The Effects of Using Artificial Sweeteners and Coffee Grounds in Chocolate Filling on Quality Characteristics and Glycemic Index (쉘 초콜릿 filling에 사용한 대체 감미료와 커피박이 품질 특성 및 당지수에 미치는 영향)

  • Chocolate is restricted for obese and diabetic patients due to high in sugar contents. We investigated shell-chocolate fillings with low calorie content and low glycemic indexes using alternative sweeteners such as tagatose and xylose and adding coffee grounds. Chocolate filling was made with sugar, cacao and butter in the original recipe. Sugar was substituted with tagatose or xylose and 3% coffee ground was added in each chocolate filling and substituted for 3% of the cocoa. Water retention and spreadability of the chocolate filling was significantly higher in the tagatose+coffee ground group in comparison to the sugar. In the chromaticity test, chocolate filling made with tagatose was darker in comparison to that made with sugar: brightness (L) was lower and redness (a) and yellowness (b) were higher than the sugar filling. Sensory evaluation revealed that chocolate filling made with tagatose had high score in 4 categories such as taste, smell, texture of foreign substance, overall acceptance. After consuming 60 g of the chocolate with different fillings, blood glucose levels of subjects at 1 and 2 h were significantly lower in the tagatose and tagatose+coffee ground groups than the other groups. In conclusion, using tagatose as the sweetener in chocolate filling has a beneficial effect on lowering blood glucose levels and increasing water retention, spreadability and sensory scores. Coffee ground did not have additive effect on quality of chocolate filling and blood glucose levels. These results indicate that shell chocolate made with tagatose can be used as a snack for diabetic patients.

DISTRIBUTION AND PHYSIOLOGICAL CHARACTERISTICS OF BACILLUS CEREUS IN RICE AND RICE PRODUCTS (미반류에 있어서 Bacillus cereus균의 분포와 생리적특성에 관한 연구)

  • Recently, Bacillus has been identified as one of food poisoning bacteria especially in products of cereal foods in foreign countries. Therefore, the quantitative distribution of Bacillus cereus in market foods, its physiological characteristics, growth rate by temperature and heat resistance of its spore were examined. Thirty two samples of cooked rice, 20 samples of kimbab(cooked rice rolled with laver), 23 samples of rice cake, 13 samples of rice ana 13 samples of barley were collected from restaurents and food stores in Busan, Korea during the period from May to November in 1980. Forty samples of 101 samples submitted to the test appeared positive for Bacillus cereus showing abut $40\%$ in detection ratio. Detection ratio of Bacillus cereus was higher than $50\%$ in barley and rice, and about $30\%$ in rice products. Average Bacillus cereus content of in the samples was $2.6\times10^6/g$ in cooked rice, $2.3\times10^6/g$in kimbab, $4.9\times10^4/g$ in rice cake while that in rice and barley was about $10^3/g$. The result of biochemical tests of the bacterium was $100\%$ positive in catalase, egg yolk reaction, gelatin hydrolysis and glucose fermentation, $100\%$ negative in xylose, arabinose and mannitol oxidation, about $90\%$ positive in acetoin production, $80.0\%$ positive in nitrate reduction and citrate utilization and $55.0\%$ positive in starch hydrolysis test. Isolation ratio of Bacillus ceresus which showed haemolysis positive and starch hydrolysis negative results, was about $38\%$ in 40 strains examined. It is known that those strains has a close relation to food poisoning accident. Growth rate and generation time of Bacillus cereus isolated from the cooked rice were $0.34hr^{-1},\;2.02hr\;at\;20^{\circ}C,\;0.73hr^{-1},\;0.95hr\;at\;30^{\circ}C\;and\;0.49hr^{-1},\;1.44\;hr\;at\;40^{\circ}C$ respectively. Heat resistance value of Bacillus cereus spores suspended in phosphate buffer solution was $D_{90}=29.0min,\;D_{95}=8.7min,\;D_{98}=3.7\;min\;and\;D_{101}=2.3\;min(z=10.5)$.

  • The effective extraction methods and chemical components of crude polysaccharides of ascidian tunics were investigated. Tow extraction conditions, autoclaving or enzyme treatment, were applied. The proximate composition of ascidian tunics was not much different between those dried in raw (containing pigments) and those acetone treated and dried (decolorized), showing $50\%$ of carbohydrate and $40\%$ of protein. It was possible to extract up to $10\%$ of crude polysaccharides from ascidian tunics regardless of the extraction methods, autoclaving or enzyme treatment. In case of the latter the extraction yield by neutrase was higher than that with alkalase (Novo co.) or mixture 2000 (Pacific chemical co.). The most effective enzyme concentration and extraction time appeared to be 24 hrs of extraction with $3\%$ neutrase. On the other hand, in autoclave treatment, 6 hrs extraction showed most desirable extraction yield, about $9.7\%$. The compositions of amino acid of decolorized ascidian tunic (acetone treated group) and the crude polysaccharide from the autoclaving (water solubles) or neutrase treatment (enzyme digestibles) were similar to each other. Histidine was the highest both in the neutrase and autoclave treatment group and the yield were $29.2\%,\;20.4\%$, respectively, followed by aspartic acid and glutamic acid. Among the minerals, the content of Ca was significantly high, followed by Mg and Na.

