Antioxidant and anti-inflammatory activities from fruiting body extracts of Lyophyllum decastes

  • Lyophyllum decastes has been used for culinary purpose. The present study was conducted to evaluate the antioxidant and anti-inflammatory effects from methanol, acetone, and hot water extracts of L. decastes fruiting bodies. The acetone and methanol extracts showed the higher 1,1-diphenyl-2-picryl-hydrazy radical scavenging activities than that of the hot water extract at 0.5-2.0 mg/mL and was comparable to the BHT, the positive control. The ferrous ion chelating effects of the mushroom extracts at 0.5-2.0 mg/mL were significantly higher than that of BHT. The reducing power of acetone extract (2.12) was significantly lower than that of BHT (2.73) at 2.0 mg/ mL. The mushroom extracts also showed inhibitory effects on production of nitric oxide (NO), and expression of inducible nitric oxide synthase (iNOS) in lipopolysaccharide-induced murine macrophage cells in a concentration dependent manner. In vivo anti-inflammatory experiment on carrageenan-induced hind-paw edema of rat model, the acetone extract of the mushroom significantly suppressed the carrageenan-induced rat hind paw edema of rats in a dose dependently. The results suggest that the fruiting bodies of Lyophyllum decastes are a good natural resource of antioxidant and anti-inflammation.

Comparison of methods for proanthocyanidin extraction from pine (Pinus densiflora) needles and biological activities of the extracts

  • Flavonoids are known to be effective scavengers of free radicals. In particular, proanthocyanidins are flavonoids that possess cardiovascular protection, antioxidative activities, and immunomodulatory activities. Here, we evaluated proanthocyanidin contents in the total polyphenolic compounds of pine needle extracts prepared by hot water, ethanol, hexane, hot water-hexane (HWH), and hot water-ethanol (HWE). Analysis of each extract indicated that the ethanol extract contained the highest proanthocyanidin concentration. The HWH and hexane extracts also contained relatively high concentrations of proanthocyanidin. On the other hand, proanthocyanidin content analyses out of the total polyphenolic compounds indicated that the HWH extract contained the highest content. These results suggest that HWH extraction is a suitable method to obtain an extract with a high level of pure proanthocyanidins and a relatively high yield. The HWH extract possessed superior activity in diverse antioxidative analyses such as 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferrous ion chelating (FIC), and ferric-ion reducing power (FRAP) assays. In addition, upon assessing the effects of the pine needle extracts on macrophages (Raw 264.7 cell), the HWH extract exhibited the highest activity. In this study, we discerned an efficient extraction method to achieve relatively pure proanthocyanidins from pine needles and evaluated the biological functions of the resulting extract, which could potentially be used for its efficacious components in functional food products.

치자(Gardenia jasminoides Ellis) 과피의 용매별 추출물의 Flavonoid 함량 및 항산화 활성 비교 (Comparison of Flavonoid Content and Antioxidant Activities of Peel Extracts from Gardenia jasminoides Ellis by Various Solvents)

  • The purpose of this study was to measure the bioactivity and antioxidant activity of peel from Gardenia jasminoides fructus Ellis (GJE) in Namhae, Korea, following some established methods. CM (Chloroform:Methanol, 2:1, v/v), 70% ethanol, and n-butanol extracts were collected. Flavonoid content and value as a functional food ingredient of GJE peel was investigated through assessing antioxidant [DPPH (1,1'-diphenyl-2-picrylhydrazyl), ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid)], and hydroxyl radical scavenging activities; superoxide dismutase like ability; ferrous ion-chelating capacity; and tannin content by solvent extraction. Solvent extract antioxidant activities significantly increased (p<0.05) at increasing concentrations (0.2, 0.4, 0.6 mg/mL). GJE peel extracts were less active than the positive control [ascorbic acid, BHA (butylated hydroxyanisole), and EDTA (ethylenediaminetetraacetic acid disodium salt dihydrate)]. Based on the results of this study, GJE peel could be used as a natural antioxidant source due to its high antioxidant activity and bioactive compound content.

