but also the exercise of lower parts of the body on a treadmill. It is a type of cross training athletic gear that has been developed for aerobic exercise throughout the entire body. It has already formed a market as big as that of the treadmill in Europe, America, etc. Recently, its demand is growing sharply in the Korean markets as well as those in Northeast Asian countries. Despite such demand increase and expansion, since most of the expensive ECTs are exclusively supplied by suppliers in only a few advanced countries, localization of the ECT is urgently required in order to enhance competitiveness of Korean manufacturers and to expand the market. The ECT development project has been in full swing for approximately two year since 2004 in order to secure independent design, as well as engineering and manufacturing processes in efforts to develop a commercially viable ECT.

Expressed Protein Ligation of 5-Enolpyruvylshikimate-3-phosphate (EPSP) Synthase: An Application to a Protein Expressed as an Inclusion Body

  • Expressed protein ligation (EPL) technique, joining recombinantly expressed proteins to polypeptides, has been widely adopted for addressing various biological questions and for drug discovery. However, joining two recombinant proteins together is sometimes difficult when proteins are expressed insoluble and unrefoldable, because ligation-active proteins via intein-fusion are obtainable when they are folded correctly. We overcame this limitation coexpressing target protein with additional methionine aminopeptidase (MAP) which enhances removal of the initiation methionine of recombinantly expressed protein. Our approach demonstrated that two domains of 46 kDa 5-Enolpyruvylshikimate-3-phosphate (EPSP) synthase, a target of herbicide glyphosate, were successfully joined by native chemical ligation, although its C-terminal domain was expressed as an inclusion body. The intein-fused N-terminal fragment of EPSP synthase (EPSPSN, residues 1-237) was expressed and the ligation-active thioester tagged N-terminal fragment (EPSPSN-thioester) was purified using a chitin affinity chromatography and mercapto-ethanesulphonate (MESNA) as intein thiolysis reagent. Its Cterminal fragment (EPSPSC, residues Met237-238CYS-427), expressed as an inclusion body, was prepared from an additional MAP-expressing strain. Protein ligation was initiated by mixing ~1 mM of EPSPSN-thioester with ~2 mM of EPSPSCCYS (residues 238CYS-427). Also we found that addition of 2% thiophenol increased the ligation efficiency via thiol exchange. The ligation efficiency was ~85%. The ligated full-length EPSP synthase was dissolved in 6 M GdHCl and refolded. Circular dichroism (CD) and enzyme activity assay of the purified protein showed that the ligated enzyme has distinct secondary structure and ~115% specific activity compared to those of wild-type EPSP synthase. This work demonstrates rare example of EPL between two recombinantly expressed proteins and also provides hands-on protein engineering protocol for large proteins.

누에나방(Bombyx mori)성충의 생존시수에 따른 지방체 세포의 미세구조 변화 (Age-Related Ultrastructural Changes in Fat Body of Silkworm Adult, Bombyx mori)

  • 본 연구는 서로 다른 수명 패턴을 갖고 있는 누에성충 중 두가지 타입을 이용하여 수행되었다. 장명품종(성충 수명이 15일 이상; LLS), 단명 품종(성충 수명이 5일 미만;SLS). 누에성충의 노화 생리를 밝히기 위해 장단명 암수에 있어서의 지방체 미세구조를 비교해 보았다. 단명품종에 있어서는, 암컷에는 조면소포체 및 글리코겐 과립이 세포질내에서 다량 확인된 반면, 수컷에는 활면소포체만이 세포질내에서 발견되었다. 또한 단명품종에 있어서는, 성충 3일째 이후 미토콘드리아의 용적이 비대해지는 경향을 보였으며, 많은 지방구 퇴화가 관찰되었다. 단명품종에 반해 장명품종에서는, 성충 5일째의 암컷은 비교적 정상적인 미토콘드리아와 핵막이 관찰되었다. 성충 15일째에 이르러서야 대부분의 세포막이 사라졌고 미토콘드리아가 비정상적으로 비대해졌다. 장명품종 수컷(성충 10일째)의 세포질내에서 다량의 지방과립이 관찰되었으며, 이 개체는 성충 15일째에 세포내용물이 모두 고갈되어 사망하였다. 따라서, 금후 수명에 관한 조직 연구를 위해서는 조직의 변화상을 관찰하기 용이한 단명품종이 적절할 것으로 사료된다.

안면부에 발생한 혈관근종의 치험례 : 증례보고 (Angiomyoma on Face : A Case Report)

  • Angiomyoma is a rare, benign smooth muscle cell tumor. These tumors may be found anywhere in the body. They frequently occur in the lower extremities except venous type. Angiomyoma in the head and neck area is very rare, and only a few cases have been reported. A 63 year-old male patient visited to outpatient clinic due to size-growing nodule-like lesion in the Lt. alar area. Excisional biopsy was done for diagnosis. The lesion was totally excised with 2 mm safety margin. Frozen biopsy of the lesion was requested, and all resection margins were proved negative. To cover the raw surface, full thickness skin grafting was performed. The graft was harvested from Rt. posterior auricular area. Tie over dressing was applyed on Lt. alar area. The graft was well taken and healed without any complication in both short term and long term follow up periods of 2 weeks, 1 month, 2 months, and 6 months. Donor site completed healed without any complications. The leiomyoma is benign tumor originated from smooth muscle, and it can be classified into solid leiomyoma, angiomyoma, and epithelioid leiomyoma. Especially, the angiomyoma consists of smooth muscle cell and blood vessel, and it is originated from the tunica media of blood vessel. Angiomyoma alone frequently occurs in the lower extremities as solitary painless subcutaneous tumor. Venous type of angiomyoma in the oral cavity was reported in other references, but on the facial surface it may be the first case reported as paper. So this report can be very meaningful.