Discrimination of Geographical Origin for Astragalus Root (Astragalus membranaceus) by Capillary Electrophoresis and Near-Infrared Spectroscopy (Capillary electrophoresis 및 근적외선분광분석기를 이용한 황기의 원산지 판별)

  • Capillary electrophoresis (CE) and near-infrared spectroscopy (NIRS) were performed to discriminate astragalus roots (Astragalus membranaceus) according to geographical origin (domestic or foreign). Two-hundred-and-four astragalus roots were extracted with 30% methanol in 0.1 M phosphate buffer (pH 2.5) and separated in a uncoated fused-silica $(50\;{\mu}m{\times}27\;cm)$ capillary. Conditions for optimal analysis included: temperature $-45^{\circ}C$, voltage -14 kV, and pressure injection time -8 sec. The optimal separation buffer was 0.1 M phosphate buffer (pH 2.5) containing 40 mM hexane sulfonic acid with 20% 2-methoxy ethanol. Raw NIR spectra were obtained using NIRS, and modified partial least square regression was used to develop the prediction model. The correlation coefficient and standard error of prediction were 0.915 and 14.3%, respectively. Under the optimal conditions established for CE and NIRS, the geographical origins of the astragalus roots were correctly identified in 80 and 97%, respectively. Astragalus roots that were not discriminated by NIRS were correctly discriminated by CE. Hence, CE and NIRS are potential methods for discriminating the geographical origins of astragalus roots that complement one another.

Effects of Green Pepper (Capsicum annuum var.) on Antioxidant Activity and Induction of Apoptosis in Human Breast Cancer Cell Lines (품종별 청고추의 항산화 효과 및 유방암 세포주에서의 세포 사멸 연구)

  • This study investigated flavonoid, total phenol, total flavonoid content, antioxidant and antiproliferative activity on human breast cancer cells (MCF-7, MDA-MB-231). Four varieties of Korean green peppers (KP: kkuri pepper, PP: phut pepper, CP: cheongyang pepper, OP: ohi pepper) and one foreign green pepper (JP: jalapeno) were used. The contents of luteolin, quercetin and apigenin, which are abundant flavonoids in green pepper, were the highest in KP. Also, the contents of total phenol, and total flavonoids were the highest in KP, followed by CP, JP, PP, and OP (KP: total phenol $13.29{\pm}0.45$ mg GAE/g D.W., total flavonoid $7.02{\pm}0.13$ mg QE/g D.W. In DPPH ABTS radical-scavenging activity, KP showed the most potent antioxidant activity. In the result of viability in human breast cancer cells, KP had the highest antiproliferative effect. These results suggest that green peppers have significant antioxidant activity and can be a possible candidate for treatment of breast cancer.

  • Biogasification is a technology that produces environmentally friendly fuel using methane gas generated in the process of stably decomposing and processing organic waste. Biogasification is the most used method for energy conversion of organic waste with high moisture content, and is a useful method for organic waste treatment following the prohibition of direct landfill (2005) and marine dumping (2013). Due to African Swine Fever (ASF), which recently occurred in Korea, recycling of wet feed is prohibited, and consumers such as dry feed and compost are negatively recognized, making it difficult to treat food waste. Accordingly, biogasification is attracting more attention for the treatment and recycling of food waste. Korea's energy consumption amounted to 268.41 106toe, ranking 9th in the world. However, it is an energy-poor country that depends on foreign imports for about 95.8% of its energy supply. Therefore, in Korea, the Renewable Energy Portfolio Standard (RPS) is being introduced. The domestic RPS system sets the weight of the new and renewable energy certificate (REC, Renewable energy certificate) of waste energy lower than that of other renewable energy. Therefore, an additional incentive system is required for the activation of waste-to-energy. In this study, the operation of an anaerobic digester that treats food waste, food waste Leachate and various organic wastes was confirmed. It was intended to be used as basic data for preparing the waste-to-energy incentive system through precise monitoring for a certain period of time. Four sites that produce biogas from organic waste and use them for power generation and heavy gas were selected as target facilities, and field surveys and sampling were conducted. Basic properties analysis was performed on the influent sample of organic waste and the effluent sample according to the treatment process. As a result of the analysis of the properties, the total solids of the digester influent was an average of 12.11%, and the volatile solids of the total solids were confirmed to be 85.86%. BOD and CODcr removal rates were 60.8% and 64.8%. The volatile fatty acids in the influent averaged 55,716 mg/L. It can be confirmed that most of the volatile fatty acids were decomposed and removed with an average reduction rate of 92.3% after anaerobic digestion.

