Fluctuations in the Abundance of Common Squid, Todarodes pacificus and Environmental Conditions in the Far East Regions during 52 Years

  • Environmental variables, fishing and biological data of the common squid, Todarodes pacificus were used to describe changes in structure, migration and abundance of the squid population in relation to ocean climate shifts. It was possible to consider the main groups of the squid (autumn and winter-spawned groups) as a single population to aid conservation in the waters around Korea and Japan (TWC and KOC regions). The patterns of yearly fluctuations in abundance of the squid population in the two regions were the same during 52 years of $1952{\sim}2003$. The abundance of the squid began to decrease in both regions in the early 1970s, remained low in the 1980s and the main squid groups synchronously increased in the 1990s coincident with favorable changes of thermal conditions and plankton production in those ecosystems. The mechanisms of changes in the structure, distribution and abundance of common squid population in relation to current-mediated migration circuits are explained on the basis of phenological variables responding to climate shifts.

봄철 제주 남부해역 난·자치어의 수직 분포 (Vertical Distribution of Icthyoplankton in the Southern Waters of Jeju Island During Spring)

  • The vertical distribution and abundance of icthyoplankton in the southern waters of Jeju Island during June 2020 were investigated. Fish eggs and larvae were identified using the mitochondrial DNA cytochrome c oxidase subunit I (mtDNA COI) and the 16S rRNA gene. During this period, fish eggs of 23 taxa belonging to 21 families and larvae of 27 taxa belonging to 25 families were collected. Fish eggs were located mostly from the surface to 30 m depth of the water column. Larvae were located from the surface to 80 m depth of the water column. Vertical distributions of fish eggs and larvae were influenced by oceanography conditions such as temperature, salinity, and thermocline depth. No discernible difference in mean thermocline depth was observed between day and night.

Establishment of Conditions for Long-Term Maintenance of Primary Embryonic Cell Cultures from Olive Flounder Paralichthys olivaceus

  • Primary cell culture is a sufficient method frequently used to study the cellular properties and mechanisms of isolated cells in a controlled environment. In this study, an embryonic cell line (FGBC8) derived from the blastula stages of embryos of olive flounder Paralichthys olivaceus was developed. Furthermore, conditions for optimal long-term maintenance of this primary embryonic cell culture were investigated. Morphologically, FGBC8 cells were composed primarily of epithelial-like cells. FGBC8 cells were subcultured for >160 passages over ~830 days. The doubling time of FGBC8 cells was 73.8 h, and the modal diploid chromosome number was 48. FGBC8 cells transfected with green fluorescence protein (GFP)-expression plasmid exhibited a strong signal 48 h after transfection. Consequently, we demonstrated that fish serum is a crucial supplement for the long-term survival and maintenance of comparable morphology in these primary embryonic cells. Our results can be used as a guide for primary embryonic cell cultures for other fish species and may be useful for cell biotechnological applications.

동해 중부연안에 서식하는 대문어(Enteroctopus dofleini) 흘림낚시의 어획실태에 관한 연구 (Catch status of the giant pacific octopus, Enterocotpus dofleini, by drift line fishery in the central coast of the East Sea, Korea)

  • Giant pacific octopus, Enteroctopus dofleini, is a large mollusk distributed in the East Sea of Korea. In this study, the catch status of giant pacific octopus by drift line fishery and the effect of sea temperature on fishing ground were investigated in Goseong-gun, Gangwon-do, which is the central coast of the East Sea. The average catch of giant pacific octopus in Gangwon-do was 1,570 tons over the past ten years, and it accounted for 21% in 2008 and 44% in 2021 compared to the total catch in the East Sea during the same period. Such data indicates that the catch in Gangwon-do has recently increased. In this study area, giant pacific octopus weighing 1.1-5.0 kg dominated accounting for 56% of the total individuals, and followed by those weighing 1 kg or less. However, the ratio of catch of giant pacific octopus over 5.1 kg tended to increase in 2021, which is thought to be related to the sea temperature that affected the fishing ground. The main depth of fishing ground was from 21 m to 50 m in this area and fishing grounds were widely distributed throughout the season except summer. Fishing ground was formed with the conditions of bottom sea temperature under 10℃ and was diminished by moving of giant pacific octopus to deeper depth with conditions of bottom sea temperature over 18℃.

