Phrixocephalus umbellatus (Copepoda : Lernaeidae) from Marine Fish, Branchiostegus japonicus of the Korea Southern Sea

  • A species of the parasitic copepods Phrixcephalus unbellatus (Lernaeide ; Cyclopoidea) from Branchiostegus japonicus is described and reported for the first time in Korea. The parasite was recovered from the eye of host. P. umbellatus was easily identified by the body shape extensive ramification of the antennal processes and numerous branches on the thoracic horns, The parasite inserted its head and the anterior portion of thorax up to the 4th segment in the eye ball of the host through a narrow hole which it usually burrowed near the upper margin of the cornea above the crystalline lens. Prevaklence of the parasite increased from 3.3% January to 11.9% in June.

Current Knowledge of Small Flukes (Digenea: Heterophyidae) from South America

  • Fish-borne heterophyid trematodes are known to have a zoonotic potential, since at least 30 species are able to infect humans worldwide, with a global infection of around 7 million people. In this paper, a 'state-of-the-art' review of the South American heterophyid species is provided, including classical and molecular taxonomy, parasite ecology, host-parasite interaction studies and a list of species and their hosts. There is still a lack of information on human infections in South America with undetected or unreported infections probably due to the information shortage and little attention by physicians to these small intestinal flukes. Molecular tools for specific diagnoses of South American heterophyid species are still to be defined. Additional new sequences of Pygidiopsis macrostomum, Ascocotyle pindoramensis and Ascocotyle longa from Brazil are also provided.

Morphological and Molecular Characterization of Philometroides seriolae from Japanese Amberjack Seriola quinqueradiata caught in East Sea, Republic of Korea

  • The Japanese amberjack Seriolae quinqueradiata is one of the most consumed fish species among the Koreans. However, information regarding parasitic infection in Japanese amberjack is scarce. This study described the morphological and molecular characteristics of a species of philometrid nematode, Philometroides seriolae, which was recovered from Japanese amberjack. This fish was caught in the sea of Goseong-gun, Gangwon-do, Republic of Korea (Korea). Six P. seriolae (Nematoda: Philometridae) were recovered from 2 Japanese amberjacks. These parasites were subgravid female which were 325-420 mm long and 2.95-3.27 mm wide. Furthermore, they had typical papillae distributed on their body surface with 14 papillae at the apical view. Sequence analysis of the small subunits of ribosomal RNA (SSU rRNA) showed high sequence identity (99.8%, 1,607/1,611-bp) with that of P. seriolae (GenBank accession no. FJ155811). This nematode species has been newly added to the Korean nematode fauna.

Peritricha ciliate infection of cultured snakehead, Channa argus (양식 가물치, Channa argus에 폐사를 일으키는 Petrichida 목 섬모충류의 감염)

  • Mortality and gill lesions in snakehead, Channa argus (body length range, 20.3-22.2 cm) reared in a aqua-farm in Busan, Korea, were associated with a dense bloom of Apiosoma-like ciliate, Order Petrichida. The size of parasite was $48.83{\pm}7.75{\mu}m{\times}14.29{\pm}2.66{\mu}m$. Histological examination revealed that a severe edema and collapse of the gill tissue were observed in a number of samples of snakehead. The mechanism of gill damage was likely due to physical irritation by the parasite. It is believed that this is the first report of Apiosoma-like ciliate in cultured fish in Korea.

Occurrence and Molecular Identification of Microcotyle sebastis Isolated from Fish Farms of the Korean Rockfish, Sebastes schlegelii

  • Microcotyle sebastis is a gill monogenean ectoparasite that causes serious problems in the mariculture of the Korean rockfish, Sebastes schlegelii. In this study, we isolated the parasite from fish farms along the coasts of Tongyeong, South Korea in 2016, and characterized its infection, morphology and molecular phylogeny. The prevalence of M. sebastis infection during the study period ranged from 46.7% to 96.7%, and the mean intensity was 2.3 to 31.4 ind./fish, indicating that the fish was constantly exposed to parasitic infections throughout the year. Morphological observations under light and scanning electron microscopes of the M. sebastis isolates in this study showed the typical characteristics of the anterior prohaptor and posterior opisthaptor of monogenean parasites. In phylogenetic trees reconstructed using the nuclear 28S ribosomal RNA gene and the mitochondrial cytochrome c oxidase I gene (cox1), they consistently clustered together with their congeneric species, and showed the closest phylogenetic relationships to M. caudata and M. kasago in the cox1 tree.

