Changes in Food Composition of Tenebrio molitor by Life Stage (생애주기에 따른 갈색거저리의 식품성분 변화)

  • Purpose: This study was aimed to compare the nutritional content of the life stages of Tenebrio molitor. Methods: Nutritional element analysis comprised 12 minerals, 18 amino acids, 4 vitamins, and 12 fatty acids in larval, pupal, and adult stages. Nutritional element content was expressed as a percentage of 100 g of sample and determined in raw and freeze-dried samples to assess the food value of the insect. Results: Moisture, crude protein, and crude fat contents in raw samples were 63.5%, 23.19%, and 3.96%, respectively. Moisture content was significantly higher in the adult stage than in larval and pupal stages, whereas crude fat and crude ash contents were lower. Mineral composition of the mealworm showed K, P, and Mg in decreasing order of content, with no differences among life stages. Total amino acid content increased from adult, pupal to larval stages, in order with each stage's growth, without difference in amino acid composition. Seventy-six to 78% of total fatty acid corresponded to unsaturated fatty acids, with oleic acid as the most abundant. In addition, fatty acid content was high in the adult stage, and clear differences were observed between life stages. Finally, vitamins B1 and B3 showed similar content between life stages, whereas vitamin B2 showed a higher content the adult than larval and pupal stages. A higher folate content was observed in pupal and adult stages than in the larval stage. Although freeze-dried samples showed a higher nutritional content than raw samples, both samples were similar in composition and content between life stages, suggesting the suitability of freeze-dried samples for use as food materials. Conclusion: These results may provide a basis for more diversified industrial applications of the Tenebrio molitor.

Comparison of Quality Characteristics of Melania snails in Korea (한국산 다슬기의 식품학적 성분 및 품질특성)

  • To identify the food characteristics of seven melania snail species in Korea, the detailed proximate, fatty/amino acid, mineral composition and chlorophyll contents were investigated. The content ranges of moisture, crude protein, crude lipid and ash were 70.1-81.0%, 11.6-16.9%, 0.4-1.6% and 1.9-3.4%, respectively. K. nadifila showed the significant mineral contents Fe, P, Ca than other species. Total Chlorophyll level was highest in K. globus ovalis ($1.2\pm0.2$ mg/g). The major fatty acids were 16:0, 22:1, 20:1, 18:1, 20:5 and 22:6 were identified as small amounts (1.0-3.3%) of total fatty acids. Citrulline was shown to be the major free amino acids (20.4-53.1%).

Recovery of Norovirus Surrogate in Seawater using an Electropositive and Electronegative Filter (양전하 및 음전하 필터를 이용한 해수 중 Norovirus Surrogate의 회수)

  • Recently coastal seawater has been contaminated by enteric viruses such as the norovirus via untreated groundwater globally. Accordingly, the consumption of molluscan shellfish from seawater that has been contaminated with fecal material has become an important issues. The levels of enteric viruses in seawater are low and recovery and concentration of the virus from large volumes of water is difficult. We compared the effectiveness of two representative method of concentrating virus using negatively and positively charged filters. The mean retention of seeded FCV by HAMF and NCCF was 48% and 78%, respectively. Overall, the recovery of NCCF was 43.3$\pm$11% better than that of HAMF. However, the eluate obtained by using beef extract solution in the NCCF procedure caused an inhibitory effect on the RT-PCR; therefore, it was necessary to employ a PCR inhibitor removal procedure. The HAMF eluate contained no PCR inhibitors, but HAMF was not an effective method of concentrating the virus from large volumes of natural seawater due to clogging.

Survey on Food Characteristic Preferences for Rural Healing Tourism for Adults in Urban Areas (도시지역 성인의 농촌치유관광 음식 특성 선호도 조사)

  • This study was conducted to investigate food needs for dining out in the context of rural healing tourism by surveying food preference characteristics. A total of 600 adults in an urban area completed an online questionnaire on food characteristics for rural healing tourism. Regarding the purpose of rural healing tourism, 61.2% of respondents answered 'relaxation' while 6.3% answered 'promotion of health'. Exactly 66.3% of respondents selected Korean foods (rice) as the preferable food type for dining out in the context of rural healing tourism. Respondents considered 'regionality' and 'ingredient' as more important among food quality characteristics, whereas 'professionality' was less important. Females perceived 'nutrition' and 'cooking' as more important than males. Among food characteristics of professionality, regionality, and traditionality, respondents most preferred 'using regional specialty food'. Preference for 'using traditional food' significantly increased with subject's age. Respondents rated 'balanced nutrition' most highly among food characteristics of health and nutrition. 'Seasonal food' was the most preferred food characteristic among cooking and ingredients. Females showed higher interest in nutrition, cooking, and ingredients and higher preferences related to food characteristics of nutrition, cooking, and ingredients than males. In conclusion, Korean foods (rice) or Korean full course menu items using local specialty foods or seasonal foods with balanced nutrition are needed for dining out in the context of rural healing tourism.

