Effect of Acibenzolar-S-methyl and Rahnella aquatilis (Ra39) on Chitinase and β-1, 3-glucanase Activities and Disease Resistance of Apple Plants

  • The effect of Acibenzolar-S-methyl (ASM) and Rahnella aquatilis Ra39 against apple fire blight disease caused by Erwinia amylovora were tested as a possible alternative to streptomycin. In vitro studies, no inhibition effect against the pathogen was found when ASM was tested. Under greenhouse conditions, application of R. aquatilis Ra39 with the highly susceptible M26 rootstock resulted in a marked disease suppression. Application of ASM and strain Ra39 caused a high decrease of the disease, 82% and 58% respectively; this was correlated with a reduction of the growth of the pathogen within host plants up to 64% and 49.5% respectively. Further studies in the field under artificial infection condition during full bloom revealed that application of ASM and R. aquatilis Ra39 with Gala variety resulted in a control effect up to 21 and 29% respectively. In physiological studies, enhanced activities of PR-proteins (chitinase and $\beta$-1, 3-glucanase) were detected, which are well known as biochemical markers for systemic acquired resistance. Application of ASM to apple shoots caused the highest chitinase activity followed by strain Ra39. The enzyme activity was increased after 2, 4 and 6 days from application. In addition, ASM-treatment caused the higher $\beta$-1, 3-glucanase activity than strain Ra39. Maximum enzyme activity was recorded after 6 days from application and then decreased after 8 and 10 days from application.

Survey of Oxolinic Acid-Resistant Erwinia amylovora in Korean Apple and Pear Orchards, and the Fitness Impact of Constructed Mutants

  • Fire blight caused by Erwinia amylovora (Ea) is a devastating disease in apple and pear trees. Oxolinic acid (OA), a quinolone family antibiotic that inhibits DNA gyrase, has been employed to control fire blight in South Korea since 2015. The continuous use of this bactericide has resulted in the emergence of OA-resistant strains in bacterial pathogens in other countries. To investigate the occurrence of OA-resistant Ea strains in South Korea, we collected a total of 516 Ea isolates from diseased apple and pear trees in 2020-2021 and assessed their sensitivities to OA. We found that all isolates were susceptible to OA. To explore the possibility of emerging OA-resistant Ea by continuous application of OA, we exposed Ea stains to a range of OA concentrations and constructed OA-resistant mutant strains. Resistance was associated with mutations in the GyrA at codons 81 and 83, which result in glycine to cysteine and serine to arginine amino acid substitutions, respectively. The in vitro growth of the mutants in nutrient media and their virulence in immature apple fruits were lower than those of wild-type. Our results suggest that OA-resistance decreases the fitness of Ea. Future work should clarify the mechanisms by which OA-resistance decreases virulence of this plant pathogen. Continuous monitoring of OA-resistance in Ea is required to maintain the efficacy of this potent bactericide.

  • M.7 rootstock is moderately resistant to fire blight. However, M.7 is generally too vigorous for high-density apple systems, but it can be grafted onto cultivars that exhibit weak tree growth, such as 'Sansa'. This study investigated the vegetative growth, yield, and fruit quality of 'Sansa' apple trees grafted on M.7 or M.9 rootstocks to assess the feasibility of establishing domestic high-density apple systems using M.7 and to determine the optimum exposure length for rootstocks. Trees were planted with exposed rootstock lengths of 5, 10, and 15 cm. The vegetative growth of apple trees grafted onto M.7 was greater than that of M.9 and vegetative growth tended to decrease as the exposed length of rootstock increased. However, the differences in yield per tree, average weights, soluble solids contents, and titratable acidity due to the rootstock and its exposure length varied. The accumulated yield over a 10 year period and the yield efficiency of M.7 were lower than that of M.9 and the yield efficiency tended to decrease as the exposed length of rootstock increased. When apple trees were grafted onto M.9, biennial bearing and tree vigor weakening occurred if the exposed length of the rootstock was over 10 cm. Conversely, when apple trees were grafted onto M.7, vegetative growth was excessive if the exposed length of rootstock was below 10 cm. Based on the results from this study, the optimum M.7 and M.9 exposure lengths for 'Sansa' were 15 cm and 5 cm, respectively.

