Expression and Purification of Extracellular Solute-Binding Protein (ESBP) in Escherichia coli, the Extracellular Protein Derived from Bifidobacterium longum KACC 91563

  • Bifidobacterium longum KACC 91563 secretes family 5 extracellular solute-binding protein via extracellular vesicle. In our previous work, it was demonstrated that the protein effectively alleviated food allergy symptoms via mast cell specific apoptosis, and it has revealed a therapeutic potential of this protein in allergy treatment. In the present study, we cloned the gene encoding extracellular solute-binding protein of the strain into the histidine-tagged pET-28a(+) vector and transformed the resulting plasmid into the Escherichia coli strain BL21 (DE3). The histidine-tagged extracellular solute-binding protein expressed in the transformed cells was purified using Ni-NTA affinity column. To enhance the efficiency of the protein purification, three parameters were optimized; the host bacterial strain, the culturing and induction temperature, and the purification protocol. After the process, two liters of transformed culture produced 7.15 mg of the recombinant proteins. This is the first study describing the production of extracellular solute-binding protein of probiotic bacteria. Establishment of large-scale production strategy for the protein will further contribute to the development of functional foods and potential alternative treatments for allergies.

Enhanced Production of Exopolysaccharides by Fed-batch Culture of Ganoderma resinaceum DG-6556

  • The objectives of this study were to optimize submerged culture conditions of a new fungal isolate, Ganorderma resinaceum, and to enhance the production of bioactive mycelial biomass and exopolysaccharides (EPS) by fed-batch culture. The maximum mycelial growth and EPS production in batch culture were achieved in a medium containing 10 g/l glucose, 8 g/l soy peptone, and 5 mM $MnCl_2$ at an initial pH 6.0 and temperature $31^{\circ}C$. After optimization of culture medium and environmental conditions in batch cultures, a fed-batch culture strategy was employed to enhance production of mycelial biomass and EPS. Five different EPS with molecular weights ranging from 53,000 to 5,257,000 g/mole were obtained from either top or bottom fractions of ethanol precipitate of culture filtrate. A fed-batch culture of G. resinaceum led to enhanced production of both mycelial biomass and EPS. The maximum concentrations of mycelial biomass (42.2 g/l) and EPS (4.6 g/l) were obtained when 50 g/l of glucose was fed at day 6 into an initial 10 g/l of glucose medium. It may be worth attempting with other mushroom fermentation processes for enhanced production of mushroom polysaccharides, particularly those with industrial potential.

  • Urban parks are progressing but are in chaos in the twenty-first century. Therefore the purposes of this study are to consider critically and classify the new paradigm of urban parks. Urban parks are one of the space products, and progressing aspects can be divided into three parts; supply, demand and market aspects. In the abstract, urban parks' progress represents process, openness or voidness, general and cultural ecology, productivity, experience program, identity or sense of place, carriers of urban regeneration, urban infrastructure, community space, multi-layered activity, active space, communication with urban space, tool of low carbon strategy and consilience. But urban parks have come under increased criticism about the long period development on trees growth, covering open space, limitation of general and cultural ecology, production, activity programs, identity and community space, visible urban regeneration, economic validity, urban sprawl, not using as the low carbon strategy, and finally negative consilience with contiguous fields. We collected these critical consideration about progressing urban parks, and proposed urban agricultural park as one of the alternative urban parks. This is closely connected with sustainable region development, low-carbon society, local food, well-being, Lohas paradigm and amenity of urban life.

  • Recently, production-consumption quantity and market size of environment-friendly agricultural products(EFAP) have been increased. Consumers have demanded food safety and nutrition of EFAP, but in the near future they will need many kinds of item, relatively low price and high quality. So producers have to make an effort for down-cost, increasing items and developing technical know-how, which can be attained by producer/region organization. Correlation coefficient between the recognition degree for certification system and purchasing amounts at a time is higher than other ones. Therefore public information works using the TV and newspaper must be promoted all the more about certification system. And if the consumer’s satisfaction level of EFAP purchased gets higher, the quantity demanded will be increased more. And consumer’s trust on EFAP is derived from certification system or close relation between producers and consumers. Thus the most important thing of marketing strategies for EFAP is continuously to focus on explanation and information works on certification system by region in metropolitan area. Also, it necessary to introduce green marketing principles and apply to strategies item type. Namely those need to establish target market segmentation And marketing strategy stages according to the green degrees of market And commodity.

  • MSMEs encountered several issues during the COVID-19 pandemic, including decreased sales and capital, difficulties in distribution, trouble obtaining raw materials, decreased production, and employee layoffs. These issues posed a threat to the national economy. The final effect was the company's bankruptcy as a result of its failure to survive a pandemic, which led to the company's downfall. This research aims to find out and analyze MSMEs' business strategy and innovation to maintain their business during the COVID-19 pandemic in Indonesia. This research is qualitative. Researchers got data using interviews, observation, and documentation. The analysis of this research is descriptive. To draw customers during the COVID-19 pandemic, MSMEs use social media or digital marketing, followed by the delivery of orders, promotions, or discounts and the creation of freebies. MSMEs develop new products as part of their ongoing efforts to expand their businesses while upholding standards for quality, value, and customer service. They also adapt to changing consumer tastes and popular food trends while showcasing some of their most intriguing products.

