Expermient were conducted to determine the effect of phosphorus stress on bacteroid content and energy status of soybean (Glycine max [L.] Merr.) nodules. Plants inoculated with Bradyrhizobium japonicum MN 110 were grown with P-stressed (0.05 mM-P) and control (1 mM-P) treatment in the greenhouse. Phosphorus stress decreased nodule mass per plant and nodule mass to whole plant mass ratio. Phosphorus concentration in leaf, stem and root tissues were reduced by 75% but in nodule tissue was reduced only by 40% under phosphorus stress during 3 week experimental period. The bacteroid content per unit nodule mass and the distribution of total nitrogen and total phosphorus among the bacteroid and plant cell fractions of nodule were not affected significantly by phosphorus stress. Regardless of phosphorus treatment, 22% of the nitrogen and 27% of the phosphorus in whole nodules were associated with the bacteroid fraction. The ATP and total adenylate concentrations in and energy charge of whole nodule were decreased 77%, 46% and 37%, respectively, by phosphorus stress. The ATP concentration in and energy charge of the host plant cell fraction of nodules were decreased 86% and 59%, respectively, but these parametres in bacteroid in nodules were not affected by phosphorus stress. These results indicated that nodule is a strong phosphorus sink and that nodule growth and development are regulated to maintain a high phosphorus and energy content in bacteroid even when the host plant is subjected to phosphorus deficiency.
This study was performed to investigate the average molecular weight range of chitosan hydrolysates showing maximum effect in macrophage activation. Nitrite production by continuous macrophage cell line J774A.1 was the highest at $10\;{\mu}g/ml$ concentration of intact chitosan. Hydrogen peroxide production by J774A.1 showed the high value of $894\;{\mu}M/mg$ macrophage protein at $1,000\;{\mu}g/ml$ concentration of chitosan hydrolysate fraction 5 and $1,044\;{\mu}M/mg$ macrophage protein at $100\;{\mu}g/ml$ concentration of the fraction 6. Chitsan hydrolysate fraction 4, fraction 6 and intact chitosan enhanced $IL-1{\alpha}$ production, while the others did not. The production of tumor necrosis factor showed the high value at $1,000\;{\mu}g/ml$ concentration of chitosan hydrolysate fraction 4, $100\;{\mu}g/ml$ concentration of the fraction 5 and fraction 6, and $10\;{\mu}g/ml$ concentration of intact chitosan. In conclusion, fractions 4, 5 and 6 of the chitosan hydrolysatets with average molecular weight of $24,000{\sim}64,000$ calculated by HPLC analysis are the most effective in macrophage activation tested in this study.

The effects of chemical binders (ladle slag, ordinary portland cement (OPC), hydroxyapatite and calcium hydroxide) on the solidification/stabilization of heavy metals (Cd, Cu, Ni, Pb, Zn) in sewage sludge were evaluated by chemical leaching tests such as EDTA extraction, TCLP and sequential extraction. The results of EDTA extraction showed that heavy metal concentrations in sewage sludge were highly reduced after solidification/stabilization with slag, cement or calcium hydroxide. However, EDTA interrupted solidification/stabilization of heavy metals by hydroxyapatite. The TCLP-extracted heavy metal concentrations in sewage sludge after solidification/stabilization with chemical amendments were highly reduced. However, Cu concentration in the sewage sludge solidified/stabilized with slag, cement or calcium hydroxide increased because the pH of TCLP solution was higher than 7. Mixtures of sludge 1 : slag 0.2 : calcium hydroxide 0.1 (wt ratio) showed the least leachability in batch TCLP and EDTA extraction. The results of sequential extraction (SM&T, formaly BCR) indicated that the distribution of heavy metals changed from exchangable and carbonate fractions to strongly bound organic fraction. It was found that maximum leachate concentrations of Ba, Cd, Cr and Pb from sewage sludge amended with slag and calcium hydroxide were far below US EPA TCLP regulations.

