Free Radical Scavenging Activity of Butanol Fraction from the Fruit of Citrus junos

  • In this study, we investigated the free radical [1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical (${\cdot}OH$) and superoxide anion (${O_2}^-$)] scavenging activity of MeOH extract and 3 fractions of Citrus junos. Of the tested fractions, the BuOH fraction showed the strongest DPPH scavenging activity, showing the $IC_{50}$ values of 63.4 mg/mL. Therefore, we continuously carried out DPPH, ${\cdot}OH$ and ${O_2}^-$ scavenging activity tests of BuOH fraction of Citrus junos. The BuOH fraction of Citrus junos inhibited DPPH radical to 97.5% at a concentration of 1000 mg/mL and the scavenging activities were increased concentration-dependently. In addition, BuOH fraction from Citrus junos also scavenged ${\cdot}OH$ in a concentration dependent manner from 5 to 1000 mg/mL. Furthermore, BuOH fraction showed about 56% ${O_2}^-$-scavengimg activity at 25 mg/mL concentration but, the scavenging activities were not enhanced in a dose dependent manner. The present results suggest that BuOH fraction of Citrus junos would have the protective potential from oxidative stress induced by free radicals.

The Nutritional Composition and Antioxidant Activity from Undariopsis peterseniana (넓미역의 영양성분 및 항산화 활성)

  • The proximate composition, free amino acid composition and mineral contents from Undariopsis peterseniana were determined, and the antioxidant activities of ethanol (EtOH) and hot water extracts of U. peterseniana were investigated using DPPH (1,1-diphenyl-1-picrylhydrazyl), ABTS (2,2'- azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)) radical scavenging effects and reducing power. The contents of moisture, ash, protein, lipid and carbohydrate were 12.5%, 23.1%, 9.7%, 0.2% and 54.5%, respectively, and alginic acid content was 12.3%. The major free amino acid contents were alanine, phenylalanine, aminoethanol, valine, glutamic acid and phosphoserine. Ca (1589.1 mg) was the largest mineral followed by Na (344.6 mg), Mg (74.3 mg), Zn (10.2 mg) and Fe (1.5 mg). The total phenolic contents of EtOH and hot water extracts were exhibited at 15.7 and 4.3 mg GAE/g sample, respectively. The antioxidant activity of EtOH extract exhibited strong ABTS and DPPH radical scavenging activities with reducing power, and hot water extract also demonstrated strong ABTS radical scavenging effects. Therefore, the results of this study suggest that U. peterseniana contained an abundance of naturally occurring nutrients (free amino acids and minerals), and the strong antioxidant activities of EtOH and hot water extracts from U. peterseniana could be good sources of natural antioxidants for healthcare products.

Antioxidative Activities and Tyrosinase Inhibitory Effects of Korean Medicinal Plants

  • To discover the sources with antioxidant and tyrosinase inhibitory activities in Korean traditional medicines, 10 extract of medicinal plants were screened for their potential to scavenge stable 1,1-diphenyl-2-picryhydrazyl (DPPH) free radical, inhibit hydroxyl radical (${\cdot}OH$), total phenolic content, and inhibition of tyrosinase. The potency of DPPH radical scavenging activity was shown in the extract of Ulmus davidiana var. japonica Nakai that has a greater effect with $IC_{50}$ values of $6.49{\pm}5.43{\mu}g/mL$, than BHA ($IC_{50}=20.99{\pm}0.74{\mu}g/mL$), L-ascorbic acid $(IC_{50}=20.59{\pm}1.06{\mu}g/mL),\;and\;{\alpha}-tocopherol\;(IC_{50}=25.55{\pm}0.26{\mu}g/mL)$ as a positive control. The ${\cdot}OH$ scavenging activities were observed in the plants tested. Acanthopanax senticosus, Cirsium setiders, U. davidiana exhibited scavenging activity of more than 60% at $500{\mu}g/mL$. The scavenging activity(%) of BHA and a-tocopherol were 64.32 and 55.87% at $500{\mu}g/mL$, respectively. The total phenolic content was determined, in order to assess its effect on the extract's antioxidant activity. The total phenoic content of $33.37{\pm}0.52mg/g$ was conformed by methanolic extract of U. davidiana. The U. davidiana and Morus bombycis exhibited tyrosinase inhibitory activity with a $34.28{\pm}1.32\;and\;75.57{\pm}1.10%$, respectively. In particular, M. bombycis has stronger tyrosinase inhibitory activity than albutin with $36.48{\pm}3.56%$ as a positive control. This work showed that the inhibitory abilities of Korean medicinal plants, such as U. davidiana and M. bombycis, on DPPH free radical, inhibit hydroxyl radical (${\cdot}OH$), and inhibition of tyrosinase and total phenolic content, can be useful in the prevention and treatment of free radical-relate disease. Investigations into further isolation of inhibitory principles of U. davidiana and M. bombycis are now in progress.

