Gamma-Irradiation Provides Microbiological Protection While Maintaining Sensory Quality Change of Fresh Kale Juice During Storage

  • The effect of gamma-irradiation on microbiological growth in kale juice during storage was studied. Fresh kale juice was prepared and irradiated at 0, 1, 3, 5, 7, 10, and 15 kGy. D values for total bacteria, yeast and mold, Salmonella, E. coli, and Pseudomonas were 3.6, 4.0, 3.2, 1.4, and 1.6 kGy, respectively. E. coli and Pseudomonas were eliminated completely at 5 and 7 kGy, respectively. Gamma-irradiation also reduced total viable bacteria during storage. Therefore, these results indicate gamma-irradiation can prevent microbial spoilage of fresh kale juice by inactivating pathogenic microorganisms.

  • Various microorganisms play important roles in food fermentation, food spoilage, and agriculture. In this study, the biotype of 54 yeast and bacterial strains having high potential for utilization in food and agriculture, including Candida spp., Lactobacillus spp., and Acetobacter spp., were characterized by matrix-assisted laser desorption/ionization time-of flight mass spectrometry (MALDI-TOF MS). This characterization using a fast and robust method provides much-needed information on the selected microorganisms and will facilitate effective usage of these strains in various applications. Importantly, the unique protein profile of each microbial species obtained from this study was used to create a database of fingerprints from these species. The database was validated using microbial strains of the same species by comparing the mass spectra with the created database through pattern matching. The created reference database provides crucial information and is useful for further utilization of a large number of valuable microorganisms relevant to food and agriculture.

식품부패 및 식중독성 미생물에 대한 쇠비름(Portulace oleracea) 메탄올 추출물의 항균활성과 성분분석 (Antimicrobial Activity of Methanol Extract from Soibirhym (Portulace oleracea) against Food Spoilage or Foodborne Disease Microorganisms and the Composition of the Extract)

  • 본 연구에서는 쇠비름을 메탄올로 추출하여 식품의 부패나 식중독성을 유발하는 세균과 곰팡이에 대한 추출물의 항균활성을 검토하고 그들의 최소생육저해농도를 결정하였으며 쇠비름 추출물의 구성성분과 함량을 분석하였다. 쇠비름은 특히 P. citreonigrum 균주에 대해 매우 높은 항균효과를 보였고 S aureus, K. pneumoniae에 대해서는 benzoic acid에 상응할 만한 항균효과를 나타내었다. 쇠비름 메탄올 추출물은 200mg/$m\ell$ 농도에서 S. aureus와 P. citreonigrum의 생육을 억제하였고, 250 mg/$m\ell$ 농도에서 K. pneumoniae의 생육을 억제하였으며, 300 mg/$m\ell$ 농도에서 P. aeruginosa와 E. coli의 생육을 억제하였다. 따라서 이들 균주에 의한 식품의 부패 및 식중독이 우려되는 식품의 항균제로서 쇠비름의 사용 가능성을 볼 수 있었다. 쇠비름 메탄올 추출물의 분석결과 전체 147가지 성분이 분석되었으며 acid 및 ester류가 41.06%로 추출물의 가장 많은 부분을 차지하고 있었다. 또한 세균, 곰팡이, 효모와 같은 미생물에 항균활성을 나타내는 것으로 보고되어 있는 2,3-dihydro-benzofuran, 4-hydroxy-3-methoxy- benzoic acid와 4-hydroxy benzeneethanol 등의 성분들이 검출되었다.

  • 가자 물 추출물과 70% 에탄올 추출물을 acetone, hexane, chloroform, butanol 용매로 순차 분획하였다. 용매 순차 분획물중 butanol분획물이 E. coli, S. typhimurium, P. aeruginosa, B. subtilis, S. aureus, L. monocytogenes균종에 대해 가장 좋은 항균활성과 추출수율을 나타내었다. 또한 polyphenol 표준물질 중 pyrogallol이 항균활성 실험결과 가장 좋은 활성을 나타내었다. 가자의 물 추출물과 에탄올 추출물을 13가지 표준품을 사용하여 HPLC 분석한 결과, pyrogallol의 수치가 가장 높게 나타났다.

