An Open Top Chamber for Forage Maize to Study the Effect of Elevated Temperature by Global Warming

  • The increase in temperature due to climate warming is predicted to affect crop yields in the future. Until now, various types of OTC (open top chamber) that simulate the future climate condition have been developed and used to study the effect of temperature increase due to global warming on maize growth. However, in most OTCs, high equipment and maintenance costs were required to artificially increase the temperature. This study was carried to develop a cost-effective and simple OTC suitable for climate warming experiments for forage maize. Three octagonal OTCs with a height of 3.5 m × a diameter of 4.08 m and a partially covered top were constructed. The lower part of OTC covered film was opened at a height of 26 cm (OTC-26), 12 cm (OTC-12) from the ground surface, or not opened (0 cm, OTC-0). Mean air temperatures during the daytime on a sunny day in OTC-0, OTC-12 and OTC-26 increased to 3.23℃, 1.33℃, and 0.89℃, respectively, compared to the ambient control plot. For a pilot test, forage maize, 'Gwangpyeongok' was grown at OTCs and ambient control plots. As a result, in the late maize vegetative growth phase (July 30), the plant height was increased more than 45% higher than the ambient control plot in all OTC plots, and the stem diameter also increased in all OTC plots. These results indicate that it is possible to set the temperature inside the OTC by adjusting the opening height of the lower end of the OTC, and it can be applied to study the response of forage maize to elevated temperature. An OTC, with its advantages of energy free, low maintenance cost, and simple temperature setting, will be helpful in studying maize growth responsiveness to climate warming in the future.

기아가 간세포막결합장치에 미치는 영향에 관한 Freeze Fracture Replica법에 의한 관찰 (Some Observations on the Intercellular Junctions between the Hepatocytes in Fasting States as Revealed by Freeze Fracture Replica)

  • 세포막의 결합장치들은 형태학적으로나 기능적으로 분화된 특이한 구조이며 각기 인접한 세포막 사이의 특징적인 부위에 위치한다. 그러나 이들 결합장치가 간세포의 기능상태에 연관하여 미세구조적으로 어떠한 변화를 보이는지에 관해서는 아직 잘 알려져 있지 않다. 본 연구는 간세포의 기능적 변동에 따르는 간세포막 사이 결합장치의 형태학적 변화를 알아보기 위하여 정상군과 8일간 기아 시킨 기아군으로 구분한 휜쥐 ($Wister,\;{\uparrow},\;250{\sim}280g$)에서 절취된 간조직을 박절편과 급속동결할 단복제법에 의거 표본을 제작하여 이들 결합장치의 미세구조상을 비교 고찰하였다. 그 결과 담세관주위의 폐쇄띠는 기아상태에서 현저히 감축되거나 소실되어 있었는데 이러한 소견은 박절편과 급속동결할단복제에서 다 같이 관찰되었으며 기아상태하에서 결합반과 교통반의 현저한 증가를 볼 수 있었다. 또한 정상상태하에서 이들 결합장치가 비교적 일정한 구조를 유지하고 있을 것이라는 일반적인 견해와는 달리 본 연구에서는 박절편이나 급속동결할단복제에서 다 같이 폐쇄띠가 부위에 따라 현저한 구조적 차이를 보였다. 본 연구에서 얻어진 이러한 결과로 미루어 폐쇄띠의 감축이나 소실은 기아로 인한 간세포의 기능저하가 담즙물질의 생성을 저해하게 됨으로서 담세관의 내압이 저하된 데 기인된 현상으로 생각된다. 또한 결합반의 증가는 기아로 인하여 감축된 간세포를 지지하므로서 간조직의 유지를 도우려는데 있었을 것이며 교통반의 증가는 기아로 인한 간세포의 기능적 저하가 간세포상호간의 물질교환을 필요로 하였기 때문이었을 것으로 추정된다. 특히 정상상태하에서도 부위에 따라 폐쇄띠의 미세구조적 변화가 있었던 것은 이 결합장치가 간세포의 기능이나 담세관의 상태에 민감하게 반응하기 때문일 것으로 생각된다.

