Structural Characterization for N-Terminal Domain of Caveolin-l

  • Caveolin-1 is a principal protein in the plasma membrane microdomains called caveolae. Caveolae play an important role in the transcytosis and pinocytosis. Therefore, caveolin-1 is most likely to work for the membrane dynamic events. In addition, caveolin-1 interacts with various signaling molecules. Although caveolin-1 possesses a variety of physiological functions, its structural properties were little construed. Here we analyzed the structural dynamics of the N-terminal caveolin-1 (residues 1-101), in order to better understand the structural properties in terms of its versatile functionality. We first analyzed its oligomeric form using GST-fused N-terminal domain, revealing that it equilibrates between a dimer and monomers in av concentration-dependent manner. The N-terminal domain of caveolin-1 was previously found to form a heptamer, so that our data suggest the dimeric form as an intermediate structure for the heptamer formation. Then, we obtained the folding profile, which indicated that $\DeltaG_{H2O}\;is\;about\;0.5\;\pm0.03$ kcal/mol. The stability of N-terminal domain is relatively low, indicating that N-terminal domain may not be crystalline. Conclusively, the dynamic and flexible structure of N-terminal domain appears more favorable to maintain the versatile functions of caveolin-1.

양형 IMS 모듈 튜블러 샤프트의 스플라인 가공 개선 (Improvement of Tubular Shaft Yoke Spline Machining in Both Side IMS Module)

  • The objective of this study is to solve a problem that is occurred during the spline machining of tubular shaft yoke in both side IMS module. In order to simulate the problem, the movement direction of upper die was set as standard case and error case. The material of tubular shaft yoke was set to S20C as refer to the analysis library. The movement directions of upper die were separated with standard case and error case. The error case was set to simulate the problem in the spline machining of tubular shaft yoke. In order to solve the problem, the outer radius of upper die were modelled from 9.40mm to 9.44mm. The simulation results were analyzed and compared in terms of effective stress, metal flow line and folding phenomena characteristics. In case of the outer radius of upper die was 9.42mm, it was observed a relatively uniform effective stress distribution and had a straight metal flow line.

유비퀴틴 단백질의 부분적으로 폴딩된 구조에 대한 분광학적 분석 (Spectroscopic Analysis of Partially Folded State of Ubiquitin)

  • Hydrophobic core가 변이된 유비퀴틴 단백질이 pH 2 용액에서 보이는 구조적인 특성을 여러 분광학적 방법으로 측정하였다. 낮은 pH값을 갖는 용액에서 이 변이 유비퀴틴의 intrinsic tryptophan fluorescence emission spectrum은 unfolded 상태보다 약간 blue shift되어 있고 또한 그 intensity도 상당히 낮게 나타났다. 이는 이 용액 조건에서 이 변이 유비퀴틴의 삼차구조가 약간 남아 있는 것을 의미한다. 같은 용액에서 이 변이 유비쥐틴의 far-UV circular dichroic spectrum은 native 상태나 unfolded 상태의 spectrum과 현저히 달랐으며 220 nm 에서의 molar ellipticity 값을 통하여 볼 때 pH 2인 용액에서 상당량의 이차구조를 지니고 있었다. 또한 같은 용액에서 이 변이 유비퀴틴은 hydrophobic dye인 8-anilinonaphthalene-1-sulfonic acid(ANS)외 fluorescence emission intensity를 증가시키고 fluorescence emission maximum이 짧은 파장에서 나타나게 하였다(blue shift). 이러한 현상은 pH 2 용액에서 이 변이 유비퀴틴의 hydrophobic core가 느슨하여져서 hydrophobic dye인 ANS가 결합할 수 있는 구조를 띠고 있음을 나타낸다. 이러한 분광학적인 관찰은 이 변이 유비귀틴이 pH 2인 용액에서 상당량의 이차구조를 지니고 있지만 hydrophobic core는 느슨하게 형성된 molten globule과 같은 형태를 지니고 있음을 나타낸다. 이 변이 유비퀴틴의 molten 히obule 형태는 단백질 폴딩 반응의 경로를 연구할 수 있는 좋은 모델이 될 수 있을 것으로 생각된다.

