Improvement of Reproductive Efficiency of Artificial Insemination Following Estrus Induction in Dog II. Semen Freezing and Artificial Insemination in Dog

  • Considerable attention has been focused on the cryopreservation of semen and estrus induction in dog, as consequence of poor productivity caused by long anestrus period, in order to enhance the productivity of youngs and to preserve the breeds. The objectives of this study were to evaluate semen quality after cryopreservation and to evaluate the Pregnancy rate after insemination (AI). Fifty infertilie dogs (age 2∼3 years) were selected for the study and divided into three different estrus induction treatment groups. Group 1: dogs (n=15) were given clomifene (0.1 mg/kg) orally for five days at 12 hr intervals. Group 2: dogs (n=15) were given bromocriptine (50 $\mu\textrm{g}$/kg) orally for five days at 12 hr intervals, followed by single injection intravenously of 500 IU GnRH (Group 3, n=20) when pro-estrus occurred. The rates of pregnancy in estrus inducted dogs mated naturally compared to those inseminated artificially with ejaculated fresh semen and frozen-thawed semen. Estrus detection was performed using the method of vaginal smear and confirmed by the plasma progesterone assay. The ejaculated semen to freeze was exposed to a mixture of Tris extender with cryoprotectant (Trisma, 81 mM: TES, 209 mM: citric acid, 6 mM; glucose, 5 mM; glycerol, 8%) and cryopreserved gradually by slow-cooling at 17 cm above the surface of liquid nitrogen (LN$_2$) for 23 min. The motility of frozen-thawed spermatozoa was assessed by phase-contrast microscopy. To assess their viability and acrosome content, spermatozoa were stained with a vital stain and Fluorescence conjugated lectin Pisum Savitum Agglutinin (FITC/PAS), respectively. Pregnancy was confirmed by ultrasonograpy on day 25, 35 and 55 post insemination. The use of fresh semen, the pregnancy rates were observed 66.6, 66.6, 75.0 and 83.3% in natural estrus, clomifene induced, bromocriptine induced and a combination of GnRH and bromocriptine, respectively. The use of frozen-thawed semen, the pregnancy rates were observed 66.6, 33.3, 50.0 and 60.0% in natural estrus, clomifene induced, bromocriptine induced and a combination of GnRH and bromocriptine, respectively. No difference was observed in the number of offspring produced among natural estrus and treated groups inseminated with fresh or frozen-thawed semen. In conclusion, the pregnancy rate of dogs treated with a combination of GnRH and bromocriptine was more effective than use of clomifene or bromocriptine only. In addition, frozen-thawed semen can be used successfully far artificial insemination in dog.

Factors Affecting on Pregnancy Rate of Recipients Following Transfer of Hanwoo Embryos Produced In Vivo (한우 체내 수정란 이식 후 수태율에 미치는 요인)

  • This study was performed to improve the pregnancy rates of recipients following transfer of bovine embryos produced in vivo. Superovulation response didn't showe significant differences between each season (4.18 in spring; 4.36 in summer; 5.50 in fall; 4.38 in winter). Pregnancy rate was significantly different (p<0.05) between fresh (43.4%) and frozen embryos (17.2%). In administration of hCG to recipients, the pregnancy rate of fresh embryos (45.7%) was slightly higher than that of control (35.3%), but the pregnancy rates of frozen embryos in control group (25.0%) was higher than that of hCG group (16.0%). When synchrony of recipient and embryo was -2, -1, 0 and 1, the pregnancy rates were 20.0, 45.0, 30.3 and 26.3%, respectively. The pregnancy rates of recipients synchronized by naturally or $PGF_{2{\alpha}}$, CIDR/$PGF_{2{\alpha}}$ and E/P/CIDR/$PGF_{2{\alpha}}$/E treatments were 35.3, 48.0, 29.0 and 40.0%, respectively. Gestation lengths and birth weights of female and male calf were 288 and 290.5 days, 28.3 and 30.0 kg, respectively. The results were showed that the superovulation response was not affected by seasons, and also pregnancy rate didn't increase by administration of hCG, synchrony of embryo and recipients, synchrony methods. Further study and concern should be focused on improving the embryo freezing and pregnancy rate for commercial embryo transfer.

