Protective Effect of Processed Panax ginseng, Sun Ginseng on UVB-irradiated Human Skin Keratinocyte and Human Dermal Fibroblast

  • In this study, we investigated the protective effects of processed Panax ginseng, sun ginseng (SG) against the UVB-irradiation on epidermal keratinocytes and dermal fibroblasts. Pretreatment of SG in HaCaT keratinocytes and human dermal fibroblasts reduced UVB-induced cell damage as seen by reduced lactate dehydrogenase release. We also found that SG restored the UVB-induced decrease in anti-apoptotic gene expression (bcl-2 and bcl-xL) in these cells, indicating that SG has an anti-apoptotic effect and thus can protect cells from cell death caused by strong UVB radiation. In addition, SG inhibited the excessive expression of c-jun and c-fos gene by the UVB in HeCaT cells and human dermal fibroblasts. We also demonstrated that SG may exert an anti-inflammatory activity by reducing the nitric oxide production and inducible nitric oxide synthase mRNA synthesis in HaCaT keratinocytes and human dermal fibroblasts. This was further supported by its inhibitory effects on the elevated cyclooxygenase-2 and tumor necrosis factor-${\alpha}$ transcription which was induced by UVB-irradiation in HaCaT cells. In addition, SG may have anti-aging property in terms of induction of procollagen gene expression and inhibition of the matrix metalloprotease-1 gene expression caused by UVB-exposure. These findings suggest that SG can be a potential agent that may protect against the dermal cell damage caused by UVB.

Effects of Pharmacological Modulators of $Ca^{2+}-activated\;K^+$ Channels on Proliferation of Human Dermal Fibroblast

  • Employing electrophysiological and cell proliferation assay techniques, we studied the effects of $Ca^{2+}$ -activated $K^+$ channel modulators on the proliferation of human dermal fibroblasts, which is important in wound healing. Macroscopic voltage-dependent outward $K^+$ currents were found at about -40 mV stepped from a holding potential of -70 mV. The amplitude of $K^+$ current was increased by NS1619, a specific large-conductance $Ca^{2+}$-activated $K^+$ (BK) channel activator, but decreased by iberiotoxin (IBTX), a specific BK channel inhibitor. To investigate the presence of an intermediate-conductance $Ca^{2+}$-activated $K^+$ (IK) channels, we pretreated the fibroblasts with low dose of TEA to block BK currents, and added 1-EBIO (an IK activator). 1-EBIO recovered the currents inhibited by TEA. When various $Ca^{2+}$-activated $K^+$ channel modulators were added into culture media for 1∼3 days, NS1619 or 1-EBIO inhibited the cell proliferation. On the other hand, IBTX, clotrimazole or apamin, a small conductance $Ca^{2+}$-activated $K^+$ channel (SK) inhibitor, increased it. These results suggest that BK, IK, and SK channels might be involved in the proliferation of human dermal fibroblasts, which is inversely related to the channel activation.

加味托里消毒飮이 瘡傷 治癒에 미치는 影響 (The Effects of GamiTakliSodocyum on Wound Healing)

  • In order to investigate the effects of GamiTakliSodocyum(GTS) on wound healing, migration of epidermis, formation of granulation tissue and number of capillary within the granulation tissue were measured in diabetic mice by local application and NZW rabbits by local application and prescription of medicine in vivo, and proliferation of human epidermal keratinocytes and human dermal fibroblasts and composition of extracellular matrix were measured in vitro. The results were summerized as follows. 1. $2\%,\;10\%$ GTS remarkably increased migration of epidermis in diabetic mice by local application. 2. $2\%,\;10\%$ GTS remarkably increased formation of granulation tissue, number of neovascularization within the granulation tissue in diabetic mice by local application. 3. 5\%,\;10\%$ GTS remarkably increased migration of epidermis in NZW rabbits by local application. 4. $5\%,\;10\%$ GTS remarkably increased fonnation of granulation tissue, number of neovascularization within the granulation tissue in NZW rabbits by local application. 5. $5\%,\;10\%$ GTS increased migration of epidennis in NZW rabbits by prescription of medicine. 6. $5\%,\;10\%$ GTS increased formation of granulation tissue, number of neovascularization within the granulation tissue in NZW rabbits by prescription of medicine. 7. GTS didn't show effect on the proliferation of human epidermal keratinocytes. 8. GTS increased the proliferation of cultured human dermal fibroblasts. 9. GTS increased the expression of procoliagen ${\alpha}1(I) mRNA in cultured human dermal fibroblasts. 10. GTS increased the expression of fibronectin mRNA in cultured human dennal fibroblasts according to dosage of GTS using northern blot hybridization but didn't increase, using RT-PCR. From the above results, it is conclude that GTS might use on wound healing.

