반응표면분석법을 이용한 쌀귀리 단백질의 알칼리 추출 공정 최적화 (Optimization of Alkali Extraction for Preparing Oat Protein Concentrates from Oat Groat by Response Surface Methodology)

  • 알칼리 추출법을 이용하여 쌀귀리로부터 단백질을 추출하고 추출수율을 최적화하기 위한 알칼리 농도 조건 및 침전 pH 조건을 반응표면분석을 통해 검토하였다. 추출수율은 추출용매인 알칼리 농도가 증가할수록 그리고 침전 pH 4.9 부근에서 높은 경향을 보였으며, 알칼리 농도가 침전 pH에 비하여 보다 큰 영향을 미치는 것으로 나타났다. 알칼리 추출 공정으로 얻어진 귀리 단백질 농축물의 질소용해지수는 등전점 부근인 pH 5에서 최소값을 나타내었고, pH 3 이하, pH 7 이상에서는 급격히 증가하였다. 알칼리 추출 공정의 최적화를 위한 회귀분석 결과, 알칼리 농도 0.06 N, 침전 pH 4.7에서 최대 수율 85.89%로 예측되었으며 예측모델식과 실제 측정값을 비교하였을 때 유의적인 차이가 크게 나타나지 않아 설계된 실험모델식은 적합한 것으로 나타났다. 따라서 기존의 연구들은 알칼리 추출법을 이용한 귀리 단백질 추출 시 알칼리 농도와 침전 pH가 각기 다름을 보였지만, 본 연구를 통하여 귀리 단백질 농축물 제조를 위한 알칼리 추출 공정의 최적 조건은 알칼리 농도 0.06 N, 침전 pH 4.7인 것으로 확인되었다.

  • This study concentrates on a new idea that is formulated as a creative space recognition in contemporary architecture and painting. It is stimulated from Deleuze's philosophy, which is expressed with the philosophical instrument, "Non-Representation". After discussing the concept of non-representation used in Deleuze's philosophy, case example in contemporary architecture are analysed to comparatively Investigate the relationship between the architectures by Rem Koolhaas and painting by Francis Bacon. Therefore, he founds expressions of "diagram" and "movement" in painting of F. Bacon. Diagram meansit is a power that is comprised of various points of wiew with the kind experiences that one has in life. Movement that were extracted form the Instrument in space recognition of Deleuze's panting theory were of topology, insertion of geometry, reiteration of layers, transparency and translucency, reflection effect.

Nutrient analysis and in vitro rumen fermentation of commercial formulated concentrates for finishing Hanwoo steers

  • The objective of this study was to evaluate the nutritional value of commercial compound feeds for late finishing Hanwoo steers using detailed chemical analysis and an in vitro rumen fermentation trial. A total of 4 different feeds were selected and used to conduct a chemical analysis for their nutrient contents. The largest variation in nutrients contents among experimental feeds was found in ether extract and the smallest one was found in total digestible nutrients. Commercial feeds C and D had a higher energy value than the others. Even if C and D had a similar feed energy value, the components used to increase energy differed between them (non-fiber carbohydrate [NFC] for C; ether extract for D). In the in vitro trial, no significant difference was observed in dry matter in vitro digestibility and gas production between treatments. However, the highest ammonia concentration (p < 0.05) was observed in C and D feeds. The low acetate to propionate ratio observed in C feeds (p < 0.01) suggested that this feed had high starch based carbohydrates that NFC degrading bacteria used to produce more propionate. It is important to provide nutritional information to farmers so that they can select the appropriate commercial feeds to suit their own feeding strategies. This study might give supporting information to farmers for a more educated, and better, selection of feeds. Further in vivo studies should be conducted to evaluate the effects of different commercial feeds on growth performances in late finishing Hanwoo steers.