Antioxidant and antimicrobial activities of different enzymatic hydrolysates from desalted duck egg white

  • Objective: The objective of this study was to look for optimal preparation of hydrolysates of desalted duck egg white powder (DDEWP) by the three different proteases and to investigate their antioxidant and antimicrobial properties. Methods: DDEWP was hydrolyzed by three proteases, including pepsin (PEP), Bacillus spp. (BA) and natokinase (NAT) with three different enzyme concentrations (0.1%, 0.3%, and 0.5%), individually. The important key hydrolysis parameters such as hydrolysis degree, yield, antioxidant and antimicrobial activity were evaluated in this experiment. Results: The results showed that the degree of hydrolysis (DH) of all treatments increased with increasing hydrolysis time and protease concentrations. The antioxidant and antimicrobial activities of the hydrolysates were affected by type and concentration of protease as well as hydrolysis time. Hydrolysis of PEP significantly (p<0.05) obtained the highest yield of hydrolysates, however, both of BA and NAT showed substantially lower DH values and still did not exceed 5% by the end of hydrolysis. Among the different hydrolysates, PEP exhibited significantly higher 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity than BA and NAT. All DDEWP hydrolysates from PEP had low ferrous ion chelating activity (<37%) that was significantly lower than that of NAT (>37% to 92%) and BA (30% to 79%). Besides, DDEWP hydrolysates of PEP presented significantly higher reducing power than BA and NAT. In antimicrobial activities, Escherichia coli, Salmonella typhimurium, and Pseudomonas aeruginosa were not effectively inhibited by any DDEWP hydrolysates of PEP except for Staphylococcus aureus. Especially, the excellent antibacterial activity against S. aureus only was displayed in DDEWP hydrolysates of PEP 0.1%. Conclusion: DDEWP hydrolysates from PEP demonstrated significantly better DH, yield, DPPH radical scavenging activity and reducing power, furthermore, had excellent inhibitory on S. aureus due to large clear zone and moderated inhibitory in bactericidal inhibition.

Optimization of ultrasonic-assisted enzymatic hydrolysis conditions for the production of antioxidant hydrolysates from porcine liver by using response surface methodology

  • Objective: The objective of this study was to optimize ultrasonic-assisted enzymatic hydrolysis conditions, including enzyme-to-substrate (E/S) ratio, pH, and temperature, for producing porcine liver hydrolysates (PLHs) with the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity by using response surface methodology (RSM). Methods: The study used RSM to determine the combination of hydrolysis parameters that maximized the antioxidant activity of our PLHs. Temperature ($40^{\circ}C$, $54^{\circ}C$, and $68^{\circ}C$), pH (8.5, 9.5, and 10.5), and E/S ratio (0.1%, 2.1%, and 4.1%) were selected as the independent variables and analyzed according to the preliminary experiment results, whereas DPPH free radical scavenging activity was selected as the dependent variable. Results: Analysis of variance showed that E/S ratio, pH, and temperature significantly affected the hydrolysis process (p<0.01). The optimal conditions for producing PLHs with the highest scavenging activity were as follows: E/S ratio, 1.4% (v/w); temperature, $55.5^{\circ}C$; and initial pH, 10.15. Under these conditions, the degree of hydrolysis, DPPH free radical scavenging activity, ferrous ion chelating ability, and reducing power of PLHs were 24.12%, 79%, 98.18%, and 0.601 absorbance unit, respectively. The molecular weight of most PLHs produced under these optimal conditions was less than 5,400 Da and contained 45.7% hydrophobic amino acids. Conclusion: Ultrasonic-assisted enzymatic hydrolysis can be applied to obtain favorable antioxidant hydrolysates from porcine liver with potential applications in food products for preventing lipid oxidation.

한국산 구아바 열매 추출물을 첨가한 구아바편의 품질특성 및 항산화활성 (Quality Characteristics and Antioxidative Activities of Guavapyun Added Korean Guava Fruit Extract)

  • This study was conducted to investigate the quality characteristics of guavapyun after the addition of different ratios of extract (0.25, 0.5, 0.75, 1.0%), non-extract and vitamin C. The quality if the pyun containing 0.5% of the guava fruit extract (guavapyun) and vitamin C was higher compared with the quality of the control pyun. In the results of the proximatecomposition, the content of water was high in the control pyun relative to the vitamin C pyun and guavapyun and the carbohydrate, ash, crude lipid and protein content was high in guavapyun. The sweetness, pH and color (L, a, b value) were very high in the samples. The texture, hardness, chewiness and gumminess were significantly high in the control pyun and the adhesiveness and cohesiveness were high in guavapyun. However, there were no significant differences in springiness between the control and the added samples. The total phenolic content was higher in guavapyun (23.57 mg GAE/100 g) than the control pyun (18 mg GAE/100 g) and vitamin C pyun(15.05 mg GAE/100 g). The antioxidant activities determined by the DPPH method and ABTS method was higher in guavapyun (41.37 mM TE/g, 15.35 mM TE/g) than the control pyun (4.43 mM TE/g, 2.17mM TE/g) and vitamin C pyun (11.33 mM TE/g, 4.51 mM TE/g). Using the FRAP method, guavapyun(9.06 mM TE/g) was shown to exhibit a stronger ferrous ion chelating activity than the control pyun (4.49mM TE/g) and vitamin C pyun (7.03 mM TE/g). Thus, the studied indigenous guavapyun was high in both antioxidative activity and total phenolic content.