Acid and Chemical Induced Conformational Changes of Ervatamin B. Presence of Partially Structured Multiple Intermediates

  • The structural and functional aspects of ervatamin B were studied in solution. Ervatamin B belongs to the $\alpha+\beta$ class of proteins. The intrinsic fluorescence emission maximum of the enzyme was at 350 nm under neutral conditions, and at 355 nm under denaturing conditions. Between pH 1.0-2.5 the enzyme exists in a partially unfolded state with minimum or no tertiary structure, and no proteolytic activity. At still lower pH, the enzyme regains substantial secondary structure, which is predominantly $\beta$-sheet conformation and shows a strong binding to 8-anilino-1-napthalene-sulfonic acid (ANS). In the presence of salt, the enzyme attains a similar state directly from the native state. Under neutral conditions, the enzyme was stable in urea, while the guanidine hydrochloride (GuHCl) induced equilibrium unfolding was cooperative. The GuHCl induced unfolding transition curves at pH 3.0 and 4.0 were non-coincidental, indicating the presence of intermediates in the unfolding pathway. This was substantiated by strong ANS binding that was observed at low concentrations of GuHCl at both pH 3.0 and 4.0. The urea induced transition curves at pH 3.0 were, however, coincidental, but non-cooperative. This indicates that the different structural units of the enzyme unfold in steps through intermediates. This observation is further supported by two emission maxima in ANS binding assay during urea denaturation. Hence, denaturant induced equilibrium unfolding pathway of ervatamin B, which differs from the acid induced unfolding pathway, is not a simple two-state transition but involves intermediates which probably accumulate at different stages of protein folding and hence adds a new dimension to the unfolding pathway of plant proteases of the papain superfamily.

간흡충(肝吸蟲)의 제이중간숙주(第二中間宿主)에 관한 실험적(實驗的) 연구(硏究) Ⅱ. 참붕어, 묵납자루 및 큰납지리 체내(體內)에 있어서 간흡충(肝吸蟲) 피낭유충(被囊幼蟲)의 운명(運命)에 관하여 (Experimental Studies on the Second Intermediate Hosts of Clonorchis sinensis II. Observations on the fate of encysted cercariae of Clonorchis sinensis in the fish hosts, Pseudorasbora parva, Acheilognathus signifer and Acanthorhodeus asmussi)

  • The present experiments proposed to pass judgement upon the suitability as the second intermediate host of Clonorchis sinensis, of three kinds of fresh-water fish by having them infected with the cercaria of Clonorchis sinensis and then observing the penetrating ability of the cercaria, and maturity, the process of degeneration and extinction, and infectivity of the metacercaria of Clonorchis sinensis. The following is a brief summary of the leading facts gained through the experiments; 1. P. parva was easily subject to invasion of the cercaria, A. signifer was much less subject to the invasion, and finally A. asmussi was hard to invade. And the infectivity of the cercariae was in proportion to their penetrating ability mentioned in the above. 2. The examination of the cercariae after having three kinds of fresh-water fish infected with them disclosed that 24 hours after the infection, all of the cercariae formed their cysts in muscle and the metacercariae kept growing, and that 7 days later the metacercariae were found folding their bodies twice, and that in 15 days the characteristic structure of the metacercariae was complete and they made a vigorous rotary movement intermittently. 3. Then the metacercariae came to a state of maturity and beyond this stage some metacercariae in P. parva started the process of degeneration and extinction in 133 days; some in A. asmussi, in 140 days; and A. signifer, in 70 days. As more days elapsed, their degeneration and extinction increased in number, and in 269 days all of them in A. signifer became dead while those in A. asmussi were all dead in 460 days. However almost all of them in P. parva survived even after 770 days. The results shown above revealed that P. parva was the most suitable as the second intermediate host among three kinds of fresh-water fish and that A. signifer and A. asmussi were not quite recommendable as the second intermediate host. The ability of the cercaria to invade fresh-water fish, and life span of the metacercaria within fresh-water fish vary outstandingly according to species of fresh-water fish. An explanation as to the mechanism must wait as the subject to be further pursued.

