Antioxidant Component and Activity of Dropwort (Oenanthe javanica) Ethanol Extracts (미나리 에탄올 추출물의 항산화성분과 항산화활성)

  • This study was conducted to investigate the antioxidant compounds and antioxidant activity on the dropwort (Oenanthe javanica) and its solvent fraction. The dropwort was extracted with 70% (v/v) ethanol, and then partitioned using the solvents of hexane, chloroform, ethyl acetate, butanol, and aqueous. The ethyl acetate fraction contained the highest phenolic and flavonoid of 240.61 mg GA eq/g and 105.57 mg catechin eq/g, followed by ethanol extract of 37.50 mg GA eq/g and 26.50 mg catechin eq/g, respectively. The DPPH radical scavenging activity ($IC_{50}$) on the solvent fractions increased in the order of ethyl acetate, butanol, ethanol extract, chloroform, aqueous, and hexane with 0.08, 0.58, 1.07, 2.43, 2.47, and 3.31 mg/mL, respectively. The ABTS radical scavenging activity was the highest value of 382 mg AA eq/g in ethyl acetate fraction. Reducing power and chelating effect on the ethanol extracts and its solvent fraction were in range of 0.23~0.75 and 0~32.01%, respectively. Hydroxyl radical scavenging activity ($IC_{50}$) was the lowest value of $26.71\;{\mu}g$/mL in ethyl acetate fraction.

Bioactive Compounds and Antioxidant Activity of Jeju Camellia Mistletoe (Korthalsella japonica Engl.) (제주 동백나무 겨우살이의 용매별 기능성 성분 및 항산화 작용)

  • Mistletoes are hemi-parasitic plant growing on different host tree and shrubs. They are traditionally used in folkloric medicine for the treatment of diarrhea, cough, diabetes, hypertension, cancer and skin infection. The purpose of this study was to determine the contents of phenolics and antioxidant activity of 70% ethanol, 100% methanol and hot water extracts of Jeju camellia mistletoe (Korthalsella japonica Engl.). Ethanol was most effective in extracting total phenols (7,427 mg gallic acid equivalent (GAE)/100 g) and flavonoid (1,777 mg rutin equivalent (RE)/100 g). The free radical scavenging activity, 1,1-diphenyl-2-picryl hydrazyl (DPPH) (EC50 = 7.8 mg/ml) and hydrogen peroxide (H2O2) (EC50 = 1.4 mg/ml), and the capacity for chelating metal ions (EC50 = 8.0 mg/ml) and reducing power (EC50 = 14.9 mg/ml) of the samples also higher in ethanolic extracts. The strong correlation (r2 = −0.996~−0.881) between antioxidant capacities and the phenolic contents implied that phenolic compounds are a major contributor to the antioxidant activity of the ethanolic extracts of Jeju camellia mistletoe. As conclusions, Jeju camellia mistletoe contains bioactive substances with a potential for reducing the physiological as well as oxidative stress and this could explain the suggested cancer preventive effect of these plants as well as their protective role on other major diseases.

Comparison of Antioxidant Effects by Different Extraction Methods in Flowers of Aster scaber, Aster maackii, Coreopsis lanceolata and Coreopsis tinctoria (참취, 좀개미취, 큰금계국 및 기생초 꽃의 추출방법에 따른 항산화 활성 비교)

  • Antioxidant substances were extracted from flowers of Aster scaber, Aster maackii, Coreopsis lanceolata and Coreopsis tinctoria using 2 types of extraction methods- ultrasonic wave and reflux, and antioxidant effects were compared. Higher yield of extract was obtained by ultrasonic wave method in all 4 species. Extraction time was different depending on species, but in all 4 species DPPH and ABTS free radical scavenging and ferrous ion chelating activity were higher or similar by ultrasonic wave than reflux method. Content of total polyphenol and flavonoid was mainly higher by ultrasonic wave than reflux method. Longer the treatment, the more the antioxidant activity and bioactive substances. Anti-lipid peroxidation of ultrasonic and reflux extracts obtained from flowers of Aster scaber and Coreopsis lanceolata were compared. Flower extract of Aster scaber obtained by reflux method showed the highest inhibition effect against peroxidation of linoleic acid, but extract of Coreopsis lanceolata obtained by 15-minute ultrasonic extract showed the highest effect. Ultrasonic wave extraction was more economical and efficient method than reflux extraction.