농길리 메탄올 추출물의 세포독성 (Cytotoxicity of the Methanol Extract of Crotalariae sessiliflorae L.)

  • The cytotoxic activity of Cratalariae sessiliflorae on cultured NIH 3T3 fibroblasts and human oral epithelioid carcinoma cells (KB) were evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) colorimetric method These fractions of methanol extract of Cratalariae sessiliflorae showed inhibitory effect in vitro in the milligram range against KB cell lines. In general, the antitumor activities of these fractions were does-dependent over the milligram range. The comparison of IC50 values of these fractions in tumor cell lines showed that their susceptibility to these fractions decrease in the following order: Fr. 4> Fr. 6> Fr. 10> Fr. 2> Fr. 11> Fr. 3> Fr. 8> Fr. 7> Fr. 9> Fr. 1> Fr. 5 by the MTT assay. These fractions were tested for their cytotoxic effects on NIH 3T3 fibroblasts using MTT assay. They exhibited potent cytotoxic activities in vitro in the milligram range against NIH 3T3 fibroblasts. In general, the cytotoxic activities of these fractions were does-dependent over the milligram range. The comparison of CD50 values of these fractions in NIH 313 fibroblasts shows that their susceptibility to these fractions in decrease the following order: Fr. 10> Fr. 9> Fr. 2 = Fr. 4> Fr. 8> Fr. 11> Fr. 1 = Fr. 7> Fr. 3> Fr. 5 = Fr. 6 by the MTT assay. These results suggests that fraction 5 has the most growth - inhibitory activity against KB cell lines.

Comparative N-Linked Glycan Analysis of Wild-Type and α1,3-Galactosyltransferase Gene Knock-Out Pig Fibroblasts Using Mass Spectrometry Approaches

  • Carbohydrate antigens expressed on pig cells are considered to be major barriers in pig-to-human xenotransplantation. Even after ${\alpha}1,3$-galactosyltransferase gene knock-out (GalT-KO) pigs are generated, potential non-Gal antigens are still existed. However, to the best of our knowledge there is no extensive study analyzing N-glycans expressed on the GalT-KO pig tissues or cells. Here, we identified and quantified totally 47 N-glycans from wild-type (WT) and GalT-KO pig fibroblasts using mass spectrometry. First, our results confirmed the absence of galactose-alpha-1,3-galactose (${\alpha}$-Gal) residue in the GalT-KO pig cells. Interestingly, we showed that the level of overall fucosylated N-glycans from GalT-KO pig fibroblasts is much higher than from WT pig fibroblasts. Moreover, the relative quantity of the N-glycolylneuraminic acid (NeuGc) antigen is slightly higher in the GalT-KO pigs. Thus, this study will contribute to a better understanding of cellular glycan alterations on GalT-KO pigs for successful xenotransplantation.

Production of transgenic cattle by somatic cell nuclear transfer (SCNT) with the human granulocyte colony-stimulation factor (hG-CSF)

  • The hG-CSF (human Granulocyte Colony-Stimulating Factor) is a growth and stimulation factor capable of inducing the proliferation of bone marrow cells, several types of leukocytes, among other hematopoietic tissue cells. hG-CSF is used in used to treat anomalies that reder a small number of circulating white blood cells, which may compromise the immune defenses of the affected person. For these reasons, the production of hG-CSF in a bioreactor system using the mammary gland of genetic modified animals is a possibility of adding value to the bovine genetic material and reducing the costs of hG-CSF production in pharmaceutical industry. In this study, we aimed the production of transgenic hG-CSF bovine through the lipofection of bovine primary fibroblasts with an hG-CSF expression cassette and cloning these fibroblasts by the somatic cell nuclear transfer (SCNT) technique. The bovine fibroblasts transfected with the hG-CSF cassette presented a stable insertion of this construct into their genome and were efficiently synchronized to G0/G1 cell cycle stage. The transgenic fibroblasts were cloned by SCNT and produced 103 transferred embryos and 2 pregnancies, one of which reached 7 months of gestation.