  • This lecture will begin by tracing some of the history behind techniques that we nowadays take for granted in the practice of embryo transfer, and in the application of the technique to various animal biotechnologies. It will be argued that an appreciation of such history can teach us a great deal about how we need to study and teach the subject, and about the best ways to conduct and finance the research that is essential to further progress. Examples in support of this argument will be taken from the changes that have occurred in the way embryos, particularly bovine embryos, have been collected, maintained in vitro, subjected to a variety of manipulations (sexing, division to produce identical animals, combination into chimeras, transfection with foreign genes), frozen and thawed, and transferred over the past 50 years. Effect of Bovine Somatotropin (bST) Treatment on Embryo Recovery and Pregnancy Rate in Hanwoo (한우에서 bST 처리가 수정란 회수 및 수태율에 미치는 영향)

  • The purpose of this study was to determine the effect of bST treatment on embryo recovery and pregnancy rate following embryo transfer. Donor cows were superovulated with Folltropin-V and PGF$_2$$\alpha$ combination method and then inseminated with frozen semen 3 times 12 hrs interval. Donor and recipient cows were assigned to control and bST group, of which was given a single injection of bST (500 mg, im) at insemination or estrus detection. Embryo collection of superovulated cows were flushed nonsurgical method at 7 to 8 days after artificial insemination. The percentare and Mean$\pm$S.E. of transferable embryo was not significantly different between control and bST treatment (72.8%/5.9$\pm$4.5 vs. 83.7%15.1 $\pm$ 1.6). The percentage and Mean$\pm$S.E. of transferable embryo in non-summer season was significantly higher than in summer (81.8%/5.4$\pm$2.1 vs. 68.7%14.774.6; P<0.05). The pregnancy rate after embryo transfer in bST treatment was significantly higher than in control (64.0 vs. 47.1%; P<0.05). There was no significant difference in pregnancy rate between summer and non-summer (51.6 vs. 61.5%; P>0.05). The results indicated that InST treatment in recipient cows could improve the efficiency of transferable embryo production and pregnancy rate after embryo transfer, and non-summer season may be better far superovulation treatment and embryo transfer.

  • This experiment was carried out to investigate the extender, cooling rate and concentration of glycerol for freezing of boar semen. The result obtained were summarized as follows: 1. Optimal cooling rate was $0.17\~022^{\circ}C/min$ from 25 to $5^{\circ}C$ in LEY extender on the viability and normal acrosome after thawed. 2. The LEY extender was effective in protecting frozen boar semen from cold shock among the extenders(p<0.001, respectively). 3. The sperm viability and normal acrosome rates after thawing was showed greater in the 3 or $4\%$ of glycerol concentration than $2\%$ in LEY extender. 4. Viability of sperm was higher when both 15mM of fructose and 3 or $4\%$ glycerol were added to the LEY extender compared with other concentrations of fructose and glycerol were added it(p<0.001).

Studies on the Survival Rate of Rapidly Frozen Bovine Immature Oocytes (소 미성숙 난포란의 급속동결 융해후 생존성에 관한 연구)