배양 조건이 Photobacterium damselae subsp. damselae의 미생물학적 성상 및 병원성에 미치는 영향 (Effects of the microbiological properties and pathogenicity of Photobacterium damselae subsp. damselae under different culture conditions)

  • 배양 온도, 배양 배지 중의 pH, NaCl 농도, iron 농도와 같은 배양 조건이 P. damselae의 미생물학적 성상 및 병원성에 미치는 영향을 조사하였다. P. damselae는 배양 온도 15-30${^{\circ}C}$에서 배지의 pH 5-9, NaCl 농도 0-6%와 $FeCl_{3}$ 첨가시 성장이 촉진되었지만, iron-chelator 첨가 시성장이 억제되었다. Iron-limited 조건에서 배양시 ECPs protein 농도, phospholipase, 용혈능, siderophore 및 cytotoxicity 활성 이 높게 나타났고, 넙치 혈청의 살균작용 대한 생존율도 증가 되었으나, iron-added 조건에서는 감소되어, 배양 조건 중 iron 농도에 영향을 가장 많이 받는 것으로 나타났다. 따라서, 본 연구에서 적용한 iron-limited 조건은 철농도가 낮은 숙주 내와 유사한 환경이므로 iron-limited에서 P. damselae 배양 시 나타나는 병원성의 증가는 어체에 감염시 나타나는 병원성 발현 기전과 유사할 것으로 사료된다.

실시간 어장정보 생산 부이시스템 개발 및 활용연구 (Development on Real Time Application System for Fisheries Oceanography Information)

  • 동해중북부 연안역의 해황 변화에 따른 연안 양식 환경변화에 능동적으로 대처할 수 있도록 연안 어장에서 관측한 해양자료를 실시간으로 일반사용자들에게 제공하는 실시간 어장정보 제공시스템을 개발 및 운용할 수 있는 연구를 수행 하였다. 실시간 어장정보 제공시스템은 크게 어장정보 생산 부이시스템, 해양자료 수집서버, 해양자료 저장데이터베이스 서버, 인터넷을 통한 자료제공을 위한 웹 서버로 구성하였다. 또한 인터넷을 통해 최종 이용자들에게 자료를 제공할 뿐만 아니라 원하는 사용자에 대해 휴대전화서비스 빚 문자서비스(SMS)를 제공할 수 있게 하였다. 실시간 어장정보 제공시스템에서 관측한 수온, 유향 유속자료를 이용하여 연안역 저층에서 수 일 이하의 단주기적인 해황변동이 발생함을 정량화 할 수 있었다. 수일 이내 단주기적 해황변동은 어장환경에 영향을 미쳐 어장의 폐사를 유발시키는 원인 중에 하나가 되기도 한다. 따라서 실시간 자료에 의해 이상 해황이 발생하면 수하식 양식시설을 적수온대로 이동 시켜 대량폐사를 막을 수 있으리라 생각된다. 또한 수층별 장기적인 자료의 데이터베이스 구축을 활용하여 수평 및 연직의 양식장 적격지 공간을 과학적으로 도출할 수 있다.

엘리뇨/라니냐와 정상 기간 동중국해 북부해역의 자치어의 군집구조 비교 (Comparison of Community Structure of Fish Larvae in the Northern East China Sea in Normal and El Niño/La Niña Periods)