인공습지시스템에서 수리학적 조건과 수질정화특성 (Purification Characteristics and Hydraulic Conditions in an Artificial Wetland System)

  • 본 연구는 농업용저수지의 수질개선을 위한 인공습지 시스템에서 수리학적 부하조건과 수질정화 특성간의 상관관계를 평가하고, 습지의 조성과 관리에 관한 기본적이며 주요 인자들을 규명하고자하였다. 인공습지시스템은 저수지 중층수를 유입시키기 위한 양수장과 미나리, 창포, 줄, 부들, 갈대등의 정수식물을 식재한 6개의 개별습지로 구성되어 있다. 시스템은 자유수면흐름방식으로 유입처리유량 $0.012-0.122\;m^3/s$,수리학적 체류시간 0.5-2.0 hr의 수리학적 고부하조건으로 운영하였으며, 수심은 0.1-0.2m, 유입수질은 저수지를 대상으로 하여 비교적 낮은 영양염류 농도(TN 2.224-2.462 mg/L, TP 0.145-0.164 mg/L)를 가지고 있다. 본 연구기간 중 각 개별습지의 평균 수질정화효율은 TN 12.1-14.3%로 갈대조에서 높게 나타났으며, TP는 6.3-9.5%로 식물 종에 따른 른 차이가없었다. SS는 17.4-38.5%, Cht-a는 12.6-20.2%로 미나리조에서 높게 나타났는데, 이는 유입수 농도가 다소 높은 때문으로 판단된다. 시간당 정화량은 TN $1.299-2.343\;g{\cdot}m^{-2}{\cdot}d^{-1}$ TP $0.85-1821\;g{\cdot}m^{-2}{\cdot}d^{-1}$, SS는 $17.9-111.6\;g{\cdot}m^{-2}{\cdot}d^{-1}$. Chl-3는 $0.011-0.094\;g{\cdot}m^{-2}{\cdot}d^{-1}$로 정화효율에서와 달리 TN은 줄에서 가장 높았고, TP는 창포에서 높았다. 침강성 물질인 SS와 Chl-a는 미나리에서 높게 나타났으며, 미나리는 BOD, COD, TN, TP등 다른 수질항목에서도 높은 값을 보여주고 있어 정화효율에서와 같이 유입수농도가 습지내 물질제거에 영향이 있음을 보여준다. 정화효율 및 시간당 정화량과 수리학적 조건간의 상관관계는 수심, 체류시간, 일유입량, 수리학적 부하량, 유입수 농도, 온도 등 다양한 변수에 의한 영향으로 비교적 낮게 나타났다. 정화효율과 수리학적조건간의 상관계수($R^2$)는 수리학적 체류시간과 0.016-0.731,일처리유량과는 0.015-0.868을 나타내었으며, 시간당 정화량과 수리학적 조건간의 상관계수($R^2$)는 수리학적 체류시간과는 0.173-0.763,일처리유량과는 0.209-0.770의 범위를 나타내었다. 정화효율과 수리학적 부하조건간의 상관계수($R^2$)Tt 0.5 이상을 나타내는 각 수생식물 습지별 수질항목은 체류시간과 일처리유량에 대해각각 20%,정화속도와 수리학적 조건간의 상관계수는 체류시간에 대해 53%, 일처리유량에 대해73%가 0.5이상을 보이고 있어 시간당 정화량과 수리학적 조건간의 상관관계가 정화효율과의 상관관계보다 좀더 유의성 있게 나타났다. 이것은 높은 수리학적 부하조건이 영양염류 등의 정화효율에는 크게 영향을 미치지 않음을 보여주고 있으며, 따라서 비교적 낮은 농도의 영양염류를 가지고 있고, 많은 처리수량을 요구하는 부영양화된 저수지의 수질개선을 위해서는 높은 수리학적 부하조건에서 시간당 정화량을 늘리는 관리방법이 경제적이며, 이에 초점을 맞추어 나가야 할 것으로 사료된다.

