Antioxidative Constituents from Paeonia lactiflora

  • The ethanol extract of the peony root (Paeonia Lactiflora Pall, Paeoniaceae) as well as its major active components including gallic acid and methyl gallate were evaluated for their protective effects against free radical generation and lipid peroxidation. In addition, the protective effects against hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line were examined. The ethanol extracts of the peony root (PREs) and its active constituents, gallic acid and methyl gallate, exhibited a significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and had an inhibitory effect on lipid peroxidation, as measured by the level of malondialdehyde (MDA) formation. The PREs did not have any pro-oxidant effect. They strongly inhibited the hydrogen peroxide-induced DNA damage from NIH/3T3 fibroblasts, as assessed by single cell gel electrophoresis. Furthermore, the oral administration of 50% PRE (50% ethanol extract of peony root), gallic acid and methyl gallate potently inhibited the formation of micronucleated reticulocytes (MNRET) in the mouse peripheral blood induced by a $KBrO_3$ treatment in vivo. Therefore, PREs containing gallic acid and methyl gallate may be a useful antigenotoxic antioxidant by scavenging free radicals, inhibiting lipid peroxidation and protecting against oxidative DNA damage without exhibiting any pro-oxidant effect.

  • The primary aim of this study is to investigate new semi-abrasive free slurry including acid colloidal silica and hydrogen peroxide for copper chemical-mechanical planarization (CMP). In general, slurry for copper CMP consists of colloidal silica as an abrasive, organic acid as a complex-forming agent, hydrogen peroxide as an oxidizing agent, a film forming agent, a pH control agent and several additives. We developed new semi-abrasive free slurry (SAFS) including below 0.5% acid colloidal silica. We evaluated additives as stabilizers for hydrogen peroxide as well as accelerators in tantalum nitride CMP process. We also estimated dispersion stability and Zeta potential of the acid colloidal silica with additives. The extent of enhancement in tantalum nitride CMP was verified through anelectrochemical test. This approach may be useful for the application of single and first step copper CMP slurry with one package system.

추출 조건에 따른 고추 수용액의 가용성 성분의 변화 (Changes of Soluble Solid Content in Red Pepper by Different Extraction Conditions)

  • The soluble solid of red pepper was extracted by water in order to investigate changes of soluble solid content by different extraction temperature $(4{\sim}90^{\circ}C)$ and time $(1/2{\sim}3\;hrs)$, and the contents of carotenoid, capsaicinoids, free sugar, organic acid, free amino acid in soluble solid were measured. Most of soluble solid in red pepper was extracted within the first 2 hrs and $93{\sim}98%$ of total soluble solid was extracted during the first 30 min. The contents of carotenoid increased by increasing extraction time and temperature, but decreased by increasing extraction time at $60^{\circ}C$ and $90^{\circ}C$. ${\beta}$-carotene content was sharply decreased after 2 hrs at $90^{\circ}C$. The content of capsaicinoid was sharply increased between 1 hr and 2 hr. Fructose and glucose in red pepper were extracted in the range of $83.8%{\sim}96.4%$ and the contents of free sugar gradually increased by increasing extraction time and temperature. The content of organic acid was gradually increased by increasing extraction time and temperature and the greatest amount of organic acid was extracted during the first 30 min of extraction time. The content of free amino acid was decreased by increasing extraction temperature.

  • 한국백삼의 유리페놀성 분획(free phenolic acid fr.)에서 phenolic acid의 분리 정제방법을 검토하였다. 먼저 silicic acid column에 의해 소분획으로 나누고 prep TLC 및 HPLC하여 정색반응 및 Pf차에 의해 phenolic acid로 추정되는 두 개의 화합물을 순수 분리하고, 결정을 얻어 UV, IR, MS, NMR로 그 구조를 밝힌 결과 ferulic acid와 vanillic acid로 확인되었다.