Chilodonella hexasticha(Protozoa, Ciliata) from Korean freshwater fish (한국산 담수 어류에 기생하는 Chilodonella hexaticha에 관하여)

  • During a study to document more fully the parasitic ciliates of Korean freshwater fishes, we found numerous small ciliates in the gills and skin of crucian carp, Carassius carassius. Cursory examination indicated that the ciliate were very similar with Chilodonella sp. In this study, we described the morphometrics of this ciliate in detail and compared with other species of Chilodonella in the world. Numerous small ciliates were observed in the body surface, fins and gills of infected fish and excessive mucus production were seen in those fish. Sometimes ulcer was observed in the body of moribund fish. From the scrutinized observation of the parasite, it was identified as Chilodonella hexasticha. The parasite was dorsoventrally flattened body, without a notch in the posterior margin, and it measured 30-$45{\mu}m$ long and 15-$30{\mu}m$ wide. In number of kineties, the right band ciliature was 3-5 and the left band ciliature was 3-5. The right ventral ciliary band shaped arch and was longer than the left, straight band. It had a single oval macronucleus, 8-$15{\mu}m$ in diameter, a single micronucleus, 2-$4{\mu}m$ in diameter. The cytopharynx was reinforced by 10-16 conspicuous nematodesmata, which shaped like a tube and curved at its inner end. Two contractile vacuoles, one anteriorly at right and the second posteriorly at left were observed in wet mounts. This parasite reproduced by a simple division at $22{\pm}1^{\circ}C$.

Study on Extermination of Copepoda, Caligus curtus Parasited to Culture Paralichthys olivaceus (양식 넙치에 기생한 갑각류, Caligus curtus의 구제에 관하여)

  • The number of maximum 63 Caligus curtus was counted culturing P. olivaceus body surface of 20cm mean body length. respectively. Parasite sites were consist of 63.4% head, 26.9% caudal peduncle, 8.3% operculum and 1.4% the other sites. Also, the sites were secreted in larage quantity of mucus with a very small bleeding. This Caligus parasite was died 15 min to 5ppm and 5min to 10ppm masoten but 28.57% was exterminated to fresh water during the four hours. On the other hand, these Caligus parasites were not exterminated during the 30 min to 200ppm formalin.

Isopod Parasite Induced Secondary Microbial Infection in Marine Food Fishes

  • Isopods are parasitic crustaceans that pose serious threat to fisheries. Several studies have tried to explore the host-pathogen relationship between marine fishes and isopods. The present study aims to understanding the secondary infections in marine fishes pertaining to isopods. To assess the secondary infection in infected fishes, parasite infested and healthy tissues of fishes were collected. The samples were subjected to standard microbiological procedure to identify the presence of pathogenic bacteria and fungi. Our results showed the branchial region had the higher microbial load of non-sporulating cenocytic fungi in infected fishes. Moreover, fungal strains isolated from the parasitic lesion confirmed that the parasitation and body lesion facilitates the entry of several pathogenic microbes at the damaged host tissue. More over the immune regulation of fish fights back by producing minute cysts, trying to encapsulate the growing fungus. But this may eventually lead to systemic infestation and death of the fish.

Characterization of proteases isolated from Kudoa septempunctata

  • Proteases play important roles in parasite development and host parasite interactions. The protease of Kudoa spp. has been recognized as a key factor of severe proteolysis of fish muscle post-mortem; however, there is little information available regarding the protease of Kudoa (K.) septempunctata, which was recently identified as a cause of food poisoning in humans. The present study was conducted to isolate and characterize proteases to elucidate the type of protease contained in the parasite and determine the optimal pH for protease activity. We confirmed the cysteine protease and metalloprotease produced by K. septempunctata. While the cysteine protease showed optimal activity at pH 5 that decreased rapidly with increasing pH, the optimal activity of metalloprotease was pH 7, and it remained stable from pH 6 to pH 8. These results indicate that the pH of cysteine protease is not proper for fish muscle postmortem, and that metalloprotease can act in human intestines. Overall, the present study provides important information that improves our understanding of the role of protease physiology and the subsequent food poisoning caused by K. septempunctata.

Intestinal parasite infections in the inhabitants along the Hantrm River, Chorwon-gun (강원도 철원군내 한탄강 유역의 장내 기생충 역학조사)

  • The prevalence of Intestinal parasite Infections In Inhabitants at the reaches of the Hantan River, Chorwon-gun, Korea, was observed from August 12 to September 14, 1993. Of 465 people observed by cellophane thick smear and formalin-ether concentration method, 2 Ascots lumbricoides (unfertilized), 1 Trichwis Dichturn, 39 Clonorchis sinensis and 16 Metagonimus egg positive cases were found. After treatment, the Metogonimus egg positive cases passed out flukes of Metogonimus Mlyata type. Of 68 fish caught In the Hantan River, 14 (20.6%) were Infected with metacercanae of Metogontmn, while no metacercana of C. sinenis was found. At this area, soul-transmitted nematodes are very low, but clonorchlasis and metagonlmlasis are prevalent by modernate endemicity.