A Routine System for Generation of Fertile Transgenic Rice Plants Using Biolistic Method

  • A routine system based on particle bombardment of embryogenic callus for recovery of fertile transgenic rice (Oryza sativa L.) plants was developed. Embryogenic callus was established within 2-3 months from calli derived from mature seeds of Korean rice cultivar, Nagdongbyeo. The callus was bombarded with the plasmid pRQ6 containing the $\beta$-glucuronidase gene (gusA) and hygromycin phosphotransferase gene (hph, conferring resistance to hygromycin B), both driven by CaMV 35S promoter. Placement of cells on an osmoticum-containing medium (0.2 M sorbitol and 0.2 M mannitol) 4 hrs prior to and 16 hrs after bombardment resulted in a statistically significant increase with 3.2-fold in transient expression frequency gusA. In five independent experiments, the average frequency of transformation showing GUS activities was $8.86\%$. A large number of morphologically normal, fertile transgenic rice plants were obtained. Integration of foreign gene into the genome of $R_0$ transgenic plants was confirmed by Southern blot analysis. GUS and HPT were detected in $R_1$ progeny and Mendelian segregation of these genes was observed in $R_1$ progeny.

Determination of Heavy Metal Contents in Various Packaging Boards (지류 포장재 종류에 따른 중금속 함량 측정)

  • This study was performed to evaluate the heavy metal contents in various packaging board. Domestic and foreign OCC (old corrugated containers) and old duplex boards were used as raw materials. Tests were made for the printed and unprinted parts of the sample. Heavy metal contents of old food packaging boards made from virgin pulp fibers were also evaluated. The contents of heavy metals including lead (Pb), cadmium (Cd), barium (Ba), arsenic (As), antimony (Sb), selenium (Se), and mercury (Hg) were determined using ICP-AES (Inductively Coupled Plasma - Atomic Emission Spectrometer), and CV-AAS (Cold vapor-atomic absorption spectrometer) after digesting the samples in a microwave oven. The contents of heavy metals contained in domestic packaging board were higher than those in overseas samples, and OCC showed higher contents of heavy metals than old duplex boards. Printed parts gave greater heavy metal contents than unprinted parts. Results indicate that recycling of paper and paperboard products increases the heavy metal contamination of the paper packaging products and this derives mostly from the heavy metals contained in printing inks. Recycling processes that decrease heavy metals in recycled fibers and new printing inks that contains less heavy metals should be developed to solve the problem associated with the heavy metals in packaging paper products.

Overview of the Development of the Korean Exposure Factors Handbook

  • A set of exposure factors that reflects the characteristics of individual behavior capable of influencing exposure is essential for risk and exposure assessment. In 2007, the Korean Exposure Factors Handbook was, therefore, issued, driven by the need to develop reliable exposure factors representing the Korean population. The purpose of this study was to overview the development process of the Korean Exposure Factors Handbook and major recommended exposure values for the Korean population to allow information exchanges and comparison of recommended values among nations. The researchers reviewed the domestic data that could be used in the development of exposure factors, confirmed a knowledge gap, and set a priority of development by phases. A methodology to measure exposure factors was established to develop measuring techniques and test their validity. Data were processed or a survey was conducted according to the availability of data. The study thus produced recommended values for 24 exposure factors grouped by general exposure factors, food ingestion factors, and activity factors by setting up a database of exposure factors and carrying out statistical analysis. The study has significantly contributed to reducing the potential uncertainty of the risk and exposure assessment derived by the application of foreign data or research findings lacking representativeness or grounds by developing a set of exposure factors reflecting the characteristics of the Korean people. It will be necessary to conduct revisions in light of the changing statistical values of national data and the exposure factors based on Korean characteristics.

The Educational Needs of a Mother when Nurturing Children (아동 양육에 관한 어머니의 교육 요구도)

  • The purpose of this study was to investigate the educational needs of a mother when nurturing children from neonates to the schoolage. A total of 657 subjects responded to the survey about the level of educational needs when nurturing children. The subjects of the study constituted of 401 mothers who visited the health center for immunization and 256 mothers who visited the pediatric outpatient department or whose children were hospitalized in pediatrics. This instrument had 64 items about nurturing children from neonates to the schoolage and one item had a score range of one to four. In data analysis, SPSSWIN 9.0 program was utilized for descriptive statistics. The results were as follows. 1) Mothers who had the neonates represented the highest educational needs about parental-neonates attachments with 3.47 of mean score compared to neonatal convulsion(3.44), management of common colds(3.44), nutrition (3.44), fever control (3.42). 2) Mothers who had infancy represented the highest educational needs about management of common colds with 3.34 of mean score compared to psychosocial developments (3.23), management of foreign bodies (3.22), feeding the food(3.19), playing with the infant(3.16). 3) Mothers who had toddlers represented the highest educational needs about psychosocial developments with 3.35 of mean score compared to discipline for children(3.34), management of teeth (3.29), management of common colds (3.21), management of accidents(3.20). 4) Mothers who had the a child in preschool represented the highest educational needs about psychosocial developments with 3.53 of mean score compared to management of accidents(3.23), discipline for children (3.00). 5) Mothers who had the child in secondary school represented the highest educational needs about psychosocial developments with 3.42 of mean score compared to management of teeth(3.13), management of accidents (3.05).
