Immunomodulatory characteristics of Streptococcus parauberis isolated from infected olive flounder, Paralichthys olivaceus (넙치, Paralichthys olivaceus 병어에서 분리된 Streptococcus parauberis의 면역 저항 특성)

  • Streptococcal diseases are known as serious problems in fresh water and marine fish culture industry worldwide. Recently, the importance of the infection of Streptococcus parauberis has been increased among the streptococcicosis since severe outbreaks in cultured olive flounder (Paralichthys olivaceus) were recorded in Korea. The aims of the present study were to investigate immunomodulatory differences between S. parauberis and S. iniae. In this study, S. parauberis isolated from diseased olive flounder was investigated on the characteristics of morphological and immune responses. Immune response of the fish to the pathogen was characterized by bactericidal activity in the serum, phagocytic activity and reactive oxygen intermediaters (ROIs) production of olive flounder. The capsules of both bacteria, S. parauberis and S. iniae seem to be associated with the ability to resist killing activity of normal sera. Also, capsulated strains could survive in the phagocytes and induce to lower ROIs production.

  • This study was conducted to determine dietary threonine (Thr) requirement for juvenile olive flounder (Paralichthys olivaceus). A total of 450 juvenile fish (23.2 ± 0.4 g) were randomly distributed to 18 tanks (215 L) with 25 fish per tank. Experimental diets included with graded levels of Thr at 0.0%, 0.4%, 0.8%, 1.2%, 1.6%, and 2.0% were assigned for triplicate groups of fish and fed two times daily to apparent satiation for 12 weeks. Weight gain, specific growth rate, feed intake, feed utilization and survival were significantly (p < 0.05) increased in fish fed with dietary Thr levels over 0.8%, and no significant differences were observed between 0.8% to 2.3% levels. Non-specific immune parameters of serum lysozyme, myeloperoxidase activity, antiprotease activity, and total immunoglobulin were significantly increased by dietary Thr over 0.8%. Based on the broken-line regression analysis, the Thr requirement for the optimum growth and immune response in olive flounder is likely to be 1.03% in the diet.

The Effects of yeast $\beta$-glucan in the Diet on Immune Response of Japanese eel, Anguilla japonica, by Oral Administration (Yeast $\beta$-glucan 첨가 사료가 뱀장어의 비특이적 면역 반응에 미치는 영향)

  • The effects of dietary yeast β-glucan administration on growth, nonspecific immune responses, serum lysozyme, skin mucous lysozyme, NBT (nitroblue tetrazolium) reduction by phagocytes, and disease resistance against Edwardsiella tarda in Japanese eel, Anguilla japonica were evaluated. Fish were fed the diets supplemented with 0%, 0.1% and 0.5% of yeast β-glucan to a commercial diet for 6 weeks. The body weight gain from the fish fed on the 0.5% supplemented diet for 6 weeks was significantly higher than the control. Both serum and skin mucous lysozyme were significantly higher in the all experimental groups except 2 weeks of 0.5% group. The bactericidal activity of serum was slightly increased at 6 weeks. Also, The intracellular superoxide anion production of kidney phagocytes was significantly higher in the all experimental groups. The diet supplemented with 0.1% were also found to raise the relative percent survival (RPS) of Japanese eel after an artificial challenge with 1×107 cells of Edwardsiella tarda per fish. The results suggested the potential of yeast β-glucan to activate some innate immune responses and to improve the growth in Japanese eel.

  • In the present paper, the immunostimulatory effects of the extracellular products (ECP) from Mycobacterium spp. and various adjuvants on the non-specific immune responses of Nile tilapia, Oreochromis nilotica, were examined. Nile tilapia were immunized by injecting ECP of Mycobacterium spp. (strain TB40, TB267 or the type strain Mycobacterium marinum) into their swim bladders. A variety of adjuvants like as Freund s complete adjuvant (FCA), Freund's incomplete adjuvant (FIA) and Titremax were similarly injected into additional groups of tilapia. The number of nitroblue tetrazolium (NBT)-positive cells observed in the swim bladder of the immunized fish was signigicantly increased by the fourth day post-immunization. By day 8, the numbers of NBT-positive cells were fewer in fish immunized with ECP from mycobacteria strains TB40 or TB267 than those immunized with ECP from M. marinum or fish injected with FCA or FIA. The level of Iysozyme activity detected in the serum of fish 40 alter immunization with ECP from various Mycobacterium spp. was also significantly higher than that found in the serum of the control fish. Head kidney macrophages showed enhanced reduction of NBT when cultured in vitro with 1 $\mu$ g/ml of ECP. Concentrations greater than this (10 or 100 $\mu$g/ml) were found to suppress the reduction of NBT by the macrophages. ECP from Mycobacterium spp. and the various adjuvants used in the study all appear to be good activators of the non-specific immune responses of Nile tilapia.

Expressed sequence tags analysis of immune-relevant genes in rock bream Oplegnathus fasciatus gill stimulated with LPS

  • We constructed a rock bream (Oplegnathus fasciatus) gill cDNA library and a total of 1450 expressed sequence tag (EST) clones were generated. Gene annotation procedures and homology searches of the sequenced ESTs were locally done by BLASTX for amino acid similarity comparisons. Of the 1450 EST clones, 1022 EST clones showed significant homology to previously described genes while 428 ESTs were unidentified, and 259 clones were hypothetical, or unnamed proteins. Encoding 313 different sequences were identified as putative bio-defense genes or genes associated with immune response.

