Effects of PEG on Embryo Production in Superovulated Hostein Cows (젖소 과배란 처리시 PEG(Polyethylene Glycol) 처리가 수정란 생산에 미치는 영향)

  • This study was conducted to improve the efficiency of embryo recovery and to establish the protocols of superovulation in Holstein cows. Sixteen Holstein cows were used the test the efficacy of three superovulation regimens using Folltropin. In the case of regimen 1, CIDR plus with E2 capsule was inserted in cows at the random stage of estrous cycle and the total of 400 mg Folltropin V was adminstered twice a day for 4 days(Folltropin V group). In regimen 2, CIDR was inserted and 3.0 mg estradiol benzoate was administered i.m. next day and the total of 400 mg Folltropin was adminstered twice a day for 4 days(Folltropin V+EB group). For regimen 3, CIDR insertion was same as in the regimen 2 and the total of 400 mg Folltropin diluted with $10\%$ PEG 8,000 was administered once(Folttropin V+PEG 8,000 group). In all the regimens, CIDR were removed on 12th day and 45 mg dinoprost was administered i.m. simultaneously. The heat detected donors were administered 200 ug LH-RH and inseminated twice with 2 straws of frozen semen 12 hours apart. Embryo were collected using Foley catherter in each uterine homs on 6${\~}$8 days after inseminations. The evaluation of collected embryos were according to the IETS manual. The CL responses according to the superovulation treatments were 5.8, 20.6, 24.0 in the Folltropin V, Folltropin+EB and Folltropin V+PE 8,000 groups, respectively and there were significant different among the treatments(p<0.01). Transferable embyos collected were 3.6$\pm$2.4, 3.3$\pm$l.8 and 2.8$\pm$2.3, in the Folltropin V, Folltropin+EB and Folltropin V+PE 8,000 groups, respectively. Degenerated and unfertilized embryos in regimen 2 and 3 than regimen 1. These results indicates that superovulation treatments with both multiple injections and a single injection using PEG of Folltropin combined with CIDR insertion at the random stage of estrus cycle can be used to produce Holstein embryos.

Superovulation in Korea Cattle with a Single Subcutaneous Injection of Folltropin-V Dissolved in Polyethyleneglycol (한우에 있어서 PEG에 용해시킨 Folltropin-V의 1회 피하주사에 의한 다배란 유기)

  • This study was conducted to determined if single subcutaneous (s.c.) injection of Folltropin-V dissolved in polyethyleneglycol (PEG) can replace as the standard multiple intramuscular (i.m.) injection. The results suggest that the s.c. treatment produced more corpora lutea, embryos recovered and transferable embryos as compared to the i.m. treatment (p<0.05). This study indicates that a single s.c. injection of Folltropin-V dissolved in PEG was effective for superovulatory response and embryo yield in Korean cattle.

Superovulation in Rabbits with a Single Injection of FSH Dissolved in Polyvinylpyrrolidone (토끼에서 FSH제의 Single Injection에 따른 과배란유기 효과)

  • A series of experiments were designed to determine the feasibility of single dose of FSH as an effective superovulation method in rabbits. The superovulation treatments examined comparatively were as follows: 1) a single injection of 100 IU PMSG, 2) Multiple injections of 1 IU Super-OV of 2X daily for 3 days, 3) Multiple injections of 6.6 rng Folltropin-V 2X daily for 3 days, 4) Multiple injections of 13.3 rng Folltropn-V l$\times$ daily for 3 days, 5) Single injection of 40 mg Folltropin-V solved in 10% PVP, and 6) Single injection of 40 mg Folltropin-V dissolved in 25% PVP. The results obtained were as follows: The nurnher of ovulation points in the single injection of 40 mg Folltropin-V dissolved in 25% PVP and multiple injections of 6.6 mg Folltropin-V 2X daily for 3 days was averaged 44.6 and 39.2, respectively, which were significantly (P<0.05) more than in a single dose of PMSG or multiple doses of Super-OV. The number of total ova recovered in the single injection of 40 mg Folltropin-V dissolved in 25% PVP and multiple injections of 6.6 mg Folltropin-V 2X daily for 3 days was averaged 30.4 and 25.9, respectively, which were significantly (P<0.05) more than in the single dose of PMSG or multiple doses of Super-OV. A similar result was obtained for the number of fertilized ova recovered from the above six treatments. From the above results it was concluded that a single injection of 40 mg Folltropin-V dissolved in 25% PVP could be an effective and simple method for superovulation in rabbits.

