Analysis of Functional Components of Leaves of Different Mulberry Cultivars (뽕잎 품종별 기능성 성분 분석)

  • Quantification of functional components, such as [l -deoxynojirimycin (DNJ), ${\gamma}$-aminobutyric acid (GABA) and flavonoids] of four different cultivars (YK-209, Cheongil, Yongchon and Kaeryang) and parts (upper, middle and lower) of mulberry leaves harvested in different areas and periods was performed by HPLC to select the best Quality of mulberry leaf suitable for manufacture of functional foods. Among four mulberry cultivars, YK-209 mulberry leaf had the highest DNJ, GABA and flavonoids levels. YK-209 mulberry leaf harvested in "Yeongcheon" area had greater DNJ, GABA and flavonoid contents than those harvested in "Sangju" area. Four major flavonoids of mulberry leaf were rutin, isoquercitrin, kaempferol-3-Ο-rutinoside, astragalin, and their levels varied greatly with cultivars. YK-BO9 mulberry leaf had the highest flavonoids content among four cultivars. DNJ, GABA and flavonoid contents of young YK-209 mulberry leaf were relatively higher in the upper part than those in the middle and lower parts. Additionally, levels of DNJ, GABA and flavonoids were higher in mulberry leaves harvested in the eariler growing season (May) than in the late growing seasons (June and August) . Particularly, among four flavonoids, rutin content had the highest in the mulberry leaf harvested in May, while isoquercitrin content did the highest in the mulberry leaf harvested in June and August. These results suggest that YK-209 mulberry leaf having high amount of functional components, such as DNJ, GABA and flavonoids, may be useful as potential source of beverages and tablets.verages and tablets.

Anti-adipogenic effect of the flavonoids through the activation of AMPK in palmitate (PA)-treated HepG2 cells

  • Background: Flavonoids are natural polyphenols found widely in citrus fruit and peel that possess anti-adipogenic effects. On the other hand, the detailed mechanisms for the antiadipogenic effects of flavonoids are unclear. Objectives: The present study observed the anti-adipogenic effects of five major citrus flavonoids, including hesperidin (HES), narirutin (NAR), nobiletin (NOB), sinensetin (SIN), and tangeretin (TAN), on AMP-activated protein kinase (AMPK) activation in palmitate (PA)-treated HepG2 cells. Methods: The intracellular lipid accumulation and triglyceride (TG) contents were quantified by Oil-red O staining and TG assay, respectively. The glucose uptake was assessed using 2-[N-(7-Nitrobenz-2-oxa-1,3-diazol-4-yl)amino]-2-deoxy-d-glucose (2-NBDG) assay. The levels of AMPK, acetyl-CoA carboxylase (ACC), and glycogen synthase kinase 3 beta (GSK3β) phosphorylation, and levels of sterol regulatory element-binding protein 2 (SREBP-2) and 3-hydroxy-3-methylglutaryl-CoA reductase (HMGCR) expression were analyzed by Western blot analysis. The potential interaction between the flavonoids and the γ-subunit of AMPK was investigated by molecular docking analysis. Results: The flavonoid treatment reduced both intracellular lipid accumulation and TG content in PA-treated HepG2 cells significantly. In addition, the flavonoids showed increased 2-NBDG uptake in an insulin-independent manner in PA-treated HepG2 cells. The flavonoids increased the AMPK, ACC, and GSK3β phosphorylation levels and decreased the SREBP-2 and HMGCR expression levels in PA-treated HepG2 cells. Molecular docking analysis showed that the flavonoids bind to the CBS domains in the regulatory γ-subunit of AMPK with high binding affinities and could serve as potential AMPK activators. Conclusion: The overall results suggest that the anti-adipogenic effect of flavonoids on PA-treated HepG2 cells results from the activation of AMPK by flavonoids.

Optimal Ethanol Extraction Conditions of Domestic Propolis and Its Chemical Composition (국내산 프로폴리스의 에탄올 최적추출조건 및 그 화학성분)

  • To determine the characteristics of propolis fermented with microorganisms, the optimal conditions for propolis extraction with ethanol were first reviewed, and the physicochemical properties of the extracted propolis were then investigated. The shaking and stationary extraction methods were tested. The flavonoid content was 6.40 mg/g when the stationary extraction method was used and 6.95 mg/g when the shaking extraction method was used. The ethanol concentration was then varied, and the use of 50, 60, 70, and 80% ethanol resulted in the total flavonoid content in the propolis samples reaching 6.75, 8.45, 7.45, and 7.75 mg/g, respectively. This indicated that a 60% (v/v) concentration was optimal. As the extraction time was increased from 1 to 3 hr, the flavonoid content slightly increased; however, extending the extraction step to more than 3 hr did not significantly affect the flavonoid content. In terms of the extraction temperature, the highest concentration of flavonoids (8.63 mg/g) was detected at 50℃. In the propolis samples, 5.80, 2.61, and 1.32 mg/g of fructose, sucrose, and rhamnose were detected, respectively. As for the amino acid content, 17 amino acids, including glutamic acid, were detected, and the total amino acid content was 2.49 mg/g.

