The present study was conducted to evaluate the usefulness of common Korean legumes as a high-fiber supplement in therapeutic diets for diabetic patients. Streptozotocin-induced diabetic rats were used as animal models and four kinds of legumes, black soybean (BS), yellow soybean (YS), green pea (GP) and soybean curd residue (SCR) were tested as high-fiber supplements. Seven groups of normal and streptozotocin-induced diabetic rats were fed isocaloric experimental diets containing 8% dietary fiber from one of four legumes or purified cellulose and pectin for 6 weeks. The effects of the legumes on the glucose and lipid metabolism of diabetic rats was examined and compared with the effects of cellulose and pectin. The legume supplementation did not show any beneficial effect on glucose tolerance, however, it exhibited a plasma cholesterol-lowering effect in diabetic rats. The cholesterol-lowering action was especially strong in BS and the degree of the effect was comparable to that of pectin. The levels of total lipids, cholesterol, and triglyceride in the hepatic tissues of rats fed legume diets were similar to those of the pectin group. All legume supplements induced an increase in fecal steroid excretion. The fecal cholesterol contents were significantly high following the supplementations of YS and SCR (p < 0.05). The excretion of fecal bile acids in the BS and YS groups was significantly higher than it was in the pectin group (p < 0.05). Concentration of lipid peroxidation products in the blood and urine of diabetic animals was lower in the legume groups than in the cellulose group. The levels of hepatic lipid peroxidation products were significantly lower in the BS and YS groups than in the pectin group (p < 0.05). From the results of this study, the plasma cholesterol-lowering effect of BS is possibly due to the significant (p < 0.05) in-crease in fecal steroid excretion, which suggests that BS could be beneficial in improving abnormal lipid metabolism in diabetic rats. The present study was conducted to evaluate the usefulness of four kinds of legumes(black soybean: BS, yellow soybean: YS, green peas: GP, soybean curd residue: SCR) as a high-fiber supplement in the therapeutic diet for diabetic patients. Seven groups of normal and streptozotocin-induced diabetic rats were fed isocaloric experimental diets containing 8% dietary fiber from one of legumes for 6 weeks. The effects of legumes on the gastrointestinal function and diabetic symptoms in diabetic rats were also examined and compared with the effects of cellulose or pectin diet. Legume supplementations had no significant effects on alleviating typical diabetic symptoms such as polydipsia, polyphasia, polyuria, and urinary glucose excretion when compared with cellulose or pectin supplementations. The supplementations of legumes all resulted in remarkable changes in gastrointestinal functions: shortening of GI transit time and increases in fecal volume, fecal water and crude fat contents. Legume supplementations appeared to supress the damages of the small intestine villi and the enteritis symptoms in diabetic rats. GP led to regular villus patterns closer to normal villus. BS, YS and GP led to the enlargement of cecum in diabetic rats. In conclusion, the present studies have demonstrated that legumes may possess a significant physiological actions in the control of various diabetic conditions.
In vitro experiments were conducted to evaluate the suitability of several mixtures of high tanniniferous non legumes with low tanniniferous legumes on in vitro gas production (IVGP), dry matter degradation, Ammonia-N, methane production and microbial population. Eight treatments were examined in a randomized complete block design using four non-legumes and two legumes (Carallia integerrima${\times}$Leucaena leucocephala (LL) (Trt 1), C. integerrima${\times}$Gliricidia sepium (GS) (Trt 2), Aporosa lindeliyana${\times}$LL (Trt 3), A. lindeliyana${\times}$GS (Trt 4), Ceiba perntandra${\times}$LL (Trt 5), C. perntandra${\times}$GS (Trt 6), Artocarpus heterophyllus${\times}$LL (Trt 7), A. heterophyllus${\times}$GS (Trt 8). The condensed tannin (CT) content of non legumes ranged from 6.2% (Carallia integerrima) to 4.9% (Ceiba perntandra) while the CT of legumes were 1.58% (Leucaena leucocephala) and 0.78% (Gliricidia sepium). Forage mixtures contained more than 14% of crude protein (CP) while the CT content ranged from 2.8% to 4.0% respectively. Differences (p<0.05) were observed in in vitro gas production (IGVP) within treatments over a 48 h period dominated by C. perntandra${\times}$G. sepium (Trt 6). The net gas production (p<0.05) was also high with Trt6 followed by A. heterophyllus${\times}$L. leucocephala (Trt 7) and A. heterophyllus${\times}$G. sepium (Trt 8). Highest (p>0.05) NH3-N (ml/200 mg DM) production was observed with the A. heterophyllus${\times}$G. sepium (Trt 8) mixture which may be attributed with it's highest CP content. The correlation between IVGP and CT was 0.675 while IVGP and CP was 0.610. In vitro dry matter degradation (IVDMD) was highest in Trt 8 as well. Methane production ranged from 2.57 to 4.79 (ml/200 mg DM) to be synonimous with IVGP. A higher bacteria population (p<0.05) was found in C. perntandra${\times}$G. sepium (Trt 6) followed by Artocarpus heterophyllus+G. sepium (Trt 8) and the same trend was observed with the protozoa population as well. The results show that supplementing high tannin non leguminous forages by incremental substitution of legume forage increased gas production parameters, NH3-N, IVDMD and microbial population in the fermentation liquid. Methane production was not significantly affected by the presence of CT or different levels of CP in forage mixtures. Among non legumes, Ceiba perntandra and Artocarpus heterophyllus performed better in mixture with L. leucocephala and G. sepium.