Inhibitory Effect of Mugwort(Artemisia asiatica Nakai) on the Growth of Food Spoilage Microorganisms and Identification of Antimicrobial Compounds

  • The antimicrobial activity of mugwort(artemisia asiatica Nakai) was investigated. The methanol extract or dried mugwort was fractionated to hexane, chloroform, ethylacetate, butanol, and aqueous fractions. The hexane fraction among these fractions showed the hifhest inhibitory effect on the growth of microorganisms such as Bacillus subtilis, Escherichia coli, Staphylococcus aureus and Lactobacillus plantarum. Bacillus subtilis, Escherchia coli, and Staphylococcus aureus were completely inhibited at a concentration of 250, 500 , and 750$\mu\textrm{g}$/ml respectively. The hexane fraction was further fractionated into 16 subfractions by silica gel column and thin layer chromatography(TLC). The subfraction No. 8, 9, and 10 on TLC exhibited high antimicrnial activity. At 3rd fractionation, subfraction No. 2 inhibited the growth of microorganisms at 500$\mu\textrm{g}$/ml. Heptadecane, dodecamethyi cyclohexasiloxane, (E,E)-2,4-decadienal, dodecamethul pentasiloxane, coumarin, 5,6,6,6a-tetrahydro-4,4,7a-trimethyl-2(4H)-benzofuranone, neophytadiene, tridecanoic acid, methyl ester, 2-methyl-4,5-nonadiene, (Z,Z)-9-12-octadecadienoyl chloride, and bis(2-ethylhexyl) were identified from this antimicrobial fraction by GC-MS.

A Study of Antioxidant and Antibacterial Activates of the Extraction of Perscaria hydropiper L.

  • This purpose of this study is to examine the antioxidant and antimicrobial activities of Persicaria hydropiper L. extract in 70% ethanol and in water, a medicinal herb, as an effort to examine the potential of medicinal herbs for development of antioxidants and natural preservative substitutes. The total poly-phenol content in the 70% ethanol extract and in the water extract were 19.88 mg/g and 7.46 mg/g, respectively. The DPPH radical scavenging activity was 90.99% and 64.98% in the 70% ethanol extract and water extract, in which 70% ethanol extract showed a higher activity. The antioxidant effect of Persicaria hydropiper L. extract appears to be very good and due to its excellent growth inhibitory effect on food-poisoning-causing microorganisms in the food, it is thought to be utilized as a potential natural preservative substitute in many areas.

유제품에 이용되는 주요 열처리 조건 (Heat Treatments Used in the Dairy Industry)

  • Heat treatment is a fundamental processing technology in the dairy industry. The main purpose of heat treatment is to destroy pathogenic and spoilage promoting microorganisms to ensure milk safety and shelf life. Despite the development of alternative technologies, such as high-pressure processing and pulse field technology for microbial destruction, heat treatment is widely used in the dairy industry and in other food processes to destroy microorganisms. Heat treatment has contributed greatly to the success of food preservation since Pasteur's early discovery that heat treatment of wine and beer could prevent their deterioration, and since the introduction of milk pasteurization in the 1890s. In Korea, food labeling standards do not stratify heat treatments into low temperature, high temperature, and ultra-high temperature methods. Most milk is produced in Korea by pasteurization, with extended shelf life (ESL : 125--140℃ / 1-10 s). Classification based on temperature (i.e. low, high, and ultra-high), is meaningless.

