Understanding the Phenomenon of "Clear Qi Below, Turbid Qi Above" with Reference to Symptom Patterns of the Sanghallon (Treatise on Cold Damage 傷寒論) ("청기재하(淸氣在下), 탁기재상(濁氣在上)"에 대한 고찰(考察) - 『상한론(傷寒論)』 병증(病症)과의 비교 -)

  • Objective : Describe the phenomenon of "clear qi below, turbid qi above" as found in the Somun Eumyangeungsangdaelon (Major Essay on Yinyang Resonances and Appearances 素問 陰陽應象大論) and compare this pattern with water-grain dysentery and flatulence symptom patterns in the Sanghallon (Treatise on Cold Damage). Method : Study the annotation of the Hwangjenaegyeong (Yellow Emperor's Internal Classic 黃帝內經) and compare the results with the Sanghallon's water-grain dysentery and flatulence. Conclusions and Results : The causes of water-grain dysentery and flatulence are associated with the ascending and descending properties of Yin and Yang. Additionally, these symptoms can also be caused by pathogenic heat, turbid pathogenic factors, and interruption of the movement of clear and turbid qi. Aspects of water-grain dysentery resemble several patterns found in the Sanghallon. If caused by a weakness of yang qi, it resembles Sayeoktang (四逆湯) syndrome. Weakness of spleen qi resembles Ijungtang (理中湯) syndrome. Flatulence is similar to fullness in the chest syndrome, which in the Sanghallon is caused by an obstruction of cold qi. If there is excessive cold, water-grain dysentery is similar to the syndrome of Gyeolhyung (結胸). If the qi is not scattered, deficiency syndrome is similar to Gyejigejagyaktang (桂枝去芍藥湯) syndrome and excess syndrome is similar to Mahwangtang (麻黃湯) syndrome. When flatulence is caused by fever in chest, it is similar to Chijasitang (梔子?湯) syndrome. When caused by heat and phlegm build up in chest, it is similar to Sipjotang (十棗湯) syndrome.

Quantitative Determination of Flatulence Factors in Legume Seeds and Soy Products (두류 및 대두제품중 가스발생인자의 함량 분석)

  • The contents of flatulence factors such as raffinose and stachyose were determined by two-dimensional thin-layer chromatography using a guide-strip technique for some legume seeds and soy products consumed in Korea. Raffinose contents in bean samples were 1.34% in domestic soybean, 1.17% in imported soybean, 0.65% in kidney bean, 0.49% in red bean and 0.49% in mung bean. Stachyose contents were 4.1% in doemstic soybean, 3.6% in imported soybean, 3.0% in kidney bean, 2.9% in red bean and 1.9% in mung bean. The flatulence factors were not detected in soy sauce whereas soy paste contained 46.6mg% of raffinose and none of stachyose. The contents of raffinose and stachyose were 26.7mg% and 53.7mg% in soy curd and 26.5mg% and 41.7mg%, respectively, in soy milk.

  • This study was undertaken to investigate for the possible use of enzymes with ${\alpha}-galactosidase$ and protease activities to remove flatulence factors as well as to improve the yield and protein digestibility in soymilk preparation. The volume and protein yield were not increased significantly by enzyme treatment. The solids yield increased by raising treatment pH 6 to 10, the temperature $30^{\circ}C\;to\;60^{\circ}C$ Enzyme treatment brought about a remarkable increase in TCA-soluble nitrogen compounds and a decrease in the contents of flatulence factors raffinose and stachyose.

  • The contents of the several antinutritional factors and lipoxygenase activity of 19 soybean varieties recommended for cultivation in Korea were analyzed. The ranges of raffinose and stachyose contents, which are the flatulence factors, were $0.74{\sim}1.58%\;d.b.\;and\;3.34{\sim}5.30%d.b.$, respectively and the total amount of these sugars was high in the varieties of Hill, Baekun and Jangbaek. The contents of trypsin inhibitor and phytate phosphorus in the soybean varieties ranged from 21.2 to 37.0 TI/g, d.b. and from 337 to 605mg%, d.b. respectively. The lipoxygenase activity of $163.6{\sim}403.5unit$ was shown in the 19 soybean varieties, and Padal, Jangbaek and S-133 were the varieties with the higest activity.