Amycolatopsis sp. KCTC 29142로부터 유래된 siderochelin A의 다제 내성 균주에 대한 항균활성 (Anti-multi drug resistant pathogen activity of siderochelin A, produced by a novel Amycolatopsis sp. KCTC 29142)

  • 본 연구에서는 신규 Amycolatopsis 균주 KCTC 29142를 분리하여 형태학적 관찰, 계통분석 및 화학분류학적 분석 등 다상 분류분석을 통해 분석하였다. KCTC 29142 균주의 에틸아세테이트추출물은 강한 항균활성을 나타났고, 활성물질은 철 이온 킬레이트 물질인 siderochelin A로 동정되었다. 본 연구에서 분리된 siderochelin A는 다제내성균인 Acinetobacter baumanii, methicillin-resistant Staphylococcus aureus (MRSA), vancomycin-resistant Staphylococcus aureus (VRSA), 및 Escherichia coli (E. coli)에 대해 강한 활성을 보였고, 임상에서 분리된 다제내성균에 대한 MIC를 결정하였다.

꾸지뽕열매 발효 추출물의 항산화 특성과 Elastase 및 Tyrosinase 저해활성 (Antioxidant Activities of Extracts from Fermented Mulberry (Cudrania tricuspidata) Fruit. and Inhibitory Actions on Elastase and Tyrosinase)

  • 발효 꾸지뽕열매의 기능성 소재으로서의 개발 가능성을 알아보고자 70% ethanol 추출물에 대한 항산화 활성과 elastase 및 tyrosinase 저해활성을 측정하였다. 실험군은 꾸지뽕열매 분말 70% ethanol 추출물(ECT), $40^{\circ}C$에서 48시간 발효시킨 분말의 70% ethanol 추출물(FECT), B. lichenifornis 균주로 $40^{\circ}C$에서 48시간 발효시킨 분말의 70% ethanol 추출물(FECTL), B. subtilis 균주로 $40^{\circ}C$에서 48시간 발효시킨 분말의 70% ethanol 추출물(FECTS) 각각을 동결건조하여 실험에 사용하였다. 수율은 ECT, EFCT, EFCTL 및 EFCTS가 각각 54.22%, 54.43%, 57.71% 및 57.23%로 높은 수율을 나타내었으며, 균주 발효 시 증가하는 경향을 보였다. L*, a* 및 b* 값은 발효 처리구에서 감소하였다. Total polyphenol 함량은 ECT 8.13 mg/g (w/w), EFCT 9.53 mg/g (w/w)로 EFCT가 ECT에 비하여 17.22%가 증가하였고, 균주발효군인 EFCTL (11.03 mg/g, w/w) 및 EFCTS (11.90 mg/g, w/w)는 ECT에 비하여 각각 35.67% 및 46.37%가 증가하였다. Total flavonoid 함량 역시 ECT 1.54 mg/g (w/w), EFCT 1.73 mg/g (w/w)로 EFCT가 ECT에 비하여 12.34%가 증가하였으며, EFCTL (1.94 mg/g, w/w) 및 EFCTS (1.85 mg/g, w/w)는 ECT에 비하여 각각 25.97% 및 20.13%가 증가하였다. ABS 라디칼 소거능은 ECT (94.61%), EFCT (95.85%), EFCTL (94.36%), EFCTS (96.69%)로 유사하였다. SOD 유사활성은 ECT (22.00%), EFCT(23.30%), EFCTL (32.60%), EFCTS (27.10%)로 EFCTL에서 높았으며 아질산염소거능에서도 유사한 경향을 나타내었다. Ferrous ion chelating 효과에서는 EFCTL (67.34%) > EFCTS (60.36%) > ECT (52.34%) > EFCT (51.73%) 순이었다. 발효처리군 EFCT, EFCTL 및 EFCTS는 무처리군 ECT에 비하여 xanthine oxidase 저해활성은 각각 9.53%, 38.66% 및 49.78%가, elastase는 19.93%, 86.19% 및 77.60%가, tyrosinase는 23.94%, 89.14% 및 78.86%가 증가하였으며 균주발효군에서 그 활성이 증가하였다. 이상의 결과, 발효 꾸지뽕열매 70% 에탄올 추출물은 항산화활성이 우수하여 기능성 증진용 소재 활용에 효과가 있을 것으로 판단된다.