EFFECTS OF VARYING DIETARY LEVELS OF TOTAL DIGESTIBLE NUTRIENTS, PROTEIN AND FIBER ON THE GROWTH OF CROSSBRED HOLSTEIN HEIFERS FED UREA-TREATED RICE STRAW DIETS UNDER TWO FEEDING SYSTEMS

  • This experiment was carried out to examine the effects of urea-treated rice straw feeding on the growth performance of crossbred Holstein heifers under different feeding conditions. In the first experiment, the animals were given diets having 2 levels of TDN and CP and 3 levels of crude fiber (22, 30 and 36%) which were formulated with urea-treated rice straw and concentrates. Daily weight gain of heifers was not different between 22% and 30% CF diets, but the reduction of TDN or CP level to 90% of the requirements decreased the weight gain. Fiber content of 36% also reduced the body weight gain. The reduction of TDN significantly reduced DM intake and increased feed conversion ratio. Feed cost per kg weight gain was significantly increased by an increase in CF to 36%. In the second experiment, separate feeding and total mixing feeding were compared. There were no significant differences between the two feeding systems in body weight gain although the possibility of superiority in SF to TMF remained. DM intake was not affected by the feeding system, but 30% CF diet gave higher DM intake. Feed cost per kg weight gain was lower in the 30% CF diet.

Seismic behavior of frames with innovative energy dissipation systems (FUSEIS 1-1)

  • After strong earthquakes conventional frames used worldwide in multi - story steel buildings (e.g. moment resisting frames) are not well positioned according to reparability. Two innovative systems for seismic resistant steel frames incorporated with dissipative fuses were developed within the European Research Program "FUSEIS" (Vayas et al. 2013). The first, FUSEIS1, resembles a vertical Vierendeel beam and is composed of two closely spaced strong columns rigidly connected to multiple beams. In the second system, FUSEIS2, a discontinuity is introduced in the composite beams of a moment resisting frame and the dissipative devices are steel plates connecting the two parts. The FUSEIS system is able to dissipate energy by means of inelastic deformations in the fuses and combines ductility and architectural transparency with stiffness. In case of strong earthquakes damage concentrates only in the fuses which behave as self-centering systems able to return the structure to its initial undeformed shape. Repair work after such an event is limited only to replacing the fuses. Experimental and numerical investigations were performed to study the response of the fuses system. Code relevant design rules for the seismic design of frames with dissipative FUSEIS and practical recommendations on the selection of the appropriate fuses as a function of the most important parameters and member verifications have been formulated and are included in a Design Guide. This article presents the design and performance of building frames with FUSEIS 1-1 based on models calibrated on the experimental results.

길초근(Valeriana fauriei Briquet) 및 양강근(Alpinia galangal Swartz) 정유의 유제 및 입제제형의 잉어(Cyprinus carpio)에 대한 급성독성 (Acute Toxicities of Emulsifiable Concentrates and Granules of Valeriana fauriei Briquet and Alpinia galangal Swartz Essential Oils against Cyprinus carpio)

  • 길초근 및 양강근정유를 유제 및 입제로 제형화한 후 이들의 급성어독성을 측정하였다. 잉어를 이용한 급성독성에 사용된 길초근 및 양강근 정유는 수증기증류법, 핵산을 이용한 용매추출법 및 초임계추출법을 이용하여 추출하였고 이들의 유제 및 입제의 급성어독성은 반수치사농도($LC_{50}$)를 구하여 평가하였다. 길초근 및 양강근 원제 중 용매추출법으로 추출한 정유만이 어독성을 나타내었다. 길초근 및 양강근 추출법별 정유 이용한 유제의 급성어독성 평가한 결과 길초근의 경우 모든 추출법 대하여 독성을 나타내지 않은 반면에 양강근 정유는 추출법에 관계없이 모두 잉어에 대한 어독성이 관측되었다. 입제의 경우 길초근은 용매로 추출한 정유에서 독성이 나타났고, 양강근 정유의 경우 초임계추출법으로 추출한 정유 입제에서만 어독성이 관측되었다. 이러한 결과는 3급 어독성에 해당되는 것으로 환경 중 사용에는 문제가 없다고 사료된다. 살충효과가 뛰어난 양강근 정유의 경우 수증기증류법 및 초임계추출법으로 추출한 정유의 $10mg\;L^{-1}$ 수준에서 노출된 잉어의 분자적 반응을 확인하여 cytochrome P450 1A 효소 및 glutathione S-transferase의 발현이 급격히 증가하였음을 확인하였고 이들은 수계환경 중 양강근 정유의 동태를 관찰하는 바이오마커로서 사용 가능하다고 사료된다.