  • This study was carried out to investigate On in vitro fertilization, survival rate and developmental rate of rapidly frozen bovine immature oocytes. Immature oocytes cultured for 1, 12, 24, 48 hours in 20% FCS + TCM-199 medium and thereafter rapidly freezing-thawed oocytes inseminated with capacitated sperm. The immature oocytes following dehydration by 1.5M DMSO + 2.0M glycerol + 0.25M sucrose + TCM 199 media + 20% FGS were directly plunged into liquid nitrogen and thawes in 3$0^{\circ}C$ water. Rapid freezing embryos co-cultured in 20% FCS + TCM-199 media containing hormones(21U/mL PMSG, 21U /mL hGG and 1 $\mu$g /mL 17$\beta$-estradiol) and cumulus cells(1 x 105-6 cells). Survival rate was defined as development rate on in vitro culture or FDA-test. The results are summarized as follows ; 1. The in vitro maturation and fertilization rate of immature bovine oocytes on in vitro maturation period(1, 12, 24, 48 hrs) before rapid freezing4hawed were 57.1%, 45.7%, 37.1%, 25.7% and 40.0%, 31.4%, 20.0%, 11.4%, respectively. 2. The survival rate of immature bovine oocytes on in vitro maturation period(1, 12, 24, 48 hrs) before rapid freezing-thawed were 33.3%, 26.7%, 20.0%, and 10.0%, respectively. The survival rate of rapid freezing4hawed immature oocytes was significantly lower than that of non-freezing oocytes. 3. The survival rate of rapid freezing4hawed excellent and good bovine embryos co-cultured in 20% FCS + TCM-199 media containing hormones(PMSG, hCG, 17$\beta$-estradiol) and cumulus cells 4 to 5 hrs and 20 to 24 hrs were 35.0%, 15.0% and 25.0%, 15.0% and 40.0%, 20.0% and 30.0%, 15.0%, respectively. The survival rate of embryos co-cultured in TCM-199 media containing hormones and cumulus cells was significantly higher than that of non co-culture.

The Motility of Bovine Sperm and Fertilization Rate of Oocytes during in-vitro Fertilization following Oviduct Epithelial Cell Co-culture (소 정자의 운동성 향상에 따른 수정율 증진 연구)

  • This study examined the motility of either the unattached(upper) or attached(lower) Hanwoo sperm to bovine oviduct epithelial cell(BOEC) monolayers to determine whether there are any changes in their motility during co-culture. The cleavage and blastocyst development rate were compared among different preincubation methods in-vitro, after oocytes were fertilized in-vitro with Hanwoo sperm on BOEC monolayers. The motility of frozen-thawed sperm in BOEC co-culture group was significantly higher than controls, especially at 5 hours and 6 hours (p<0.05) of incubation, in sperm treatment medium without heparin and caffeine. The motility of frozen-thawed sperm in BOEC co-culture group was significantly higher than controls, especially at 3 hours (p<0.05) and 6 hours (p<0.01), in sperm treatment medium containing heparin and caffeine. The motility of the attached( lower) sperm was significantly higher than the unattached(upper) sperm during co-culture with BOEC at all times(p<0.01 or p<0.05), except for 6 hours. After Hanwoo oocytes were fertilized in-vitro with the sperm that had been co-cultured with BOEC in sperm treatment medium containing heparin and caffeine, we determined the cleavage and blastocyst development rate, according to the preincubation methods. Both the cleavage and blastocyst development rate from 2 hour preincubation group were the highest, but significant difference was not recognized. These results show that BOEC plays an important role on sperm hyperactivation related to capacitation regardless of heparin and caffeine in sperm treatment medium. However, oviduct epithelial cell had no significant effect on the development of embryos after in-vitro fertilization in the presence of added heparin and caffeine in sperm treatment medium.

Semen Quality of the Black Bengal Bucks Used at Commercial Artificial Insemination

  • Only an optimum number of viable spermatozoa in a frozen-thawed insemination dose can ensure conception at artificial insemination (AI). We report here the percentages of normal, abnormal and viable spermatozoa present in the frozen-thawed semen of 20 Black Bengal bucks used for commercial AI. Bucks in this experiment were of 19.3~46.1 months old and 25~42 kg body weight. Four semen straws (0.25 ml) from each buck were collected for evaluation of their kinetic parameters. Scrotal circumference was measured by using a scrotal tape, sperm motility was estimated on eye estimation and sperm concentration was determined by using a haemocytometer. Sperm morphology was studied in paraformaldehyde fixed spermatozoa under differential interference contrast (DIC) microscope. To determine the proportion of live (plasma membrane intact) spermatozoa, semen was stained with SYBR-14 and propidium iodide and examined under fluorescent microscope. Scrotal circumference, post-thaw sperm motility, sperm concentration per insemination dose and proportion of normal spermatozoa were $21.5{\pm}0.7\;cm$, $43.5 {\pm}5.4%$, $83.5{\pm}6.7$ million and $88.3{\pm}4.1%$, respectively. The percentages of spermatozoa with head shape and acrosome abnormalities were lower ($2.7{\pm}1.1$ and $1.4{\pm}1.3$, respectively), whereas higher percentages of abnormalities ($7.0{\pm}1.8$) were observed in mid piece and tail portion. The proportion of live spermatozoa was $28.5{\pm}5.4$. It is concluded that although a good number of morphologically normal spermatozoa are present in the insemination dose, the proportion of live spermatozoa is low, which warrants further improvements of buck semen freezing procedures to ensure good quality at AI.

