Prevention from microbial post-harvest injury of fruits and vegetables by using grapefruit seed extract, a natural antimicrobial agent (천연항균제처리에 의한 과채류의 선도유지 및 병해방지에 관한 연구 -저장중 병리적 장해 방지를 중심으로-)

  • In order to retain the freshness of fruits and vegetables and to reduce the rate of disease damage, grafruit seed extract (GPSE), natural microorganism control agent, was applied during the preservation process of fresh fruits and vegetables. GFSE showed an effective inhibitory action against plant putrefactive bacteria and fungi which were involved in the decay of fruits and vegetables. Minimal inhibitory concentrations for GFSE against the microbes were in the range of 50 to 2,000 ppm. Direct observation of microbial cells and spores using electron microscopy showed their function was destroyed by the treatment of the dilute solutions of GFSE. Fresh Welsh onions, onions and red peppers treated with GFSE and stored in polyethylene film (0.1 mm) retained better quality in color and texture than the non-treated control. GFSE was efficient in controlling the germination of potatoes. It was observed that GFSE would reduce disease damages and have bactericidal and fungicidal properties during the storage of such fruits and vegetables as zucchinis, cucumbers, tomatoes and mandarin oranges.

Antioxidant and tyrosinase inhibitory activities of immature fruits of Malus pumila cv. Fuji (미성숙 사과의 항산화 및 tyrosinase 저해 활성 평가)

  • In this study, the antioxidant and tyrosinase inhibitory activities of aqueous ethanolic extract from the immature fruits of Malus pumila cv. Fuji were evaluated. The antioxidant capacities of the extract was investigated employing radical scavenging assays using DPPH and $ABTS^+$ radicals. The skin-whitening effect of M. pumila cv. Fuji extract was tested using mushroom tyrosinase assay. In addition, the total phenolic content was measured by a spectrophotometric analysis. All tested samples showed a dose-dependent radical scavenging and tyrosinase inhibitory activities. Among the tested samples, the ethyl acetate (EtOAc)-soluble portion from the immature fruits of M. pumila cv. Fuji was showed the significant DPPH and $ABTS^+$ radicals scavenging activities. Also, the tyrosinase inhibitory activity of the ethyl acetate (EtOAc)-soluble portion from immature apples was higher than other solvent-soluble portion. These results suggest that unripe fruits of M. pumila cv. Fuji could be considered as a new valuable source of natural antioxidant and skin-whitening agents. Systematic investigation of immature fruits of Malus pumila cv. Fuji will be performed for the further development of its biological properties.

Antioxidant Activity of Phenol Compounds from Mulberrry Fruits (오디로부터 분리한 페놀성 물질의 항산화 효과)

  • For the purpose of developing natural antioxidant, the antioxidative activity of phenolics isolated from Korean Mulberry(Morus) fruits was determined. Optimum extracting condition for phenolics were 60$\%$(v/v) ethanol. The content of rutin and isoquercetrin as flavonoids were 82.83 mg/100 g and 43.23 mg/100 g respectively. The 2,2'-azinobis-3-ethyl benzothiazoline-6-sulfonic acid radical decolorization and antioxidant protection factor (PF) were determined with extracts from Mulberry fruits. Results showed 98$\%$ inhibition on ABTs by 80$\%$ ethanol extract and 1.25 PF on antioxidant protection factor by 60$\%$ ethanol extract. Electron donation ability on DPPH was higher 60$\%$ ethanol extract than other ethanol extracts. Also, hydroxyl radical scavenging activity of extracts was higher 60$\%$ ethanol extract than other extracts.

Antioxidative Activity of the Extracts from the Leaves and Fruits of Acer ginnala

  • The antioxidative effect of the extracts from the leaves and fruits of Acer ginnala against free radicals was studied by two different methods using DPPH radical-generating system, and hydroxyl radical-generating system $(Cu^{++}/H_2O_2\;system)$ which induces DNA strand breaking. Compared with well known antioxidative plants, green tea, Scutellaria baicalensis, the Acer ginnala extracts showed excellent radical-scavenging activity in DPPH radical-generating system and inhibited effectively hydroxyl radical induced-DNA strand breaking in a concentration-dependent manner in $Cu^{++}/H_2O_2$ system whereas the green tea extract stimulated the strand breaking at a low concentration. These results suggest that he extracts from the leaves and fruits of Acer ginnula could be good antioxidative agents.