Development of Bile Salt-Resistant Leuconostoc citreum by Expression of Bile Salt Hydrolase Gene

  • Probiotic bacteria must have not only tolerance against bile salt but also no genes for antibiotic resistance. Leuconostoc citreum is a dominant lactic acid bacterium in various fermented foods, but it is not regarded as a probiotic because it lacks bile salt resistance. Therefore, we aimed to construct a bile salt-resistant L. citreum strain by transforming it with a bile salt hydrolase gene (bsh). We obtained the 1,001 bp bsh gene from the chromosomal DNA of Lactobacillus plantarum and subcloned it into the pCB4170 vector under a constitutive P710 promoter. The resulting vector, pCB4170BSH was transformed into L. citreum CB2567 by electroporation, and bile salt-resistant transformants were selected. Upon incubation with glycodeoxycholic acid sodium salt (GDCA), the L. citreum transformants grew and formed colonies, successfully transcribed the bsh gene, and expressed the BSH enzyme. The recombinant strain grew in up to 0.3% (w/v) GDCA, conditions unsuitable for the host strain. In in vitro digestion conditions of 10 mM bile salt, the transformant was over 67.6% viable, whereas only 0.8% of the host strain survived.

Antioxidant Activities of Five Melania Snails of the Genus Semisulcospira in Korea (한국산 다슬기류(Semisulcospira spp.) 5종의 항산화 활성)

  • The in vitro antioxidant activities of five melania snails in the genus Semisulcospira (S. coreana, S. forticosta, S. libertina, S. tegulata and S. gottschei) were tested in detail. The total phenolic contents of the snails ranged from $32.3{\pm}1.0$ to $87.9{\pm}6.9$ mg gallic acid equivalent (GAE)/g dry sample. The $EC_{50}$ values for the 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities were $2.245{\pm}0.179$ mg/mL for S. coreana and $9.319{\pm}1.276$ mg/mL for S. gottschei and differed significantly (P<0.05) among the tested species. The superoxide dismutase (SOD)-like activity was highest for S. gottschei at 67.2% and lowest for S. forticosta at 4.7%. However, no significant differences among the species were recognized for the peroxynitrite anion scavenging activity. Comparing the correlation coefficients between the total phenolic contents and the DPPH radical and peroxynitrite anion scavenging activities, there was a low level relationship between each activity.

Development and Evaluation of a Next-Generation Sequencing Panel for the Multiple Detection and Identification of Pathogens in Fermented Foods

  • These days, bacterial detection methods have some limitations in sensitivity, specificity, and multiple detection. To overcome these, novel detection and identification method is necessary to be developed. Recently, NGS panel method has been suggested to screen, detect, and even identify specific foodborne pathogens in one reaction. In this study, new NGS panel primer sets were developed to target 13 specific virulence factor genes from five types of pathogenic Escherichia coli, Listeria monocytogenes, and Salmonella enterica serovar Typhimurium, respectively. Evaluation of the primer sets using singleplex PCR, crosscheck PCR and multiplex PCR revealed high specificity and selectivity without interference of primers or genomic DNAs. Subsequent NGS panel analysis with six artificially contaminated food samples using those primer sets showed that all target genes were multi-detected in one reaction at 108-105 CFU of target strains. However, a few false-positive results were shown at 106-105 CFU. To validate this NGS panel analysis, three sets of qPCR analyses were independently performed with the same contaminated food samples, showing the similar specificity and selectivity for detection and identification. While this NGS panel still has some issues for detection and identification of specific foodborne pathogens, it has much more advantages, especially multiple detection and identification in one reaction, and it could be improved by further optimized NGS panel primer sets and even by application of a new real-time NGS sequencing technology. Therefore, this study suggests the efficiency and usability of NGS panel for rapid determination of origin strain in various foodborne outbreaks in one reaction.

Amygdalin Contents in Peaches at Different Fruit Development Stages

  • Amygdalin contents of the seeds, endocarps, and mesocarps from three peach cultivars (i.e., Stone Peach, Hikawa Hakuho, and Bakhyang) were measured at three stages of fruit development (stone-hardening, fruit enlargement, and ripening). The peach samples were dried and defatted with a Soxhlet apparatus, reflux extracted with methanol, and analyzed using reverse phase high-performance liquid chromatography. During all fruit development stages, the amygdalin contents in the seeds were higher than those in the endocarps and mesocarps. The amygdalin contents of the Stone Peach were comparatively higher than the Hikawa Hakuho and Bakhyang (P<0.05). Further, the amygdalin contents during ripening were very low or not detected. Overall, the amygdalin contents of the three peach cultivar samples (seed, endocarp, and mesocarp) increased until the fruit enlargement stage and either remained constant or decreased during ripening.
