Effect of Evodiae Fructus on the ovarian function and gene expression of caspase-3, MAP kinase and MPG in female mice (오수유 투여가 자성생쥐의 생식능력과 caspase-3, MAPK 및 MPG유전자 발현에 미치는 영향)

  • Purpose: These experiments were undertaken to evaluate the effect of administration of Evodiae Fructus on ovarian functions and differential gene expressions related caspase-3, MAPK and MPG in female mice. Methods: We administered the Evodiae Fructus to 6-week-old female ICR mice for 4, 8, or 12 days. With different concentration of Evodiae Fructus, the female mice were injected PMSG and hCG for ovarian hyperstimulation. The mice divided into 3 groups for each experiment. We chose the caspase-3 for cell apoptosis, MAPK and MPG genes for cell viability and DNA repair. Results: In case of 4, 8, 12 day of Evodiae Fructus, we were examined the mean number of total ovulated oocytes and the number of morphologically normal oocytes. We were also examined the embryonic developmental competence in vitro. In addition we were also examined the differential expression of cell viability related genes, caspase-3, MAPK and MPG according to concentration and duration of Evodiae Fructus administration. MPG gene expressions for cell viability and DNA repaie were increased in dose dependent manner than that of control group in 4-day administration group. Conclusion: It is suggested that the medication of Evodiae Fructus has beneficial effect on reproductive functions of female mice via promotion of cell proliferation.

Fetal safety of medicinal herbs and food ingredients during pregnancy: Recommendations from traditional Korean medicine based on expert opinions

  • Objectives: This study aimed to establish and provide reliable information for general public, based on expert consensus, on the risks of misuse of medicinal herbs for food and pure food ingredients for the fetus during pregnancy. Methods: A panelist of seven traditional Korean medicine (TKM) gynecologists responded to a questionnaire summarizing the fetal safety literature for twenty-five medicinal herbs for food and pure food ingredients derived from medicated diet (藥膳, Yaksun) recipes during three online Delphi rounds anonymously. Results: Ginkgonis Semen (Ginkgo nut), Illici Veri Fructus (Star anise), lavender, bitter gourd, and parsley were agreed at the level 1 of "Do not consume". These five ingredients were recognized as having significant risks both in the literature evidence and in expert opinion. Rosemary, Citri Unshius Pericarpium, Discoreae Rhizoma, lemongrass, Schisandrae Fructus, Cassiae Semen, Foeniculi Fructus, Mori Fructus, Cinnamomi Cortex, and Astragali Radix were agreed at the level 2 of "consultation with TKM practitioner is required". Conclusion: Based on the consensus of a seven-member expert panel of TKM gynecologists, consumption of Ginkgonis Semen (Ginkgo nut), Illici Veri Fructus (Star anise), lavender, bitter gourd, and parsley should be avoided by pregnant women. For Rosemary, Citri Unshius Pericarpium, Discoreae Rhizoma, lemongrass, Schisandrae Fructus, Cassiae Semen, Foeniculi Fructus, Mori Fructus, Cinnamomi Cortex, and Astragali Radix, the level 2 advisory may be recommended to use with caution and to consult a TKM practitioner for advice on consumption, dose, and duration.

  • Sedative activity of Zizyphi fructus was evaluated by potentiation of hexobarbital-induced hypnosis test and its active principles have been characterized as nornuciferine and lysicamine. A new cyclopeptide alkaloid, daechucyclopeptide-1 was isolated together with zizyphusine.

  • Magnoliae Flos, Xanthii Fructus and Menthae Herba have long been used for treatment of nasal disease in Korea. The 70%-ethanol extracts of Magnoliae Flos, Xanthii Fructus and Menthae Herba were used externally in order to investigate the effects on a few strains of microbe, nasal alergy and inflammation, rhinitis and sinusitis. When the proportion of Magnoliae Flos : Xanthii Fructus : Menthae Herba was 2 : 2 : 1, the results showed great significance compared to the control group(non-extracts) in treatment of various nasal disorder.

  • Gardeniae Fructus (Gardenia jasmmoides Ellis) is widely used as a natural food colorant and as a traditional medicine for the treatment of hepatic and inflammatory diseases. The quality evaluation of Gardeniae Fructus was carried out by HPLC using geniposide as a marker. The geniposide was isolated from commercial Gardeniae Fructus on ODS column using a mixed solvent of acetonitrile-water (v/v 9:91) as mobile phase and a detection wavelength 240 mm. The content of geniposides in twenty different samples was in a range of 1.20% to 7.17% of total tissues and the average was 4.97%.