개머루덩굴 추출물의 식품부패 및 병원성 미생물에 대한 항균효과 (Antimicrobial Effect of Ampelopsis brevipedunculata Extracts On Food Spoilage or Foodborne Disease Microorganism)

  • 본 연구는 천연 식품보존료 개발의 일환으로 한약 재료로 이용되고 있는 개머루덩굴을 에탄올로 추출하여 식품부패 및 병원성 미생물에 대한 항균활성을 조사하였다. 개머루덩굴 에탄올 추출물은 식품부패 및 병원성 미생물에 대해 농도 의존적으로 항균효과를 보였으며, 그 중에서 Escherichia coli O157:H7 균에 대해 가장 높은 항균효과를 보였다. 또 개머루덩굴 에탄올 추출물이 식중독 유발세균의 성장에 미치는 효과를 검정하기 위해 Salmonella typhimurium, Yersinia enterocolitica, Staphylococcus aureus, Escherichia coli O157:H7 균들의 배양액에 개머루덩굴 에탄올 추출물의 농도를 250, 500, 1,000, 2,000 mg/L 각 농도별로 첨가하여 생육을 조사한 결과 250 mg/L까지는 대조군과 별 차이를 보이지 않았으나 500 mg/L 이상에서는 유의적인(p<0.05) 감소를 관찰할 수 있었다. 본 연구결과는 개머루덩굴 에탄올 추출물이 식중독을 유발시키는 세균에 대하여 우수한 항균작용을 나타내고 있으며, 따라서 개머루덩굴이 효과적인 천연보존료로서 이용될 수 있음을 시사하고 있다.

살균 Target 미생물 사멸효과 예측 프로그램 개발 (Development of a Computer Program Predicting Sterilization Effects on Target Microorganisms)

  • 가열살균 처리시의 온도값을 이용하여 다양한 미생물들의 가열살균 효과를 D값, z값 등의 미생물 내열성 특성치와 kinetic 모델 및 수식을 이용하여 살균치 계산을 컴퓨터를 이용하여 수행함으로써 target 미생물의 살균정도를 예측하고자, Excel과 Visual Basic을 연동하여 작성된 시스템으로 Excel을 기본으로 사용한 프로그램을 개발하였다. 가열살균 target 미생물을 선택하고자 야채식품을 기반으로 하는 식품에서의 미생물을 검토하여, 내열성인 가장 큰 호기성 포자형성균인 Bacillus spp. 계통이 산을 형성하여 맛 등의 품질을 저하시켜 변패를 유발하며, 위생적인 면에서 포자수를 줄이거나 사멸할 필요가 있어서 선택하였다. 병원성 균 중에서 $D_{121.1}$이 0.0065분, z값이 7.8$^{\circ}C$인 Bacillus cereus와 부패현상 관련 균 중에서 $D_{121.1}$이 0.5분, z값이 12$^{\circ}C$인 Bacillus subtilis를 대상으로 해서 개발되었으며, 필요시에는 D값과 z값을 직접 대입하므로서, 여러 가지 미생물에 대해서도 적용이 가능하게 하였다.

Application of bio-preservation to enhance food safety: A review

  • Consumers and industry experts frequently have negative perceptions of most chemical preservatives. Although most people concede that they cannot resolve global food waste issues without preservatives, they prefer products without chemical preservatives. Numerous emerging technologies is now surpassing conventional methods for mitigating microbial food deterioration in response to consumer demand and fundamental health and safety considerations, including biological antimicrobial systems such as using food-grade microorganisms and their metabolites primarily originating from microorganisms, plants, and animals. Microbial compounds, including bacteriocins, bacteriophages, and anti-fungal agents, plant extracts such as flavonoids and essential oils; and animal-originated compounds, such as lysozyme, chitosan, and lactoferrin, are considered some of the major bio-preservatives. These natural compounds can be used alone or with other preservatives to improve food safety. Hence, the use of microbes or their metabolic byproducts to extend the shelf life of foods while maintaining safety standards is known as bio-preservation. To manufacture and consume foods in a safe condition, this review primarily aims to broaden knowledge amongst industry professionals and consumers regarding bio-preservation techniques, bio-preservatives, their classifications, and distinctive mechanisms to enhance food safety.
