Metacercariae of Digenetic Trematoda, Neascus spp. in Chiselmouth, Acrocheilus alutaceus from the Willamette River

  • During a survey of the helminth parasites of fresh water fishes in Willamette River, metacercariae of Neascus spp. were found on examination of muscle, spinal cord, fin and skin of Chiselmouth, Acrocheilus alutaceus. One hundred percent of 19 Chiselmouth examined were found to be infected with digenetic trematodes. The prevalence of infection was higher in muscle, varying from $95\%\;and\;100\%$. Higher levels were present in muscle, such that the $100\%$ level being found in upper anterior, lower anterior and posterior. Small numbers of metacercariae were found on the fin and skin. The commonest sites of infection were the musculature and spinal cord. Intensities of Neascus spp. metacercariae tended to be and high levels in muscle and spinal cord. Those were low levels occurred in the fin and skin. There was a high degree of Neascus spp. in the occurrence of digenetic trematode from Chiselmouth.

Exposing Zebrafish to Silver Nanoparticles during Caudal Fin Regeneration Disrupts Caudal Fin Growth and p53 Signaling

  • Zebrafish were exposed to commercial silver nanoparticles (${\sim}$10-20 nm) at 0.4 and 4 ppm during cadual fin regeneration. The silver was in the $Ag^+$ ionic form. Fin regeneration was slow in the group exposed to the lower concentration. The cadual fin, gill, and muscle were assayed after 48 hours and subjected to histological analysis. In all tissues sampled, fish exposed to nanoparticles exhibited infiltration, large mitochondria with empty matrices, and accumulation of nano-sized silver in blood vessels. The results suggested mitochondrial damage and induction of inflammation. Microarray analysis of RNA from young zebrafish (52 hours post-fertilization) that were exposed to nanometer-sized silver particles, showed alteration in expression of the p53 gene pathway related to apoptosis. Gene expression changes in the nanoparticle-treated zebrafish led to phenotypic changes, reflecting increased apoptosis.

Lipid Class and Fatty Acid Composition of Muscle of Common Squid Todarodes pacificus (살오징어 (Todarodes pacificus)의 근육부위별 지질 Class 및 지방산 조성)

  • The chemical components, lipid class, and fatty acid composition of muscle from male and female common squid, Todarodes pacificus, were examined to evaluate the potential utilization of muscle from fin, head, arms, and tentacles, which consumers usually like less than the mantle. The mantle was found to constitute 47-49% of the total muscle and the proportion was slightly higher in females than in males. For the remaining 51-53% of the muscle, the only gender difference was that the arms of males contained approximately 3% more muscle than those of females (P<0.05). The protein content was higher in the mantle, arms, and tentacles than in the fin and head in both males and females (P<0.05), and was slightly higher in males (15.7-20.7%) than in females (15.1-19.2%). By contrast, the lipid content was slightly higher in females (1.82-2.54%) than in males (1.01-2.37%), and the fins in both males and females contained the most lipids (2.37-2.54%) of all muscle. The prominent lipid classes in the muscles were free sterol (males 81.5-91.9% vs. females 84.9-91.8% for the non-polar lipid content), phosphatidylcholine (PC, males 59.3-62.4% vs. females 49.2-57.8% for the phospholipid content) and phosphatidylethanolamine (PE, males 22.0-28.8% vs. females 25.6-33.8% for the phospholipid content). The percentage of PC was approximately 5-10% higher in males (P<0.05), especially in the fin, while that of PE was approximately 3-5% higher in female (P<0.05), especially in the head. All of the squid muscle contained 52.1-54.9% of n-3 polyunsaturated fatty acids (PUFA), such as docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA). Males contained slightly more DHA, whereas female contained more EPA. The total percentage of n-3 PUFA differed little among muscles within the same gender.

Toxicity of Puffer Fish, Takifugu poecilonotus (Heuinjeombok) and Takifugu vermicularis (Gukmaeribok) from Coastal Water of Korea (연안산 흰점복 (Takifugu poecilonotus)과 국매리복 (Takifugu vermicularis)의 독성)

