Antioxidative and cytoprotective effects of tectorigenin and glycitein isolated from the pueraria thunbergiana and its derivative, genistein, were determined. Among these three compounds, tectorigenin and glycitein bearing 6-methoxyl groups in both isoflavones showed significant free radical scavenging activities on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and xanthine/xanthine oxidase (XOD) generating superoxide anion radical. Tectorigenin only showed a slight inhibitory effect on XOD. We further studied the inhibitory effects of these isoflavones on the lipid peroxidation in rat liver microsomes induced by enzymatic and non-enzymatic methods. Each of them exhibited inhibitory effect on both ascorbic $acid/Fe^{2+}-{\;}and{\;}ADP/NADPH/Fe^{+3}-induced$ lipid peroxidation. Moreover, tectorigenin exhibited the highest protection of hydrogen peroxide damage on HepG2 and Vero cells among the three isoflavones, in the cytoprotective assay. It was suggested that the pattern of antioxidative and cytoprotective effect of isoflavones could be crucially by the aromatic substitution of oxygen-containing groups.
To investigate the cyclohexane and xylene mixture treatment on the liver damage, the rats were treated by the mixture of cyclohexane and xylene (CH+X) and then, liver damage was demonstrated by liver function findings based on liver weight/body weight, serum level of alanine aminotransferase (ALT), xanthine oxidase (XO) and then compared with cyclohexane treated group (CH group) and xylene-treated group (X). The CH+X group showed merely severer liver damge than CH or X group. On the other hand, CH+X group showed lower activity of hepatic cytochrome P-450 dependent aniline hydroxylase (CYPdAH) than CH or X group, but no statical differences were demonstrated among three experimental groups. Especially the hepatic GSH content was merely declined than CH or X group and the activity of hepatic GST was higher in CH+X group than CH or X group. In conclusion, cyclohexane and xylene mixture treated animals showed merely severer liver damage than cyclohexane or xylene treated group and such a fact may be caused by inhibition of cyclohexane or xylene metabolism and oxygen free radical.
A growing body of evidence supports lipid peroxidation as having a role in the pathogenesis of liver disease. Although the probable cause of damage to human hepatocytes may be multifactorial, free radicals have been implicated in a variety of liver diseases, particularly in the presence of iron overload and toxic substances such as ethanol. Consequently, antioxidants, particularly those of plant origin such as flavonoids, may help to reduce the risk of developing these diseases. Silybum (S.) marianum, a medicinal plant widely used in traditional European medicine for the treatment of liver disorders, was evaluated for antioxidant activity. Thin layer chromatography and High Performance Liquid Chromatography analyses of crude extract of the plant confirmed the presence of a number of flavonoids reported in the literature. The antioxidant activity of these flavonoids was measured through inhibition of lipid peroxidation and 1, 1-diphenyl-2- picrylhydrazyl radical scavenging. The crude plant extract showed marked antioxidant activity in both assays. These results suggest that S. marianum contains flavonoids with antioxidant activity, capable of inhibiting or scavenging free radicals, thus supporting its traditional use as a hepatoprotective agent.
Excess free iron generates oxidative stress that may contribute to the pathogenesis of various causes of neurodegenerative diseases. In this study, we assessed the modification of ferritin induced by $H_2O_2$. When ferritin was incubated with $H_2O_2$, the degradation of ferritin L-chain increased with the $H_2O_2$ concentration whereas ferritin H-chain was remained. Free radical scavengers, azide, thiourea, and N-acetyl-$_L$-cysteine suppressed the $H_2O_2$-mediated ferritin modification. The iron specific chelator, deferoxamine, effectively prevented $H_2O_2$-mediated ferritin degradation in modified ferritin. The release of iron ions from ferritin was increased in $H_2O_2$ concentration-dependent manner. The present results suggest that free radicals may play a role in the modification and iron releasing of ferritin by $H_2O_2$. It is assumed that oxidative damage of ferritin by $H_2O_2$ may induce the increase of iron content in cells and subsequently lead to the deleterious condition.