Effect of Replacing Corn and Wheat Bran With Soyhulls in Lactation Cow Diets on In Situ Digestion Characteristics of Dietary Dry Matter and Fiber and Lactation Performance

  • An in situ digestion trial (Experiment 1) and a lactation trial (Experiment 2) were conducted to determine the effects of replacing corn and wheat bran with soyhulls (SH) in lactating dairy cow diets on the extent and kinetics of digestion of DM and NDF, and lactation performance. In experiment 1, five mixed feeds consisting of mixed concentrate and roughages (50:50 on a DM basis) were formulated on isonitrogenous and isoenergetic bases to produce five levels (0, 25, 50, 75 and 100%) of SH replacement for corn and wheat bran. SH had high in situ digestion (92 and 89% for potentially digestible DM and NDF) and fairly fast digestion rate (7.2 and 6.3 %/h for DM and NDF). Increasing level of SH replacement resulted in increased NDF digestibility (linear, p=0.001-0.04) and similar DM digestibility (beyond 12 h incubation, p=0.10-0.41). As level of SH replacement increased, percentage of slowly digestible fraction (b) of DM increased (linear, p=0.03), percentage of rapidly digestible fraction (a) of DM tended to decrease (linear, p=0.14), and DM digestion lag time tended to be longer (linear, p=0.13). Percentage of potentially digestible fraction (a+b) and digestion rate (c) of slowly digestible fraction of dietary DM remained unaltered (p=0.36-0.90) with increasing SH in the diet. Increasing level of SH for replacing corn and wheat bran in the diet resulted in increases in percentages of b (quadratic, p<0.001), a (linear, p=0.08), a+b (quadratic, p=0.001) and a tendency to increase in c for NDF (linear, p<0.19). It was also observed that there was a satisfactory fit of a non-linear regression model to NDF digestion data ($R^2=0.986-0.998$), but a relatively poor fit of the model to DM digestion data ($R^2=0.915-0.968$). In experiment 2, 42 lactating Holstein cows were used in a randomized complete block design. SH replaced corn and wheat bran in mixed concentrates at 0, 25, and 50%, respectively. These mixed concentrates were mixed with roughages and fed ad libitum as complete diets. Replacing corn and wheat bran with SH at 0, 25 and 50% levels did not influence (p=0.56-0.95) DM intakes (18.4, 18.6, and 18.5 kg/d), milk yields (27.7, 28.4 and 27.6 kg/d), 4% fat-corrected-milk (FCM) yields (26.2, 27.6, and 27.3 kg/d) and percentages of milk protein (3.12, 3.17 and 3.18%), milk lactose (4.69, 4.76 and 4.68%) and SNF (8.50, 8.64, and 8.54%). On the other hand, milk fat percentges linearly increased (3.63, 3.85 and 3.90% for SH replacement rates of 0, 25 and 50% in the diet, p=0.08), while feed costs per kg FCM production were reduced.