도라지 잎과 줄기의 화학성분 및 항산화활성 (Chemical Composition and Antioxidative Activities of Platycodon grandiflorum Leaves and Stems)

  • 도라지 잎과 줄기를 기능성식품 재료로 이용하기 위한 기초자료를 제공하기 위하여 화학성분 및 항산화 활성을 조사하였다. 도라지 잎과 줄기의 수분함량은 각각 84.31%와 5.91%였으며, 도라지 잎과 줄기의 무기성분 중 Ca이 351.49 mg%와 217.56 mg%로 함량이 많았다. 도라지 잎의 주요 유리당은 glucose로 1,729.8 mg%가 함유되어 있었고, 줄기에서는 fructose가 734.87mg%였다. 도라지 잎의 주요 아미노산으로는 glutamic acid(242.91 mg%)와 arginine(228.60 mg%)이었으며, 줄기에서는 lysine(110.08 mg%)과 glutamic acid(80.40 mg%)였고, 도라지 잎과 줄기의 주요 지방산은 oleic acid와 linoleic acid였다. 도라지 잎과 줄기의 용매 분획물을 이용하여 DPPH radical 소거활성을 측정한 결과 분획물의 농도가 증가함에 따라 DPPH radical 소거활성이 증가하였으며, 환원력도 농도의존적인 경향을 보였고, 특히 부탄올 분획물에서 환원력이 2.25로 가장 높았다.

참당귀의 무기성분, 유리당, 아미노산 및 지방산 함량 특성 (Characterization of Inorganic Components, Free Sugars, Amino Acids, and Fatty Acids in Angelica gigas Nakai)

  • Background : The major compounds of Angelica species are decursin, decursinol angelate, nodakenin, umbelliferone and ${\beta}$-sitosterol, which act anti-inflammatories, reduce pains, protect the liver and enhance the immune system. This study investigated the chemical compositions, minerals, metals, sugars and overall amino acid composition in Angelica gigas Nakai. Methods and Results : Powder of Angelica roots smaller than 30 mesh were used. Physico-chemical analysis revealed the presence of carbohydrates (62.0%), crude proteins (13.9%), moisture (11.4%), crude fats (7.3%) and ash (5.4%). Results showed that potassium was present in the highest amount (1,859 ppm), followed by magnesium (214.5 ppm), calcium (147.3 ppm) and sodium (6.0 ppm). Free sugar profiles showed the presence of sucrose (29.3 g/100 g). The total amino acids concentrations was 9,752 mg/100 g, the most common and dominant amino acids were arginine (2,181 mg/100 g), glutamic acid (1,212 mg/100 g) and aspartic acid (834 mg/100 g). The total free amino acids contents was 1,476 mg/100 g, in which the most common amino acid were arginine (932 mg/100 g), glutamic acid (127 mg/100 g), and ${\gamma}$-aminobutyric acid (80.4 mg/100 g). The fatty acid composition of A. gigas showed a higher concentration of unsaturated fatty acids such as linoleic acid (443.9 mg/100 g) and palmitic acid (181.3 mg/100 g) according to gas chromatography. Conclusions : These results showed that Angelica roots can be used in various fields of foods and medicines, and in the preparation of cosmetics.

  • The present study investigated the effect of diets supplemented with different levels (0, 1, 3, and 5 %) of fig leaf ethanol extract (FLEE) on the nutritional composition of black rockfish Sebastes schlegeli. Fish ($10.05{\pm}0.44g$) were fed to apparent satiation twice daily for 8 weeks. Adding FLEE decreased crude lipid levels and increased crude protein and ash. Among the three organic acids in the whole body of black rockfish, lactic acid was predominant, followed by citric acid and oxalic acid. Five free sugars were found in all groups. Fucose and glucose were the dominant free sugars in the FLEE-added group. The abundant fatty acids in the FLEE-added group were C16:0, C18:1-cis (n9), and C22:6n-3. The major amino acids in the samples were glutamic acid, aspartic acid, glycine, leucine, alanine, lysine, and arginine. The abundant free amino acids in the FLEE-added group were taurine, glutamic acid, alanine, leucine, and arginine.

오염중의 유이지방산이 세척에 미치는 영향(제일보 팔미트산의 세척 특성) (Studies on the Detergency Characteristics of Free Fatty Acid in Oily Soil. Part 1. Detergency of Palmitic Acid.)

  • The effects of surfactants and concentration of NaOH in surfactant solution on the removal of free fatty acid soil from cotton fabrics were investigated. Cotton fabrics were soiled with palm itic acid which is the most common fatty acid found in natural oily soil and washed in Lauder-ometer with various types of surfactant with or without NaOH. The rate of soil removal was estimated by analyzing palmitic acid contents in fabric before ar d after washing. Analysis of palmitic contents was made by extracting palmitic acid with azeotropic mixture of alcohol-benzene and the extracts were titrated with standard NaOH solution. It was shown that the types of surfactant are important factor in free fatty acid removal and the efficiency increases in the following order: SLS$90\%$ of initial sorption. In relation to the mechanism of detergency, the suspending and emulsifying power of surfactants were also examined. From the results of this experiments, it could be concluded that the soap formation with alkali and the suspending power of surfactant arc significant factors in free fatty acid soil removal, but the emulsifying power of it is neglizible.

