Cladocera are important food organism for seed production of finfishes. freshwater cladocera such as Daphnia and Moina are well known food organisms for the larval rearing of freshwater fishes and are easy for mass culture. However, mass culture technique for marine cladocera are not yet developed, The only mass produced food organisms available these days for the larval production of marine finfishes are rotifer and Artemia. An estuarine cladoceran, Diaphanosoma celebensis, has a high possibility of being used as a food organism for the larval rearing of marine finfishes because this species is much easier to mass culture than marine ones. Therefore many studies are needed for this species. In this study, the effects of the volumes of culture container, 40, 1,500 and 15,000 ml, on the stable production of this species were tested and results are as follow: The maximum densities of this species in each of the culture volumes were reached after 14 days in 40 ml, 12 days in 1,500 ml, and 21 days in 15,000 ml with values of 3.4 $\pm$ 0.4, 14.2 $\pm$ 2.1 and 2.5 $\pm$ 1.6 per ml, respectively. The relative population growth index (RPGI) was stable in the culture volume of 1,500 ml. Moreover, possible harvesting number(individual/ml/day) was much higher in the 1,500 ml container than the other culture volumes. Therefore, optimum culture volume among the tested volumes for mass production of this species was 1,500 ml.
Alien species are known to threaten regional biodiversity globally, which has increased global interest regarding introduction of alien species. The Ministry of Environment of Korea designated species that have not yet been introduced into the country with potential threat as alert alien species to prevent damage to the ecosystem. In this study, potential habitats of Esox lucius and Maccullochella peelii, which are predatory and designated as alert alien fish, were predicted on a national basis. Habitat suitability was evaluated using EHSM (Ecological Habitat Suitability Model), and water temperature data were input to calculate Physiological Habitat Suitability (PHS). The prediction results have shown that PHS of the two fishes were mainly controlled by heat or cold stress, which resulted in biased habitat distribution. E. lucius was predicted to prefer the basins at high latitudes (Han and Geum River), while M. peelii preferred metropolitan areas. Through these differences, it was expected that the invasion pattern of each alien fish can be different due to thermal preference. Further studies are required to enhance the model's predictive power, and future predictions under climate change scenarios are required to aid establishing sustainable management plans.
The objective of this study was to evaluate biodiversity and natural habitat environment of freshwater ecosystem. Our aim was therefore to produce a set of biodiversity and habitat indicators based on multi-parameters of water quality and biodiversity by analyzing the characteristics of the results by indicators. We selected four indicators a) anthropogenic disturbance, b) habitat diversity, c) biodiversity, d) ecosystem structure. The fishes cohabiting with Brachymystax lenok tsinlingensis were Rhynchocypris kumgangensis and Zacco koreanus. As a result of the anthropogenic disturbance evaluation, it was analyzed that vegetation embankment showed a more stable environment in the tributary than the main stream, and other disturbance was not confirmed As a result of the habitat diversity evaluation, it was analyzed that habitat evaluation index showed a high score of 200 more on average, showing an optimal habitat condition. As a result of the biodiversity evaluation, it was analyzed as a clean habitat condition with a high proportion of sensitive species, abundant dissolved oxygen, and little pollutants. As a results of the ecosystem structure, the ecological health condition metrics and appearance of endangered species showed large score deviation, but it was analyzed that the stream ecosystem health was generally excellent. There was a slight correlation between the habitat environment and the results of the nature habitat evaluation according to the appearance of the B. lenok tsinlingensis.
In an effort to develop the biomarker for monitoring the contamination of xenoestrogen in the freshwater environment of Korea, reverse transcription-polymerasechain reaction (RT-PCR) analysis of vitellogenin (VTG) gene expression was optimized in Hearisarsus Iaseo, Based on the homology of the VTG cDNA sequences between the common carp and zebra fish, a set of PCR primers for VTG mRNA amplification for H; labo was designed. VTG mRNA level in livers from female and male fishes was analyzed by RT-PCR following single injection of 17 beta estradiol($E_2$ 10 mg $kg^{-1}$ B.W.). As an internal control, beta actin mRNA was amplified. One us of total liver RNA was subjected to RT-PCR. In female the amount of PCR productof VfC gradually increased in the range from 16 to 34 cycles of amplification. On the contrary, in control male, PCR product first detected at 32 cycles of amplification and linearly increased up to 40 cycles of amplification. In $E_2$ injected male liver, the VTC mRNA level was similar to that in the female. Taken together, this result suggests that liver of male H. labo expresses minute amount of VTG mRNA which are2-l6 equivalent of female and that induction of VTG mRNA occurs in male liver after estrogen treatment. In conclusion, the optimized protocol for RT-PCR analysis of VTG mRNA expression in liver of male H. labo will provide the environmental monitoring method for the xenoestrogen contamination in the rivers in Korea.