Dose-related Effects of Follicle Stimulating Hormone on Superovulation in Indigenous Cows of Bangladesh

  • The present study aimed at determining the effective dose of Folltropin, a follicle timulating hormone (FSH), on superovulation in indigenous cows of Bangladesh. Fifteen regularly cycling 5~7 years old dry cows, weighing 200~250 kg with 2.5~3.0 body condition scores (BCS) were divided into three groups (n=5). Individual groups were superovulated with 100, 200 or 300 mg of Folltropin per animal. The superovulation treatment was initiated at Day 10 or Day 11 of the estrous cycle (Day 0=day of estrus). Alfaprostol (6 mg) was injected to each cow 72 h after the initiation of superovulation treatment to induce eestrus. After confirming standing estrus, the cows were inseminated 2~3 times, 12 h apart, depending on the duration of estrus. At Day 6 or Day 7, individual horns of the uterus were flushed with 150~200 $m\ell$ of phosphate buffered saline supplemented with BSA (0.2%), penicillin (100 IU/$m\ell$) and streptomycin (100 $\mu\textrm{g}$$m\ell$) using a two-way foley catheter. The embryos were concentrated, removing the excess medium through an embryo filter, and identified under a stereomicroscope. The identified embryos were collected, washed four times, evaluated and graded as excellent, good, fair or poor. The excellent, good and fair embryos were considered as transferable quality embryos. The mean (range). numbers of embryos collected vs. transferable quality embryos far 100, 200 and 300 mg of Folltropin were 4.5 (1~10) vs. 3.5 (1~8); 2.5 (1~4) vs. 1 (0~2) and 0.0 (0~0) vs. 0.0 (0~0), respectively, Folltropin at a dose of 100 or 200 mg produced suitable ovarian stimulation for superovulation in indigenous zebu cows of Bangladesh. A dose of 300 mg or more Folltropin consistently caused preovulatory corpora lutea formation in the ovaries and resulted in zero embryo recovery.

Ovarian Follicular Populations Prior to and during Superovulation in Cattle: Relationship with Superovulatory Response

  • The present study examined the follicular populations prior to and during superovulation and investigated their relationship with superovulatory response in crossbred cattle. Eleven animals were administered i.m. 8 doses of Folltropin of 2.5 ml each (1.75 mg/ml) spread over 4 days beginning on Day 10 of oestrous cycle, and 30 and 20 mg Lutalyse was given alongwith the 5th and 6th injections of Folltropin, respectively, to induce luteolysis. The animals were artificially inseminated 48, 60 and 72 h after the first Lutalyse injection. The number of corpora lutea (CL) was recorded by palpation per rectum and embryos were recovered non-surgically on Day 6 (Day 0 day of superoestrus). The ovarian follicular population was examined by transrectal Ultrasonography 15 h prior to and 52 h after the first FSH injection, and then on the day of superoestrus and the day of flushing. The follicles were classfied on the basis of diameter as small (3-5 mm), medium (6-9 mm) and large (${\geq}10mm$). The total number of follicles increased significantly (p < 0.01) from $2.45{\pm}0.35$, 15 h prior to the first FSH injection to $8.09{\pm}1.12$, 52 h after the first FSH injection and then further to $13.27{\pm}1.89 $ on the day of superoestrus. A positive correlation was observed between the number of small follicles 15 h prior to the first FSH injection (r = 0.60, p < 0.05), the number of large follicles 52 h after the first FSH injection (r=0.59, p < 0.05) and the number of CL. The follicular population prior to and during superovulation was, however, not significantly different between high (> 6 CL) and low responders (${\leq}6CL$). The present study suggests that the follicular populations undergo dynamic changes during superovulation and that follicular populations prior to superovulation have a limited application as an indicator of the superovulatory response.

Successful Superovulation and Recovery of Embryos in Repeat Breeding Crossbred Cows

  • The present study was undertaken to find out possibilities for superovulation and recovery of freezable/transferable embryo from repeat breeding crossbred cows. For this study a total of 10 Holstein crossbred cows having history of failure of conception even after more than 4~5inseminations were taken and superovulated using Folltropin-V at 100 to 140% dose schedule. The results indicated that out of 10 donors, 8 responded to superovulatory treatment and yielding a total of 94 embryos, out of which 45 were of transferable/freezable quality. The mean ovulations and mean transferable embryos were 11.5$\pm$2.91 and 4.5$\pm$1.66, respectively. These observations suggests that the repeat breeding cows having conception failure after several artificial inseminations can be used as donors for production of embryos and calves through embryo transfer technology.

