마이크로웨이브 추출공정에 의한 양파의 올레오레진 함량 및 기능적 특성 (Oleoresin Content and Functional Properties of Fresh Onion by Microwave-Assisted Extraction)

  • 양파를 conventional extraction(CE) 방법과, micro-wave-assisted extraction(MAE) 방법에 의하여 각각 물과 에탄올로 추출하고 추출조건에 따른 올레오레진 수율과 생리활성을 비교하였다. 추출 방법에 따른 올레오레진 함량의 차이는 거의 없었고 최적 추출시간은 CE의 경우 2시간인 반면 MAE에서는 5분 내외로 추출시간이 크게 단축되었다. 전자공여작용은 물 추출의 경우 CE 및 MAE 에서 각각 44% 및 50%이었고 에탄올 추출의 경우 각각 45% 및 51%로 추출방법에 의한 차이는 거의 없었다. 아질산염 소거작용은 모든 추출물에서 pH가 증가함에 따라 감소하는 경향을 보였는데, 특히 pH 1.2에서는 모든 추출물에서 85% 이상의 높은 소거작용을 보였다. Tyrosinase 저해작용은 물 추출물의 경우 CE와MAE에서 모두 20% 수준이었고 에탄올 추출물도 40% 수준의 비교적 낮은 저해작용을 나타내었다. ACE 저해작용에서는 CE보다 MAE 추출물에서 더욱 강한 활성을 나타내어 CE 방법으로 추출했을 때는 60% 수준이었고 MAE 방법으로 추출했을 때는 70% 수준이 었다. 또한, 추출방법에 따른 피르브산 함량 변화에서도 물 추출물의 경우 CE와 MAE 각각 양파 1g당 6.8 및 $6.4\;{\mu}moles$이었고 에탄올 추출물에서는 이보다 낮은 4.3과 $5.6\;{\mu}moles$이었으며 추출방법에 따른 유의적인 차이는 보이지 않았다.

Effects of the slaughter weight of non-lean finishing pigs on their carcass characteristics and meat quality

  • The present study aimed to assess the feasibility of increasing the slaughter weight (SW) of non-lean finishing pigs to improve their meat quality. A total of 36 (Landrace × Yorkshire) × Duroc gilts and barrows were slaughtered at 115 (Av), 125 (Hi), or 135 (XHi) kg, followed by physicochemical analyses and sensory evaluation on their longissimus dorsi (LD) and Semimembranosus (SM) muscles. Backfat thickness was greater (p < 0.05) for the XHi (31.2 mm) and Hi (29.3 mm) groups than for Av (25.0 mm). Dressing percentage and yield of the belly per whole carcass were also slightly greater for XHi and Hi vs. Av. The intramuscular fat (IMF) content of SM was greater for XHi (2.64%) than for Av (1.83%) and Hi (2.04%) and also was correlated with SW (r = 0.55). The pH value, lightness, redness, drip loss, shear force, and moisture and protein contents of LD and SM, as well as IMF content of LD, were unaffected by SW. Percentages of 14:0, 16:0, and total saturated fatty acids (FA) were less for Hi and XHi vs. Av in SM, those of total unsaturated FA, 18:2, 20:4, and n-6 being opposite; FA composition of LM was not influenced by SW except for a reduced 18:0 percentage for XHi vs. Av. The sensory score was less for XHi vs. Av for odor in fresh LD and SM, and less for Hi and XHi vs. Av for aroma in fresh LM; scores for color, drip loss, marbling, and acceptability were unaffected by SW. As for cooked muscles, none of the scores for color, aroma, flavor, juiciness, tenderness, and acceptability was affected by SW, except for a greater LD color score for Hi and XHi vs. Av. Collectively, the results suggested that the increased yield of the carcass and belly due to increased SW is outbalanced negatively by excessive backfat deposition in production efficiency, whereas the SW increase exerts little influence on overall sensory quality of fresh or cooked meat. Production of non-lean market pigs overweighing 115 kg therefore will be uneconomical unless consumers pay a substantial premium for the over-fattened pork.