Quality and Antioxidant Characteristics of Granule Tea Prepared with Sea Tangle (Laminaria japonica) and Sea Mustard (Undaria pinnatifida) Powder as Affected by Extraction Method (추출방법을 달리한 미역 및 다시마 과립차의 품질 및 항산화 특성)

  • This study was conducted to evaluate the quality and anti-oxidant characteristics of different granule tea variants (WSMGT: granule tea prepared from sea mustard hot water extract; ASMGT: granule tea prepared from sea mustard autoclave extract; WSTGT: granule tea prepared from sea tangle hot water extract; and ASTGT: granule tea prepared from sea tangle autoclave extract) from sea tangle and sea mustard powder. The Color of WSTGT showed the highest $L^*$, $b^*$ values, and the lowest $a^*$ value. The water absorption binding agents in ASMGT were higher than those in WSMGT, WSTGT. and ASTGT. Binding agents on water absorption in ASMGT were higher than those of WSMGT, WSTGT, and ASTGT. The alginic acid content of ASMGT was highest. The total polyphenol and total flavonoid contents of ASMGT and ASTGT were highest. The electron donating ability and ferrous ion chelating activity in WSTGT greatly increased compared to those in the other granules tea variants. These results suggest that pressure extraction can be used in functional foods.

Antioxidants of Pine Needle Extracts According to Preparation Method (제조방법별 솔잎추출물의 항산화성 검토)

  • This study was carried out to investigate the effects of pine needle extracts on lipid oxidation and free radical reaction in iron sources reacted with active oxygen species. The results were summarized as follow; the catalytic effects of active oxygen on lipid oxidation in oil emulsion tended to be showed $OH,\;H_2O_2\;and\;KO_2$ in order. At the same time, pine needle extracts itself were tended to be showed a little catalytic effects. Active oxygen scavenging ability of pine needle extracts didn't show, but pine needle extracts played role as a strong chelating agents to bind iron ion if $Fe^{2+}$ ion exist in oil emulsion. The content of $Fe^{2+}$ ion and total iron in CPNP were higher than those of HPNP and FPN. The content of ascorbic acid of FPN showed the highest (87.77 ppm) among several pine needle extracts. Electron donating ability of HPNP and CPNP were 81% and 78%, respectively, which were showed higher content than those of FPN. The SOD-like activity of HPNP showed 44.30%, compared to other pine needle extracts which means the most strong antioxidant reaction. The nitrite scavenging effects were tended to be different, depending on pH value as pH value was increased. Especially, they didn't show the nitrite scavenging effect in pH6.0.

Chemical Interaction of L-Ascorbic Acid with Ferritin Molecule (가시부영역의 흡수 스펙트럼 변화에 의한 L-Ascorbic Acid와 Ferritin과의 상호작용)

  • This study consisted of designing a sensitive assay to measure the rate of L-ascorbic acid (AsA)-prompted release of ferritin iron, the use of ferrozine as a chelating agent to trap releases Fe(II). The initial rate of iron release was measured in the appearance of Fe(ferrozine)$_3$$^{2+}$ at 562 nm. The release of iron from ferritin by AsA was dependent on time and AsA conditions under aerobic and anaerobic conditions. Effect of oxygen on the release of iron from ferritin was also confirmed. It was suggested that the release of iron from ferritin was participate not only AsA but also $O_2$$^{[-10]}$ . In this study, it was found that iron can be released from ferritin and chelate as Fe(ferrozine)$_3$$^{2+}$ and the release was more than 50% in the presence of AsA without $O_2$$^{[-10]}$ . Based on the findings, the following can be assumed (1) AsA is diffused into ferritin (2) ferric ion is reduced to ferrous ion (3) is diffused from ferritin.tin.

Antioxidant and Antimicrobial Activities of Fruiting Bodies of Phellinus gilvus Collected in Korea (국내에서 수집된 마른진흙버섯 자실체의 항산화 및 항균 효과)

  • This study was initiated to evaluate the antioxidant and antimicrobial activities of methanol extract (ME) and hot water extract (HWE) obtained from the fruiting bodies of medicinal mushroom, Phellinus gilvus. The free radical scavenging activity of ME from P. gilvus on 1,1-diphenyl-2-picrylhydrazyl (DPPH) were 93.65% at 2 mg/mL, which was comparable with the positive control, butylated hydroxytoluene (BHT, 96.97%) at the same concentration. The ferrous ion-chelating ability of ME and HWE was significantly higher than that of BHT at all concentration levels. The antimicrobial assay of ME was performed against six bacteria and one species of fungus. ME exhibited antibacterial activity against 5 out of 6 bacteria: Staphylococcus aureus, Streptococcus mutans, Bacillus subtilis, Escherichia coli, and Pseudomonas aeruginosa; whereas, ME did not show antimicrobial activity against gram-negative bacterium Vibrio vulnificus and fungal species Candida albicans. The minimum inhibitory concentration (MIC) of ME against 5 strains of bacteria, such as S. aureus, S. mutans, B. subtilis, E. coli, and P. aeruginosa, was 100, 100, 50, 100, 200 mg/mL, respectively. The results suggest that good antioxidant and microbial activities of P. gilvus fruiting bodies might be used for natural antioxidant and antimicrobial agents.