The authentication of the physico-chemical properties of ginsenosides reference materials as well as qualitative and quantitative batch analytical data based on validated analytical procedures is a prerequisite for certifying good manufacturing practice (GMP). Ginsenoside Rb1 and Rg1, representing protopanaxadiol and protopanaxatriol ginsenosides, respectively, are accepted as marker substances in quality control standards worldwide. However, the current analytical methods for these two compounds recommended by Korean, Chinese, European, and Japanese pharmacopoeia do not apply to red ginseng preparations, particularly the extract, because of the relatively low content of the two agents in red ginseng compared to white ginseng. In manufacturing fresh ginseng into red ginseng products, ginseng roots are exposed to a high temperature for many hours, and the naturally occurring ginsenoside Rb1 and Rg1 are converted to artifact ginsenosides such as Rg3, Rg5, Rh1, and Rh2 during the heating process. The analysis of ginsenosides in commercially available ginseng products in Korea led us to propose the inclusion of the (20S)- and (20R)-ginsenoside Rg3, including ginsenoside Rb1 and Rg1, as additional reference materials for ginseng preparations. (20S)- and (20R)-ginsenoside Rg3 were isolated by Diaion HP-20 adsorption chromatography, silica gel flash chromatography, recrystallization, and preparative HPLC. HPLC fractions corresponding to those two ginsenosides were recrystallized in appropriate solvents for the analysis of physico-chemical properties. Documentation of those isolated ginsenosides was achieved according to the method proposed by Gaedcke and Steinhoff. The ginsenosides were subjected to analyses of their general characteristics, identification, purity, content quantification, and mass balance tests. The isolated ginsenosides showed 100% purity when determined by the three HPLC systems. Also, the water content was found to be 0.534% for (20S)-Rg3 and 0.920% for (20R)-Rg3, meaning that the net mass balances for (20S)-Rg3 and (20R)-Rg3 were 99.466% and 99.080%, respectively. From these results, we could assess and propose a full spectrum of physico-chemical properties of (20S)- and (20R)-ginsenoside Rg3 as standard reference materials for GMP-based quality control.
In this study, changing characteristics of bed materials downstream of Daecheong Dam by flushing flow was investigated. Flushing flow affected the downstream environment in such a way that the mean diameter of bed material was increased and the standard deviation was decreased. The vertical sorting of bed materials composed of mixed sediment in the sediment box was observed. The surface layer was composed of gravels, and uniform sand material was buried below it. Relative fractions (Di/D50) of the bed material trapped in the box was nearly 1.0 for the bed material coarser than D$_{50}$ at Hyundo gauging station, and was between two values on the bed surface before and after the flushing flow at Bugang gauging station. Ratio of driving force to resisting force for each fraction ($\tau_i^*$) with the faction size maintained constant for the bed material coarser than D$_{50}$ at Hyundo station. At Bugang station, however, it was rapidly decreased. It means that pavement of the surface layer of bed material at Bugang station was developed, with the sediment particles moving downstream uniformly by flushing flow.
To study characteristics of the heavy metal pollution, sediment samples were collected at 67 sites on the roadside of Jeonju city during summer and winter, 2002. The total concentration of metals including Cd, Co, Cr, Cu, Ni, Pb, Zn, and Mn in the sediment samples were determined. The results indicate that the roadside sediments in Jeonju city have lower (1/2 to 1/7 times) concentrations of Zn, Cu, Pb and Cd than the metal concentrations previously reported for roadside soil, dust and sewage sludges in Seoul. However, the metal concentrations are higher than environmental quality criteria in soil suggested from several countries, and Zn, Cu, Pb and Cd contents are usually 2-7 times higher than the world average contents of the metals in natural soil. Although pollution index and concentrations of Cr, Ni, Pb and Zn in the roadside sediments at industrial area were usually higher than those of downtown and residential area, the metal having small vehicle- and steel-related industries had high concentrations of metals. The results of chemical partitioning analysis showed that Pb, Zn and Mn are mainly associated with carbonate/adsorbed and Fe-Mn oxide phases but that Cu is largely associated with the organic and sulfide fractions. It thus indicates that both large and small (vehicle- and steel-related) industries are main sources of heavy metal contamination. Due to high solubility of the carbonate phases by natural leaching episodes, the carbonate/adsorbed Cd, Co, Ni, Pb, Zn and Mn in the roadside sediments may serve as a potential source of contamination.
This study examined the changes of microstructural compositions in cement matrix according to the depth from the surface of a reinforced concrete (RC) column exposed to fire. The RC column was exposed to a standard fire for 180 minutes. After the fire test, core samples passing through the column section were obtained. Using the core samples, the remaining fractions of calcium-silicate-hydrates (C-S-H) and calcium hydroxide in cement matrix at the surface, the depth of 40 mm and 80 mm and the center (175 mm) were examined using thermal gravimetric analysis (TGA) and X-ray diffraction analysis (XRDA). Using nuclear magnetic resonance (NMR) technique, the silicate polymerization of C-S-H in cement matrix was also evaluated. The experimental results indicated that the amount of C-S-H loss at the center of column experiencing the transferred fire temperature of $236^{\circ}C$ has been underestimated as the TGA results showed the highest C-S-H contents are located at the depth of 80 mm, where the transferred fire temperature is $419^{\circ}C$. Moreover, the destruction of silicate connections at the center was observed as similar as that at the depth of 40 mm, where the transferred fire temperature was $618^{\circ}C$. This might be attributed to the temperature changes during cooling time after the fire test was neglected. Due to the relatively low thermal conductivity of concrete, the high temperature, which can affect the change of microstructure in cements, will hold longer at the center of the column than other depth.
