Anti-oxidative Properties of Lipids Extracted from Rainbow Trout (Oncorhynchus mykiss) Fed with Carotenoids and Conjugated Linoleic Acid

  • A commercial diet supplemented with carotenoids and conjugated linoleic acid was fed to rainbow trout (Oncorhynchus mykiss) for 8 weeks. To investigate the anti-oxidative properties of these compounds, lipids from the muscle and viscera of the fish were subjected to different assays. At $10\;{\mu}g/mL$, L-ascorbic acid exhibited 95% 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activity, while the tissue lipids showed little radical scavenging activity. At 50 and $100\;{\mu}g/mL$, the lipids of the muscles and viscera showed 11.7-22.6% and 11.3-24.9% DPPH radical-scavenging activity, respectively. A lipid peroxidation inhibitory assay using the ferric thiocyanate method was also performed in comparison with $\alpha$-tocopherol at a concentration of 6.0 mg/mL. Our results indicate that the anti-oxidative property of the lipids in fish muscle, which was 85.2% compared to 85.3% for the visceral lipids, was stronger than that of $\alpha$-tocopherol (74.3%) following 3 days of storage at $40^{\circ}C$.

Nutritional composition and antioxidant activity of pink oyster mushrooms (Pleurotus djamor var. roseus) grown on a paddy straw substrate

  • Pleurotus djamor var. roseus is an edible mushroom isolated from the wild and cultivated on paddy straw substrates. The present study was carried out to compare the nutritional composition and antioxidant properties of P. djamor var. roseus at different growth stages (primordia, basidiomata, and mycelia). The protein content was is in the range of 31.48 to 35.50 g/100g dw. The crude fiber content ranged from 8.0 to 14.60 g, and that of total carbohydrates ranged from 44.75 to 48.90 g. Sodium, magnesium, and calcium reached the maximum levels in basidiomata, and selenium was detected in basidiomata and mycelia (0.47 - 0.22 mg/Kg). The amino acid profile showed that all essential and nonessential amino acids and glycine showed maximum levels in basidiomata and 15.98 ± 0.01 g/100g. The fatty acid profile showed the presence of saturated and unsaturated fatty acids; the unsaturated fatty acid content was maximum in all of the samples, ranging from 76 - 40.41%. The total phenol and flavonoid contents as well as the scavenging (DPPH), ferric thiocyanate (FTC), and thiobarbituric acid (TBA) activities in the context of methanol and water extracts from primordia, basidiomata, and mycelium were determined. Among them, basidiomata and mycelial methanol extracts exhibited significant antioxidant activity. Overall, these findings show that P. djamor var. roseus can be used as a functional food for daily consumption.

Antioxidant Effect of Crataegi Fructus Extract on the Oxidative Stress of Reactive Oxygen Species in Cultured Human Skin Fibroblast (배양 인체피부섬유모세포에 있어서 활성산소의 산화적 손상에 대한 산사(山査)추출물의 항산화 효과)

  • This study was done to evaluate the antioxidant effect of Crataegi Fructus (CF) extract on the oxidative stress induced by reactive oxygen species (ROS), The human skin fibroblasts (Detroit 551) were cultured with various concentrations of hydrogen peroxide $(H_2O_2)$. The cytotoxicity of $H_2O_2-induced$ oxidative stress was performed by XTT assay for the cell viability according to the dose- and time-dependent treatment. For the protective effect of CF extract on $H_2O_2-mediated$ oxidative stress, cell viability, lactate dehydroganase (LDH) activity, and ferric thiocyanate (FTC) assay for the inhibitive activity of lipid peroxidation on CF extract were carried out. In this study, $H_2O_2-mediated$ oxidative stress was decreased cell viability dose-, and time-dependent manner and increased LDH activity compared with the control in these cultures. In the protective effect, CF extract increased cell viability and decreased LDH activity on $H_2O_2-mediated$ oxidative stress, especially, CF extract has antioxidant effect by the showing the inhibitive activity of lipid peroxidation by FTC assay. From these results, It is suggested that $H_2O_2-mediated$ oxidative stress was highly toxic, and also, CF extract showed the protective effect on $H_2O_2-mediated$ oxidative stress by showing the increased cell viability, decreased LDH activity and lipid peroxidation inhibition in these cultures.