간흡충 제이중간숙주에 관한 실험적 연구 IV. 치리체내에 있어서 간흡충 피낭유충의 운명 (Experimental Studied on the Second Intermediate HoBts of Clonorchis sinensis IV. Observations on the fate of metacercariae of Clonorchis sinensie in the fish hast, CuffricuEus eigenmunni)

  • In order to judge appropriately the suitability as the second intermediate host of Clonorchis sinensis, the present experiments were proposed to survey the metacercaria of C. sinensis in the fish host, Cuztriculus eigenmanni, in the 6eld, and to observe, by experimental infection of the fish with Clonorchis cercaria, the penetrating ability of the cercaria, maturity, the process of degeneration and extinction, and infectivity of the metacercaria. The following is a brie summary of the leading facts gained through the experiments. 1. All the ten fish (mean body weight $13.4{\pm}3.439$) which were caught in Ok-ku irrigation reservoir were in acted with the metacercariae and the average number of the metacercariae found in the fish body/gram was 4.55. 2. The Bish was easily subject to invasion of the cercaria, and the examination of the cercariae after having the fish infected disclosed that 5:l hours after the infection, all of the cercariae formed their cysts in muscle and the metacercariae kept growing, that 7 days later the metacercariae were found folding their bodies twice, and that in 15 days the characteristic structure of the metacercariae was complete and made a vigorous rotary movement intermittently. 3. In the case of control fish, Pseudorasbora parka, both penetrating ability of the cercariae and the infective degree of the metacercariae to control host were stronger as compared with the 5sh host because that the control fish had higher suceptibility to C. sinensis. 4. Afterwards, the metacercariae in the fish host came to a state of maturity and beyond this stage some metacercariae started the Process of degeneration and extinction in as early as 30 days, and as more days elapsed, mortality of the metacercariae was not uniformly increased but difference of mortality appeared to be according to the individual of Ssh host. That is, in 390, 540 and 586 days, all of the metacercariae in the fish hosts became dead, meanwhile almost all of those found in the fish host of 596 days after the infection survived (mortality; 8, 51%) , Percentage of orally administered metacercariae developing to maturity in rabbit is 10.465 in the last case. 5. As the days were progressed the shapes of the metacercariae in the fish host tended to change from ellipsoid to almost round shapes, the length became shorter while the width was wider. The results shown above revealed that C. eigenmanni was, though it could not be proper host than p. larva. suitable as the second intermediate host in addition to our previous observation results.

충주층군(忠州層群)과 서산층군(瑞山層群)의 비교연구(比較硏究) (Comparative Studies between Chungju and Seosan Groups)

  • The Chungju and Seosan Groups have been known usually as Precambrian formations in Korea. But their relative and absolute ages have been controvericial problem in relation with other geologic system such as so-called Ogcheon and Yeoncheon Systems in Korea. This study has mainly focused on the corelation of the Chungju Group with the Seosan Group in their stratigraphy, structure, metamorphism, and iron ore deposits. In the process of study, the auther surveyed and reclassified the Chungju and Seosan Groups and corelated with Gyeonggi and Ogch cheon metamorphic belts and got some new data. The Chungju iron-bearing formations showing transtitional relation with the Gyeonggi Gneiss Complex and the Jangamri Formation consisting mainly of pebble bearing calcarious phyllite, should be seperated from the Gyemyeongsan formation which is mainly composed of metavolcanic rocks. The Jangamri Formation and the coaly phyllite, which can be corelated respectively with the Hwaggangri Formation and Changri Formation in Ogcheon Group, are repeated in the Gyemyeonsan and Munjuri Formations with the overturned anticlinal folding(F1). So the Chungju Group which was defined as an indipendant geologic unit from the Ogcheon Group should be limited only on the Chungju iron Formation. The Seosan Group can be classified stratigraphically such as Seosan Formation consisting of iron-bearing quartzite and mica schist, Daesan Formation overlying unconformably on the Seosan Formation and Gyeonggi Gneiss Complex. Taean Formation overlying unconformably on the Daesan Formation should be seperated from Seosan Group. There are many similarity in the stratigrphy, structure, and metamorphic facies between Chungju and Seosan Groups exept the metavolcanic rocks in the Gyemyeongsan and Munjuri Formations and the pebble bearing calcareous phyllite in the Jangamri Formation. The two Groups were deformed with two kinds of differant stages, the first shows $N30^{\circ}-40^{\circ}E$ trend of fold axis, the second $N70^{\circ}-80^{\circ}W$ respectively. The Seosan Formation, which is the lowest formation in Seosan Group and bearing the iron formation, was metamorphosed at 2500 m. y. before. These age is similar with the metamorphic age of Gyeonggi metamorphic belt and with the age of Algoman and Kenoran Orogenies which devide the Precambrian into Archean and Proterozoic Era. So the Seosan Formation, which is included in some migmatitic rocks of Gyeonggi Gneiss Complex, is the oldest formation in Korea and can be corelated with the Anshan Group which bears the oldest iron formation in China. The metamorphic facies of the Precambrian metamorphism in Seosan area is simillar with that of Chungju area, showing high temperature-low pressure amphibolite facies which is corelated with the Gyeonggi metamorphic belt, the oldest metamorphic belt in Korea ($650^{\circ}-680^{\circ}C$, 3.2-4.4 Kb). The high temperature intermediate pressure amphibolite facies in Seosan area with the low temperature-intermediate presure greenschist facies of Taean formation is corelated with that of Ogcheon Group ($590^{\circ}-640^{\circ}$ C, 5.2-6.3 Kb). The Chungju and Seosan iron formations were deposited in Archean, showing geochemical composition of Precambrian iron formations. The Chungju iron formation was mainly formed by the chemical precipitation, on the other hand, the Seosan iron formation was formed by alternated action of chemical and detrital depositions.