사람 중성호성 백혈구의 기능에 있어서 Arachidonic Acid의 활성화 기전 (Activation Mechanism of Arachidonic Acid in Human Neutrophil Function)

  • 칼슘을 함유하는 반응액에서 arachidonic acid는 뚜렷하게 superoxide와 $H_2O_2$의 생성을 자극하였고 NADPH oxidase를 활성화하였다. 칼슘이 없는 반응액에서 NADPH oxidase에 대한 arachidonic acid의 자극 작용은 나타나지 않았다. Arachidonic acid에 의하여 자극된 respiratory burst는 EGTA, TMB-8, verapamil, diltiazem, nifedipine, dibucaine, lidocaine, CCCP, 2,4-dinitrophenol, sodium arsenate, chlorpromazine, theophylline, $HgCl_2$, PCMB와 PCMBSA에 의하여 억제되었으나, tetrodotoxin, tetraethyl ammonium chloride와 procaine의 영향은 받지 않았다. EGTA는 거의 완전히 arachidonic acid에 의한 ${\beta}-glucuronidase$의 유리를 억제하였으며, verapamil, CCCP와 theophylline은 약간 억제하였으나, 이에 반하여 dibucaine은 유의한 효과를 나타내지 않았다. Arachidonic acid는 중성호성 백혈구로 부터 칼슘을 유리시켰으며, 이는 TMB-8에 의하여 감소되었다. Arachidonic acid에 의한 세포내 유리 칼슘 농도의 상승은 EGTA와 CCCP에 의하여 억제되었고 TMB-8에 의하여 약간 억제되었다. Arachidonic acid와 verapamil 또는 arachidonic acid와 dibucaine의 동시 첨가에 의하여 상승된 세포내 유리 칼슘 농도의 양은 arachidonic acid 단독에 의한 것보다 현저하였다. 이상의 결과로 부터 다양한 생화학적 변화가 arachidonic acid에 의하여 활성화된 중성호성 백혈구의 기능 표현에 관여할 것으로 시사된다. Arachidonic acid는 세포내 칼슘 저장고로부터 칼슘유리를 자극하여 세포내 유리 칼슘 농도를 상승시킬 것으로 추정된다. 중성호성 백혈구의 활성화증에 칼슘 유입과 유출은 동시에 이루어질 것으로 추정된다.

국내산 마늘의 향미성분 (Flavor Composition of Garlic from Different Area)

  • 산지별 마늘의 향미성분으로서 향기, 유리당, 유리아미노산 및 유기산 함량을 비교, 분석하였다. 마늘의 향기성분을 GC로 분리한 결과 28개의 피크를 얻었고 이중 GC/MS를 이용하여 25종의 화합물을 동정할 수 있었다. 향기성분을 총피크면적을 기준으로 할 때 서산산이 가장 높았으며 함평산은 낮은 것으로 나타났다. 마늘의 주요 향기성분으로 알려진 황화합물에 있어서는 남해산, 서산산 및 의성산 사이에는 유의적인 차이가 없는 것으로 나타났다. 이온 크로마토그래피로 유리당을 분석한 결과 glucose, fructose, sucrose 이외에 fructooligosaccharide인 1-kestose, 1-nystose, 1-f-fructosyl nystose가 비교적 많이 함유되어 있었으며, 서산마늘은 1-nystose, 1-F-fructosyl nystose함량이 높았다. 총 유리아미노산 함량은 $2036.1{\sim}2704.0mg%$ (건조물기준)정도 함유되어 있었으며, 한지계마늘인 의성마늘과 서산마늘이 남해 및 함평산 마늘 보다 유리아미노산 함량이 비교적 높은 것으로 나타났다. 유기산 분석 결과 마늘중에는 citric acid, pyruvic acid, malic acid, lactic acid, pyroglutamic acid, succinic acid, oxalic acid, malonic acid, levulinic acid 및 fumaric acid가 존재하였고 이들 유기산의 총함량은 건조마늘 100g당 $1905.7{\sim}2359.2mg$이 함유되어 있었다. 함평산 마늘은 다른 지역의 마늘 보다 유기산 함량이 비교적 적은 것으로 나타났다.
