Long Distance Identification of Water and Oil using an Ultraviolet Fluorescence Measurement System (원거리의 물과 오일을 구별할 수 있는 UV형광측정시스템 개발과 분석에 대한 연구)

  • Owing to the rising volume of seaborne trade, oil spills damage the marine environment for over 250 yearly. Thus, various analysis methods such as the Fourier-transform infrared (FTIR), Raman spectroscope, and gas chromatography are used to monitor oil spills at sea, but these methods are expensive. Recently, to reduce operational costs, an underwater fluorometer was adopted. However, this approach is not ideal for the remote sensing of oil spills because the device gets submerged in the sea. In this study, we have designed and developed a monitoring system that uses ultraviolet fluorescence to detect spilled oil or water from a distance, as well as proposed an analyzing method defining based on water Raman signal and QF535. Each fluorescence spectrum of water, oil (crude oil), and Bunker A was obtained using the system, and was calculated and analyzed from the spectrum individually. Based on the results of the analysis, we could successfully identity water and oil at a long distance.

Effect of light and sediment grain size on the vertical migration of benthic diatoms

  • Using chlorophyll fluorescence, the vertical migration of benthic diatoms responding to light intensity and affected by sediment grain size was studied. Minimal fluorescence ($F_o$) of surface sediment was measured by imaging pulse amplitude modulated (Imaging-PAM) fluorometer, and used to monitor diatom biomass variation in surface sediments. The test diatoms, Amphora coffeaeformis (C. Agardh) K$\ddot{u}$tzing and Cylindrotheca closterium (Ehrenberg) Reimann & Lewin, migrated to the sediment surface under irradiance from 50 to 500 ${\mu}mol$ photons $m^{-2}s^{-1}$. However, the diatoms exhibited no evident increase of surface biomass under dark conditions, and even showed slightly decrease of surface biomass under irradiances over 1,000 ${\mu}mol$ photons $m^{-2}s^{-1}$. The light intensity inducing the maximum surface migration of A. coffeaeformis was 100 ${\mu}mol$ photons $m^{-2}s^{-1}$, while the light intensity producing the same effect for C. closterium was 250 ${\mu}mol$ photons $m^{-2}s^{-1}$. C. closterium showed higher motility than A. coffeaeformis. Faster diatom surfacing was observed in larger grain size sediments (125-335 ${\mu}m$) than smaller ones (63-125 ${\mu}m$). This study confirmed the significant influence of light as a main triggering factor behind migration, indicated the distinct effect of different sediment grain size, and highlighted the species-specific migratory ability.

Effects of Asian Dust (KOSA) Deposition Event on Bacterial and Microalgal Communities in the Pacific Ocean

  • Atmospheric aerosol deposition caused by Asian dust (KOSA) events provide nutrients, trace metals, and organic compounds over the Pacific Ocean that enhance ocean productivity and carbon sequestration and, thus, influence the atmospheric carbon dioxide concentrations and climate. Using dust particles obtained from the snow layers on Mt. Tateyama and the surface sand of Loess Plateau in incubation experiments with natural seawater samples on a shipboard, we demonstrate that dust-particle additions enhanced the bacterial growth on the first day of incubation. Gram-positive bacterial group and alpha-proteobacteria were specifically detected form seawater samples including the mineral particles. Although the remarkable dynamics of trace elements and nutrients depend on dust-particle additions, it is possible that organic compounds present in the mineral particles or transported microbial cells could also contribute to an increase in the quantities of bacteria. The chlorophyll concentrations at fractions of every size indicated a similar pattern of change between the seawater samples with and without the dust-particle additions. In contrast, the chlorophyll measurement using submersible fluorometer revealed that the dynamics of phytoplankton composition were influenced by the dust-particles treatments. We conclude that the phytoplankton that uses the bacterial products would increase their biomass. We show that KOSA deposition can potentially alter the structures of bacterial communities and indirectly influence the patterns of marine primary production in the Pacific Ocean.