Antioxidant and free radical scavenging activities of Cleome rutidosperma

  • The study was aimed at evaluating the antioxidant and free radical scavenging activities of ethanolic extract and its fractions of Cleome rutidosperma. The antioxidant activity, reducing power, total phenolic content, total flavonoid content, superoxide anion scavenging activity, nitric oxide anion scavenging activity, in vitro antilipid peroxidation activity and in vitro non-enzymatic hemoglobin glycosylation were studied. The results obtained in the study indicate that Cleome rutidosperma is a potential source of natural antioxidant. All the parameters were found to be concentration dependent and increased with increasing amounts of sample. Flavonoids, phenolic compound like tannins, terpenoids may be responsible for the antioxidant activity of the plant. Variation of solubility parameters in various models may be attributed to non-linearity of activity of ethanol extract fractions models. Further investigation on the isolation and identification of antioxidant component(s) in the plant may lead to chemical entities with potential for clinical use.

Free Radical Scavenging and Lipid Peroxidation Inhibition Potential of Hygrophila auriculata

  • Hygrophila auriculata (K. Schum.) Heine is a wild herb commonly found in moist places on the banks of tanks, ditches and paddy fields throughout India and is one of the main sources of Ayurvedic 'Rasayana' drug. The free radical scavenging potential of 50% aqueous alcoholic extract of H. auriculata (HAEt) was studied for DPPH scavenging activity, nitric oxide, hydroxyl radical and ferryl bipyridyl complex scavenging activity along with lipid peroxidation and total antioxidant capacity inhibition using Thiobarbituric acid reactive substances (TBARS) in rat liver homogenate. Integral antioxidative capacity was determined by photochemiluminescence assay. HAEt showed good radical scavenging activity at various concentrations $(200-1000\;{\mu}g/ml)$ against 1,1 diphenyl, 2-picrylhydrazyl (DPPH) (32.32-77.02%) with moderate scavenging activity against Nitric oxide (12.46-52.84%), hydroxyl radical (11.69-55.26%), ferryl bipyridyl complex (17.66-58.67%) and lipid peroxidation (0.829-0.416nmoles/mg protein). The above results indicate HAEt to be very effective antioxidant. It was suggested that the varied therapeutic activities claimed for the plant in the Indian indigenous systems of medicine may be in the part due to the free radical scavenging and/ or potent antioxidant activity.

Synthesis of Heterocyclic Chalcone Derivatives and Their Radical Scavenging Ability Toward 2,2-Diphenyl-1-Picrylhydrazyl (DPPH) Free Radicals

  • A series of heterocyclic chalcone derivatives bearing heterocycles such as thiophene or furan ring as an isostere of benzene ring were carefully prepared, and the influence of heterocycles on 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities was systematically investigated. Structure-activity relationships (SAR) analysis showed that the activities of thiophene ring-containing chalcones were higher than those of furan ring-containing chalcones, and the presence of methyl substituent of heterocyclic ring distinctly affected the activities compared with non-substituted heterocycles in an opposite manner, with the 4'-methyl group of thiophene ring increasing activity and the 3'-methyl group of the furan ring decreasing activity. The distinct isosteric effect of heterocycles (i.e., thiophene or furan ring) on radical scavenging activities of heterocyclic chalones was distinctly demonstrated in our work.