Effects of different levels of molasses in a concentrate supplement on milk yield of cows grazing Setaria grass pastures were investigated. Thirty Friesian cows, 6-7 years old with mean pre-experimental body weight of $428{\pm}6.5$ kg, in early stage of lactation were randomly allotted to five dietary treatments in a completely randomized design experiment that lasted for 126 days. Experimental dietary treatments were forage alone and forage/concentrate mixtures with molasses included at 0, 5, 10 or 15% levels designated as $T_1$, $T_2$, $T_3$, $T_4$ and $T_5$, respectively. The parameters studied were voluntary dry matter (DM) intake, average daily live weight change (LWC), milk yield, body condition score (BCS) and apparent nutrient digestibility coefficients. The DM and energy contents of Setaria grass were low compared to the concentrate diets. Fibre fractions-NDF, ADF, ADL, hemicellulose and cellulose; and gross energy were higher in concentrate mixtures than in the forage. Total DM intake (forage+concentrate mixtures) was significantly higher (p<0.001) in cows on the concentrate mixtures. LWC was not significantly different (p>0.05) between the cows. Average milk yields were significantly different (p<0.05) between cows. Fat corrected milk (FCM) was similar among cows in the treatments. BCS was better (p<0.001) in cows on concentrate mixtures. Digestibilities of DM, CP, NDF, ADF, ADL, OM, and energy were significantly higher (p<0.001) in cows on $T_2$, $T_3$, $T_4$ and $T_5$ than in those on $T_1$. There were no significant differences in the digestibility of DM, CP, NDF, ADF and ADL (p>0.001) in cows on concentrate mixtures. This study therefore demonstrated that lactating dairy cows in Fiji need a level of readily fermented energy source such as molasses in their diets; however, a level above 10% is not nutritionally suitable for lactating dairy cows. Based on data on production parameters-milk yield, fat corrected milk, body condition score and apparent nutrient digestibility coefficients-molasses levels that range between 5-10% are recommended, however, 10% is the best and therefore recommended for inclusion in the concentrate mixture of lactating dairy cows on a basal diet of Setaria sphacelata in Fiji.

Introduction: A new third version of the Copenhagen Psychosocial Questionnaire (COPSOQ III) has been developed in response to trends in working life, theoretical concepts, and international experience. A key component of the COPSOQ III is a defined set of mandatory core items to be included in national short, middle, and long versions of the questionnaire. The aim of the present article is to present and test the reliability of the new international middle version of the COPSOQ III. Methods: The questionnaire was tested among 23,361 employees during 2016-2017 in Canada, Spain, France, Germany, Sweden, and Turkey. A total of 26 dimensions (measured through scales or single items) of the middle version and two from the long version were tested. Psychometric properties of the dimensions were assessed regarding reliability (Cronbach α), ceiling and floor effects (fractions with extreme answers), and distinctiveness (correlations with other dimensions). Results: Most international middle dimensions had satisfactory reliability in most countries, though some ceiling and floor effects were present. Dimensions with missing values were rare. Most dimensions had low to medium intercorrelations. Conclusions: The COPSOQ III offers reliable and distinct measures of a wide range of psychosocial dimensions of modern working life in different countries; although a few measures could be improved. Future testing should focus on validation of the COPSOQ items and dimensions using both qualitative and quantitative approaches. Such investigations would enhance the basis for recommendations using the COPSOQ III.
A tomato (Lycopersicon esculentum cv. Naomi F1) juice was separated into ascorbate and phenolic portions using a Sep-pak $C_{18}$ cartridge, and its each portion was tested for inhibition of N-nitrosodimethylamine (NDMA) formation. Ascorbate and phenolic portions of tomato juice inhibited NDMA formation by $81.37{\pm}0.25%$ and $72.03{\pm}0.25%$, respectively. The phenolic portion was further fractionated by prep-HPLC and inhibitory effects of NDMA formation by 4 fractions $(1{\sim}4)$ from tomato juice was tested under the different pH conditions (pH 1.2 and 4.2). Fraction 2 inhibited NDMA formation by $50.10{\pm}0.46%$ (pH 1.2) and $64.30{\pm}0.20%$ (pH 4.2), respectively. Fraction 2 was further separated into 4 subfractions $(2a{\sim}2d)$. Subfraction 2b especially inhibited NDMA formation by $70.62{\pm}0.45%$ (pH 1.2) and $75.30{\pm}0.45%$ (pH 4.2). This subfraction was confirmed o-coumaric acid through the analysis of GC-Mass spectrum, $^1H-NMR$ and $^{13}C-NMR$.