  • Zea mays, known as maize or corn, is a major staple crop and an important source of energy for humans and animals, thus ensuring global food security. Approximately 9.4% of the loss of total annual corn production is caused by pathogens including fungi, bacteria, and viruses, resulting in economic losses. Although the use of fungicides is one of the most common strategies to control corn diseases, the frequent use of fungicides causes various health problems in humans and animals. In order to overcome this problem, an eco-friendly control strategy has recently emerged as an alternative way. One such eco-friendly control strategy is the use of beneficial microorganisms in the control of plant pathogens. The beneficial microorganisms can control the plant pathogens in various ways, such as spatial competition with plant pathogens, inhibition of fungal or bacterial growth via the production of secondary metabolites or antibiotics, and direct attack to plant pathogens via enzyme activity. Here, we reviewed microorganisms as biocontrol agents against corn diseases.

Industrial Policy as a Development Strategy: Cuba' s Experience and Policy Implications (개발전략으로서 산업정책: 쿠바의 경험과 정책적 시사점)

  • This paper analyzes Cuba's market-oriented reforms to alleviate essential problems with socialist countries such as soft budget constraints and incentive problems. It also discuss about effectiveness of industrial policy as a development strategy. The soft budget constraints and incentive problems resulted in the collapse of Soviet bloc and COMECON in early 1990s. After the collapse, Cuban economy suffered a steep dive, and national income tumbling down rapidly. Cuban faced serious shortages of food, gasoline, and other basic necessities of life. To halt and partially reverse economic downturn and dire austerity in the 1990's, the Cuban government made some partial reforms to the inherited Soviet system of cental planningand faced severe shortage in food, energy, and daily necessities. In response to the economic crisis. Cuba introduced economic reforms and implemented industrial policy as a development strategy as long as Cuba maintained a strong socialist country. Cuban government established the economic free zone law and attempted to induce foreign direct investment by implementing export-led industrial policy. Fiedel Castro approved the Law No. 165 "Free Zones and Industrial Parks", in 1996. However, Cuba's ESZ strategy seems to have failed because of the U.S. sanctions, but also because of Cuba's own policies, which do not allow foreign investors to hire workers directly and impose a high implicit tax on wages. By limiting advanced techniques of personnel and organization management, indirect employment can result in lowering work efforts and productivity of workers, and aggravating production efficiency in the ESZs. Another reason to fail comes from the double wage structure due to the double monetary-exchange rate system. Most of the high non-wage costs result from the double exchange rate system. Due to Cuba's imbalanced industry and production structures, concentrated labor force, and urbanization and centralization of agriculture production, the industrial transformation development model suggested by Lewis has not been successful unlike other Asian agriculture-led development model. Cuba has to overcome many difficulties in implementing industrial policy as a development strategy.

Production of transgenic potato exhibiting enhanced resistance to fungal infections and herbicide applications

  • Potato (Solanum tuberosum L.), one of the most important food crops, is susceptible to a number of devastating fungal pathogens in addition to bacterial and other pathogens. Producing disease-resistant cultivars has been an effective and useful strategy to combat the attack of pathogens. Potato was transformed with Agrobacterium tumefaciens strain EHA101 harboring chitinase, (ChiC) isolated from Streptomyces griseus strain HUT 6037 and bialaphos resistance (bar) genes in a binary plasmid vector, pEKH1. Polymerase chain reaction (PCR) analysis revealed that the ChiC and bar genes are integrated into the genome of transgenic plants. Different insertion sites of the transgenes (one to six sites for ChiC and three to seven for bar) were indicated by Southern blot analysis of genomic DNA from the transgenic plants. Expression of the ChiC gene at the messenger RNA (mRNA) level was confirmed by Northern blot analysis and that of the bar gene by herbicide resistance assay. The results obviously confirmed that the ChiC and bar genes are successfully integrated and expressed into the genome, resulting in the production of bialaphos-resistant transgenic plants. Disease-resistance assay of the in vitro and greenhouse-grown transgenic plants demonstrated enhanced resistance against the fungal pathogen Alternaria solani (causal agent of early blight).

  • A strategy for the automation of glutamic acid fermentation has been developed by the use of $CO_2$ analyzer together with a controller. It was found that a linear relationship existed between growth and $CO_2$ level in the exit gas. Therefore penicillin addition at an appropriate biomass concentration to excrete glutamate could be achieved automatically. In addition, an automatic batch feeding method (fed-batch culture) provided a means of overcoming substrate inhibition effects on growth and glutamic acid production in batch culture, thereby increasing productivity and product yield.

  • Strategic alliance is increasingly becoming a popular strategy in the family restaurant industry. In general, strategic alliance can be defined as several brands collaborating in technology development, marketing, or production while keeping their independence as separate business entities. This study identified segments on the basis of sales promotion resulting from strategic alliances between family restaurants and card companies. This study further investigated how brand image, brand value, price fairness, customer loyalty and demographics are different among the segments. From the statistical analysis, three segments were found; 'short-period benefits oriented' segment, 'intangible and discount benefits oriented' segment and 'free benefits oriented' segment. Among the three segments, the 'free benefits oriented' and 'intangible and discount benefits oriented' segments had greater perceived brand image, brand value and customer loyalty than the 'short-period benefits oriented' segment.