Red brownish p-pheylenediamine (PPD) has been widely used hair dye for women. The dye was known to cause systemic anaphylaxis, dermatitis and bladder cancer. But the effect of PPD toxicity with oxygen free radical has not been studied. This study investigated the degree of skin injury by PPD. PPD ($2.5\%$ PPD in $2\%\;NH_{4}OH$) was applied to the rat skin ($25 mg/16.5\;cm^2$) 3 or 5 times every other day. Histopathological findings demonstrated the proliferation of epithelial cells and the increased keratinization by PPD. The activities of glucose 6-phosphatase (G6Pase) was decreased and acid phosphatase (ACP) was increased in PPD-applied rat skin. Groups in which PPD was applied 5 times were more damaged than groups applied 3 times. To examine the relationship between tissue damage and oxygen free radicals, effect of PPD on xanthine oxidase (XO) activity was measured and XO activity was more significantly increased in the group treated with PPD 5 times than 3 times. However, reduced glutathione (GSH) content, and the activities of catalase (CAT), super-oxide dismutase (SOD) and glutathione S -transferase (GST) were more decreased in PPD-applied groups than in controls. Even though the activities of XOD was not changed in the group treated with PPD 3 times, the decreased activities of oxygen free radical system and the damaged skin tissue were observed. This result might be caused by the production of toxic PPD metabolites in rat skin. In conclusion, topical PPD application led to skin injury in a dose-dependent manner, probably due to the generation rate of oxygen free radical.

Here, we showed that Acanthocoris sordidus extract inhibited both cell and DNA damage caused by oxidative stress. In a radical scavenging assay, the scavenging activity of the A. sordidus extract against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl radicals was 48.9% and 37.8%, respectively, that of ascorbic acid, which was used as a positive control. The ferrous iron chelating activity of the A. sordidus extract was 80.0% compared to that when ethylenediaminetetraacetic acid (EDTA) was used a control. To verify the inhibitory effect of the extract on oxidative cell damage induced by reactive oxygen species (ROS), a lipid peroxidation assay was performed. The results showed that peroxidation was completely inhibited in an extract-treated group compared to a radical-treated group. The level of p21 protein expression was 68.1% that of a control sample. The DNA cleavage-inhibiting property of the A. sordidus extract-treated group was 53.3% that of a control group. Moreover, the phosphorylation of the H2AX protein was reduced to 39.0% of that treated with radical agents, indicating that the extract might inhibit the DNA damage that causes radical oxidation. Taken together, our findings suggest that the A. sordidus extract is effective not only in repressing oxidation by free oxygen radicals and hydroxyl radicals but also in decreasing cell and DNA damage caused by oxidative stress.
The purpose of this study is to investigate the appropriate management of severe radionecrotic wounds of the anterior chest wall associated with infection of the soft tissues and ribs and exposure of vital structures(heart and lung), and present our strategies for reconstruction of these complicated patients. 9 patients have undergone radical debridement and immediate microsurgical reconstruction for severe radionecrotic wounds of the anterior chest wall over last 7 years. All patients had extensive osteomyelitis of the ribs or sternum, and chronic infection or cutaneous fistulae. 2 patients had pericardial effusions due to longstanding inflammation, and 6 patients had pleural effusions. 2 patients had ipsilateral lung collapse. 10 free flaps were performed for coverage of the huge defects. One patient required 2 free flaps to control the inflammation. 8 free TRAM flaps were used for coverage of the defects and in addition, the rectus abdominis muscle was packed into any deep cavity. 1 patients underwent latissimus dorsi muscle free flap because of previous abdominal surgery. After extensive debridement of the infected, radionecrotic wounds, all 10 free flaps were successful. All the extensive radionecrotic defects of the anterior chest wall were completely healed. Free flaps successfully covered the exposed vital structures of the heart and lungs. Patients with severe radionecrotic defects of the anterior chest wall after ablative breast cancer surgery and radiotherapy were successfully treated by radical debridement and immediate free flap surgery. The TRAM flap together with the rectus muscle is the treatment of choice for these huge defects. The latissimus dorsi muscle flap was the second choice in patients with previous abdominal surgery. The recipient vessel should be carefully selected because of possible radiation damage and inflammation.