The Effect of Lactobacillus Mixture Culture Fluid Extracts on Atopic Dermatitis Chemokine Expression of in HaCaT Cells (HaCaT 세포에서 Lactobacillus 혼합배양액 추출물이 아토피관련 케모카인 발현에 미치는 효과)

  • Objectives : Recently the case of lactobacillus mixture culture fluid appliment was reported. In this study, anti-inflamation effects and anti-allergy effects were studied by stimulus of lactobacillus mixture culture fluid extracts in HaCaT cells. Methods : The atopic dermatitis were induced by TNF-${\alpha}$ and interferon-${\gamma}$ in HaCaT cells. TARC/CCL17, MDC/CCL22, RANTES/CCL5 and ROS production were investigated to explain anti-inflamation and allergy effects of lactobacillus mixture culture fluid with cell-enzyme-linked Immunosorbent assay in 450 nm, 485 nm, 535 nm with spectro-fluorometer. Results : The extracts of lactobacillus mixture culture fluid were decreased TARC/CCL17, MDC/CCL22, RANTES/CCL5 expressions and ROS production with a concentration dependent manner. Conclusions : The effects mechanism of Lactobacillus mixed culture fluid for atopic dermatitis symptoms were considered to be explain anti-inflamation and allergy effects via control of cytokine, chemokine and ROS production, and the fluid could be applied in skin cells directly. But classified AD symptom degrees reported in clinical case before as Reaction Period, Reduction Period, Effect Period, Reproduction Period and Rebound Period could not be explained. Further study will be expected.

Physiological Responses of Porphyra yezoensis Ueda (Bangiales, Rhodophyta) Exposed to High Ammonium Effluent in a Seaweed-based Integrated Aquaculture System

  • Porphyra yezoensis is known to act as a biofilter against nutrient-rich effluent in seaweed-based integrated aquaculture systems. However, few studies have examined its physiological status under such conditions. In this study, we estimated the photosynthetic activity of P. yezoensis by chlorophyll fluorescence of PSII (${\Delta}F/F'm$ and relative $ETR_{max}$) using the Diving-PAM fluorometer (Walz, Germany). In addition, bioremediation capacity, tissue nutrients, and C:N ratio of P. yezoensis were investigated. The ammonium concentration in seawater of seaweed tank 4 decreased from $72.1{\pm}2.2$ to $33.8{\pm}0.4{\mu}M$ after 24 hours. This indicates the potential role of P. yezoensis in removing around 43% of ammonium from the effluents. Tissue carbon contents in P. yezoensis were constant during the experimental period, while nitrogen contents had increased slightly by 24 hours. In comparison with the initial values, the ${\Delta}F/F'm$ and $rETR_{max}$ of P. yezoensis had increased by about 20 and 40%, respectively, after 24 hours. This indicates that P. yezoensis condition improved or remained constant. These results suggest that chlorophyll fluorescence is a powerful tool in evaluating the physiological status of seaweeds in a seaweed-based integrated aquaculture system.

The Electrochemical Chlorination for Marine Plankton Community Disinfection (해양 플랑크톤 군집의 전기분해 염소소독 효과)

  • To confirm whether or not the Electrochemical Disinfection System (EDS) meet with the D-2 regulation established by IMO (International Maritime Organization), the biological treatment efficacy of the EDS was assessed using three groups of natural marine plankton (bacteria, $10-50\;{\mu}m$ and $>50\;{\mu}m$ sized organisms). Influent water was passed through the EDS under the flow velocity ($23.8\;m^3/hr$) and test design was consisted of control (no treatment) and experimental (10 ppm and 30 ppm) condition for total residual chlorine (TRC). And the biological condition of the influent water followed the standards established by the guidelines for the approval of ballast water management systems. The disinfection efficacy of the $10-50\;{\mu}m$ sized organisms (phytoplankton) was assessed by three kinds of measurements using photomicroscope, epifluorescence microscope and fluorometer (fumer Designs 10-AU). After being passed through the EDS, all motile phytoplankton lost their motility under photomicroscope, the colour of chlorophyll fluorescence fumed from red into green under epifluorescence, and the high chlorophyll fluorescence (Expt. 1: 6.95, Expt. 2: 7.11) detected by fluorometer decreased into value not detected. These results indicated phytoplankton community was totally killed after electrochemical disinfection treatment. Survivorship of the larger organisms than $50\;{\mu}m$ was determined based on the appendage's movement under a stereomicroscope. Natural assemblage collected from ambient seawater was killed shortly after being passed through the EDS, whereas some Artemia remained alive. However, no live Artemia was found after 24 hour further exposure to each TRC concentration (10 and 30 ppm) under darkness. After electrochemical treatment, the target bacteria such as aerobes, coliform and Escherichia coli were completely killed on the basis of CFU (colony forming unit) on Petrifilm plate ($3\;M^{TM}$) after 48 hr incubation. Moreover, no regrowth was found in the three groups of plankton during five days under additional exposure to the treated water. These results indicated that the disinfection efficiency of the EDS on the three groups of plankton satisfy D-2 regulation.