  • Seven antioxidative flavonoids were isolated from Jindalrae flowers (Rhododendron mnonulatum Turcz.), an edible plant in Korea. These compounds were identified as afzelin, ampelopsin, catechin, myricetin, myricitrin, quercetin and quercitrin on the basis of IR, UV, FAB-MS, $^1H\;NMR,\;and\;^{13}C\;NMR$ data. These compounds were consisted of two flavonols, three flavonol glycosides, a flavane, and a dihydroflavonol. The flavonol glycosides (14.4 g) present in th ethyl ether and ethyl acetate fractions comprised up to 82% of their total flavonoid amount (17.6 g) finally recovered by means of polyamide C-200 column chromatography, preparative TLC, recrystallization, and Sephadex LH-20 column chromatography. The antioxidant activities were measured in an ethanol solution of linoleic acid in the presence of ferric thiocyanate. The antioxidant efficiency increased in the order of afzrlin<$\alpha-tocopherol$

  • Six phenolic acids were isolated from Jindalrae flowers (Rhododendron mucronulatum Turcz.), an edible plant in Korea. These compounds were identified as chlorogenic acid, 3,5-O-dicaffeoylquinic acid, 4,5-O-dicaffeoylquinic acid, caffeic acid, ferulic acid, and p-coumaric acid on the basis of IR, UV, $^{1}H$ and $^{13}C$ NMR, FAB-MS, ES-MS and/or El-MS data. Phenolic Acid Composition and Antioxidative Activity of Chestnut Endoderm (밤 삽피의 페놀산 조성과 항산화 활성)

  • Free, soluble esterified and insoluble bounded phenolic acids were separated from Eungi chestnut endoderm. The composition and contents of phenolic acid were analyzed by gas chromatography, and their antioxidant activity was examined by DPPH assay, 2-deoxyribose oxidation, and ferric thiocyanate method. Gallic, ellagic, salicylic, and gentisic acids in free phenolic acid fraction, gallic, ellagic, and protocatechuic acids in soluble esterified fraction, sianpic and gentisic acids were the major phenolic acids in insoluble bounded fraction. Marked differences were observed in the phenolic acid composition and contents among the fractions. Free phenolic acid fraction showed the strongest antioxidant activity. Results revealed chestnut endoderm could be a potential antioxidant source containing gallic and ellagic acids.

Comparative Evaluation of Antioxidant Activities of Ethanol Extracts and Their Solvent Fractions Obtained from Selected Miscellaneous Cereal Grains (잡곡 유래 에탄올 추출물 및 이의 유기용매 분획들의 항산화 활성 비교평가)

  • To examine the antioxidant activities of 11n selected miscellaneous cereal grains (proso millet, yellow glutinous proso millet, hwanggeumchal sorghum, glutinous sorghum, white glutinous sorghum, yellow glutinous foxtail millet, nonglutinous foxtail millet, green glutinous foxtail millet, golden foxtail millet, barnyard millet, and adlay), the free radical-scavenging activities of 80% ethanol extracts of the individual grains were investigated using 1,1-diphenyl-2-picryl-hydrazl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) methods. The ethanol extracts of hwanggeumchal sorghum, glutinous sorghum, and barnyard millet grains exhibited more potent free radical-scavenging activities as compared to the other grains. When these three ethanol extracts were sequentially fractionated with n-hexane, methylene chloride, ethyl acetate, and n-butanol, the majority of the antioxidant activities were detected in the ethyl acetate and butanol fractions in which phenolic ingredients were abundant. The ethyl acetate and butanol fractions of hwanggeumchal sorghum and the ethyl acetate fraction of glutinous sorghum showed higher antioxidant activity than that of ${\alpha}$-tocopherol. Both ferric thiocyanate (FTC) and thiobarbituric acid (TBA) methods demonstrated that these organic solvent fractions could inhibit lipid peroxidation. The ethyl acetate fractions from hwanggeumchal sorghum, glutinous sorghum, and barnyard millet grains could suppress tertiary-butyl hydroperoxide (TBHP)-induced apoptotic events, including sub-G1 peaks, ${\Delta}{\Psi}m$ loss, activation of caspase-9 and caspase-3, and cleavage of PARP and lamin B, in human HL-60 cells. These results show that the grains of hwanggeumchal sorghum (Sorghum bicolor L. Moench cv. Hwanggeumchalsusu), glutinous sorghum (Sorghum bicolor L. Moench cv. Chalsusu), and barnyard millet (Echinochloa esculenta) possess efficient antioxidant activity, which could protect cells from oxidative stress-mediated cytotoxicity.