Antioxidant and Cell Activity Using Extracts of Mume Fructus (오매 추출물들의 항산화 및 세포 활성)

  • Mume fructus is the roasted fruits of Prunus mume and has been used as traditional chinese medicine. In this study, mume fructus extracts were prepared by three different methods including hot water extract (sample1), fermentation extract using Lactobacillus (Sample2-LP and sample-LA) and ethanol extract (sample3). Total polyphenol and flavonoid contents were improved by fermentation process, compared to water extract. Sample 3 showed the highest activity in DPPH radical scavenging. The cytotoxicity of the sample 2-LP was in the range of 83.3% cell viability at $300{\mu}g/m{\ell}$ concentration. Mume fructus extracts in a concentration-dependent manner inhibited melanogenesis and NO synthesis was inhibited. Specifically, the extracts fermented by L. plantarum (sample2-LP) showed higher anti-oxidant activity, anti-inflammatory and skin whitening activity than other extracts. It suggests that sample 2-LP could be potentially used as a resource of materials for functional cosmetics.

Antioxidative Activity and Active Compound Analysis of the Extract and Fractions of Corni Fructus (산수유 추출물 및 분획물의 항산화 활성과 활성성분 분석)

  • In this study, analysis of active compounds that are believed to be highly relevant to antioxidant activity was carried out on the methanol extract and its solvent fractions of Corni fructus. The DPPH radical scavenging activity for the comparison of antioxidant activity was higher in order of aqueous fraction > methanol extract > ethyl acetate fraction > n-hexane fraction. It is similar to the order of total polyphenol contents in the samples. As a result of LC-MS analysis, phenolic acid compounds such as caffeic acid, gallic acid and chlorogenic acid and lognin, which is known as a representative active ingredient of Corni fructus, were identified as active compounds. And the antioxidative activity and the total polyphenol content of the extracts and solvent fractions were found to be related to the contents of the compounds. Particularly, it was confirmed that phenolic acid such as caffeic acid contributes to the antioxidative activity of the aqueous fraction of Corni fructus methanol extract.

1H NMR-based metabolomic study of Cornus officinalis from different geographical origin

  • Cornus officinalis (Cornaceae) is primarily grown in Asian countries. The pericarp of C. officinalis (Corni Fructus) is a well-known traditional medicine with tonic, analgesic, and diuretic properties. We analyzed methanolic extracts of Corni Fructus (grown in Korea and China) by $^1H$ NMR spectroscopy. Metabolite profiling was performed to characterize the metabolic difference between different Corni Fructus origins (Korea or China). Principal components analysis revealed significant separation between Comus Fructus from different origins. The metabolites responsible for differences were identified using loading plots, coefficients plots, and variable influence on projection followed by t-tests. As a result, 16 metabolites were identified and quantified; tyrosine, acetate, sucrose, and malate differed the most between origins. These data suggest that NMR-based metabolomics can be used to identify differences between Corni Fructus samples obtained from different regions.

Anti-hyperlipidemic Effect of Sophorae Fructus on Hyperlipidemic Rat Induced by High Fat-rich Diet (회화나무 열매의 고지혈증 개선 효능)

  • Anti-hyperlipidemic effect of Sophorae Fructus in male Spague-Dawley rats induced by high-rich diet were studied. Oral administration of Sophorae Fructus extract significantly decreased serum total cholesterol (TC), LDL-cholesterol, triglyceride (TG), GOT and GPT levels, and incresed serum HDL-cholesterol level and also decreased the level of liver TC, TG, GOT and GPT. These results suggest that Sophorae Fructus extract is effective for the hyperlipidemia.

The Change of Ginsenosides Composition from Decocted Ginseng with Citric Acid and Schisandrae Fructus (인삼 탕액 제조시 구연산과 오미자에 의한 진세노사이드 조성 변화)

  • In order to observe the change of ginsenosides, two simplified decoctive conditions are set up aqueous citric acids (0, 12.5, 25, 50, 75, and 100 mM) with ginseng powder (3 g) and Schisandrae Fructus (0, 1, 2, 3 g) with ginseng powder (3 g). Decoctive temperature and duration are $95^{\circ}C$ and 1.5 h. The contents of major ginsenosides are decreased by increasing concentration of citric acid. But the contents of converted ginsenosides have not been a noticeable increase. In the decoctive condition with Schisandrae Fructus, the contents of major protopanaxdiol ginsenosides seriously decreases that compared with major protopanaxtriol ginsenosides. Therefore, Schisandrae Fructus provides the condition that predominantly converts protopanaxdiol ginsenosides in ginseng decoction.