Enhanced PHB Accumulation in Photosystem- and Respiration-defective Mutants of a Cyanobacterium Synechocystis sp. PCC 6803 (Synechocystis sp. PCC 6803의 에너지 대사 결함 돌연변이 균주에서의 Poly(3-hydroxybutyrate) 축적량 증진)

  • Photoautotrophic bacteria are promising candidates for the production of poly(3-hydroxybutyrate) (PHB) since they can address the critical problem of substrate costs. In this study, we isolated 25 Tn5-inserted mutants of the Synechocystis sp. PCC 6803 which showed enhanced PHB accumulation compared to the wild-type strain. After 5-days cultivation under nitrogen-limited mixotrophic conditions, the intracellular levels of PHB content in these mutants reached up to $10-30\%$ of dry cell weight (DCW) comparable to $4\%$ of DCW in the wild-type strain. Using the method of inverse PCR, the affected genes of the mutants were mapped on the completely known genome sequence of Synechocystis sp. PCC 6803. As a result, the increased PHB accumulation in 5 mutants were found to be resulted from defects of genes coding for NADH-ubiquinone oxidoreductase, O-succinylbenzoic-CoA ligase, photosystem II PsbT protein or histidine kinase, which are involved in photosystem in thylakoid inner membrane of the cell. The values of $NAD(P)H/NAD(P)^+$ ratio in the cells of these mutants were much higher than that of the wild-type strain as measured by using pulse-amplitude modulated fluorometer, suggesting that PHB synthesis could be enhanced by increasing the level of cellular NAD(P)H which is a limiting substrate for NADPH-dependent acetoacetyl-CoA reductase. From these results, it is likely that NAD(P)H would be a limiting factor for PHB synthesis in Synechocystis sp. PCC 6803.

Prevalence of Fumonisin Contamination in Corn and Corn-based Feeds in Taiwan

  • The purpose of this study was to investigate the prevalence of fumonisin contamination in corn and corn-based feeds in Taiwan. A total of 233 samples was collected from 8 feed mill factories located in four different regions in Taiwan. The presence of fumonisin $B_1$ ($FB_1$) and $B_2$ ($FB_2$) was determined by thin layer chromatograph, while the total fumonisin content was determined using immuno-affinity column cleanup and fluorometer quantitation. Our results showed that 55 samples of swine feeds had the highest percentage of incidence of $FB_1$ and $FB_2$ (41.8% and 41.8%, respectively), followed by 66 samples of duck feeds (40.9% and 37.8%). However, the percentage of incidence of $FB_1$ and $FB_2$ was much lower in 43 samples of broiler feeds (23.2% and 13.9%) and 69 samples of corn (17.3% and 10.1%). Corn and duck feeds were found to have a significant higher level of means of total fumonisins ($5.4{\pm}1.5$ and $5.8{\pm}0.6$ ppm, respectively) than swine feeds ($2.9{\pm}0.4$ ppm) and broiler feeds ($3.0{\pm}0.5$ ppm). Comparing fumonisins distribution in different regions, the highest percentage of $FB_1$ incidence (39.2%) was found in the eastern region of Taiwan, and total fumonisins level ($4.5{\pm}0.7$ ppm) was significantly higher than other regions. However, the highest percentage of $FB_2$ incidence (32.0%) was found in the central region of Taiwan. Trimonthly analysis of data showed that both high percentage of $FB_1$ and $FB_2$ incidence (39.3% and 37.7%) and total concentration of fumonisin ($5.7{\pm}0.4$ ppm) were found in the period of Jan. to Mar., The incidence and concentration were significantly higher than other trimothly periods. These results indicate that fumonisin B mycotoxins are both widespread and persistent in feed-grade corn and corn-based feeds in Taiwan.