Quality and Acceptability of Meat Nuggets with Fresh Aloe vera Gel

  • Aloe vera has been used worldwide for pharmaceutical, food, and cosmetic industries due to its wide biological activities. However, quality improvement of low fat meat products and their acceptability with added Aloe vera gel (AVG) is scanty. The aim of this study was to explore the feasibility of using fresh AVG on physicochemical, textural, sensory and nutritive qualities of goat meat nuggets. The products were prepared with 0%, 2.5%, and 5% fresh AVG replacing goat meat and were analyzed for proximate composition, physicochemical and textural properties, fatty acid profile and sensory parameters. Changes in lipid oxidation and microbial growth of nuggets were also evaluated over 9 days of refrigerated storage. The results showed that AVG significantly (p<0.05) decreased the pH value and protein content of meat emulsion and nuggets. Product yield was affected at 5% level of gel. Addition of AVG in the formulation significantly affected the values of texture profile analysis. The AVG reduced the lipid oxidation and microbial growth in nuggets during storage. Sensory panelists preferred nuggets with 2.5% AVG over nuggets with 5% AVG. Therefore, AVG up to 2.5% level could be used for quality improvement in goat meat nuggets without affecting its sensorial, textural and nutritive values.

Effect of Fluorescent Light Treatment during Imbibition and Culture on Growth of Soybean Sprout

  • The lateral root formation in soybean sprout culture declines its quality. This study was done to measure the effect of fluorescent light treatment during 24 hour imbibition and 6-day culture on seed germination and growth of soybean sprout. After 6 day culture, the sprouts were sorted as normal (>4cm), abnormal (<4cm) and non-germination by their hypocotyl lengths, and lateral roots, fresh and dry weights were measured. Lateral roots were less formed in the fluorescent light treatment lasted during the whole period of the imbibition than in the treatment for 50 minutes a day during the culture. The fluorescent light treatment during the imbibition mainly affected the germination and growth compared to the treatment done during the culture. Compared to the dark imbibition, the light treatment during the imbibition resulted in more normal sprouts, thicker diameters of hypocotyl and hook, and more fresh weights in cotyledon, hypocotyl, whole sprout, and economic yield. However, these results were reverse in lengths of hypocotyl and root, and fresh and dry weights of roots. It is concluded that the fluorescent lamp mainly irradiating red and blue lights can be used for the sprout production as an alternative light replacing blue and red lights treated during the imbibition because it blocked the lateral root appearance and stimulated growth of the sprout.

수입 생약(生藥)의 국산화(國産化)에 관한 연구-황연(黃蓮)의 순화재배(馴化栽培)에 대하여- (Studies on Domestic Production of Imported Botanical Drugs-On Domesticational Cultivation of Coptis japonica-)

  • A study was carried out to investigate the possibility of acclimation of Coptis japonica Makino in Korea. Coptis japonica could be grown within 4 years as a medicinal crude drug if it is cultivated in the area of mountain facing north(300m above sea level) with 70% light interception and appropriate soil texture. It is to be noted that the propagation of the plant is more important than the yield of berberine because the berberine contents do not change in different conditions. The results were as follows; 1. The germination rates of the seeds by covering with the pine tree leaves were the highest and averaged 63%. 2. The germination rates of dry seeds were not affected in spite of low temperature and $GA_3$ treatment. 3. The germination rates of the seeds stored in moistured sand were averaged 85%, but in dry state, they were not germinated. 4. The number of leaves and fresh weight of Coptis japonica from the area of mountain facing north were increased significantly. 5. The growth and fresh weight of Coptis japonica were reduced in clay. 6. The best growth was obtained under 70% light interception. It is possible to intercrop between evergreen trees. 7. The foliage dressing did not affect the growth. 8. The number of leaves and fresh weight were increased from the 3rd year, and the average weight of rhizomes was 5.7g in 4th year. 9. The berberine contents of the rhizomes of Coptis japonica under various treatment conditions ranged from 9 to 10%. Coptidis Rhizoma from this study and from Korean market showed much higher berberine contents than those from Japanese market.

Relation of Calcium Activity in Milk and Milk Production of Holstein Cows in Hot Season