Parasitism of Pseudocaligus sp. in wild grass puffer Takifugu niphobles in the coast of Yeosu in spring, 2007 was identified. The preadult and adult stages of Pseudocaligus sp. were observed, which consisted of cephalothorax, genital segment and abdomen. Lunules were present on the anterior margin of the frontal plate. In order to develop the treatment method, we examined the effects of formalin and fresh water on the parasite. The Pseudocaligus sp. were detached from the host fish by bathing at concentrations of 50 to 400 ppm formalin within 60 min, but 57% was exterminated to fresh water for 90 min.
We described here the morphological characteristics for the species identification and fish hosts of Isoparorchis sp. (Digenea: Isoparorchiidae) in the Republic of Korea (Korea). Total 1,371 freshwater fishes collected in Yangcheon (Stream) in Sancheong-gun, Gyeongsangnam-do were examined by the artificial digestion methods to survey the infection status of digenetic trematode metacercariae for 4 years (2013-2016). Adult and larval worms of Isoparorchis sp. were detected in 38 (8.4%) out of 451 fish in 4 species, i.e., Pungtungia herzi, Acheilognathus koreensis, Squalidus japonicus coreanus and Odontobutis platycephala, examined. The infection density was 1.1 worm per fish infected. They were mainly found in the subcutaneous tissues and abdominal cavities. Nodules with worms in the subcutaneous tissues were revealed as the blue ink-colored bulges. Adults leaf-like, $21.6{\times}9.84mm$ in average size. The ratio of body length to body width was 2.20: 1. Oral sucker subterminal, $1.03{\times}1.22mm$. Pharynx muscular, $0.55{\times}0.54mm$. Esophagus very short. Ceca convoluted, terminated near the posterior end. Ventral sucker anterior 1/3.75, $1.99{\times}2.10mm$. The ratio of ventral sucker to oral sucker was 1.74: 1. Testes round to elliptical, both sides of ventral sucker, $1.43{\times}1.33mm$. Vitellaria highly dendritic, posterior 1/3 level. Eggs operculated, embryonated, $52{\times}32{\mu}m$ in size. By the present study, 4 fish species aforementioned are to be listed as the fish hosts of Isoparorchis sp. in Korea and additionally the morphological characteristics are to be described for the species identification.

Background: The flathead grey mullet Mugil cephalus has the widest distribution among mugilid species. Recent studies based on mitochondrial DNA sequences showed that the species comprises at least 14 different groups, three of which occur in the northwest Pacific. We analyzed the otolith microchemistry of M. cephalus at several locations in Korea to improve understanding of migration pattern and population origin. Results: We collected 123 sagittal otoliths from seven locations and determined their concentrations of eight elements (7Li, 24Mg, 55Mn, 57Fe, 60Ni, 63Cu, 88Sr, and 138Ba) using laser ablation inductively coupled plasma mass spectrometry. Mean otolith elemental ratios differed significantly among the locations. The Sr:Ca, Fe:Ca, and Ba:Ca ratios were significantly higher than others, and useful chemical signatures for investigating the habitat use of M. cephalus populations. We identified five diverse and complicated migration patterns using the otolith data that we collected: estuarine resident (type I), freshwater migrant (type II), estuarine migrant (type III), seawater resident (type IV), and seawater migrant (type V). A canonical discriminant analysis plot revealed separation of two groups (type II in the Yellow Sea vs. other types in remaining locations). Two locations on Jeju Island, despite their close proximity, had fish with quite different migration patterns, corroborating previous molecular studies that distinguished two groups of fishes. Conclusion: We successfully showed that the migration patterns of the Korean mullet varied by location. Only fish from the western sector of Jeju had a unique migration pattern, which is likely confined population in this area. Among the eight otolith elements measured, the Sr:Ca ratio was found to be the best indicator of migration pattern and population origin.
Five strains of gram-negative and yellow-pigmented bacteria were recently isolated from diseased freshwater fishes in Korea. All isolates were confirmed as a known fish pathogen of columnaris disease, Cytophaga columnaris based on their colonial and cellular morphology, and on physiological, biochemical and antigenic characteristics. Although the isolates were from different fish species, their characteristics of them were very similar to those of the reference strains of C columnaris (NCMB $2248^T$ and EK 28). Also, profiles of OMPs of Korean isolates were similar to those of the reference strain, C columnaris NCMB $2248^T$ when analyzed by SDS-PAGE.

In this study, the molecular structures of tet(M) and tet(G) carried by tetracycline (Tc) resistant bacteria in intestinal microflora from the imported ornamental fish were characterized and compared with each other depend on the imported countries. Of the total isolates, approximately 8.9% of the Ent-lac+(lactose fermentative bacteria on coliform media) Tc resistant isolates in fish from three different countries, Singapore, Taiwan and Brazil, were appeared to contain tet(M). Three representative isolates of different countries, Aeromonas spp. JSM-1 (Singapore), JTM-1 (Taiwan) and JBM-1 (Brazil), were isolated and analyzed the molecular structures of tet(M) gene. Interestingly, partial sequence of tet(M) genes (1099 bp) in JBM-1 (Brazil) showed 99.5% homology with the tet(M) found in the Vibrio spp. RV16 isolate, obtained from marine fish in Korea and known to carry Tn1545 parent type of tet(M). In contrast, tet(M) gene in JSM-1 and JTM-1 showed mosaic structure of Tn1545 and Tn916, and 100% homology with each other. It may suggest the presence of various characteristics in terms of tet(M) gene structure. The determined sequence of the tet(G) from Aeromonas spp. JSG-1 and JBG-1 isolated from Singapore and Indonesia ornamental fish respectively showed similar nucleotide sequence homology but revealed a few nucleotide changes in comparison with the sequence of the prototype tet(G) gene (S52437 in GenBank).