Evaluation and Application of Algae Online Analyzer for Alarming Algal Bloom and Water Quality Management of Korean Reservoirs (조류발생 경보 및 호수 수질관리를 위한 엽록소 자동측정기의 적용 및 타당성 연구)

  • In order to evaluate the application of Algae Online Analyzer (AOA), an instrument of automatic measurement of chlorophyll a concentration, was tested and compared with the acetone extraction method on the basis of microscopic counting of phytoplankton in field water (Paltang Reservoir). We simultaneously conducted AOA operation and extraction method with the same water sample, to compare both results of chlorophyll a measurement. Phytoplankton were enumerated by inverted microscope with the Sedgwick-Rafter chamber, and classified into the genus or species. According to the AOA measurement, the diatom most (83.6%) strongly contributed to the total chlorophyll a concentration, followed by chlorophyceae> cyanophyceae>cryptophyceae. Overall, the results of both AOA and extraction method showed a similar trend and significant correlation (r=0.87, n=302, p<0.001), however, there were some differences according to the season and species. In particular, the relationship between AOA Chl-a density of the diatom (r=0.73, p=0.010) and cyrptophyceae (.=0.83, p=0.00154) were siginificant, while chlorophyceae (r= -0.13) and cyanophyceae (r= -0.16) showed no clear relationship during the study period. Although we can not fully understand why there was difference between both mothods, AOA application for alarming algal bloom and water quality management during the algal bloom appears to be very relevant. However, the further study or technical upgrade of AOA measurement is required, especially in the case of low density of phytoplankton or species-specific measurement.

The Effect of Light Intensity on the Growth and Chlorophyll Fluorescence Parameters of Three Ardisia Genus Native to Korea

  • This study investigated the growth and chlorophyll fluorescence reactions of three Ardisia genus grown under various indoor light intensity conditions with the aim of evaluating their suitability as indoor plants. Young seedlings of A. crispa (Thunb.) A.DC., A. pusilla DC., and A. japonica (Thunb.) Blume were used in the experiment. The plants were cultivated indoors for 10 weeks under different light intensities: 10, 50, 100, and 200 PPFD (μmol·m-2·s-1), and their growth was compared with that of plants cultivated in a greenhouse during the same period (mean value 236.8±20.4 PPFD at noon). Also, chlorophyll fluorescence analysis was investigated with a portable PAM fluorometer. The indoor plants were maintained at 12/12 h photoperiod, temperature at 25±1℃, and humidity at 55±3%. Irrigation frequency (once every three days) was the same for the indoors and the greenhouse. The results of growth in three Ardisia plants showed that almost all parameters except leaf number and chlorophyll content had similar levels regardless of light intensity. A. crispa and A. pusilla plants grown in 200 PPFD were investigated to have low chlorophyll contents. Meanwhile, chlorophyll fluorescence parameters differed based on light levels. In A. crispa, the Fv/Fm (0.77), DIo/RC (0.47) and Fm/Fo (4.77) parameters tended to be poor at 200 PPFD compared to those at other light intensities. Similarly, the DIo/RC, Fm/Fo, and Pi_Abs parameters of A. pusilla plant (200 PPFD) are 0.45, 4.48 and 2.42, respectively, which can be considered stress. The analysis of fluorescence in A. japonica showed that all parameters except ETo/RC had similar levels regardless of light intensity. The ETo/RC parameter was 0.49 and 0.72 in the control plants and plants 200 PPFD, respectively, which was lower than those in plants at other light intensities. Therefore, it seems that the relatively high light intensity acted as a stressor for Ardisia plants.