Ribes diacanthum Pall (RDP) is a member of the Saxifragaceae family. The plant is traditionally used in Mongolia for the treatment of various ailments associated with kidney and bladder's diseases, cystitis, kidney stone, and edema. This study was aimed to investigate antioxidant activities of different solvent extracts of whole Pall plants, based on ferric-reducing antioxidant potential (FRAP), 2,2'-azinobis(3-ethybenzothiazoline-6-sulfonic acid) ($ABTS{\cdot}+$) radical scavenging activity, 1,1-diphenyl-2-picrydrazyl ($DPPH{\cdot}$), and hydroxyl (${\cdot}OH$) radical scavenging activities. Additionally, total flavonoids and phenolic contents (TPC) were also determined. The ethyl acetate extract of RDP (EARDP) had a remarkable radical scavenging capacity with an $IC_{50}$ value of 0.1482 mg/mL. In addition, EARDP was shown to be higher in total phenolic and flavonoid contents than the methanol extract of RDP (MRDP). Moreover, the EARDP had the predominant antioxidant capacity, DPPH, hydroxyl, and ABTS radical scavenging activities and ferric reducing power. These results suggest a potential for R. diacanthum Pall extract as a functional medicinal material against free-radical-associated oxidative damage.
An antioxidant activity of Rosa rugosa extract and its solvent-partitioned fractions was determined not only by measuring lipid peroxide produced when a mouse liver homogenate was exposed to the air at 37$^{\circ}C$, using thiobarbituric acid (TBA) but also by evaluating the free radical scavenging effect against DPPH radical, authentic peroxynitrite, and 3-morpholinsydnonimine (SIN-1). All its partitioned fractions including crude extract showed potent scavenging effect against DPPH radical, peroxynitrite, and lipid peroxidation. n-BuOH fraction, in particular, was found to be the most effective in DPPH radical scavenging ability as well as inhibition against lipid peroxidation. The 15% aqueous MeOH fraction also showed a strong potency which was slightly lower than n-BuOH fraction. Based on these results, we suggest that Rosa rugosa could be useful for preventing an oxidative damage.
Erythrina stricta, a deciduous tree widely used traditionally in indigenous system of medicine for various ailments such as rheumatism, fever, leprosy, epilepsy etc. The leaves of Erythrina stricta was extracted with ethanol (70%) and used for the evaluation of various in vitro antioxidant assays which includes H - donor activity, nitric oxide scavenging, superoxide anion scavenging, reducing ability, hydroxyl radical, hydrogen peroxide scavenging, total phenolic content, total flavonoid content, total antioxidant activity by thiocyanate and phosphomolybdenum method, metal chelating, $\beta$-carotene bleaching, total peroxy radical assays. The pro-oxidant activity was measured using bleomycin-dependent DNA damage. Ex vivo models like lipid peroxidation and erythrocyte haemolysis were also used to study the antioxidant property of the extract. The various antioxidant activities were compared with suitable standard antioxidants such as ascorbic acid, butylated hydroxyl toluene, $\alpha$-tocopherol, curcumin, quercetin and Trolox. The generation of free radicals viz. $O_2^{{\cdot}-}$, $OH^{\cdot}$, $H_2O_2$, $NO^{\cdot}$ and peroxyl radicals were effectively scavenged by the ethanolic extract of Erythrina stricta. In all the methods, the extract offered strong antioxidant activity in a concentration dependent manner. The total phenolic content, flavonoid content and total antioxidant activity in Erythrina stricta were determined as microgram (g) pyrocatechol, quercetin and $\alpha$-tocopherol equivalent/mg respectively. The extract did not exhibit any prooxidant activity when compared with ascorbic acid. The results obtained in the present study clearly indicates that Erythrina stricta scavenges free radicals and reduces lipid peroxidation, ameliorating the damage imposed by oxidative stress in different disease conditions and serve as a potential source of natural antioxidant.