  • The content of Ca in milk exceeds the typical saturation level of Ca salts, which is necessary for neonate growth. This calcium is distributed between the casein micelles in the colloidal and aqueous phases. Information on the properties of calcium activity in the aqueous phase is limited compared with that on the properties of bound or sequestrated calcium. The objectives of this study were to evaluate the changes in calcium activity in fresh milk using an ion-selective electrode and to assess the relationship between calcium activity and milk production in hot season. Milk samples collected from 10 cows at the National Agricultural Research Center for Kyushu Okinawa Region in June to October (Min-Max: 7.2-$35.2^{\circ}C$, 24.3-100% RH) were analyzed on total calcium concentrations and calcium activity. We observed that the rectal temperature of the cows increased according to elevation of ambient temperature but that the pH of the collected milk ($6.61{\pm}0.01$ (Mean${\pm}$SEM)) was not significantly influenced by rectal and ambient temperature. Total calcium concentrations and calcium activity in fresh milk decreased in July (Min-Max: 21.1-$33.5^{\circ}C$, 48.9-100.0% RH) compared with the values after August (Min-Max: 18.1-$35.0^{\circ}C$, 26.5-96.2% RH) (p<0.05); however, there was no significant correlation between the two parameters. The ratio of calcium activity to total calcium concentration decreased after August compared with the values in June and July (p<0.05). The calcium activity in fresh milk was positively correlated with milk yield (r = 0.45, p<0.01) and negatively correlated with milk lactose content (r = -0.53, p<0.01). These results suggest that the calcium activity in milk could be affected by ambient temperature and might be associated with milking production in hot season.

Genetic variability, associations, and path analysis of chemical and morphological traits in Indian ginseng [Withania somnifera (L.) Dunal] for selection of higher yielding genotypes

  • Background: The study was carried out to assess the genetic variability present in ashwagandha and to examine the nature of associations of various traits to the root yield of the plant. Methods: Fifty-three diverse genetic stocks of ashwagandha (Withania somnifera) were evaluated for 14 quantitative characteristics. Analysis of variance, correlation, and path coefficient analysis were performed using the mean data of 2 years. Results: Analysis of variance revealed that the genotypes differed significantly for all characteristics studied. High heritability in conjunction with high genetic advance was observed for fresh root weight, 12 deoxywithastramonolide in roots, and plant height, which indicated that selection could be effective for these traits. Dry root weight has a tight linkage with plant height and fresh root weight. Further, in path coefficient analysis, fresh root weight, total alkaloid (%) in leaves, and 12 deoxywithastramonolide (%) in roots had the highest positive direct effect on dry root weight. Conclusion: Therefore, these characteristics can be exploited to improve dry root weight in ashwagandha genotypes and there is also scope for the selection of promising and specific chemotypes (based on the alkaloid content) from the present germplasm.

Uniconazol 및 Paclobutrazol의 葉面散布 처리가 더덕의 생육 및 수량에 미치는 영향 (Effect of Foliar Application of Uniconazol and Paclobutrazol on Growth and Yield of Codonopsis lanceolata)

  • 더덕의 재배기술 개선을 목적으로 Uniconazol 및 Paclobutrazol을 더덕의 생장기별로 엽면산포(葉面散布)를 행했다. Uniconazol 및 Paclobutrazol 처리는 처리시기에 관계 없이 농도가 높을수록 초장은 억제되었으며 엽면적(葉面積)의 감소를 나타냈다. Uniconazol 처리에 의한 지하부의 형질은 3절(節) 분화시의 처리를 제외한 모든 처리구에서 대조구와의 유의차가 인정되지 않았다. 그러나12절 분화시의 10mg $liter^1$ 와 100mg $liter^1$ 농도처리에서는 대조구보다 생체중(生體重)이 각각 64%, 100%의 증가를 보였다. 세포독성(細胞毒性) Screening의 결과는 대조구 및 6절 분화시 10mg $liter^1$ 처리구에서만 활성을 나타했다. Paclobutrazol 처리에 의한 지하부의 생체중(生體重)은 3절(節) 분화시의 처리를 제외한 모든 처리구에서 대조구와의 유의차가 인정되지 않았다. 그러나 12절 분화시의 10mg $liter^1$ 농도처리에 의해 조금의 증가를 보였다. 세포독성(細胞毒性) Screening의 결과는 대조구 및 모든 처리구에서 약한 활성치를 나타냈다.