철근 및 프리스트레스트 콘크리트 구조물의 비선형 유한요소 해석 (Nonlinear Finite Element Analysis of Reinforced and Prestressed Concrete Structures)

  • 본 논문은 콘크리트 구조물의 재료 비선형과 시간의존적 거동을 고려한 유한요소 해석에 관한 것으로 회전균열모델에 토대를 둔 면내균열모델을 사용하여 하중에 따른 균열의 영향을 모사하였으며 콘크리트는 직교이방성 거동을 한다고 가정하였다. 특히 휨거동 시의 인장보강효과(tension stiffening effect)를 고려하기 위해 파괴에너지 개념에 토대를 둔 한계식을 제안하였고 이를 통해 수치해석시 나타나는 유한요소의 크기에 따른 수치해석 오차의 최소화를 도모하였다. 또한 embedded model을 사용하여 철근의 거동을 모사할 경우 콘크리트의 변위장에 따라 효율적으로 대처할 수 있도록 구성방정식을 체계화 하였으며 비균열단면, 균열단면 등 여러 상황에서 구조물의 시간에 따른 거동을 살펴보기 위해 평면응력 상태에서 재령보정 탄성계수법에 따른 구성방정식을 유도하고 이를 토대로 예제해석을 수행하였다.

현대 건축과 회화에서 힘의 자유로운 분산에 관한 연구 - 프랭크 게리의 건축과 프란시스 베이컨의 회화에 있어 바로크적인 것을 중심으로 - (A Study on the free dispersal of the Power in Contemporary Architecture and Painting - Focused on the baroque in the Architecture of Frank O. Gehry and the Paintings of Francis Bacon -)

  • This study concentrates on a new idea that is formulated as a creative spatialization in contemporary architecture and painting. It is stimulated from the classical baroque, which is expressed with the conceptual instruments, 'excessive', 'decorative', 'accidential' Suggested from that, this study tries to open the actual sense area of 'the baroque' by analysing the architectural ideas of wrenching and folding by Frank O. Gehry and in the same context by elucidating the dynamic deformalising of the figures by Francis Bacon. The redefined 'new baroque' operates in the chain of the transformable and flexible images that must be recognized as 'the real' in the sense of 'the most empirical' and are vulnerable to dispersal. The typical instrument of Gehry's 'new baroque architecture' is the enclosing of the space and its folding, and Bacon, whose painting can be called as the modern baroque, takes as the expressing methods a wrenching of the figures, a relieving of the colored background against the figure and a disappearing of figures into the background. These different ideas meet in the theoretical gravitation field of the philosophy of Deleuze, in which the reconstituted baroque conception means the tension between the folds, on expansion and contraction of which the variable space is dependent, Through the general principle of 'the new baroque', a free dispersal of power, to architect and painter is offered the possibility, not only to make the real world intimate to the real life, but also to produce from there a vivid new space. Finally it brings to enjoyer and viewer the possibility, to perceive the operating power in and around oneself.

프랭크 게리의 건축에 있어 바로크적인 것 (On the Baroque in the Architecture of Frank O. Gehry-in relation to the baroque concept of Deleuze)

  • 본 연구는 현대건축에서 창조적인 공간화로 요약될 수 있는 새로운 개념에 대한 해명을 목표로 한다. 고전적 바로크 개념의 기능가치를 기반으로 하여 본 연구는 현대 건축가인 프랭크 게리에 의해 시도된 주름과 비틀기 건축을 분석함으로써, '바로크적인 것'의 현재적인 의미를 드러내고자 한다. 새로이 정의된 '바로크적인 것'은 가장 경험적인 것이라는 의미에서 실재적인 것으로 인식되어야 하며 또한 분산적인 특징을 지니는 변형적이고 탄력적인 이미지들의 사슬 속에서 작용한다. 게리의 신 바로크 건축의 전형적인 도구는 공간을 에워싸고 주름잡는 것이다. 이 같은 건축이념은 들뢰즈의 철학적인 바로크 개념에 맞닿아 있다. 들뢰즈의 바로크 개념은 주름들 간의 긴장관계를 의미하는 것으로, 공간의 변화는 이러한 긴장관계의 수축과 확장에 의존해있는 것이다. 힘의 자유로운 분산이라는 신 바로크 개념의 일반원리를 통해 건축가에게는 실재세계를 현실적인 삶과 가장 밀접하게 만들 가능성이 제공되며, 향유자와 관찰자에게는 자기 내부와 자기 주위에서 작용하는 생동적인 힘을 감지할 가능성이 부여되는 것이다.