Effects of Osmolality Step during Vitrification on Survival of Vitrified IVP Embryos in Korean Native Cattle (Hanwoo)

  • Solution of glycerol, ethylene glycol, sucrose, dextrose (GESD) and cryotop methods were carried out to investigate the survivability on vitrification of embryos. Embryos cultured in vitro were vitrified by GESD of 10 or 8 step and cryotop methods of 6 step, from cryopreservation step to frozen-thawed and culture step. Survival rate and ICM, TE cells of embryos were investigated after frozen-thawed 24 h. As a results, cryotop method was significantly (p<0.05) higher ($85.76{\pm}5.3$ vs. $66.71{\pm}2.4$, $44.80{\pm}2.1%$) than GESD 10 or 8 step methods on survivability. Also, In ICM cell number, cryotop method was significantly (p<0.05) higher to $45.67{\pm}4.7$ cells than GESD 8 step method. TE cell number was significantly (p<0.05) highest to $111.00{\pm}11.0$ cells in cryotop method. On the other hand, survival rate, TE and total cell number were all the significantly (p<0.05) high, except ICM in GESD 10 step method between GESD 10 step method and GESD 8 step method. In conclusion cryotop method was to be most effective, but it is considered necessary to study vitrification method for step-by-step freezing and thawing process.

Modification of Efficient Vitrification Method by Using Open Pulled Straw (OPS) and EM Grid as Vehicles in Human Embryonic Stem Cell (인간 배아 줄기세포의 OPS와 Grid를 이용한 유리화 동결법의 효율성 비교)

  • Human embryonic stem (hES) cell lines have been derived from human blastocysts and are expected to have far-reaching applications in regenerative medicine. The objective of this study is to improve freezing method with less cryo-injuries and best survival rates in hES cells by comparing various vitrification conditions. For the vitrifications, ES cells are exposed to the 4 different cryoprotectants, ethylene glycol (EG), 1,2-propanediol (PROH), EG with dime-thylsulfoxide (DMSO) and EG with PROH. We compared to types of vehicles, such as open pulled straw (OPS) or electron microscopic cooper grids (EM grids). Thawed hES cells were dipped into sequentially holding media with 0.2 M sucrose for 1 min, 0.1 M sucrose for 5 min and holding media for 5 min twice and plated onto a fresh feeder layer. Survival rates of vitrified hES cells were assessed by counting of undifferentiated colonies. It shows high survival rates of hES cells frozen with EG and DMSO (60.8%), or EG and PROH(65.8%) on EM grids better than those of OPS, compared to those frozen with EG alone (2.4%) or PROH alone (0%) alone. The hES cells vitrified with EM grid showed relatively constant colony forming efficiency and survival rates, compared to those of unverified hES cells. The vitrified hES cells retained the normal morphology, alkaline phosphates activity, and the expression of SSEA-3 and 4. Through RT-PCR analysis showed Oct-4 gene expression was down-regulated and embryonic germ layer markers were up-regulated in the vitrified hES cells during spontaneous differentiation. These results show that vitrification method by using EM grid supplemented with EG and PROH in hES cells may be most efficient at present to minimize cyto-toxicity and cellular damage derived by ice crystal formation and furthermore may be employed for clinical application.