방울 토마토 재배 시 퇴비단 여과 액비의 이용가능성 (Availability of Slurry Composting and Biofiltration for Cultivation of Cherry Tomato)

  • 본 실험은 SCB(slurry composting and biofiltration) 액비의 방울 토마토(Solanum lycopersicum L. 'Unicon') 수경재배 이용가능성을 알아보기 위해 실시 되었다. 배양액처리 대조구로는 방울 토마토 전용 배양액(Control 1N)을 사용하였고, 이 배양액의 질소량($218.32mg{\cdot}L^{-1}$)을 기준으로 시판양액 1N(CNS 1N), SCB 1/2N, SCB 1N, SCB 2N을 타이머 제어에 의해 하루에 440~520mL을 공급하였다. 31일 동안 재배한 후 지상부 생체중과 건물중, 엽면적, 초장, 줄기직경, SPAD값, 마디수를 조사하였으며, 그 후 주 1회 총 7번의 과실을 수확하여 총 생산량과 상품성 과실 비율, 과실중, 당도와 산도, 과실의 총 페놀 농도 및 항산화도, 배꼽썩음병 발생률을 조사하였다. 그 결과 SCB처리 중에는 SCB 1/2N은 지상부 생체중 건물중, 엽면적, 줄기직경, 마디수 및 SPAD값에서 Control 1N 및 CNS 1N과 유의적 차이를 보이지 않으면서 우수한 생육을 유도하였다. SCB는 질소 농도가 증가하면서 점차로 생육의 저해현상을 보였다. 과실의 총 생산량은 Control 1N에서 가장 높았으며, SCB 처리중 생장이 가장 좋았던 SCB 1/2N 처리구는 Control 1N 총 생산량의 47%를 기록하였다. 상품성 과실 비율도 SCB 1N과 SCB 1/2N 처리구는 약 57~58%로 낮게 나타났다. 당도와 산도, 총 페놀 농도와 항산화도에서는 SCB 2N에서 가장 높은 수치를 보였고, 나머지 처리구와 대조구에서는 유의적으로 차이를 보이지 않았다. 배꼽썩음병은 Control 1N, CNS 1N에서는 발생하지 않았지만, SCB 2N, 1N, 1/2N 처리구는 각각 약 7, 13, 19%의 발생률을 나타내었다. 결론적으로 SCB 1/2N 처리는 Control 1N와 CNS 1N과 비교해서 방울 토마토의 영양생장에는 양호한 결과를 보였지만, 과실의 수량과 품질을 유지하지는 못했다. 따라서 SCB 액비는 방울토마토 시설 수경 재배에서 배양액으로의 가능성은 확인하였지만, 과실의 생산량과 상품성을 고려한다면 개화기 및 과실 비대기에 추가적인 무기 양분의 공급이 필요할 것으로 판단되었다.

육묘과정 중 배양액 추비시용과 정식방법에 따른 '설향' 딸기 생육 및 수량에 미치는 영향 (Effect of Fertilizer Application and Planting Method on Growth and Yield of 'Seolhyang' Strawberry in Seedling Stage)

  • This study was performed to identify the effect of fertilizer application and planting method on growth and yield of 'Seolhyang' strawberry during seedling raising. According to the concentration of fertilizer applied, the height of daughter plants was the highest at an EC of $0.8dS{\cdot}m^{-1}$. Leaf number and crown diameter were greatest at an EC of $0.6-0.8dS{\cdot}m^{-1}$. In the first measurement, root number was highest in non-fertilizer application, while root weight was heaviest in non-fertilizer application and EC $0.4dS{\cdot}m^{-1}$. The higher the concentration of fertilizer applied as culture media, the lower the growth rate. Thirty days after planting on the main field, plant height and number of new leaf were highest at an EC of $0.8dS{\cdot}m^{-1}$. However, no significant difference was found in leaf length and width and chlorophyll content according to fertilizer application. Marketable yield of 25 g or higher was greatest in EC $0.6dS{\cdot}m^{-1}$. In contrast, no significance was found in total marketable yield at an EC of $0.4-0.8dS{\cdot}m^{-1}$. Aconsistent pattern was exhibited in the growth of 1-5 harvesting flower clusters according to planting method. The length of leaf and flower cluster was short and chlorophyll content was low, when bed soil was removed 100% in harvesting of the first flower cluster. In all treatment, leaf length was shortened until harvesting of the second and third flower cluster, but rapidly lengthened in harvesting of the third and fourth flower clusters. Moreover, the length of flower cluster had a increasing tendency from harvesting of the third flower cluster. However, chlorophyll content was reduced continuously until harvesting of the fifth flower cluster, and was lowest in harvesting of the fourth flower cluster without removal of bed soil. Total yield was greatest in treatment of crown removal in bed soil between November and May. Late marketable yield between March and May was highest in treatment of 100% bed soil removal, followed by treatment of crown removal. Marketable yield of 25 g or higher was greatest in treatment of crown removal between December and February, while greatest in treatment of 100% bed soil removal between March and May.