Production of ${\alpha}$- and ${\beta}$-Galactosidases from Bifidobacterium longum subsp. longum RD47

  • Approximately 50% of people in the world experience abdominal flatulence after the intake of foods containing galactosides such as lactose or soybean oligosaccharides. The galactoside hydrolyzing enzymes of ${\alpha}$- and ${\beta}$-galactosidases have been shown to reduce the levels of galactosides in both the food matrix and the human gastrointestinal tract. This study aimed to optimize the production of ${\alpha}$- and ${\beta}$-galactosidases of Bifidobacterium longum subsp. longum RD47 with a basal medium containing whey and corn steep liquor. The activities of both enzymes were determined after culturing at $37^{\circ}C$ at pH 6.0 for 30 h. The optimal production of ${\alpha}$- and ${\beta}$-galactosidases was obtained with soybean oligosaccharides as a carbon source and proteose peptone no. 3 as a nitrogen source. The optimum pH for both ${\alpha}$- and ${\beta}$-galactosidases was 6.0. The optimum temperatures were $35^{\circ}C$ for ${\alpha}$-galactosidase and $37^{\circ}C$ for ${\beta}$-galactosidase. They showed temperature stability up to $37^{\circ}C$. At a 1 mM concentration of metal ions, $CuSO_4$ inhibited the activities of ${\alpha}$- and ${\beta}$-galactosidases by 35% and 50%, respectively. On the basis of the results obtained in this study, B. longum RD47 may be used for the production of ${\alpha}$- and ${\beta}$-galactosidases, which may reduce the levels of flatulence factors.

${\alpha}$-Galactosidase from Bacillus megaterium VHM1 and Its Application in Removal of Flatulence-Causing Factors from Soymilk

  • A bacterial strain capable of producing extracellular ${\alpha}$-galactosidase was isolated from a sample of sugarcane industrial waste. Microbiological, physiological, and biochemical studies revealed that the isolate belonged to Bacillus sp. Furthermore, based on a 16S rDNA sequence analysis, the new isolate was identified as Bacillus megaterium VHM1. The production of ${\alpha}$-galactosidase was optimized based on various physical culture conditions. Guar gum and yeast extract acted as the best carbon and nitrogen sources, respectively. The optimum pH was 7.5 and the enzyme remained stable over a pH range of 5-9. The enzyme was optimally active at $55^{\circ}C$ and thermostable with a half-life of 120 min, yet lost 90% of its residual activity within 120 min at $60^{\circ}C$. One mM concentrations of $Ag^2$, $Cu^2$, and $Hg^{2+}$ strongly inhibited the ${\alpha}$-galactosidase, whereas the metal ions $Fe^2$, $Mn^{2+}$, and $Mg^{2+}$ had no effect on the ${\alpha}$-galactosidase activity, and $Zn^{2+}$, $Ni^{2+}$, and $Ca^{2+}$ reduced the enzyme activity slightly. When treated with the B. megaterium VHM1 enzyme, the flatulence-causing sugars in soymilk were completely hydrolyzed within 1.5 h.

  • ${\alpha}$-Galactosidase from Aspergillus niger as a possible enzyme for removal of flatulence factors in soybean foods was produced the highest in 120 hours in either Czapeck-Dox liquid medium or wheat bran solid medium. The most efficient carbon and nitrogen sources in Czapeck-Dox medium were raffinose and sodium nitrate, respectively, whereas the addition of the sources showed negative effects in wheat bran. pH optima for enzyme activity and stability were 4.0-5.0 and 3.5-6.5, respectively, and optimum temperature for stability was $40-50^{\circ}C$. Upon reaction on p-nitrophenyl-${\alpha}$-D-galactoside, Michaelis constant was 0.42 mM and maximum velocity was 152 ${\mu}moles$ substrate/minute/kg solid medium. Mercuric chloride acted as a strong noncompetitive inhibitor and p-chloromercuribenzoate, even in low concentration, acted as a competitive inhibitor. Crude ${\alpha}$-galactosidase hydrolyzed raffinose and stachyose completely, giving spots of monosaccharides only on thin-layer chromatogram.