현장실증시험에 의한 단일 및 이중필터층 우물의 해수 여과 특성 연구 (A Study on the Seawater Filtration Characteristics of Single and Dual-filter Layer Well by Field Test)

  • 국내에서는 바닷가 주변의 상가나 수협 등의 활어 위판장 등에 많은 양의 해수가 사용되고 있다. 이러한 해수는 기상악화나 태풍 등에 의해 관련시설이 수시로 파손되고 조기 폐색되어 시간적, 경제적 비용이 많이 소요되고 있는 실정이다. 또한 오염된 해수가 그대로 유입되는 경우가 많아 환경적인 문제를 야기하기도 한다. 본 연구는 직접적인 해수취수의 대안으로 이중필터취수정을 적용한 해변여과 방식의 해수취수 적용성을 평가하고자 수행하였다. 본 연구에서는 토층으로 이루어진 해변의 자유면대수층에서 필터조건을 달리하여 실규모의 이중필터취수정과 단일필터취수정을 각각 설치하고 설치된 우물에서 단계 및 연속대수성시험을 실시하여 필터재 조건에 따른 투수특성 및 적정양수량을 평가하였다. 단계대수성시험 분석결과 이중필터 취수정에서 우물 개량의 정도가 양호하여 단일필터 대비 110.3%의 투수계수 상승효과가 발생하는 것으로 분석되었다. 동일 양수량 대비 이중필터의 투수계수가 높게 나타나며, 이는 이중필터가 단일필터에 비해 투수성이 개선된 영향으로 판단된다. 연속대수성시험 분석결과 관측정 및 양수정을 이용한 투수계수 분석에서 이중필터취수정(SD1200)이 단일필터취수정(SS800)에 비해 높은 투수특성을 나타내는 것으로 평가되었으며, 이중필터가 단일필터 대비 평균 110.7%의 투수계수 상승효과가 발생하는 것으로 분석되었다. 수위강하량 분석을 통한 양수량 평가결과, 수위강하 2.0 m일 때 이중필터 취수정이 단일필터 취수정 대비 122.8% 양수량이 증대되는 것으로 분석되었으며, 비수위강하량을 이용하여 적정양수량을 산정한 결과 이중필터취수정이 단일필터취수정에 비해 136.0% 높은 양수량을 보이는 것으로 분석되었다. 또한 변곡점을 이용한 양수량 평가결과 이중필터가 단일필터에 비해 160.0% 높은 적정양수량을 보이는 것으로 분석되었다. 적정양수량의 경우 분석방법에 따라 단일필터 대비 122.8~160.0%의 개선효과가 있는 것으로 평가되었으며, 이를 평균한 단일필터 대비 이중필터의 양수량 개선율은 139.6%이다. 즉, 이중필터 설치만으로 일반적인 우물 대비 취수효율이 40% 정도 개선될 수 있음을 확인하였다. 변곡점을 이용한 이중필터취수정의 적정양수량은 2843.3 L/min로써 이중필터취수정 단일공의 일 해수취수량은 약 $4,100m^3/day$ (${\fallingdotseq}4094.3m^3/day$)에 달하는 것으로 평가되었다. 이와 같이 공당 다량의 취수가 가능하기 때문에 해수취수의 적용성이 높을 것으로 기대된다. 또한 이중필터취수정을 이용하여 해수취수를 수행하는 경우 기존에 문제시되었던 기상악화나 태풍 등에 의한 파손우려를 해소할 수 있으며, 해변의 모래층이 필터재 역할을 하여 오염정도를 개선하는 효과가 있을 것으로 기대된다. 따라서, 기존 해수취수기술에 대한 환경적인 문제 해결의 대안이 될 수 있고, 설치비용 및 파손등과 관련된 장기적인 유지관리비용 면에서도 유리하여 경제적 측면에서 그 적용성이 매우 높을 것으로 판단된다. 본 연구의 결과는 추후 수행예정인 이중필터취수정의 강변여과수 적용을 위한 현장실증시험의 기초자료로 활용될 예정이며, 그 결과를 종합하여 강변여과 및 해변여과기술과 관련된 우물의 설계 및 시공에 대한 표준화된 기준을 제시할 수 있을 것으로 기대된다.