Gene annotation procedures and homology searches of the sequenced ESTs were analyzed using BLASTX. Of the 1533 EST clones, 1165 different ESTs showed significant homology to previously described genes while 368 ESTs were unidentified, hypothetical, or unnamed proteins. Encoding 106 different sequences were identified as putative bio-defense genes or genes associated with immune response.

Effect of Short-term Water Restriction on Body Weight, Egg Production, and Immune Response of Local and Commercial Layers in the Late Phase of Production

  • Forty-five Hisex commercial layers and forty-five local Saudi breed layers were used to determine the acceptable limit of short-term water restriction in the late phase of production, when the problem of high feed and water consumption is expected. The experiment was performed under hot and arid environmental conditions when the layers were at fifty weeks of age. Layers from each breed were randomly assigned in groups of five into nine floor pens. The average environmental temperature was 37.2-$38.6^{\circ}C$, and the relative humidity was between 20 to 37%. The trial was divided into 3 periods; control (1 week), water restriction (2 weeks) and rehydration (1 week). During the restriction period, layers from each breed were divided into three groups that received 20, 40, and 0% restriction of drinking water relative to their consumed water during the control period. During the study, feed and water consumption, body weight, changes in body weight, egg production, primary antibody response to SRBC, and rectal temperature were evaluated. Water restriction did not result in any clear effect on feed intake in either breed, however, commercial layers tended to consume less feed compared to the local breed. Body weight declined with water restriction during the first week of restriction in the commercial breed regardless of rate of restriction, but it was delayed until the second week in the local breed. Water restriction of 40% decreased egg production in both breeds but with a delay of 1 week in the local breed. Antibody level to SRBC was not affected by water restriction in the commercial line while it was highly affected in the local breed. A water restriction of 20% is considered to be an acceptable limit under the current experimental conditions without a negative effect on egg production in both breeds and considering the immune status of the local breed. Whereas, 40% restriction had a negative effect on egg production, and varied effects in the other traits in both breeds.

  • The effects of extacts of Omae Prunus mume were tested on non-specific immune response of Japanese eel Anguilla japonica. Lysozyme activity in the serum of the administrated group was presented higher than control, whereas no difference was lysozyme in the mucus or bacterial reaction of the serum. The lysozyme activity of the serum was increased 2 weeks after administrated and highest of that was 5 weeks later. Results of the experimental challenge of Edwardsiella tarda and Aeromonas hydrophila were presented administrated group was higher than control. The results of both groups in the growth rate were same.

  • Interferon regulatory factors (IRFs) are a family of transcription factors essential to the control of antiviral immune response, cell growth, differentiation and apoptosis. IRF10 of zebrafish (Danio rerio) was negative regulation of the interferonΦ1 and 3 response in vitro. In this study, we analyze the induction of in vivo immune response activation from the IRF10 gene of zebrafish and the protective effect against VHSV. As the results, the group inoculated with IRF10 expression vectors, there was no expression of IFNΦ1, suggestion that IRF10 may function as a negative regulator of IRF3, which binds to the IFNΦ1 promoter. And other types of interferon genes (IFNΦ2-4) are thought to have been activated, inducing to the expression of pro-inflammatory cytokine and Mx genes. As the results of challenge test performed at 14 days after inoculation of the expression vectors, the maximum survival rate [50% (1㎍ DNA) and 42.5% (10㎍ DNA)] for IRF10 group were recorded. Meanwhile, the survival rates of pcDNA3.1 and PBS as the control groups were 10% and 15%, respectively. This study suggests that the possibility that activation of IRF10 molecule could be exploited as a VHS control method.

  • Two consecutive studies were conducted to evaluate the effects of dietary supplementation with fermented garlic powder (FGP) or fermented garlic fluid (FGF) on growth performance, immune responses, and disease resistance of olive flounder Paralichthys olivaceus. In experiment I, olive flounder (BW: 65 g) were fed four experimental diets formulated to contain 0%, 0.5%, 1%, and 1.5% FGP (designated as FGP-0, FGP-0.5, FGP-1, and FGP-1.5, respectively). After the 10-weeks feeding trial, feed intake was significantly lower in fish fed the FGP-0.5 and FGP-1.0 diets, as compared to those fed the control diet. Fish fed the FGP-0 and FGP-0.5 diets showed significantly lower survival, as compared to the other treatments. Dietary supplementation with FGP resulted in higher non-specific immune responses than the FGP-0 group. Plasma cholesterol and triglyceride levels decreased as dietary FGP level increased. In experiment II, olive flounder (BW: 65 g) were fed four experimental diets for 10 weeks. The diets were prepared with a commercial expanded pellet to have 0%, 0.25%, 0.5%, and 1% FGF (designated as FGF-0, FGF-0.25, FGF-0.5, and FGF-1, respectively) by adsorption. At the end of the second feeding trial, feed intake was significantly lower in fish fed the FGF-0 diet, as compared to other treatments. Fish fed the FGF-0.25 and FGF-0.5 diets exhibited significantly lower cholesterol levels, as compared to other treatments. Lysozyme activity significantly increased with increases in dietary FGF. Cumulative mortality in a challenge test with Streptococcus iniae was significantly lower in the fish groups fed FGF-supplemented diets than in fish fed the control diet. The results of this study indicated that dietary supplementation with FGP or FGF can enhance the non-specific immune responses and disease resistance of olive flounder against S. iniae.