건조방법에 따른 꾸지뽕열매 추출물의 항산화활성과 Angiotensin Converting Enzyme I 저해활성 (Antioxidant and Angiotensin Converting Enzyme I Inhibitory Activities of Extracts from Mulberry (Cudrania tricuspidata) Fruit subjected to Different Drying Methods)

  • 건조 꾸지뽕나무 열매의 기능성 소재으로서의 개발 가능성을 알아보고자 70% ethanol 추출물에 대한 생리활성을 측정하였다. 실험군은 꾸지뽕나무 열매 동결건조분말 70% ethanol 추출물(ECFD), 열풍건조분말 70% ethanol 추출물(ECHD), 적외선건조분말 70% ethanol 추출물(ECID)을 각각 동결건조하여 실험에 사용하였다. 수율은 적외선건조에서 높은 수율을 나타내었다. 색도는 $L^*$, $a^*$, $b^*$$H^{\circ}$ 값 모두 열풍건조 및 적외선건조에서 감소하였으며, 총 폴리페놀 및 플라보노이드 함량은 적외선건조에서 가장 높은 함량을 나타내었다. 전자공여능은 적외선건조에서 높은 활성을 나타내었으며, 환원력 및 ABTS 라디칼 소거활성에서도 유사한 경향을 나타내었다. 아질산염 소거능은 적외선건조, 열풍건조, 동결건조 순으로 높은 활성을 나타내었다. SOD 유사활성은 동결건조에서는 611.97 mUnit, 열풍건조에서는 723.02 mUnit, 적외선건조에서는 891.93 mUnit로 열풍건조 및 적외선건조가 동결건조에 비하여 각각 18.15% 및 45.75% 높은 활성을 나타내었다. 반면 ferrous ion chelating 효과에서는 동결건조(52.36%)>적외선건조(47.16%)>열풍건조(30.04%)의 순으로 동결건조 처리구에서 높은 활성을 나타내었다. ACE 저해활성에서는 열풍건조 및 적외선건조가 동결건조에 비하여 각각 9.30% 및 30.88%가 증가하였으며, XO 저해 활성에서는 열풍건조 및 적외선건조가 동결건조에 비하여 각각 5.09% 및 9.02%가 증가하였다. 이상의 결과 적외선 건조 처리한 꾸지뽕나무 열매 추출물은 다양한 항산화 활성 증대효과를 나타냄으로써 산화적 손상 예방 소재개발에 활용가치가 있다고 사료된다.

산딸나무의 부위별 추출물 및 용매 분획물의 생리활성 비교 (Comparison of Biological Activities of Extracts from Different Parts and Solvent Fractions in Cornus kousa Buerg)

  • 산딸나무의 꽃, 잎, 열매, 가지 등 부위별 추출물과 용매 분획물을 제조하여 각각의 생리활성물질 함량, 항산화활성 그리고 tyrosinase 억제활성을 분석하였다. 총 폴리페놀 및 플라보노이드 함량은 다른 부위보다 꽃 추출물에서 169.638 $mg{\cdot}g^{-1}$과 25.418 $mg{\cdot}g^{-1}$으로 가장 높게 나타났다. DPPH 및 ABTS 라디칼 소거활성과 $Fe^{2+}$ chelating 효과 역시 꽃 추출물에서 가장 우수한 것으로 조사되었다. Ferric thiocyante 방법으로 지질 과산화 억제효과를 조사한 결과, 꽃, 잎, 가지의 경우는 BHT보다 높은 억제활성을 보였다. Tyrosinase 억제활성은 오직 꽃 추출물에서만 나타났다. 높은 생리활성을 보인 꽃과 잎 추출물을 n-hexane, chloroform, ethyl acetate, n-butanol의 순으로 용매분획한 결과, 폴리페놀함량은 꽃과 잎 모두 water 분획물에서 각각 67.006 $mg{\cdot}g^{-1}$ 및 66.739 $mg{\cdot}g^{-1}$으로 가장 많았다. 반면 플라보노이드 함량은 꽃의 경우 ethyl acetate 분획물에서 가장 높았고, 잎은 butanol 분획물에서 가장 높게 조사되었다. 자유라디칼 소거활성은 꽃 추출물의 경우 ethyl acetate 분획물에서 가장 높았고 잎 추출물은 butanol 분획물에서 가장 높게 조사되었다. 꽃과 잎의 각 용매 분획물이 tyrosinase의 활성에 미치는 영향을 조사한 결과, 모두 water 분획물에서 각각 49.24% 및 31.8%로 가장 높은 억제 효과를 보였다.