Effects of 630-nm Organic Light-emitting Diodes on Antioxidant Regulation and Aging-related Gene Expression Compared to Light-emitting Diodes of the Same Wavelength

  • To investigate the aging-related physiological functions of organic light-emitting diodes (OLEDs), we examined mRNA expression changes in aging-related genes due to oxidative stress inhibition by 630-nm red light OLEDs. As a result of irradiating 630-nm OLED with an intensity of 5 mW/cm2 for 15 min, the viability of dermal fibroblasts significantly increased by 1.3-fold. In addition, reactive oxygen species generated by H2O2 were significantly reduced about 4.9-fold by irradiation with 630-nm OLED. Quantitative reverse-transcription polymerase chain reaction results showed that 630-nm OLEDs altered aging-related gene mRNA expression levels through antioxidant activity. The mRNA expression levels of matrix metalloproteinase1 (MMP1) and MMP9 decreased significantly, by about 2.2- and 2.5-fold, compared to the control group, whereas those of collagen, type I, and alpha 1 increased significantly, by 4.9-fold. The mRNA expression levels of cancer suppression genes p16 and p53 in dermal fibroblasts were also significantly reduced by 630-nm OLED irradiation, by about 1.4- and three-fold, respectively, compared to the control. Overall, it was confirmed that 630-nm OLED irradiation lowered the level of ROS formation induced by H2O2 in dermal fibroblasts, and that this antioxidant effect could regulate the mRNA expression levels of aging- and tumor suppression-related genes. This study shows a link between 630-nm OLED irradiation and anti-aging physiological functions such as antioxidant function, and suggests the potential of OLEDs as a useful light source for skin care.

기계적 표면 처리된 TITANIUM PLASMA SPRAYED IMPLANT에 대한 치은섬유아세포전개양상의 형태학적 관찰 (THE MORPHOLOGICAL OBSERVATION OF HUMAN GINGIVAL FIBROBLASTS ATTACHMENT AND SPREADING ON THE MECHANICAL TREATED TITANIUM PLASMA SPRAYED IMPLANT SURFACE)

  • 구강내 매식된 임프란트가 과도한 교합력이나 염증등의 이유로 구강내로 노출되었을 때 세균독소에 이완된 면을 제거하고 평활한 면을 형성하여 주위의 연조직, 경조직에 적합한 상태로 만들어 건강한 상태로 구강내에 유지하기 위해서 임프란트 매식체 표면을 기계적인 표면처리방법으로 처치하여 이러한 방법이 임프란트 표면성분, 치은섬유아세포의 전개양상에 미치는 영향을 알아보고자 본 실험을 실행하였다. IMZ사에서 제작한 직경 10mm, 높이 2mm의 원판 타이타늄을 이용하여 피막되지 않은 타이타늄면과 TPS면을 대조군으로 하고 기계적인 표면처리방법인 low speed stone bur처치면을 실험군으로 설정한 후 EDX로 타이타늄 표면성분을 분석하였고 주사전자현미경으로 치은섬유아세포의 전개양상을 관찰하였다. EDX에 의한 타이타늄 표면성분분석 결과 모든 실험군에서 titanium peak, 소량의 aluminum이 나타났으며 그외의 성분은 나타나지 않았다. 치은섬유아세포의 전개양상에 대한 주사전자현미경 관찰결과 평활한 타이타늄면에서 접종 30분 후 세사상돌기와 박판엽상으로 확장된 세포가 많이 관찰되며 6시간 후 신장된 치은섬유아세포가 시편에 밀착된 양상을 보였고 24시간 후 치은섬유아세포는 시편의 모든 면을 피개하며 가공시의 평행한 선을 따라 방향성을 띄었다. TPS가 잔존한 stone처치군에서 세포 접종 30분 후 세사상돌기가 적게 관찰되어 평활한 타이타늄면에 비해 초기부착이 늦은 것을 알 수 있었고 6, 24시간후 치은섬유아세포는 거친면으로 인해 시편에 밀착되지 못한 양상을 보였으나 평활한 타이타늄면과 연결되며 시편의 모든면을 피개하였다. TPS군에서 치은섬유아세포는 세포 접종 30분후 세사상돌기를 거의 찾아 볼 수 없어 초기부착이 다른군에 비해 늦으며 세포배양 6, 24시간후에도 시편에 밀착되지 못하고 박판상돌기가 가늘고 길게 돌출되어 여러면에 부착된 양상을 보였으며 세포가 부착되지 않은 TPS면이 관찰되었다.