  • The toxicity of two species of puffer fish, Takifugu poecilonotus (Heuinjeombok) and T. vermicularis (Gukmaeribok) collected from the coastal regions of Korea was determined using a mouse bioassay. In the T. poecilonotus collected in Jeju and Tongyeong, the proportion of toxic specimens containing ${\ge}10$ mouse units (MU) per gram exceeded 95% for the skin, liver, ovary, and fin, and approximately 30% for the testis and muscles. In each of the organs, the highest toxin levels were 79 MU/g in the muscle, hundreds (158-365) of MU per gram in the fin, intestine, testis, and gallbladder, but thousands (1,147-2,406) of MU per gram in the skin, liver, and ovary. In T. vermicularis collected from Incheon and Gunsan, the proportions of toxic specimens were 100% for the gallbladder, and 56-68% for the skin, fin, liver, and intestine however, no toxic muscle specimens were noted. The highest toxin scores were below 10 mouse units (MU) per gram in the muscle, 20-94 MU/g in the skin and fin, 319 MU/g in the intestine, and thousands (1,548-4,624) of MU per gram in the liver, gonad, and gallbladder. The toxicity in the muscle of T. vermicularis was deemed acceptable for human consumption, whereas the toxicities in the muscle of T. poecilonotus and the skin of both species of puffer fish were significantly high, such that special attention may be required when the fish is intended for human consumption.

Comparison of Free Amino Acids and Nucleotides Content in the Olive Flounder Paralichthys olivaceus Fed with Extruded Pellet (건조 배합사료로 사육한 넙치 어육의 유리아미노산 및 핵산 관련 성분의 비교)

  • This study analyzed the taste components of dorsal muscle and fin muscle collected from olive flounder cultured with extruded pellet (EP) compared with those of olive flounder cultured with raw fish moist pellet (MP) as a control. The olive flounder in this study were cultured for 10 months with either the formulated extrude pellets (FEP), commercial extruded pellets (CEP) or the MP feed, and the average weight was 1.15 kg. The major nucleotides and related compounds in the dorsal muscles of the fish fed with MP, CEP and FEP diets were AMP and IMP. The fin muscle of the olive flounder contained the largest amounts of IMP($5.91{\mu}mol/g$). The K value of the dorsal muscle in olive flounder fed with MP, CEP and FEP showed 9.63%, 9.83% and 5.84%, respectively. The free amino acids in the dorsal muscle of olive flounder showed significantly differences in the composition depending on the feed, but the distribution of free amino acids showed similar patterns. Among free amino acids, taurine showed the highest content in all the experimental groups, and the asparagine and citrulline contents were significantly higher in the groups fed with CEP and FEP than in the group fed with MP. The significant differences were observed in the contents of specific free amino acids in dorsal muscle and fin flesh depending on the feed for cultured olive flounder.

Mechanical Design Fabrication and Test of a Biomimetic Fish Robot Using LIPCA as an Artificial Muscle (인공근육형 LIPCA를 이용한 물고기 모방 로봇의 설계, 제작 및 실험)

  • This paper presents mechanical design, fabrication and test of a biomimetic fish robot actuated by a unimorph piezoceramic actuator, LIPCA(Lightweight Piezo-Composite curved Actuator.) We have designed a linkage mechanism that can convert bending motion of the LIPCA into the caudal fin movement. This linkage system consists of a rack-pinion system and four-bar linkage. Four types of artificial caudal fins that resemble caudal fin shapes of ostraciiform subcarangiform, carangiform, and thunniform fish, respectively, are attached to the posterior part of the robotic fish. The swimming test under 300 $V_{pp}$ input with 0.6 Hz to 1.2 Hz frequency was conducted to investigate effect of tail beat frequency and shape of caudal fin on the swimming speed of the robotic fish. At the frequency of 0.9 Hz, the maximum swimming speeds of 1.632 cm/s, 1.776 cm/s, 1.612 cm/s and 1.51 cm/s were reached for fish robots with ostraciiform, subcarangiform carangiform and thunniform caudal fins, respectively. The Strouhal number, which means the ratio between unsteady force and inertia force, or a measure of thrust efficiency, was calculated in order to examine thrust performance of the present biomimetic fish robot. The calculated Strouhal numbers show that the present robotic fish does not fall into the performance range of a fast swimming robot.