Root-nodule nitrogen-fixing bacteria are known for being specific to particular legumes. This study isolated the endophytic root-nodule bacteria from the nodules of legumes and examined them to determine whether they could be used to promote the formation of nodules in other legumes. Forty-six isolates were collected from five leguminous plants and screened for housekeeping (16S rRNA), nitrogen fixation (nifH), and nodulation (nodC) genes. Based on the 16S rRNA gene sequencing and phylogenetic analysis, the bacterial isolates WC15, WC16, WC24, and GM5 were identified as Rhizobium, Sphingomonas, Methylobacterium, and Bradyrhizobium, respectively. The four isolates were found to have the nifH gene, and the study confirmed that one isolate (GM5) had both the nifH and nodC genes. The Salkowski method was used to measure the isolated bacteria for their capacity to produce phytohormone indole acetic acid (IAA). Additional experiments were performed to examine the effect of the isolated bacteria on root morphology and nodulation. Among the four tested isolates, both WC24 and GM5 induced nodulation in Glycine max. The gene expression studies revealed that GM5 had a higher expression of the nifH gene. The existence and expression of the nitrogen-fixing genes implied that the tested strain had the ability to fix the atmospheric nitrogen. These findings demonstrated that a nitrogen-fixing bacterium, Methylobacterium (WC24), isolated from a Trifolium repens, induced the formation of root nodules in non-host leguminous plants (Glycine max). This suggested the potential application of these rhizobia as biofertilizer. Further studies are required to verify the N2-fixing efficiency of the isolates.
Intercropping cereals with legumes is known to improve forage production and crude protein yield. Sorghum × sudangrass hybrids (SSH) have excellent dry matter content and high cultivation temperatures. In this study, we investigated the growth characteristics, forage productivity, and feed value of intercropping SSH with different legumes in rice paddy fields. We used five treatments in this study SSH monocropping and four intercropping treatments of SSH with, lablab, cowpea, sesbania, and two cultivars of soybean (Chookdu 1 and 2). SSH plant height was not significantly different between the monocropping and intercropping treatments. However, the plant heights of lablab, cowpea, and sesbania were significantly higher than those of the two soybean cultivars. The total dry matter yield (kg/ha) was significantly higher in SSH monocropping than in intercropping; among the intercropping treatments, the one with SSH and Chookdu 2 yielded the highest total dry matter yield. The SSH feed value was significantly different between the monocropping and intercropping treatments, although there were no differences between the intercropping treatments. Among the intercropped legumes, lablab showed the highest neutral detergent fiber and acid detergent fiber contents, and cowpea had the highest crude protein content. These results reveal that intercropping SSH with legumes in paddy fields could be a promising cultivation technique to maintain stable forage productivity.