Alpha-1,3-galactosyltransferase-deficient miniature pigs produced by serial cloning using neonatal skin fibroblasts with loss of heterozygosity

  • Objective: Production of alpha-1,3-galactosyltransferase (${\alpha}GT$)-deficient pigs is essential to overcome xenograft rejection in pig-to-human xenotransplantation. However, the production of such pigs requires a great deal of cost, time, and labor. Heterozygous ${\alpha}GT$ knockout pigs should be bred at least for two generations to ultimately obtain homozygote progenies. The present study was conducted to produce ${\alpha}GT$-deficient miniature pigs in much reduced time using mitotic recombination in neonatal ear skin fibroblasts. Methods: Miniature pig fibroblasts were transfected with ${\alpha}GT$ gene-targeting vector. Resulting gene-targeted fibroblasts were used for nuclear transfer (NT) to produce heterozygous ${\alpha}GT$ gene-targeted piglets. Fibroblasts isolated from ear skin biopsies of these piglets were cultured for 6 to 8 passages to induce loss of heterozygosity (LOH) and treated with biotin-conjugated IB4 that binds to galactose-${\alpha}$-1,3-galactose, an epitope produced by ${\alpha}GT$. Using magnetic activated cell sorting, cells with monoallelic disruption of ${\alpha}GT$ were removed. Remaining cells with LOH carrying biallelic disruption of ${\alpha}GT$ were used for the second round NT to produce homozygous ${\alpha}GT$ gene-targeted piglets. Results: Monoallelic mutation of ${\alpha}GT$ gene was confirmed by polymerase chain reaction in fibroblasts. Using these cells as nuclear donors, three heterozygous ${\alpha}GT$ gene-targeted piglets were produced by NT. Fibroblasts were collected from ear skin biopsies of these piglets, and homozygosity was induced by LOH. The second round NT using these fibroblasts resulted in production of three homozygous ${\alpha}GT$ knockout piglets. Conclusion: The present study demonstrates that the time required for the production of ${\alpha}GT$-deficient miniature pigs could be reduced significantly by postnatal skin biopsies and subsequent selection of mitotic recombinants. Such procedure may be beneficial for the production of homozygote knockout animals, especially in species, such as pigs, that require a substantial length of time for breeding.

피부 섬유아세포에서 광자극의 효과 (The Effect of Photomodulation in Human Dermal Fibroblasts)

  • 피부는 낮 동안 태양빛과 인공 빛에 끊임없이 노출되어 있으며, 그중 5%는 UV 영역, 50%는 가시광선, 나머지 45%는 적외선 영역으로 구성되어 있다. 이중 자외선의 피부에 대한 영향은 많은 연구가 되어 왔으나, 나머지 영역에 대한 연구는 미진한 실정이다. 이에, 가시광선에서 적외선 사이의 파장이 피부 섬유아세포에 어떤 영향을 미치는지 연구하고자 하였다. 광처리에 의한 효과는 광파장, 처리 시간, 광세기, 광조합 등 다양한 파라미터들의 조합에 의해 그 효능이 결정되므로, 본 연구에서는 섬유아세포의 성장 및 콜라겐 합성과 관련된 기능을 촉진시킬 수 있는 광처리 조건을 찾아내고자 하였다. 가시광선과 적외선 영역 사이의 6개의 파장을 처리한 결과, 레드(630 nm)와 그린(520 nm) 파장에 의해 섬유아세포의 증식이 증가함을 확인하였다. 광처리 시간은 콜라겐 합성량 증가를 위해서는 10 min의 광처리가 30 min의 광처리 보다 적합한 조건이었다. 광세기는 $0.05{\sim}0.75mW/cm^2$에서 6개의 광세기로 분할하여 실험한 결과, 레드 $0.3mW/cm^2$와 그린파장 0.15, $0.3mW/cm^2$ 세기가 type I collagen의 mRNA의 양을 증가시킬 수 있었다. 마지막으로 두 개 파장을 순차적으로 조합 처리하였을 때의 효과를 확인한 결과, 레드와 그린파장의 조합 조건은 섬유아세포의 수적증가를 목적으로 할 때 효율적인 방법이며, 콜라겐 합성에는 레드 단독처리가 보다 효과적인 방법이었다. 따라서 본 연구에서 제시하는 광처리 조건을 이용시 피부 세포의 성장이나 콜라겐 합성에 긍정적 영향을 유도할 수 있으며, 재생 및 피부 미용 등에 활용할 수 있는 가능성이 클 것으로 기대된다.
