  • Because people who take more than two drugs have increases, a simple and sensitive method for the simultaneous analysis of amphetamine, methamphetamine and nalbuphine in urine was developed. After alkalinization of the urine samples with 6 N-NaOH, the analytes were extracted using ethyl acetate, derivatized with MSTFA : TSIM : TMCS (= 100 : 2 : 5) prior to gas chromatography-mass spectrometry(GC-MS) analysis with selected ion monitoring. Sensitivity study of the Yfiler® PLUS PCR Amplification Kit in forensic casework samples (법과학 현장시료에서 Yfiler® PLUS PCR Amplification Kit의 민감도 연구)

  • A variety of Y-STR analysis kits have been developed and used in the forensic field. Prior to the forensic application of a new kit, laboratory validation and sensitivity tests are essential processes in selecting suitable alternatives and for assuring that standard operating procedures are followed. In this paper, we have performed a sensitivity study of a new commercial kit, the Yfiler® PLUS PCR Amplification Kit (Yfiler plus kit, released in 2014) by comparing it with the AmpF/STR® YfilerTM PCR Amplification Kit (Yfiler kit, released in 2004). The Yfiler plus kit includes the 17 Y-STR loci of the Yfiler kit and has been supplemented with 10 new Y-STR loci. First, we analyzed the sensitivity difference between the two kits using commercial control DNA 2800M and 007. In addition, we compared the detection rate between the two kits from the 16 selected forensic casework samples of less than 0.5 ng concentrations. The results show that the sensitivity and detection rate of the Yfiler plus kit are higher than the corresponding rates of the Yfiler kit. In addition, we were able to obtain more Y-STR profiles with the use of the new kit. Thus, we suggest that Yfiler plus kit is a more effective forensic tool to detect Y-STR profiles from forensic casework samples of low concentrations.

  • Smart Phone의 OS로 많이 사용하는 Android/Windows Mobile Smart Phone의 사용이 급격히 증가하고 있다. 무료 WiFi Zone과 인터넷 사용에 대한 취약점이 존재한다. Mobile Forensic의 증거 자료를 추출하는 방법은 SYN, JTAG, Revolving 방법이 있지만, 기존 휴대폰과 달리 Smart Phone은 OS와 구조, 사용방식과 기술의 차이로 인한 Mobile Forensic 연구 방법도 달라야 한다. 본 논문에서는 Smart Phone에서는 많이 사용되는 Windows Mobile/Android Smart Phone의 OS와 구조 차이를 분석한 데이터 백업과 스펙 분석 및 증거자료 분석을 한다. 또한 무료 WiFi Zone을 통한 인터넷 사용시에 취약점을 분석한다. 그리고 Android/Windows Mobile Smart Phone의 Forensic 자료를 생성하여 증거를 추출하고, Mobile Forensic 보고서를 생성한다. 본 연구를 통하여 Mobile Forensic의 기술 발전에 초석을 제공할 것이다.

  • A small and inexpensive thermal cycler (PCR machine), known as the MiniPCRTM Mini8 Thermal Cycler (Amplyus, Cambridge, MA, USA), was developed. In this study, the performance of this PCR machine was compared with the GeneAmp® PCR system 9700 (Applied Biosystems) using four autosomal short tandem repeat (STR) kits, a Y-chromosome STR kit, and a mitochondrial DNA HV1/HV2 sequence analysis. The sensitivity and stochastic effects of the STR multiplex kits and the quality of the DNA sequence analysis were similar between the two PCR machines. The MiniPCRTM Mini8 Thermal Cycler could be used for analyses at forensic DNA laboratories and crime scenes. The cost of the PCR is so economical that school laboratories and individuals could use the machines.