In Vitro Free Radical Scavenging and Hepatoprotective Activities of Taraxacum mongolicum (포공영의 자유라디칼 소거 및 간세포 보호활성)

  • The methanol (MeOH) extract and ist fractions of Taraxacum mongolicum (Compositae) were examined for their scavenging effects on 1,1-diphenyl-2- phenylhydrazyl (DPPH) and superoxide radicals, and hepatoprotective effects on tacrine-induced cytotoxicity in human hepatoma cell line, Hep G2 cells. Both methylene chloride $(CH_2Cl_2)$ and butanol (n-BuOH) soluble fractions of the MeOH extract showed the free radicals scavenging and hepatoprotective effects. From these results, it is suggested that hepatoprotective effect of these fractions partly relies on their free radical scavenging activity.

  • The EtOAc and n-BuOH soluble fractions from the aerial part of Setaria viridis showed a strong free radical scavenging activity. Six major compounds were isolated from these fractions. They were identified by spectral data as tricin (1), p-hydroxycinnamic acid (2), vitexin 2"-Ο-xyloside (3), orientin 2"-Ο-xyloside (4), $tricin-7-Ο-{\beta}-D-glucoside$ (5) and vitexin 2"-Ο-glucoside (6). Among these compounds, 4 and 5 exhibited strong free radical scavenging activities on 1, 1-diphenyl-2-picrylhydrazyl (DPPH). We further studied the effects of these isolated compounds on the lipid peroxidation in rat liver microsomes induced by non-enzymatic method. As expected, 4 and 5 exhibited significant inhibition on $ascorbic/Fe^{2-}$ induced lipid peroxidation in rat liver microsomes.ver microsomes.

Effects of Hwagi-Jogyeong-Tang (HJT) on Human HaCaT keratinocyte and malignant melanoma cells (화기조경탕(化氣調經湯)이 피부 세포 재생 및 악성 흑색종 세포에 미치는 영향)

  • Objective : Hwagi-Jokyeong-Tang (化氣調經湯, HJT) was described in DongeuiBogam(東醫寶鑑). This remedy has been used to treat patients with Naryeok, which is similar as tuberculous cervical lymphadenitis in western medicine. Methods : In this study, the present author investigated the effects of HJT on on Human HaCaT keratinocyte and malignant melanoma cells such as SK-MEL-2 and B16F10 in terms of cell viabilities, proliferations, DPPH free radical scavenging activities, oxygen free radical productions and inhibitory action on elastase activities. Results : HJT acceleated proliferation of HaCaT keratinocytes dose-dependantly. HJT also prevented cell death of HaCaT induced by Hydrogen peroxide, which products oxygen free radicals. On the contrary, HJT did not affect proliferations of SK-MEL-2 or B16F10. In addition, HJT was shown to have DPPH free radical scavenging activities and also have inhibitory effects on elastase activities too. On the fluorescent examinations, the present author know that HJT did not affect production levels of oxygen free radicals in malignant melanoma cell, SK-MEL-2. Conclusions : These results suggest that HJT has possibilities of usage for functional cosmetics which have skin regeneration or prevention from skin tissue injury.

Anti-oxidant Activities of Punica granatum Extracts (석류 추출물의 항산화 작용)

  • Pomegranate (Punica granatum L.) which is very rich in Polyphenols and tannins was recently reported its anti-oxidant activities and phytoestrogenic activities in vivo test and many clinical studies, but the effects of them on the skin have not been reported. The experiments were tarried out in vitro to determine anti-oxidant activities of pomegranate extracts on DPPH radical scavenging assay, NBT/Xanthine Oxidase-superoxide scavenging assay, silica-induced intracellular $H_2O_2$, hydroperoxide and superoxide generation assay in RAW 264.7 cells. It showed that the methanolic extract of dried pomegranate peels have the most significant anti-oxidant activities on free radical scavenging assay and inhibitory activities on silica-induced intracellular free radical generation in RAW 264.7 cells. The concentrated juice of pomegranate showed only DPPH radical scavenging activities and inhibited hyaluronidase activity. Moreover, pomegranate seed oil inhibited specially silica-induced intracellular hydroperoxide generation in RAW 264.7 cells. These results suggest that the methanolic extract of dried pomegranate peels and pomegranate seed oil have more anti-oxidant activity than concentrated juice of pomegranate. Thus the extracts of pomegranate peels and seed oil could be developed cosmetic ingredients for anti-aging.