  • The aim of this study was to compare community structure of larval fish species in the northern East China Sea during normal meteorological conditions in autumn 2009, during the El Ni$\tilde{n}$o period in 2009-2010, and during the La Nina period in 2010. Fifty taxa were recorded during the study period; the most dominant species were Benthosema pterotum and Gobiidae spp. In October 2008 during the normal period, warm water from the Tsushima Warm Current (TWC) intruded more into the surface and middle layers, and cold water affected by the Yellow Sea Cold Water (YSCW) intruded into the bottom layer. In October 2009 during the El Ni$\tilde{n}$o period, intrusion of the China Coastal Water (CCW), which has low salinity (<32.2 psu), was more apparent than intrusion of the TWC or YSCW. In October 2010 during the La Nina period, intrusion of the TWC and CCW was relatively weak, resulting in the lowest temperature and highest salinity observed during the study period in the eastern part of the study area. Hierarchical cluster, one-way ANOSIM (analysis of similarities), and SIMPER (similarity-percentages procedure) analyses provided two main results. First, the abundance of the most dominant larval fish species in autumn of the normal period was greater than that in autumn of the El Ni$\tilde{n}$o/La Nina periods, resulting in a significant difference in ichthyoplankton community structure between the periods. The abundance of Benthosema pterotum increased in the normal period, possibly influenced by the intrusion of cold water from the YSCW; the abundance of species residing in Korean waters (e.g., Gobiidae spp.) probably decreased during the El Ni$\tilde{n}$o/La Nina periods. The second finding was that the abundance of subtropical larval fish in autumn of the normal period was generally larger than that during autumn of the El Ni$\tilde{n}$o/La Nina periods. This could have been induced by the stronger intrusion of warm water from the TWC during the normal period. Although differences in oceanographic conditions between El Ni$\tilde{n}$o and La Nina periods were observed, the differences in ichthyoplankton community structure between the two periods were not significant.

수소생산을 위한 Rhodopseudomonas sphaeroides의 최적 배양조건 (Optimum Culture Conditions for Hydrogen Production of Rhodopseudomonas sphaeroides)

  • We examined optimum culture conditions of Rhodopseudomonas sphaeroides B5 for effective utilization of substrate and sunlight for hydrogen production. The optimum concentration range of DL-lactate as electron donor for hydrogen production by resting cells was from 5 to 50mM, and optimun CN ratio (lactate/glutamat) for maintenence of hydrogen production activity by growing cultures was from 5 to 6. Hydrogen production by the cultures of low cell density (0.36mg/ml dry cells) was saturated with 10 Klux light intensity. Under constant illumination of 50Klux which was set up as the average medium value of annual variation of sunlight intensity, hydrogen production with various cell densities in the culture resulted in highest production rate (132${\mu}$l/hr/mg dry cells) up to 0.64mg/ml dry cells. However, the amount of total hydrogen production was saturated with cell density of 2.1mg/ml dry cells. In addition to these, the optimum inner thickness pervious to light of the culture vessel for hydrogen production which was measured under sunlight was 5 cm.

$F_o$-값 측정 시스템의 개발 (Development of $F_o-value$ Measuring System)

  • 식품의 가열살균공정의 최적화는 에너지 소비의 성력화를 위한 공정 자동화와 제품의 미생물학적 안전성 및 영양적, 기호적 품질을 감안하여 이루어져야 한다. 본 연구에서는 이미 개발한 마이크로컴퓨터를 이용한 자동화 시스템을 개선하여 미생물학적 안전성 확보를 위한 가열살균기준 설정에 필수적인 $F_o-값 측정 시스템을 개발하였다. $F_o-값은 가열살균 중인 명태 고기풀의 냉점에서의 온도변화에 따라 순간치사율(lethal rate)을 구하고 전체 공정에서의 치사율의 합(integrated lethality)을 $F_o-값으로 나타내었다. 또한 정확도는 냉점에서의 열침투곡선으로부터 계산된 열확산도의 정확도로써 검토하였다. $F_o-값은 다양한 온도에서의 미세한 조건의 변화에도 불구하고 정확한 측정이 가능하였다. 또한 다양한 조건에서 동시에 계산된 명태 고기풀의 열확산도는 문헌상에 보고된 추정식으로 구한 열확산도와 큰 차이를 보이지 않았으며, 그 차이는 $\pm12\%$이내였다.