Total Polyphenol Contents and Antioxidant Activities of Methanol Extracts from Vegetables produced in Ullung Island (울릉도산 산채류 추출물의 총 폴리페놀 함량 및 항산화 활성)

  • To discover new functional materials using edible plants, antioxidant activities of methanol extracts from various parts of seven wild vegetables were investigated in vitro. Total polyphenol contents, determined by Folin-Denis method, varied from 16.74 to $130.22{\mu}g/mg$. Radical-scavenging activities of methanol extracts were examined using ${\alpha},\;{\alpha}-diphenyl-{\beta}-pirrylhydrazyl$ (DPPH) radicals and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) assay. Inhibition effects on peroxidation of linoleic acid determined by ferric thiocyanate (FTC) method and on oxidative degradation of 2-deoxy-D-ribose in Fenton-type reaction system were dose-dependent. Athyrium acutipinulum Kodama (leaf and rood), Achyranthes japonica (Miq.) Nakai (seed), and Solidago virga-aurea var. gigantea Nakai (root) showed relatively high antioxidant activities in various systems.

AChE Inhibitory Effect and Antioxidative Activity of Submerged Cultured Products from Hericium erinaceum (Hericium erinaceum 액체배양 생성물의 Acetyl-cholinesterase 저해 활성과 항산화 활성)

  • The water-soluble or ethanol-soluble materials extracted from fruit bodies and cultured products (mycelium and broth) of H. erinaceum were prepared, and their inhibitory effect on acetylcholinesterase (AChE) activity from Electrophorous electricus was investigated. Inhibition of 75-85% for AChE activity at concentration of 10 mg/ml was obtained and the mechanism was due to general non-competitive inhibition. Especially, the supernatant of culture broth by ethanol treatment, exhibited a strong inhibition activity of 94% at 10 mg/ml. The samples from fruit body, mycelium and broth (supernatant and precipitate by ethanol treatment) which were extracted from H. erinaceum, were very effective to inhibit the initial stage oxidation of a linoleic acid at concentration of 0.1 mg/ml. The antioxidative activity of these samples were superior than rutin, vitamin C and tocopherol as antioxidative standards by FTC (ferric thiocyanate) method, and also showed the very strong antioxidative activity of 95% without significant difference of the samples by TBA-RS (thiobarbituric acid-reactive substance) method.

Bioactive Materials and Antioxidant Properties of Fermented Rice-bran Extract (쌀겨발효추출물의 이화학적 특성 및 항산화 작용)

  • This study suggests that fermented rice bran extract contains natural antioxidants. The contents of bioactive materials (e.g., polyphenolic compounds and flavonoids), antioxidative properties (DPPH (α,α'- diphenyl-β-picrylhydrazyl) free radical scavenging activity, Fe reducing, Cu reducing power, peroxidation of linoleic acid and rat hepatocyte microsome) were tested by in vitro experimental models using fermented rice bran (FRB) extract. The concentrations of phenolic compound and flavonoid were 19.92 mg/g and 11.56 mg/g, respectively. In oxidation in vitro models using DPPH free radical scavenging activity, (free radical scavenging activity 69.8%) Fe reducing power and Cu reducing power (effect of dose-dependent manner), Fe2+/ascorbate induced linolenic acid peroxidation by ferric thiocyanate and thiobarbituric acid (TBA) methods (inhibition activity 81%), and autooxidation of rat hepatic microsomes membrane (lipid peroxidation inhibition activity 38%), antioxidative activities were stronger in FRB extract than FRS (Fermented Rice and Soybean, positive control) extract and, these effects were dose-dependent manner. From these results, FRB extract was shown to have the most potent antioxidative properties and contain the highest amounts of antioxidative compounds such as phenolic compounds and flavonoids. Overall, these results may provide the basic data to understand the antioxidative properties of fermented rice bran for development of functional foods.