A full path assessment approach for vibration serviceability and vibration control of footbridges

  • Most of the existing evaluation criteria of vibration serviceability rely on the peak acceleration of the structure rather than that of the people keeping their own body unmoved on the structure who is the real receiver of structural vibrations. In order to accurately assess the vibration serviceability, therefore, a full path assessment approach of vibration serviceability based on vibration source, path and receiver is not only tentatively proposed in this paper, taking the peak acceleration of receiver into account, but also introduce a probability procedure to provide more instructive information instead of a single value. In fact, semi-rigid supported on both sides of the structure is more consistent with the actual situation than simply supported or clamped due to the application of the prefabricated footbridge structures. So, the footbridge is regarded as a beam with semi-rigid supported on both sides in this paper. The differential quadrature-integral quadrature coupled method is not only to handle different type of boundary conditions, but also after being further modified via the introduction of an approximation procedure in this work, the time-varying system problem caused by human-structure interaction can be solved well. The analytical results of numerical simulations demonstrate that the modified differential quadrature-integral quadrature coupled method has higher reliability and accuracy compared with the mode superposition method. What's more, both of the two different passive control measures, the tuned mass damper and semi-rigid supported, have good performance for reducing vibrations. Most importantly, semi-rigid supported is easier to achieve the objective of reducing vibration compared with tuned mass damper in design stage of structure.

정적 플랭크 운동의 동작 별 효과성 차이 검증 -근전도 분석을 중심으로- (Verification of the Difference in Effectiveness of Static Plank Exercise by Motion -Focusing on EMG Analysis-)

  • 본 연구의 목적은 정적 플랭크 운동의 동작 차이에 따른 코어근육의 근활성도를 비교하는 것이었다. 본 연구의 대상자는 "J"대학교 20대 남성 10명을 대상으로 진행하였고(연령, 23.20±0.65 세; 신장, 174.54±1.51 cm; 체중, 70.00±2.24 kg; 신체질량지수, 22.94±0.51 kg/m2), 4가지의 풀, 엘보우, 사이드 및 리버스 정적 플랭크 동작을 수행하였으며, 근전도 분석을 위한 표면전극 부착 부위는 신체 근육의 우측복직근, 외복사근, 광배근 및 척추기립근으로 설정하였다. 본 실험 결과, 광배근과 척추기립근의 근활성도는 리버스 플랭크 동작 시 가장 높게 나타났고(p<.001), 복직근과 외복사근의 근활성도는 엘보우 플랭크 동작 시 가장 높게 나타났다(p<.001). 따라서 본 연구의 결과는 추후 정적 플랭크 동작 시 효과적인 운동 프로그램의 자료가 될 것으로 기대된다.

천연염색 임부복 디자인 개발을 위한 임산부들의 선호도에 관한 연구 - 대구.경북지역을 중심으로 - (A Study on Pregnant Women's Preference for Design Development of Natural Dyed Maternity Dresses - Focusing on Daegu and Gyeongbuk Regions -)

  • This study aims to examine the actual condition of and satisfaction with purchasing common maternity dresses and natural dyed maternity dresses as well as purchasing defined factors among pregnant women and those who lately gave birth to a baby. Addressed too are involved problems and the possible solutions. Among pregnant women and those who lately gave birth to a baby living in Daegu or Gyeongbuk areas, a preliminary survey was conducted in July, 2008. After revised and complemented, the 177 copies of the questionnaire were distributed in October of the year, and investigated for two months up to December of the year. Except 6 copies of the questionnaire that were not appropriate for analysis, 171 copies were used, and the results are as follows: First, The majority of the respondents were women aged 30 to 35, full-time housewives, and having a monthly income of 2 million to 3 million won. Second, The respondents preferred one-piece dresses, leggings-type maternity dresses, and pink-tone colors. Third, The respondents regarded the feelings as the most important among the maternity dress purchasing defined factors. Fourth, They preferred maternity dresses priced less than 150,000 won. The respondents showed a lot of interests in and tended to buy natural dyed products.

후륜 조향 동력학 모델 및 제어 로직 개발 (Development of the Dynamic Model and Control Logic for the Rear Wheel Steering in 4WS Vehicle)

  • In the turning maneuver of the vehicle, its motion is mainly dependent on the genuine steering characteristics in view of the directional stability for stable turning ability. The under steer vehicle has an ability to maintain its own directonal performance for unknown external disturbances to some extent. From a few years ago, in order to acquire the more enhanced handling performance, some types of four wheel steering vehicle were considered and constructed. And, various rear wheel control logics for external disturbances has not been suggested. For this reason, in this posed rear wheel control logic is based on the yaw rate feed back type and is slightly modified by an yaw rate tuning factor for more stable turning performance. And an external disturbance is defined as a motivation of the additional yaw rate in the center of gravity by an uncertain input. In this study, an external disturbance is applied to the vehicle as a form of the additional yawing moment. Finally, the proposed rear wheel control logic is tested on the multi-body analysis software(ADAMS). J-turn and double lane change test are performed for the validation of the control logic.