Unfolding of Ervatamin C in the Presence of Organic Solvents: Sequential Transitions of the Protein in the O-state

  • The folding of ervatamin C was investigated in the presence of various fluorinated and non-fluorinated organic solvents. The differences in the unfolding of the protein in the presence of various organic solvents and the stabilities of O-states were interpreted. At pH 2.0, non-fluorinated alkyl alcohols induced a switch from the native $\alpha$-helix to a $\beta$-sheet, contrary to the $\beta$-sheet to $\alpha$-helix conversion observed for many proteins. The magnitude of ellipticity at 215 nm, used as a measure of $\beta$-content, was found to be dependent on the concentration of the alcohol. Under similar conditions of pH, fluorinated alcohol enhanced the intrinsic a-helicity of the protein molecule, whereas the addition of acetonitrile reduced the helical content. Ervatamin C exhibited high stability towards GuHCl induced unfolding in different O-states. Whereas the thermal unfolding of O-states was non-cooperative, contrary to the cooperativity seen in the absence of the organic solvents under similar conditions. Moreover, the differential scanning calorimetry endotherms of the protein acquired at pH 2.0 were deconvoluted into two distinct peaks, suggesting two cooperative transitions. With increase in pH, the shape of the thermogram changed markedly to exhibit a major and a minor transition. The appearance of two distinct peaks in the DSC together with the non-cooperative thermal transition of the protein in O-states indicates that the molecular structure of ervatamin C consists of two domains with different stabilities.

충남 대천해수욕장과 서천군 마량리 지역에 분포된 남포층군 변성퇴적암층의 변성지구조 진화 (The Tectono-metamorphic Evolution of Metasedimentary Rocks of the Nampo Group Outcropped in the Area of the Daecheon Beach and Maryangri, Seocheon-gun, Chungcheongnam-do)

  • 충남 대천해수욕장부근과 서천군 마량리 인대에 변성역암, 변성사암, 천매암 등으로 구성된 변성퇴적 암층이 노출되어 있다. 원암의 구성으로 볼 때 남포층군 중의 조계리층에 대비되며, 대표적인 변성광물군은 흑운모-백운모-석영(${\pm}$사장석${\pm}$녹니석)과 흑운모-백운모-석류석-석영(${\pm}$사장석${\pm}$녹니석)으로 중압변성의 각섬암상중 석류석대에 속한다. 지질온도압력계에 의해 계산된 온도-압력조건은 $560{\sim}595^{\circ}C$$6.9{\sim}8.2\;kb$이다. 천매암에서 분리된 흑운모의 K-Ar 연대는 $143.2{\pm}3.6\;Ma$, $122.6{\pm}2.4\;Ma$$124.8{\pm}2.4\;Ma$인데 뒤의 두 연대는 후기에 열적교란을 받은 것으로 판단된다. 기 발표된 연대측정에 의하면 대동누층군의 생성시기는 $187{\sim}175\;Ma$로 (Han et al., 2006; Jeon et al., 2007) 이 연구의 결과들과 조합하여 대보조산운동기에 일어난 이 지역에서의 변성지구조적 진화과정을 유추하였다. 남포층군 퇴적 직후인 175 Ma 부근에 습곡작용과 중첩된 트러스트운동으로 지각의 두께증가가 시작되었고, 정점변성작용을 거친 후 지각확장에 따른 정단층운동으로 흑운모의 폐쇄온도까지 빠르게 삭박되면서 냉각되는 과정을 겪었다.
