Effect of an herbal formulation on DOCA-salt and fructose induced models of hypertension in rats

  • The present study was carried out to investigate the antihypertensive effect of a folklore herbal formulation (HF) (300mg/kg/day; p.o.) in deoxycorticosterone acetate (DOCA)-salt induced and fructose induced hypertensive rats. In DOCA model, DOCA (15 mg/kg, s.c., twice a week) was administered to unilateral nephrectomized rats for 4 weeks. In fructose model, drinking water was replaced with 10% fructose solution for 6 weeks to induce hypertension. Systolic blood pressure (BP) was measured once every week during the treatment schedule. After completion of treatment schedule, BP and vascular reactivity to various agonists like Noradrenaline, Adrenaline, Phenylephrine and Serotonin (5-hydroxytrptamine; 5-HT) were recorded in rats of both models. A cumulative concentration response curve of 5-HT was carried out in isolated rat fundus strip of the DOCA-salt induced and fructose induced hypertensive rats. The results tend to suggest that HF possesses antihypertensive activity.

Activation of the renin-angiotensin system in high fructose-induced metabolic syndrome

  • High fructose intake induces hyperglycemia and hypertension. However, the mechanism by which fructose induces metabolic syndrome is largely unknown. We hypothesized that high fructose intake induces activation of the renin-angiotensin system (RAS), resulting in hypertension and metabolic syndrome. We provided 11-week-old Sprague-Dawley rats with drinking water, with or without 20% fructose, for two weeks. We measured serum renin, angiotensin II (Ang II), and aldosterone (Aldo) using ELISA kits. The expression of RAS genes was determined by quantitative reverse transcription polymerase chain reaction. High fructose intake increased body weight and water retention, regardless of food intake or urine volume. After two weeks, fructose intake induced glucose intolerance and hypertension. High fructose intake increased serum renin, Ang II, triglyceride, and cholesterol levels, but not Aldo levels. High fructose intake increased the expression of angiotensinogen in the liver; angiotensin-converting enzyme in the lungs; and renin, angiotensin II type 1a receptor (AT1aR), and angiotensin II type 1b receptor (AT1bR) in the kidneys. However, expression of AT1aR and AT1bR in the adrenal glands did not increase in rats given fructose. Taken together, these results indicate that high fructose intake induces activation of RAS, resulting in hypertension and metabolic syndrome.

  • Rats that are fed a fructose-rich diet develop hypertension, insulin resistance, and hypertriglyceridemia. To elucidate whether altered expression levels of endothelin-1 and nitric oxide synthase are related to the development of insulin-resistant hypertension, we examined the present study. Male Sprague-Dawley rats were fed a fructose-rich diet for 5 weeks. Systolic blood pressure significantly increased in fructose-fed rats. While serum free fatty acid and plasma nitrite/nitrate levels did not significantly differ between the fructose-fed and control groups, plasma insulin and serum triglyceride concentrations significantly increased in the former. Endothelin-1 mRNA expression in the aorta increased in fructose-fed rats. Neither the protein expression of constitutive nitric oxide synthase nor that of inducible nitric oxide synthase were significantly affected by fructose feeding. However, nitrite/nitrate levels in the aorta were significantly increased. These results suggest that an increase in vascular endothelin-1 is an important contributing factor to the development of hypertension in fructose-fed rats. However, the vascular nitric oxide pathway may not be causally related to the development of fructose-induced hypertension.

Antihypertensive activity of Zingiber officinale and Korean ginseng in experimentally induced hypertension in rats

  • We investigated the antihypertensive effect of Pet ether extract (PE) of ginger rhizome; its toluene fraction (TF) and Korean ginseng extract (KGE) in deoxycorticosterone acetate (DOCA) - salt induced and fructose induced hypertensive rats. In DOCA model, DOCA (25 mg/kg, once a week; s.c) was administered in uninephrectomised animals for 4 w. PE (50 mg/kg/day; p.o), TF (10 mg/kg/day; p.o) and KGE (30 mg/kg/day; p.o) were evaluated for their antihypertensive effect. In the fructose model, drinking water was replaced with fructose (10%) for five weeks to induce hypertension. PE (50 mg/kg/day; p.o) and KGE (30 mg/kg/day; p.o) were assessed for its antihypertensive effect in fructose model. After completion of the treatment schedule, vascular reactivity to various agonists like 5-HT, noradrenaline, adrenaline, phenylbiguanide and acetylcholine were recorded in rats of both the models. A cumulative dose response curve (CDRC) of 5-HT was carried out in isolated rat fundus strip of the fructose induced hypertensive rats. Chronic administration of PE (50 mg/kg/day; p.o), TF (10 mg/kg/day; p.o), and KGE (30 mg/kg/day; p.o) significantly reduced the blood pressure in DOCA salt whereas PE (50 mg/kg/day; p.o) and KGE (30 mg/kg/day; p.o) reduced the blood pressure in fructose induced hypertensive rats. Treatment with PE (50 mg/kg/day; p.o) and KGE (30 mg/kg/day; p.o) in fructose model for five weeks shifted the CDRC towards the right on rat fundus. The mechanism of action may partly involve the serotonergic antagonistic property.