  • The purpose of this study was to determine the effect of bST treatment on progesteron concentration, embryo recovery and pregnancy rate following embryo transfer. Donor cows were superovulated with Folltropin-V and PGF₂ α combination method and then inseminated with frozen semen 3 times 12 hrs interval. Donor and recipient cows were assigned to control and bST group, of which was given a single injection of bST (500 ㎎, sc) at insemination or estrus detection. Embryo collection of superovulated cows were flushed nonsurgical method at 7 to 8 days after artificial insemination. (omitted)

Influence of the Dominant Follicle on the Superovulatory Response in Cattle

  • Nine cows were superovulated by administration of 8 injections of Folltropin each (2.5 ml/injection, 1.75 mg/ml) i.m spread over 4 days, beginning on Day 10 of oestrous cycle, and 30 and 20 mg prostaglandin $F_{2{\alpha}}$ was given along with the 5th and 6th injections of Folltropin, respectively, to induce luteolysis. The animals were artificially inseminated 48, 60 and 72 h after the first prostaglandin $F_{2{\alpha}}$ injection. The number of corpora lutea was recorded by palpation per rectum and by ultrasonography on Day 6 (Day 0 = day of oestrus). The ovaries were examined daily by ultrasonography on Days 3-9 of the oestrous cycle for following the growth and regression of the largest follicle, which was considered the morphologically dominant follicle. The animals were classified into two groups depending upon the presence (n = 4) and absence of a dominant follicle (n = 5). There was a high correlation (r = 0.97, p < 0.001) between the number of corpora lutea observed by palpation per rectum and that determined by ultrasonography. Mean (${\pm}SEM$) number of corpora lutea determined by ultrasonography ($11.20{\pm}3.71$ vs $3.25{\pm}0.75$) and by palpation per rectum ($10.40{\pm}3.91$ vs $2.25{\pm}0.75$) was significantly higher (p < 0.05) in the nondominant group compared to that in the dominant group. There was no difference in the numbers of follicles 2-3 mm ($13.80{\pm}4.49$ vs $8.00{\pm}1.08$), 4-6 mm ($7.00{\pm}1.87$ vs $3.50{\pm}1.33$), and the total number of follicles ${\geq}2mm$ ($22.00{\pm}5.95$ vs $12.50{\pm}1.26$) between the two groups, one day prior to initiation of superovulation. There was, however, a significant (p<0.01) positive correlation between the number of corpora lutea with the numbers of follicles 2-3 mm (r = 0.83), 4-6 mm (r = 0.80) and the total number of follicles ${\geq}2mm$ (r = 0.89) observed one day prior to initiation of superovulation. The results of this study indicate that the presence of a dominant follicle adversely affects the superovulatory response in cattle.

Possibility of Embryo Collection by Repeatable Superovulation in Hanwoo : Case Report (한우에서 반복 과배란처리에 의한 수정란 채란 가능성 : 사례보고)

  • The purpose of this study was carried out to determine the possibility of continued embryo collection by repeatable superovulation from a Hanwoo elite cow, who was already estimated to an elite cow by DNA analysis. The donor cow was superovulated with Folltropin-V and PG $F_2$$\alpha$ combination method for 5 times, 2 months interval during 10 months. The estrus cows were inseminated with frozen semen 3 times 12 hrs interval. Embryo collection from superovulated cows was flushed non-surgical method at 7 to 8 days after artificial insemination. The superovulated cow was collected from 1 to 5$^{th}$ times as total (14, 4, 10, 6, 9 embryos; total 30 embryos) and transferable embryos (7, 4, 10, 3, 6; Mean$\pm$S.E : 6.0$\pm$2.7 embryos). Seventeen recipients were transferred 24 embryos and then detected to pregnant 10 recipients after embryo transfer (58.8%; 10/17 recipients) The results indicated that the possibility of repeatable superovulation from elite donor cow for embryo transfer could be confirmed in this study, although the replicated number was not enough.