Korean Quality Assessment Criteria for Statement Analysis Reports and Testimony (한국 진술분석 보고서 및 증언에 대한 질적 평가 기준)

  • Statement analysis is a technique that examines the credibility of a statement by scientifically analyzing problems and psychological characteristics that appear in the content of the statement. The statement analysis report is prepared, submitted, and used for legal judgments when there is a suspicion of sexual abuse for children(under 13 years of age) and persons with disabilities since it is usually difficult to secure physical evidence nor eyewitnesses. However, the criteria for evaluating the quality of a statement analysis report or testimony are not available in Korea. Although forensic experts and professional organizations in North America and Europe are providing recommendations and guidelines for preparing forensic assessment reports, qualitative analysis research studies for forensic reports revealed a number of problems such as missing or poorly described essential information and lack of logical connection between evaluation results and forensic opinions. Therefore, forensic evaluation guidelines and forensic reports submitted to the courts in the United States, as well as the Structured Quality assessment of eXpert testimony (SQX-12) developed in Sweden were examined to suggest the Korean version of quality evaluation criteria for statement analysis report and testimony. This criteria can be used to improve effectiveness of forensic reports within criminal justice system and used as a guideline to assess the quality forensic reports or expert testimony prepared by experts. However, this criteria do not guarantee the reliability of the statement itself.

Application study of PCR additives to improve the split peaks in direct PCR

  • Analysis techniques using DNA profiling are widely used in various fields including forensic science and new technologies such as the Direct PCR amplification method are being developed continuously in order to acquire the DNA profiles efficiently. However, it has a limits such as non-specific amplification according to the quality of crime scene evidence samples. Especially, split peaks caused by excessive DNA samples are one of the important factors that could cause the debate to allow researchers to interpret the DNA profile results. In this study, we confirmed the occurrence rate of split peaks in each STR (short tandem repeats) locus of the $GlobalFiler^{TM}$ kit and investigated the possibility of improving the split peaks using several PCR additives such as DMSO (dimethylsulfoxide), $MgCl_2$, Betaine and Tween-20. As a result, we could make three groups according to the occurrence rate of split peaks in Direct PCR and it was confirmed that the ratio of split peaks could be reduced by DMSO (87.4 %), $MgCl_2$ (84.5 %) and Betaine (86.1 %), respectively. These results indicate that PCR additives such as DMSO, $MgCl_2$ and Betaine can be improve the split peaks in Direct PCR and thereby facilitate subsequently a successful DNA profile results.

Reliability Evaluation of EDR Data Using PC-Crash & Vbox (Vbox와 PC-Crash를 활용한 EDR 기록정보의 신뢰성 평가)

  • The EDR(Event Data Recorder) is a part of the ACU(Airbag Control Unit) functions mounted on a vehicle. EDR data have pre-crash data and post-crash data. Pre-crash data are recorded within 5 sec from time zero(AE) with 0.5 sec resolution, and reveal vehicle speed, engine rotation speed, throttle opening, brake pedal operation, acceleration pedal position and steering angle, etc. Using this EDR data, the investigation of a traffic accident can become more objective and scientific. Crash tests of three vehicles equipped with EDR function had been performed successfully. Evaluation of EDR data reliability had also been performed using Vbox and PC-Crash's sequence table function. Based on the results, we could confirm EDR data's reliability and availability for Traffic Accident Analysis by the series of this process.

  • In the past, the domestic research on mobile forensics has been limited to cell phones. Increasing use of smart phones, studies on smart phone forensic will be conducted actively in the future. In particular, the study on Android forensic is very important because Android smart phone market share is increasing rapidly. In this paper, we describe an implementation of an Android smart phone forensic tool based on logical analysis. Compared with Oxygen Forensic Suite 2010, this tool saves time it takes to perform Android smart phone forensic because this tool provides search feature and resource links for extracted media information. So far, no smart phone forensic tool is introduced in Korea. Accordingly, this tool would contribute to the advancement of the technology on smart phone forensic.

  • The chemical element and structural characterization of different types of cements and its brick stones are been investigated under forensic aspects using X-ray florescence (XRF) and X-ray diffraction (XRD) spectrometer. The XRF provides rapid compositional data for controlling almost all stages of raw materials, clinker and cement. The decisive advantage of XRD methods is based on the unique character of the diffraction patterns of crystalline substances, the ability to distinguish between elements and their oxides, and the possibility to identify chemical compounds, polymeric forms, and mixed crystals by non-destructive examination. Therefore, combination of these examinations is useful and able to apply for the forensic analysis in comparison of cements and brick stones. There are more study remained to determine the viability of method for forensic analysis of brick stones and the limits of the discrimination that can be achieved.