  • 식물병발생은 농업 체계에서 다양한 요인에 의해 좌우된다. 최근의 광범위한 환경적인 기후 변화로 인하여 기후 온난화, 파종 시기와 품종의 선택 같은 농업 체계가 영향을 받고 있다. 포장에서의 점차적인 병 발생 양상은 직, 간접적인 환경 변화에 영향을 받을 수 있다. 본 연구에서, 콩 품종, 파종 시기, 재배의 지역적인 차이가 우리나라의 콩 병 발생에 미치는 영향을 조사하기 위하여 병발생 빈도를 평가하였다. '태광', '풍산', '청자3호', '새올', '다원' 품종을 5월 15일, 6월 1일, 6월 15일, 6월 30일 4 회에, 수원, 나주 두 지역에 파종하였을 때, 콩 들불병과 불마름병 등이 주요 병으로 발견되었다. 들불병이 '태광' 품종에서 주로 심각하게 발생한 반면, '다원' 품종은 불마름병에 대해 특히 감수성이었다. 들불병과 불마름병의 병 진전은 품종과 관계없이 파종 시기가 지연 될수록 감소하였다.

초분광 이미지를 이용한 배나무 화상병에 대한 최적 분광 밴드 선정 (Spectral Band Selection for Detecting Fire Blight Disease in Pear Trees by Narrowband Hyperspectral Imagery)

  • 화상병이란 erwinia amylovora라는 강한 전염성을 보유하고 있어 감염 시 1년 내에 과수를 고사시키며 그 중심으로 반경 500m이내에 과수 재배를 불가능하게 만드는 세균성 바이러스이다. 이 화상병은 과수의 잎과 가지를 진한 갈색 또는 검은색으로 변색시키기 때문에 분광학적으로 검출이 가능하다고 판단되며 이는 다중분광센서를 탑재한 무인기를 이용하는 것이 효율적이다. 그러나 다중분광센서는 적은 중심 파장과 함께 넓은 반치전폭(FWHM)을 가지고 있어 화상병에 가장 민감하게 반응하는 파장 대역을 파악하기 어렵다. 그렇기 때문에, 본 논문에서는 화상병에 감염된 잎과 가지와 비감염된 잎과 가지의 초분광 이미지를 5 nm FWHM으로 취득한 후 각각 10 nm, 25 nm, 50 nm와 80 nm FWHM로 평준화한 후 샘플을 7:3, 5:5와 3:7의 비율로 훈련데이터와 검증데이터로 나누어 의사결정트리 기법으로 최적의 파장을 선정하고 overall accuracy (OA)와 kappa coefficient (KC)를 이용한 분류 정확도 평가를 통해 배나무 화상병 검출가능성을 확인하였다. 화상병에 감염 및 비감염된 잎과 가지의 초분광 반사율을 비교한 결과, green, red edge 및 NIR 영역에서 차이가 두드러지게 나타났으며 첫 번째 분류 노드로 선택된 파장 영역은 대체로 750 nm와 800 nm였다. 잎과 가지 영역의 영상데이터를 의사결정트리 기법을 이용하여 분류정확도를 종합적으로 비교한 결과, 50nm FWHM 인 4개 대역(450, 650, 750, 950nm)은 10nm FWHM인 8개 대역(440, 580, 660, 680, 680, 710, 730, 740nm)의 분류 정확도 차이가 OA에서 1.8%와 KC에서 4.1%로 나타나 더 낮은 비용의 밴드패스필터인 50nm FWHM을 이용하는 것이 더 유리하다고 판단된다. 또한 기존의 50nm FWHM 파장대역들에 25nm FWHM파장대역들(550, 800nm)을 추가하는 것을 통해 화상병 검출뿐만 아니라 농업에서 다양한 역할을 수행할 수 있는 다중분광센서를 개발할 수 있다고 판단된다.

Analysis of the Stumpage Valuation for Pinus koraiensis Stands in the Research Forest of Kangwon National University

  • This paper assesses the stmpage value of Korean white pine (Pinus koraiensis) stands in the Research Forest of Kangwon National University. Assessment was done by means of the cost value method (Faustmann formula) for age class I, Glaser method for age class II-V, and inversion market method for age class over VI. Generally the value of stumpage is calculated by the inversion market method in the market. However, immature trees and middle age class trees are not assessed in market, and the Korean forest is not old enough to harvest. So, when forests are damaged by attacks from forest fire, blight and harmful insects, the forest cannot be compensated for the loss from the government or insurance company. For this reason, the value of all-age class trees are calculated by using appropriate methods. As a result, the value of age I class stands (0.3 ha) is calculated as 1,786,305 (won), age II-Vclass stands (22.1 ha) 206,677,975 (won) and age VI class and over stands (24.8 ha) 523,789,603 (won).