생물반응기를 이용한 황근 부정근의 대량증식과 추출물의 항산화 및 미백 활성 평가 (Mass Proliferation of Hibiscus hamabo Adventitious Root in an Air-lift Bioreactor, and the Antioxidant and Whitening Activity of the Extract)

  • 황근(Hibiscus hamabo Sieb. et Zucc., 노랑무궁화)은 주로 제주도 전역과 전남 남부지역에 자생하는 우리나라 유일의 야생 무궁화속의 낙엽성 반관목식물이다. 최근 무분별한 자생지 파괴에 따른 개체수 감소로 환경부 지정 한국적색목록 내 멸종위기야생생물 2급으로 지정되어 있다. 선행연구에서는 황근의 종자발아, 생태 및 유전적 특성, 내염성 등이 보고되었다. 따라서 본 연구에서는 생물반응기를 이용하여 황근 부정근의 대량증식, 항산화 및 미백효과를 조사하여 기능성 원료 활용 평가를 실시하였다. 황근 종자 및 식물체는 사전 환경부 채취 허가를 받은 제주시 구좌읍 관내지역에서 채집하였으며, 표면살균 후 MS 등 배지 조성을 조절하여 기내 세포주로 도입하였다. 그 결과, 종자의 전반적인 반응율이 지상부 신초 발생 및 지하부 뿌리 발근 측면에서 51.17~51.83%로 가장 효율적인 것으로 나타났다. 5,000 mL 생물반응기에서 황근 부정근의 증식에 유리한 배지 조건의 경우, 1/2 MS (Murashige and Skoog, 1962), 30 g/L 자당(sucrose), 2 mg/L IBA (indole-3-butyric acid)에서 8주 동안 배양한 것임을 확인하였다. 또한 총페놀 화합물 함량, 총플라보노이드 함량, DPPH 자유 라디칼 소거능 및 멜라닌 함량 분석을 통해 바이오매스 및 2차대사산물 축적의 연관관계를 비교하였다. 본 연구를 통해 황근 부정근의 대량증식, 항산화 및 미백 효능은 고부가가치 향장품 산업의 고품질 소재개발과 연계하여 추가적인 생리활성물질 연구의 가능성을 시사한다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. A photosensitizer behaved as an added absorber. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. Our department has developed an information system for routine hematology using down-sized computer system. We have used an IBM 486 compatible PC with 16MB main memory, 210 MB hard disk drive, 9 RS-232C port and 24 pin dot printer. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Optimal stimulus parameters, which can minimize muscle fatigue and can achieve effective muscle contraction were determined in slow and fast muscles in Sprague-Dawley rats. Stimulus patterns of our stimulator were designed to simulate electromyographic activity monitored during locomotion of normal subjects. Muscle types of the lower extremity were classified according to their mechanical property of contraction, which are slow muscle (msoleus m.) and fast muscle (medial gastrocneminus m., rectus femoris m., vastus lateralis m.). Optimal parameters of electrical stimulation for slow muscles were 20 Hz, 0.2 ms square pulse. For fast muscle, 40 Hz, 0.3 ms square pulse was optimal to produce repeated contraction. Higher stimulus intensity was required when synergistic muscles were stimulated simultaneously than when they were stimulated individually. Electrical stimulation for each muscle was designed to generate bipedal locomotion, so that individual muscles alternate contraction and relaxation to simulate stance and swing phases. Portable electrical stimulator with 16 channels built in microprocessor was constructed and applied to paraplegic patients due to lumbar cord injury.
