Determination of Brassinolide by HPLC equipped with Fluoresence Detector in Rice(Oriza sativa L.) (HPLC 형광분석법을 통한 벼에서 Brassinolide의 검정)

  • To determine brassinolide in rice(Oriza sativa L.) using HPLC equipped with fluoresence detector, a highly sensitive fluorescence reagent. 1-cyanoisoindole-2-m-phenylboronic acid, was synthesized from the reaction of o-phthaldehyde, m-phenylboronic acid and KCN, then was reacted with brassinolide. The formation ratio of brassinolide boronate exhibited 90% up at the ratio of $20\;:\;1({\mu}g/{\mu}g)$ of 1-cyanoisoindole-2-m-phenylboronic acid and brassinolide respectively. The detection limit of brassinolide boronate with fluoresence detector was 0.16 ng. Brassinolide was detected in heading stage(biomass : 10 g) and panicle formation stage(biomass : 100 g) of the rice(Oryza sativa L.) with quantity of $0.8\;{\mu}g\;and\;0.2\;{\mu}g$respectively. However, brassinolide was not detected in blooming and elongation stage.

  • A fiber optic pH sensor has been fabricated using nile blue entrapped in an ammonia catalyzed silica sol-gel film coated on glass substrate by dip-coating. The sensor was fixed on the end of an optical fiber. The sensor showed pH sensitivity when dipped into liquids at different pHs. Linear and reproducible responses were obtained in standard buffer solutions in the pH range $6.0{\sim}8.5$, which encompasses the clinically-relevant range. The effects of interferences on the determination of pH were also investigated. The sensors were successfully applied to the determination of pH in different commercial ionic drinks.

  • The effect of vesicular L-$\alpha$-phosphatidylcholine on the rate of formation of all-trans-N-retinylidene-n-butylamine (3) and on the regeneration kinetics of bacteriorhodopsin pigment from retinal and bacterio-opsin have been studied. An estimate of the relative positions of retinal and n-butylamine in the vesicles has been made by fluoresence quenching experiments. Partition coefficient of retinal and microviscosity of the retinal-binding region have also been determined. The results are discussed in terms of the nature of chemical interaction between retinal and amine in a lipid environment.

  • The study of the research for Random process have been recently increasing rapidly. There are many methods in generating of Random signal, however, mainly these are dependent upon utilizing of hot noise of resistance and noise of discharge tube. Consequently, it is not easy to obtain of Random Noise of stabilized low frequency. Therefore, I like to study over the result of principle and design in the method of obtaining the Random Noise with faint radiations of fluoresence materials.

  • A series of novel pseudopeptides contained coumarin skeleton were synthesized through the Ugi-four-component reaction. The 3-substituted coumarin-3-carboxamides were formed through reaction of benzaldehyde derivatives, anilines, coumarin-3-carboxylic acid and isocyanides with high yields and high bond-forming efficiency at room temperature. These novel amidated coumarins exhibit brilliant fluorescence in range of 535-547 nm in chloroform.

  • To investigate the effect of ginseng extracts on the changes of flllidity and lysis of liposome reverse phase evaporation vesicle (REV) was employed as model membrane and methylene blue was used as photosensitizer. Fluoresence polarization (P vaule) that represented fluidity of liposome was increased by photooxidation. All of the ginseng saponin inhibited the increasing rate of P value; the order of effect was ginseng water extract>biol saponin>triol saponin>crude saponin. In trapped G-6-P% measurement for lysis of liposome, ginseng water extract and crude saponin promoted the lysis of liposome. Therefore, we thought that ginseng extracts acted as both antioxidant and detergent.

  • Simultaneous measurements of turbulent velocity and concentration field in a stirred mixer tank are carried out by using PIV/LIF technique. Instantaneous velocity fields are measured with a 1K$\times$1K CCD camera adopting the frame straddle method while the concentration fields are obtained by measuring the fluorescence intensity of Rhodamine B tracer excited by the second pulse of Nd:Yag laser light. Image distortion due to the camera view-angle is compensated by a mapping function. It is found that the general features of the mixing pattern are quite dependent on the local flow characteristics during the rapid decay of mean concentration. However, the small scale mixing seems to be independent on the local turbulent velocity fluctuation.

  • Most of the previous studies on natural convection were based on the numerical analysis, and some experimental studies considered the classic case of thermal convection. In this study, an adiabatic square body was located at the center of an enclosure between the bottom hot and top cold walls. And the measurement of the detailed temperature fields was conducted by the method of two-color Laser Induced Fluorescence using a Nd:Yag laser. As a result, heat transfer characteristics of natural convection with an adiabatic body was estimated as a function of time over a range of Rayleigh numbers.
