This study described about preparation of the cross-linked silk fibroin (SF) film by ${\gamma}$-irradiation of the casted SF film, which is fabricated from aqueous solution regenerated via fibers of cocoons and their application as supports for human cell culture. The properties of cross-linked SF film were evaluated by FT-IR spectroscopy, contact angle, solubility to water, thermal analysis, surface area analyzer, and morphology via scanning electron microscopy (SEM). The cross-linked SF films were not dissolved in water and exhibited the rough surface morphology, large surface area, and good thermal properties. The human fibroblast cell (CCD-986sk) and embryo kidney-ft cell were well growed on the surface of cross-linked SF film supports prepared by ${\gamma}$-irradiation. The cross-linked SF film prepared by ${\gamma}$-irradiation can be used as biomaterials for human cell culture.

Reactive oxygen species (ROS) are known to cause oxidative modification of DNA, proteins, lipids and small cellular molecules and are associated with tissue damage and are the contributing factors for diabetes, inflammation, aging, cancer, arteriosclerosis, and hypertension. We screened the anti-oxidative effect on V79-4 hamster lung fibroblast cells induced by hydrogen peroxide with eleven extracts of combined medicinal plants. Dancheonhwankakambang and Samikangyabtang were found to show the scavenging activities of DPPH radical and intracellular reactive oxygen species, which is measured by dichlorodihydrofluorescin diacetate method (DCHF-DA).

The effect of irradiation of x-ray to developing rat mandible in the gestation stage was focused on the study of mandible development and the side effect of x-ray irradiation. The author studied the effect of x-ray irradiation with the gestated rat and their off'springs. 100 rads, 200 rads, 300 rads and 400 rads of x-ray was irradiated in regular order schematically at the lower left abdomen of gestated rat. 18½days after conception, their off'springs were sacrificed and exaimined their developing mandible with histological findings. The results were as followed. 1. In the 100-200 rads irradiated rat off'springs, bony trabeclulation was revealed irregular shape. In combine with this finding, osteoblast and fibroblast were appeared shrunken of their nucleus and location of eccentric position. 2. In the 300-400 rads irradiated rat off'springs, decrease of fibroblast and osteoblast appearance in the periosteum were prominently observed and empty lacunae were frequently appeared in their bone matrix. 3. The advent of osteoclast and resorption of cortical bone were appeared in proportion to increasing of x-ray irradiation.

The expression of genes induced by Dichloroacetate (DCA) treatment was analyzed by mRNA differential display. Purified total RNAs from rat liver treated with saline or DCA (100 mg/100 g b.w.) were reverse transcribed by using a set of oligonucleotide primers. The PCR products were resolved on a denaturing sequencing gel. PCR band representing mRNA expressed specifically in DCA-treated liver was excised and reamplified by PCR. A 120-bp c-DNA clone named IC1 was isolated and the DNA sequence of IC1 was analyzed. IC1 revealed 50% homology with 3' end of a mouse fibroblast growth factor mRNA This result indicates that DCA induces the expression of a gene which has a 50% homology with a Mouse fibroblast growth factor, and expression of this gene might be involved in non genotoxic process caused by DCA.

To investigate the role of heparin in keratinocyte growth factor (KGF) and acidic fibroblast growth factor (aFGF) high affinity binding to the KGF receptor (KGFR), a cell free system was established which utilized a secreted chimeric molecule between the KGFR extracellular domain and the immunoglobulin heavy chain Fc domain (KGFR-HFc). KGFR-HFc was purified from NIH 3T3 cells and demonstrated the binding of $[^3H]-heparin$ as well as heparin Sepharose. Scatchard analysis showed that the dissociation constant for heparin binding to KGFR-HFc was 140 nM. High affinity KGF and aFGF binding to KGFR-HFc remained unchanged after treatment with 0.6 M NaCl, which is the concentration sufficient to release any bound heparin to the KGFR-HFc. These results strongly suggest that although the KGFR interacts with heparin, the presence of heparin is not absolutely required for high affinity binding of either KGF or aFGF to the KGFR.

The purpose of this study was to development of transgenic cow using the nuclear transfer. To secrete hFSH in urea, the vector was constructed with UPII promoter. The fetal fibroblast cells (KbFF) were constructed from pregnant day 45 male fetus. The hFSH genes were cotransfected with pcDNA3 (neo) vector to KbFF cells by electroporation. (omitted)

IPSKCDNA gene(1.8 kbp) encoding rat brain IP3K enzyme contained Not I restric site in open reading frame. The Not I sequence, GCGGCCGC, was converted to GCAGCCGC by site-directed mutagenesis. The mutated IP3KcDNA was digested with EcoR I and ligated with EcoR I-restricted psp72·Not2 vector. The resulting psp72 · Not2-IP3KCDNA was digested with the Not I restriction enzyme and then subcloned into the Not I -digested PZIP · NeoSV(X) mammalian expression vector. The PZIP · NeoSV(X) -IPSKCDNA was transfected into CCL39 hamster lung fibroblast cells. The efficiency of the expressed IPSKCDNA gene was significantly higher than expected generally, not only a mean 5-fold increase in the amount of enzyme, but also 16-fold increase in enzyme activity from tractsfected CCL39 cells by the method of Western blot using anti-lP3K antibodies. Both distribution of IPSK in various rat tissues and biochemical properties were discussed.

Bovine cumulus-oocyte complexes were recovered from ovaries at a slaughter and then divided into five groups: control group(unvitrified oocytes), 0 hr. group(composed of oocytes vitrified before the onset of maturation) and 10, 14, and 20 hrs groups(vitrified respectively at 10, 14 and 20 hrs after the onset of maturation). The oocytes remained vitrified for 24 hrs, and then were thawed in 30℃ water bath. Survival and cleavage rates were defined as development rate on in vitro culture and stained with aceto-orcein or FDA test.

A plasmid vector, $RSVLTR/{\beta}G2$, containing lacZ gene under the control of the RSVLTR promoter were transfected into chicken embryo fibroblasts by three different transfection methods. Calcium phosphate, lipsome and DEAE-dextran techniques were applied for transfection of chicken cells. A histochemical assay with X-gal was used as a simple method for screening transfected cells. Plasmid $RSVLTR/{\beta}G2$ was expressed proficiently in the chicken embryo fibroblast. Calcium phosphate-DNA precipitate transfection resulted in the highest efficiency for transient expression of $RSVLTR/{\beta}G2$. Transfected cells formed colonies on the 9th day of incubation indicating stable transformation of the inserted plasmid.

Antibacterial, free radical scavenging, proliferative, and cytotoxicity effects of Doenjang extracts were examined. All samples except water extract showed strong antibacterial activity against oral bacteria, Streptococcus. pyogenes, S. mutants, S. sanguinis, S. sorbrinus, S. criceti, S. antinosus, S. gordonii, and Porphyromonas. gingivalis (MIC and MBC values: 0.08-1.25 and 0.16-2.50 mg/ml, respectively). DPPH method showed ethanol and ethyl acetate extracts are effective inhibitors of oral bacteria. Based on MTT assay, 24 h exposure to 0.31 mg/ml of all extracts, excepted water extract, resulted in strong cytotoxicity on KB cells. All extracts strongly inhibited human gingival fibroblast viability.