  • The ethanol extracts of mixed vegetables (Bioactive Vegetables, BV), mixed fruits (Bioactive Fruits, BF) and their liquid formulation (Chungpae Plus , CP) were evaluated for their antioxidative and antigenotoxic activities. They were shown to possess the significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and were revealed to show the inhibitory effect of lipid peroxidation as measured by malondialdehyde (MDA) formation although the potencies were not higher than those of vitamin C. They did not possess any pro-oxidant effect on bleomycin-Fe(III) dependent DNA degradation, whereas vitamin C showed strong pro-oxidant effect. Furthermore, oral administration of BV and BF inhibited micronucleated reticulocyte (MNRET) formation of mouse peripheral blood induced by KBrO3 treatment in vivo. CP also showed significant inhibition under same experimental condition. Therefore, the liquid formulation (CP) containing BV and BF may be a useful natural antioxidative and antigenotoxic agent by scavenging free radicals, inhibition of lipid peroxidation and protecting chromosomal damage.

  • A total of 65 isolates of Penicillium were isolated from decayed post-harvest fruits of apple pear citrus and grape. The Penicillium species isolated from the apple were idnetified as P. aurantiogriseum and P. expansum those from the pear were P. crustosum and P. expansum and those from the grape were P. aurantiogriseum and P. expansum, From decayed citrus fruits. P. digitatum and P. italicum were isolated. Citrinin and patulin from these species in the YES(yeast extract sucrose) broth were extracted with ethyl acetate and purified by thin-layer chromatography(TLC) and high performance liquid chromatography(HPLC) Among 51 isolates of Penicillium from apple pear and grape 7 isolates produced citrinin 13 isolates produces patulin and 12 isolates produced citrinin and patulin also. All 14 isolates of Penicillium from citrus produced only patulin.

  • In the growth of the Rubus coreanus Miquel fruits, the unripened (10 days) and ripened (25 days) fruits after flowering were harvested. The chemical characteristics of different maturational stages of the unripened and ripened fruits were investigated. Total amount of phenolic compounds was 4.00-7.56% in the unripened fruits and 3.78-5.57% in the ripened fruits, respectively. Furthermore, total amounts of organic acids such as malic, citric, succinic, and oxalic acids were 16.40 mg/100 g in the unripened fruits and 28.82 mg/100 g in the ripened fruits, respectively. In organic acids of the unripened and the ripened fruits, citric acid (8.76-15.47 mg/100 g) was the highest amount among other organic acids. Soluble sugars were significantly increased from 11.07 to 21.54% in the unripened and ripened fruits. Therefore, ripened fruits had the high levels of phenolic compounds, organic acids and soluble sugars. For the biological studies of R. coreanus, methanol extracts of R. coreanus leaves, fruits, and stems were evaluated for their insecticidal activities against Myzus persicae (Hemiptera: Aphididae), Plutella xylostella (Lepioptera: Plutellidae) and Spodoptera litura (Lepioptera: Noctuidae) by leaf dipping method. From these results, the extract of R. coreanus leaves revealed potent insecticidal activity against P. xylostella. However, the methanol extracts of R. coreanus fruits and stems had no any insecticidal activity against M. persicae, P. xylostella and S. litura. The R. coreanus leaves have promising potential as new insecticidal agent against P. xylostella.

Hepatoprotective Effect of Coccinia indica Against $CCl_4$ Induced Hepatotoxicity

  • The hepatoprotective effect of the ethanolic extract of Coccinia indica fruits in rats treated with carbon tetrachloride. In hepatotoxic rats, liver damage was studied by assessing parameters such as aspartate aminotransferase (AST), alanine aminotransferase (AlT), alkaline phosphatase (AIP) and gamma glutamyl transpeptidase (GGT) in serum, and concentrations of total proteins, total lipids, phospholipids, triglycerides and cholesterol in both serum and liver. The effect of co-administration of ethanolic extract on the above parameters was further investigated. Histopathological study of the liver in experimental animals was also undertaken. Hepatic damage as evidenced by a rise in the levels of AST, AIT, AIP and GGT in serum, and also changes observed in other biochemical parameters In serum and liver showed a tendency to attain near normalcy in animals co-administered with the extract. The normal values for AST (IU/L), AIP (IU/I), protein (g/100 ml) and total lipids (mg/100 ml) in serum (i.e.,20.24, 70.04, 5.72 and 135.54 respectively) were found to alter towards values 32.61, 127.11, 3.83 and 265.91 in hepatotoxic rats. These parameters Attained near normal values (I.e.,22.82, 79.30, 5.22 and 151.24 for AST, AIP protein and total lipids respectively) in ethanolic extract co-administered rats. Profound steatosis, ballooning degeneration and nodule formation observed in the hepatic architecture of $CCl_4$ treated rats were found to acquire near-normalcy in drug co-administered rats, thus corroborating the biochemical observations. Thus the study substantiates the hepatoprotective potential of ethanolic extract of Coccinia indica fruits.