The Complaints Rates of Minor Discomforts and Its Influencing Factors. (임신중 경증불편감 호소율과 영향 요인에 관한 연구)

  • Various minor discomforts are reported to be complained by the pregnant women. but what were and how much were they complained has not been revealed until recently. So, These lack of knowledge have given nurses difficulties in planning of care to promote the health during the pregnancy. Objectives of this study were to identify the rate of complaints in each minor discomforts and to explore the influencing factor on reduction of complaints of minor discomforts. The subjects were 120 mothers, who delivered their babies in hospital, from September to December 1990 and the reason why the postpartum mothers were selected as the subjects were minor discomforts could develope during the entire period of pregnancy. Data were gathered by the questionnares and analized statistically using SAS and SPSS program. Results were as follows. 1. More than one third of the subjects complained frequent urination, fatigue, increase of the vaginal discharge, morning sickness, the increased urination at night, pain in leg, backache, edema in leg, constipation, dyspnea, varicose vein, flatulence, headache. 2. Minor discomforts complained to be severe in more than one third of the subjects were frequent urination and increased urination at night. 3. The influencing factor to reduce the complaints of minor discomfort was revealed to be the perceived family support (r=0.030431, t=0.0007). We suggest that nurses should emphasize the importance of the family support to the pregnant women and their family, and to explore the relationships among the locus of control, the perceived family support and the complaints of minor discomforts.

  • ${\alpha}-galactosidase$ releases galactoside from raffinose and stachyose which are the major sugars in soybean, Although raffinose and stachyose were known as flatulence factors, these sugars were recently claimed as bifidus factors. In this experiment we studied the properties of ${\alpha}-galactosidase$ and its production from Bifidobacterium sp. Int-57. Int-57 produced higher level of ${\alpha}-galactosidase$ than other intestinal bacteria. The production of ${\alpha}-galactosidase$ was greater when grown on raffinose compared with other carbohydrates tested. Partially purified ${\alpha}-galactosidase$ was obtained after sonication of harvested cell pellet followed by DEAE-cellulose chromatography and Sepharose CL-6B gel filtration, and assayed using PNP-${\alpha}-galactosidase$ as a substrate. Optimum pH for activity was 7.0 and optimum temperature was $40^{\circ}C$. At 5 mM concentration of metal ions, $CoCl_{2}\;and\;CuCl_{2}$ and inhibited the enzyme activity by 33% and 21% respectively. The enzyme was shown to hydrolyse genuine substrates, i.e. raffinose and stachyose.

Genotype and Environment Influence on Raffinose and Stachyose Content of Soybean Seed (콩 종자의 Raffinose 및 Stachyose 함량에 대한 유전자형과 환경의 영향)

  • Soybean (Glycine max (L.) Merr.) is an important crop for protein, oil, carbohydrates, isoflavones, and many other nutrients to humans and animals. But, antinutritional factors in the raw mature soybean are exist. Raffinose and stachyose are main antinutritional factors in soybean seed. Both raffinose and stachyose are carbohydrates, belonging to the raffinose family of oligosaccharides (RFOs). RFOs are not readily digested in humans and cause flatulence or diarrhea. The objective of this research is to obtain the information on raffinose and stachyose content according to genotype and environment. A total of twenty two soybean genotypes (11 cultivars, 3 germplasms and 8 breeding lines) were selected. Each genotype was grown in the field for two years with two replications and harvested in bulk at natural maturity for two years. Content of raffinose and stachyose was detected by HPLC. The raffinose content (g/kg) of 22 genotypes was $2.68{\pm}0.21-5.87{\pm}2.43$ in year 1 and was $3.24{\pm}0.37-9.05{\pm}0.16$ in year 2. The stachyose content (g/kg) was $4.23{\pm}0.98-27.68{\pm}9.90$ at year 1 and was $5.11{\pm}1.09-25.32{\pm}0.35$ in year 2. Genotype and environment have highly significant effects on raffinose and stachyose content. Three genotypes (Da-7, 116-13, and RS-78) have low stachyose content at 5% significant level in two years. A positive correlation ($R^2=0.1985^*$) between raffinose and stachyose was observed in year 2. These informations are valuable in soybean genetics and breeding program related with raffinose and stachyose content.