Comparing Protein Expression in Erwinia amylovora Strain TS3128 Cultured under Three Sets of Environmental Conditions

  • Erwinia amylovora, the causal agent of fire-blight disease in apple and pear trees, was first isolated in South Korea in 2015. Although numerous studies, including omics analyses, have been conducted on other strains of E. amylovora, studies on South Korean isolates remain limited. In this study, we conducted a comparative proteomic analysis of the strain TS3128, cultured in three media representing different growth conditions. Proteins related to virulence, type III secretion system, and amylovoran production, were more abundant under minimal conditions than in rich conditions. Additionally, various proteins associated with energy production, carbohydrate metabolism, cell wall/membrane/envelope biogenesis, and ion uptake were identified under minimal conditions. The strain TS3128 expresses these proteins to survive in harsh environments. These findings contribute to understanding the cellular mechanisms driving its adaptations to different environmental conditions and provide proteome profiles as reference for future studies on the virulence and adaptation mechanisms of South Korean strains.

Dynamics of Bacterial Communities by Apple Tissue: Implications for Apple Health

  • Herein, we explored the potential of the apple's core microbiota for biological control of Erwinia amylovora, which causes fire blight disease, and analyzed the structure of the apple's bacterial community across different tissues and seasons. Network analysis results showed distinct differences in bacterial community composition between the endosphere and rhizosphere of healthy apples, and eight taxa were identified as negatively correlated with E. amylovora, indicating their potential key role in a new control strategy against the pathogen. This study highlights the critical role of the apple's bacterial community in disease control and provides a new direction for future research in apple production. In addition, the findings suggest that using the composition of the apple's core taxa as a biological control strategy could be an effective alternative to traditional chemical control methods, which have been proven futile and environmentally harmful.

Analysis of Endophytic Bacterial Communities and Investigation of Core Taxa in Apple Trees

  • Fire blight disease, caused by Erwinia amylovora, is a devastating affliction in apple cultivation worldwide. Chemical pesticides have exhibited limited effectiveness in controlling the disease, and biological control options for treating fruit trees are limited. Therefore, a relatively large-scale survey is necessary to develop microbial agents for apple trees. Here we collected healthy apple trees from across the country to identify common and core bacterial taxa. We analyzed the endophytic bacterial communities in leaves and twigs and discovered that the twig bacterial communities were more conserved than those in the leaves, regardless of the origin of the sample. This finding indicates that specific endophytic taxa are consistently present in healthy apple trees and may be involved in vital functions such as disease prevention and growth. Furthermore, we compared the community metabolite pathway expression rates of these endophyte communities with those of E. amylovora infected apple trees and discovered that the endophyte communities in healthy apple trees not only had similar community structures but also similar metabolite pathway expression rates. Additionally, Pseudomonas and Methylobacterium-Methylorobrum were the dominant taxa in all healthy apple trees. Our findings provide valuable insights into the potential roles of endophytes in healthy apple trees and inform the development of strategies for enhancing apple growth and resilience. Moreover, the similarity in cluster structure and pathway analysis between healthy orchards was mutually reinforcing, demonstrating the power of microbiome analysis as a tool for identifying factors that contribute to plant health.

대두의 세균성병에 관한 연구 (Studies on Bacterial Diseases of Soybean)

  • 1. 소규모접종시험을 위해서는 wire brush 접종 또는 다침접종법이 가장 효과적이었으며 포장에서의 대규모시험을 위해서는 전착제를 첨가한 세균현탁액의 살포가 좋았다. 2. 조사지역내에서의 발견된 병은 세균성 점무늬병과 세균성 불마름병 뿐이었고 담배 불마름병균에 의한 피해는 발견할 수 없었다. 3. 병원균은 이병조직속에서 겨울철인 경우 실내에서 6개월까지 생존할 수 있었고 냉장고 속에서는 10개월이상 존재했으며 토양속에서는 불마름병균이 4일, 점무늬병균이 30일간 생존하였다. 4. 종자표면에 부착된 세균은 전염원으로서 중요하지 않았으나 실험실내의 배양기 상에서는 종자의 발아를 억제했다.