Comparison of Antioxidant Potentials in Methanolic Extracts from Soybean and Rice Fermented with Monascus sp.

  • The potential antioxidant activities of methanolic extracts from soybean and rice fermented with Monascus sp. were investigated. M. pilosus IFO 480 and M. anka IFO 478 were screened as a suitable strain to promote the antioxidant activities in soybean- and rice- fermentation. The methanol extracts from soybean and rice after fermenting for 20 days at $30^{\circ}C$ resulted in a significant increase in the antioxidant capacities expressed as radical (ABTS and DPPH) scavenging assay and peroxidation inhibition (%) by thiocyanate method and increased (p<0.01) by a 2.6 to 3.1-fold compared with those of the unfermented products. The average antioxidant potentials of Monascus-fermented soybean extracts (MFSE) were significantly (p<0.01) stronger than Monascus-fermented rice extracts (MFRE). A linear correlations between free radical scavenging activity of MFSE and the total phenolics content (r=0.84) and total flavonoids content (r=0.81) were observed. These results indicated that MFSE exhibited stronger (p<0.01) antioxidant activity and contained significantly higher levels (p<0.05) of phenolics than MFRE.

Comparison of Amino Acid, Mineral Compositions and Antioxidant Activity of Apple and Crab Apple (사과와 꽃사과의 구성 아미노산, 무기질 조성 및 항산화 활성 비교)

  • To confirm the applicability of crab apple, the composition and content of amino acids and minerals with 'Fuji' apple were measured. Apple and crab apple were extracted with water, 70% and 100% ethanol, respectively. The contents of polyphenols, flavonoids, and ascorbic acid, antioxidant activities of these extracts were measured. The amino acid composition of apples comprised a total of 17 amino acids. The total amino acid contents of apple and crab apple were 2,050.45 mg/kg and 900.05 mg/kg, respectively. For minerals, the total mineral content of apple and crab apple were 489.14 mg%, 529.77 mg%, respectively. The contents of polyphenols, flavonoids, and ascorbic acid of apple and crab apple extracts were highest in 70% ethanol extracts. The content of polyphenols, the crab apple extract, showed a generally higher content than the apple extract. The content of flavonoids, apple and crab apple extracts revealed no difference between extracts. The content of ascorbic acid, apple extract showed a generally higher content than the crab apple extract, but there was no significant difference between extracts. In the case of DPPH and ABTS radical scavenging abilities, 70% ethanol extract showed the highest antioxidant activity, and crab apple showed higher activity than apple extracts.

The Anti- and Pro-oxidative Effects of Orally Administered Flavonoids in Normal Rats

  • The present study was designed to investigate the effects of genistein, daidzein, and quercetin on the antioxidative systems of normal rats. Male Sprague-Dawley rats were divided randomly into seven groups and treated with flavonoids at either 2 or 20 mg/day or through vehicle for four weeks. Lipid peroxidation in the liver was inhibited significantly following administration of quercetin. Genistein and daidzein did not have significant effects except in rats treated with 20mg daidzein/day. Genistein and daidzein treatment did not affect the content of $\alpha$-tocopherol in the serum and liver, while quercetin caused a slight increase. In hepatic glutathione and its related enzymes, genistein and daidzein treatment tended to cause a decrease in $\alpha$-tocopherol content, although no significant difference was found. However, quercetin treatment significantly decreased the content of glutathione together with the activity of glutathione reductase in all doses in the liver but there was no significant difference in the brain. Interestingly, daidzein treatment in the brain at 2mg/day significantly increased glutathione (27.1% p<0.05) compared with the control group, while at 20mg/day glutathione decreased significantly (26.6%, p<0.05). In conclusion, genistein has not antioxidant effects. Daidzein quercetin may have the capacity to produce not only antioxidants but also have adverse effects including the production of pro-oxidants. Therefore, people should consider consumption at a high dosage.