Plant characteristics of four winter crop hairy vetch, red clover, crimson clover and rye as green manure were evaluated at three harvesting time(Apri1 23, May 9, May 28) in spring to select superior winter green manure crop. Rye and hairy vetch had superior wintering ability and produced high dry matter compared with crimson clover and red clover. But N amount of rye was small because of its low plant N% despite of early dry matter accumulation. Wintering ability of crimson clover was very poor, so its dry matter decreased rapidly with low seeding rate. Hairy vetch had high N % (4%, C:N ratio 11) and produced high N yield among four plant. It is estimated that hairy vetch have high N-fixing ability because its N% was not changed largely despite of dry matter accumulation at late harvesting time contrast to the other legume. It was thought that hairy vetch was superior green manure crop in considering yield of dry matter and N. (Key words : Winter legumes, Hairy vetch, Green manure yield, Nitrogen concentration)
본 연구는 전남농업기술원 답리작 시험포장에서 2006년과 2007년 사이에 보리와 이탈리안 라이그래스와 각종 두과를 혼파비율을 달리한 처리하여 파종하였다. 주구는 보리 및 이탈리안 라이그래스와 자운영, 크림손클로버, 헤어리벳치, 사료용 완두였고 세구는 화본과와 두과를 각각 50:50, 60:40, 70:30 그리고 80:20이었다. 공중질소 고정량 및 이의 이동을 측정하기 위하여 질소동위원소($^{15}N(NH_4)_2SO_4$)를 2007년 4월 15일 처리 후 쿼드랏 안의 식물체를 6월 8일에 수확하여 화본과 및 두과로 분리한 후 안정성동위원소 질량분석기를 이용하여 분석하였다. 공중질소 고정률은 두과에서 화본과로, 질소이동은 차이법과 동위원소 희석법을 이용하여 구하였다. 본 실험결과의 주요내용은 동위원소 초과는 화본과에서는 차가 없었던 반면 보리와 두과와 혼파비율이 60:40인 처리에서 유의적인 차이가 있었다. 그 값은 보리와 혼파한 두과는 0에서 0.54 범위였다. 이탈리안 라이그래스와 두과 혼파에서는 초과 값이 더 높아 보리에서는 0.71에서 0.82, 두과는 0.37에서 1.01 사이였다. 공중 질소 이용비율은 보리와의 조합은 0%~49.5%, 이탈리안 라이그라스 조합은 0~60.5%의 범위로 다양한 결과를 나타내었다. 차이법에 의한 이동량 시험에서는 보리와의 조합에서 12.3~90.9 kg/ha, 이탈리안 라이그래스 조합에서는 보리보다 더 많아 31.7~107.8 kg 사이였다. 한편 동위원소 희석법에서는 보리와 두과 혼파조합은 0~36.1 kg/ha 그리고 이탈리안 라이그래스와 두과 혼파에서는 0~50.6 kg/ha로 이탈리안 라이그래스 조합이 더 많은 것으로 나타났다. 차이법에 의한 질소 이동량이 동위원소 희석법보다 더 많은 것으로 나타났다.
Anthracnose fungi associated with soybean (Glycine max), small red bean (Vigna angularis), and green bean (Phaseolus radiatus var. aurea) were compared with respect to cultural characteristics, morphological aspects, and pathogenicity. From the legumes, four different species of Colletotrichum were identified based on their morphological characters : C. truncatum, C. destructivum, C. trifolii and C. gloeosporioides. Colonies of Colletotrichum spp.on culture media varied in colour and growth habits depending upon the media and isolates. In general, formation of conidia, setae, and perithecia on water agar leaf medium was better than that on potato dextrose agar medium under near ultraviolet light. In cross inoculation tests, isolates of C. truncatum, c. destructivum, C. trifolii and C. gloeosporioides could infect soybean, small red bean, and green bean at different growth stages. There were some differences in pathogenicity or susceptibility depending on crops tested. C. truncatum was the most virulent and major pathogenic species among the four species.
Changes in contents of free sugars, amino acids, and fatty acids of legumes were analyzed for each phase of in vitro digestion. In addition, contents of resistant starch in raw and digested pulses were compared. Soybeans, kidney beans, cowpeas, and chickpeas were analyzed. An in vitro digestion model was used to analyze contents of nutrients using LC-MS and GC-MS. Stachyose in kidneybean, cowpea, and chickpea increased as the digestion phase progressed. In four types of legumes, raffinose slightly decreased or showed no significant difference between the Oral phase and the BBMV phase. Content of glucose, a monosaccharide, increased during the BBMV phase. During the digestion phase, levels of free amino acids and free fatty acids also increased. Content of resistant starch was reduced compared to that in the raw material. It was 0.01g/100 g food in soybean, 1.06 g/100 g food in red kidney bean, 0.77g/ 100g food in cowpea, and 0.76 g/100 g food in chickpea. It was confirmed that nutrients in the in vitro digestion model were liberated at each digestion phase with changes in the content of resistant starch. These results are expected to be used as fundamental data for obtaining bioavailability of nutrients.