Cinnamaldehyde Attenuates Cataractogenesis via Restoration of Hypertension and Oxidative Stress in Fructose-Fed Hypertensive rats

  • Objectives: Several studies have revealed that systemic hypertension is strongly associated with cataractogenesis. However, the pathophysiology and treatment is often unclear. In this study, we evaluated the anti-cataractogenic effect of cinnamaldehyde (CA), a natural organic compound, in rats with fructose-induced hypertension. Methods: The rats were divided into six groups. For six weeks, the normal group received a suspension of 0.5% carboxy methyl cellulose (10 mL/kg/day, p.o.) while five other groups received a 10% (w/v) fructose solution in their drinking water to induce hypertension. By the end of the third week hypertension had been induced in all the animals receiving fructose. From the beginning of the fourth week to the end of the sixth week, one of those five groups (control) continued to receive only 10% (w/v) fructose solution, one group (standard) received ramipril (1 mg/kg/day, p.o.) plus 10% (w/v) fructose solution, and three groups (experimental) received CA at doses of 20, 30, and 40 mg/kg/day p.o., plus 10% (w/v) fructose solution. Blood pressure was measured weekly using a non-invasive blood pressure apparatus. After six weeks, the animals were sacrificed, and the anti-cataractogenic effects on the eye lenses were evaluated. Results: Administration of fructose elevated both the systolic and the diastolic blood pressures, which were significantly reduced by CA at all dose levels. In the control group, a significant increase in the malonaldehyde (MDA) level and decreases in the total protein, $Ca^{2+}$adenosine triphosphate (ATP)ase activity, glutathione peroxidase, catalase, superoxide dismutase and glutathione levels, as compared to the normal group, were observed. Administration of CA at all doses significantly restored the enzymatic, non-enzymatic, antioxidants, total protein, and $Ca^{2+}$ATPase levels, but decreased the MDA level, as compared to the control group. Conclusion: The present study revealed that CA modulated the antioxidant parameters of the serum and lens homogenates in hypertension-induced cataractogenic animals.

고과당식이 랫드모델에서 적하수오 투여에 의한 대사증후군 개선효과 (Beneficial effect of Polygoni Multiflori Radix in high fructose diet-induced metabolic syndrome rat model)

  • Objectives : Polygoni Multiflori Radix (Jeokhasuo in Korean) is a Oriental traditional herbs widely used in East Asian countries. Overconsumption of fructose results in hypertension, dyslipidemia, obesity and impaired glucose tolerance which have documented as a risk of cardiovascular diseases. This experimental study was designed to investigate the beneficial effects of an ethanol extract from Polygoni Multiflori Radix (PMR) in high-fructose (HF) diet-induced metabolic syndrome rat model. Methods : Sprague-Dawley (SD) rats were divided into three groups; Control group, receiving regular diet and tap water, HF group, and HF + PMR group both receiving supplemented with 65% fructose (n=10), respectively. The HF + PMR group initially received HF diet with PMR (100 mg/kg/day) for 8 weeks. Results : PMR significantly prevented the metabolic disturbances such as hyperlipidemia, hypertension and impaired glucose tolerance. Chronic treatment with PMR significantly decreased body weight, fat weight and adipocyte size, suggesting a role of anti-obesity effect. PMR led to improve the hyperlipidemia through the increase in HDL cholesterol level as well as the decrease in triglyceride and LDL cholesterol level. In addition, PMR suppressed adhesion molecules and endothelin-1 (ET-1) expression in aorta resulting in the decrease of hypertension. In muscle tissue, PMR significantly recovered the HF-induced insulin resistance through increase of insulin receptor substrate-1 (IRS-1), p-$AMPK{\alpha}1/2$, and p-Akt expression. PMR improved HF-induced metabolic disorders and its action was caused by energy metabolism-mediated insulin signaling activation. Conclusions : These results demonstrate that PMR may be a beneficial therapeutic for metabolic syndrome through the improvement of hyperlipidemia, obesity, insulin resistance and hypertension.