Antioxidant Activities of Mixed Grains (혼합잡곡의 항산화 활성 비교)

  • The purpose of this study was to investigate the levels of antioxidant activities in some traditional and a few super food mixed rice. Amongst nine chosen samples, traditional five grains were analyzed and results revealed highest content of crude-protein ($8.05{\pm}1.11%$) and lowest crude-fat content ($1.74{\pm}0.29%$); however, the calories was found to be relatively low ($358.05{\pm}0.34kcal$) in the samples. Total polyphenols, flavonoids and tannins were extracted from nine samples with 80% methanol and biochemical activity was measured. The content of total polyphenols, flavonoids and total tannin was $206.5{\sim}452.0{\mu}g/mL$, $0.126{\sim}0.340{\mu}g/mL$, and $548.1{\sim}774.8{\mu}g/mL$, respectively. The traditional five grains showed the highest values except for DPPH radical scavenging activity. DPPH radical scavenging activity was 5 to 40.3% higher in eight samples than the traditional five grains sample. From these results, it is conjectured that a mixture of five grains, might exhibit equal or considerably higher effect as healthy diet when compared to super food. The results from this study would serve as basic data for the use of traditional mixed grains rice diet for good health.

Changes of Some Flavonoids in the Peel of Late Maturing Citrus during Maturation

  • Eleven flavonoids, including rutin, naringin, hesperidin, neohesperidin, quercetin, naringenin, kaempferol, hesperetin, nobiletin, 3,5,6,7,8,3',4'-methoxylated flavone and tangeretin in the peel of late maturing citrus fruit species of Mucott (smith tangerine), Singamha (C. natsudidai), Sambogam (C. sulcata), Hongpalsak (C. hassaku), Seminol (Dancy tangerine) and Jawdung (C. aurantium) harvested during from August to December were analyzed with HPLC. The mobile phase consisted of water and acetonitrile with 0.5% acetic acid. Wavelength in UV detector was determined at 254 nm. Naringin and neohesperidin content in the peel of Jawdung harvested at the early stage of maturation were 34.02 mg/g and 13.68 mg/g, respectively, and it was highest among the tested citrus fruits. Hesperidin content in the peel of Mucott harvested at the earthy stage of maturation was 12.48 mg/g. Rutin content of Sambogam harvested at the early stage of maturation was 5.13 mg/g. Quercetin, naringein, kaempferol, nobiletin, 3,5,6,7,8,3',4'-methoxylated flavone flavonoids were in trace. Flavonoid contents of Singamha, Sambogam and Jawdung were high in the peel of fruits at the early stage of maturation, after which time they decreased rapidly.

Anti-Inflammatory Effect of Quercetagetin, an Active Component of Immature Citrus unshiu, in HaCaT Human Keratinocytes

  • Citrus fruit contain various flavonoids that have multiple biological activities. However, the content of these flavonoids are changed during maturation and immature Citrus is known to contain larger amounts than mature. Chemokines are significant mediators for cell migration, while thymus and activation-regulated chemokine (TARC/CCL17) and macrophage-derived chemokine (MDC/CCL22) are well known as the typical inflammatory chemokines in atopic dermatitis (AD), a pruritic and chronic inflammatory skin disease. We reported recently that the EtOH extract of immature Citrus unshiu inhibits TARC and MDC production. Therefore, we investigated the activity of flavonoids contained in immature Citrus on TARC and MDC levels. As a result, among the various flavonoids, quercetagetin has stronger inhibitory effects on the protein and mRNA expression of TARC and MDC than other flavonoids. Quercetagetin particularly has better activity on TARC and MDC level than quercetin. In HPLC analysis, the standard peak of quercetagetin matches the peaks of extract of immature C. unshiu. This suggests that quercetagetin is an anti-inflammatory component in immature C. unshiu.

Quantitative Analysis of Flavonoids in the Unripe and Ripe Fruits and the Leaves of Four Korean Rubus species (한국산 4종 Rubus속 숙과, 미숙과 및 잎의 플라보노이드 함량 분석)

  • The five flavonoids, isoqueritrin, juglanin, astragalin and 2-O-trans-p-coumaroyl astragalin were quantitatively analyzed in the leaves, unripe- and ripe fruits of four Korean Rubus species including Rubus crataegifolius, R. pungens var. oldhami, R. parvifolius, and R. coreanus belonging to the Rosaceae family. The HPLC analysis demonstrated that flavonoids are more abundant in the leaves than in the fruits. In the four Rubus species, ripe fruits generally contained similar or more amount of flavonoids than in unripe fruits. In particular, the quantity levels of total flavonoid (49.81${\pm}$0.50 mg/g) and isoquercitrin (35.08${\pm}$0.37 mg/g) were highest in the leaf of R. crataegifolius. In addition, the content of the flavonoids were quite low in the unripe fruits of R. coreanus that is most widely used as Rubi Fructus.