Antioxidative and antimicrobial activities of Oenothera biennis extracted by different methods (추출방법을 달리한 달맞이꽃 추출물의 항산화 및 항균 활성)

  • A effect of extraction methods, including stirrer extraction method (SE), ultrasonification extraction method (USE), reflux extraction method (RE), autoclave extraction (AE) and low temperature high pressure extraction (LE) method on the antioxidative and antimicrobial activities of ethanol extracts of Oenothera biennis was investigated. The extraction yield (46.33%), total polyphenol (463.05 mg GAE/g) and flavonoid (71.71 mg RHE/g) content of Oenothera biennis extract obtained by RE were higher than those from other extraction methods. The antimicrobial activity of Oenothera biennis extract was only observed against Bacillus cereus among other tested organisms (Bacillus cereus, Staphylococcus aureus, Escherichia coli and Salmonella Typhimurium). Oenothera biennis obtained by RE showed the best DPPH radical scavenging ability (74.40%), ABTS radical scavenging ability (65.29%), reducing power (1.370 ($OD_{700}$)) and ferrous ion chelating ability (90.14%) compared with other tested extraction methods tested. The RE method was the most efficient method for extracting crude antioxidant and antimicrobial substances from Oenothera biennis. These results suggested that Oenothera biennis obtained by RE could be used as a bioactive and functional material in the food industry.

Antioxidant and physiological activities of Hijikia fusiforme by extraction methods (추출방법에 따른 톳 추출물의 항산화 및 생리활성 특성)

  • The physiological properties of extracts from Hizikia fusiforme with different extraction methods (hot water extraction, autoclave extraction and high pressure extraction) were investigated. The amounts of substances related to polyphenol and flavonoids contents were the highest in autoclave extract (30.51 mg/g and 4.78 mg/g, respectively). The DPPH radical scavenging activity was the highest in the autoclave extract (81.80%) at the 5 mg/mL. Autoclave extract exhibited the strongest ABTS radical scavenging activity and reducing power among the three extraction methods. However, ferrous ion chelating, TBA reactive substances and xanthine oxidase inhibition activity of high pressure extract were higher than those of the other extracts. Additionally, the tyrosinase and elastase inhibition activities of high pressure extract with a concentration of 1 mg/mL were higher than those of the other extracts. These results suggest that extracts of Hizikia fusiforme have a potential to act as functional materials, and autoclave and high pressure extractions are superior to hot water extraction for enhancement of the biological activity.

Flavonoid Content and Antioxidant Activities of Gardenia jasminoides Ellis Seed Extracts Produced by Ethyl Acetate, Distilled Water and Methanol (치자(Gardenia jasminoides Ellis) 씨 에틸 아세테이트, 물 및 메탄올 추출물의 플라보노이드 함량 및 항산화 활성)

  • The purpose of this study was to measure the bioactivity and antioxidant activity of seed from Gardenia jasminoides Ellis fructus (GJE). We determined tannin content ($1.517{\pm}0.003mg\;CE/g$, mg of catechin equivalents). The extraction yields of the GJE seed solvent were distilled water (DW) 36.61%, 70% methanol 30.10% and ethyl acetate (EA) 20.40%. The antioxidant activities of the extracts were significantly increased (0.2, 0.4, 0.6 mg/mL). Flavonoid contents (mg QE/g, mg of quercetin equivalents) were observed in the order of 70% methanol (0.799), DW (0.565) and EA (0.117). Antioxidant activities (DPPH, OH, ferrous ion-chelating capacity) were similar to the flavonoid content of each solvent. But ABTS scavenging activity and SOD like ability of DW extract were higher than 70% methanol extract. Comparing the yield and the antioxidant activity of each solvent of GJE seed, it seems to be preferable to use DW and 70% methanol solvent for extraction. As a result of this experiment, it has high antioxidant activity and physiological activity, which is expected to be highly valuable as a functional food and a natural antioxidant.