쪽파 에탄올 추출물이 과당-유도 고혈압 흰쥐에 미치는 영향 (Effects of the Ethanol-extract of Allium wageki on the Fructose-induced Hypertensive Rats)

  • The effects of the ethanol (EtOH)-extract of Allium wageki were studied on the changes of body weight (BW), systolic blood pressure (SBP), triacylglycerol (TG), cholesterol, and insulin levels in Sprague-Dawley rats in which high fructose feeding elicited hyperinsulinemia, hypertriglyceridemia, and hypertension. SBP and BW were measured for 7 weeks on the fructose diet, fructose/EtOH-extract, vehicle- treated, or vehicle/EtOH-treated rats. The EtOH-extract of Allium wageki blocked the increases of SBP ($119.4{\pm}3.7\;mmHg$ in fructose/EtOH-extract of Allium wageki; $141{\pm}4.3 \;mmHg$ in fructose diet, p<0.01) and BW ($368{\pm}11.4\;g$ in fructose/EtOH-extract of Allium wageki; $402{\pm}15.7\;g$ in fructose diet, p<0.05) by high fructose-diet. The increases of TG, total cholesterol, and insulin levels were also blocked by administration of EtOH-extract of Allium wageki in fructose-induced hypertensive rats. These results suggest that EtOH-extract of Allium wageki has protect effects on the increases of TG, total cholesterol, insulin level, and blood pressure in fructose-induced hypertensive rats.

고과당식이 랫드모델에서 복분자 투여에 의한 대사증후군 개선효과 (Beneficial Effect of Rubus Coreanus Miq in a Rat Model of High Fructose Diet-induced Metabolic Syndrome)

This experimental study was designed to investigate the beneficial effects of Rubus coreanus Miq.(RCM) in high-fructose diet-induced metabolic syndrome. Animals were divided into three groups; Control group fed regular diet and tap water, fructose groups were fed the 65% high-fructose (HF) diet with/without RCM 100 mg/kg/day for 8 weeks, respectively. Chronic treatment with RCM significantly decreased body weight, fat weight and adipocyte size. Moreover, RCM significantly prevented the development of the metabolic disturbances such as hyperlipidemia and hypertension. RCM also led to increase in high density lipoprotein level in the HF group. In addition, RCM suppressed vascular cell adhesion molecule-1 (VCAM-1) expression and significantly recovered the levels of endothelial nitric oxide synthase (eNOS) expression in aorta. These results demonstrates that RCM may be a beneficial therapeutic for metabolic syndrome through the improvement of hyperlipidemia, obesity, and hypertension.

  • The present study was aimed to investigate whether ethanol-extract of Allium wageki has an ameliorative effect on the renal function in high fructose-diet induced hypertensive rats .. The urine osmolality (Uosmol) was decreased in rats with high fructose-diet (60%) during the whole experiment period without change of the urine volume (UV). The urinary excretion of sodium (UNaV) and chloride (UCIV) were decrease significantly in rats with fructose-induced hypertensive rats, whereas urinary excretion of potassium (UKV) was Increased. The creatinine clearance (Ccr) and solute-free water reabsorption were also decreased by treatment of fructose-rich diet. Among these renal functional parameters, Ccr was partially restored by the administration of ethanol-extract of Allium wageki. The Uosmol was also partially restored by the administration ethanol-extract of Allium wageki at the end of the experimental period. Taken together, ethanol-extract of Allium wageki has the ameliorative effect on glomerular filtration rate in rats with high fructose-diet induced hypertension.

연근의 에탄올 추출물이 과당으로 유도된 고혈압 흰쥐의 혈압과 신장 기능에 미치는 효과 (Effects of Ethanol-Extract of Lotus Root on the Renal Function and Blood Pressure of Fructose-Induced Hypertensive Rats)

  • 연근의 에탄올 추출물이 과당 유도 고혈압 백서의 신장 기능에 미치는 영향을 관찰하고자 뇨량과 전해질 배설량, 혈장의 creatinine, creatinine 청소율, 자유-수분 재흡수량 등을 측정하였다. 그 결과 연근은 과당 유도 고혈압 백서에서 나타나는 전해질 배설량의 변화를 개선시키는 효과는 없는 것으로 나타났고 단지 삼투질 농도의 배설량에만 약간의 개선 효과를 보였다. 신장의 가장 중요한 기능중 하나는 여과 기능이고 이를 나타내는 지표는 사구체 여과율인데, 고혈압 백서에서 혈장내 creatinine 농도가 증가하였고, creatinie 청소율이 크게 억제되었으나 연근추출물 투여 후 사구체 여과율의 감소가 회복되었다. 그러므로 연근은 과당으로 유도된 고혈압 흰쥐의 사구체 여과율을 개선시키는 것으로 사료된다.