Toxicity of the Puffer Fish Takifugu porphyreus and Takifugu rubripes from Coastal Areas of Korea (한국 연안산 검복(Takifugu porphyreus)과 자주복(Takifugu rubripes)의 독성)

  • Toxicity of two species of puffer fish, Takifugu porphyreus and Takifugu rubripes, collected from coastal regions of Korea, was determined using a mouse bioassay, In T. porphyreus, the proportion of toxic specimens containing ${\ge}$ 10 MU/g was 58.3% for the ovary, 32.6% for the skin, 12.0% for the gallbladder, 11.6% for the liver and intestine, and 9.3% for the fin; no toxicity was detected in the muscle and testis using the mouse bioassay. The highest toxin levels were 531 MU/g in the liver, 253 MU/g in the intestine, 136 MU/g in the gallbladder, 118 MU/g in the skin, 116 MU/g in the ovary, and 108 MU/g in the fin. The skin, which is used for human consumption, showed significantly high toxicity with an average of $11{\pm}3\;(mean{\pm}SE) MU/g$. Takifugu porphyreus toxicity also exhibited remarkable regional variation. In T. rubripes, the proportion of toxic specimens was 25.0% for the ovary, 15.8% for the liver, 11.1% for the gallbladder, and 5.3% for the fin and intestine; no toxicity was detected in the muscle, skin, or testis. Among the organs, the highest toxin levels were 228 MU/g in the ovary, followed by 112 MU/g in the liver, 28 MU/g in the gallbladder, 18 MU/g in the intestine, 11 MU/g in the fin, and 8 MU/g in the skin. Thus, we found acceptable toxin levels in the edible muscle and skin of T. rubripes and in the muscle of T. porphyreus. However, the skin of T. porphyreus, which showed significantly high toxicity, requires special attention when used for human consumption.

Quality Characteristics of Cultured Olive Flounder Paralichthys olivaceus Fed with Extruded Pellets; I. Comparison of Fatty Acid and Amino Acid Contents (건조 배합사료로 사육한 양식산 넙치의 어체 품질평가; I. 지방산 및 구성아미노산 조성 비교)

  • This study analyzed the fatty acid and amino acid compositions of dorsal and fin muscles collected from olive flounder cultured using a formula feed(extruded pellets, EP) and compared them to those of fish cultured with a raw fish moist pellet(MP) feed as a control. The olive flounder in this study were cultured for 10 months with either the formulated extruded pellets(FEP), commercial extruded pellets(CEP), or the MP feed, and their average weight was 1.15 kg. The proximate compositions of the dorsal muscle were not different among the groups, whereas in the fin muscles of the fish fed with the MP diet, moisture content was higher and crude lipid content was lower. The major fatty acids in the dorsal and fin muscles of the fish fed with the MP, CEP, and FEP diets were palmitic acid and oleic acid. Finally, no significant differences were observed among the diet groups in the results for amino acid content or the sensory and textural properties of the muscle.

Contents of Total and Organic Mercury in Bone, Muscle and Fin of Carassius carassius Middle Steam of Nakdong River, Korea (낙동강 본류에 서식하는 붕어에서의 총수은 및 유기수은 함량)

  • This study was conducted to examine the levels of mercury in tissue of Carassius carassius collected at five areas(Andong, Sangju, Waekwan, Gangjung and Gaejin) in Nakdong river from February to March, 1993. The contents of total and organic mercury were determined by methods of Gold amalgam and Selective atomic absorption with mercury analyzer(Model SP-3A). The total mercury contents in muscle were $0.148{\pm}0.0646$ ppm by the Gold amalgam, and $0.113{\pm}0.0345$ ppm by the Selective atomic absorption, but in all parts of Carassius carassius were $0.342{\pm}0.4235$ ppm and $0.240{\pm}0.2974$ ppm respectively. These two methods were extremely high correlated(r=0.9812, p<0.001). The mean value of total and organic mercury by the two methods was high in the order of bone>fin>muscle, respectively, and the proportion of organic mercury to the total was 69.0%. Mean value of total and organic mercury in the muscle was the highest at Wakeman located in middle basin of Nakdong river, and in the bone and fin was the highest at Andong dam located in upper basin. According to sizes and parts of fish, the concentration of mercury was the highest in the muscle of 15-19 cm group, the highest in the bone of 10-14 cm group and in the fin of 20-24 cm group.

  • The outbreak of haemorrhagic speticemia and dermal lesions in cultured Rock sea breem in Jeju Island are frequently associated with keratitis. Prolonged infection leads to the symptoms accompanied by necrosis of fin and skin and the fishes died. Present study aims to study the histopathological changes of diseased fish muscle tissue. The tissue at the initial stage of infection showed normal muscular texture, however in the late stages of infection, muscular lesion had been expanded and hence, necrosis had progressed deeply into muscle tissue. Mild tissue in caudal fin was found with more necrosis, and gill tissues were also collapsed. In Kidney, renal tubules were teared along with tissue destruction (Espada, J et al, 1993). Hence in the present study, we examined the histopathological variation of the infection fishes, and this basic data would be used for future research of in haemorrhagic specticemia and dermal diseases.
