The present study was conducted with the aim to investigate the ameliorative effects of a new soybean product (cheonggukjang) fermented with Bacillus amyloliquefaciens SCGB1 (SFBA) in atopic dermatitis (AD) mouse model. Visual evaluation of AD induction in the mice indicated the remarkable control of SFBA in reducing the pathological severity of AD-like skin lesions reported as the SCORAD score of AD clinical symptoms. The results revealed that SFBA reduced dorsal skin and epidermal thickness to a similar extent with prednisolone. Further analysis revealed the dominance of SFBA in restraining mast cell infiltration in the dermis; immunoglobulin-E expression in serum; and TH2 IL-4 cytokine and itch-related IL-31 cytokine in the mice skin and serum. SFBA also suppressed scratching behaviours in mice induced by compound 48/80. Further histological findings also revealed the alleviation of collagen fiber deposition in dermal skin of the AD mice model. These actions of SFBA were examined to be mediated by its suppression of the phosphorylation activation of key signalling molecules such as $NF-{\kappa}B$ and MAPK responsible for the induction of cytokine production. Thus, SFBA can be considered as a promising functional food for managing clinical, histological and immunological spectra associated with AD.
L-5-methyltetrahydrofolate (5-MTHF) is one of the biological active forms of folate, which is widely used as a nutraceutical. However, low yield and serious pollution associated with the chemical synthesis of 5-MTHF hampers its sustainable supply. In this study, 5-MTHF production was improved by engineering the 5-MTHF biosynthesis pathway and NADPH supply in Lactococcus lactis for developing a green and sustainable biosynthesis approach. Specifically, overexpressing the key rate-limiting enzyme methylenetetrahydrofolate reductase led to intracellular 5-MTHF accumulation, reaching 18 ㎍/l. Next, 5-MTHF synthesis was further enhanced by combinatorial overexpression of 5-MTHF synthesis pathway enzymes with methylenetetrahydrofolate reductase, resulting in 1.7-fold enhancement. The folate supply pathway was strengthened by expressing folE encoding GTP cyclohydrolase I, which increased 5-MTHF production 2.4-fold to 72 ㎍/l. Furthermore, glucose-6-phosphate dehydrogenase was overexpressed to improve the redox cofactor NADPH supply for 5-MTHF biosynthesis, which led to a 60% increase in intracellular NADPH and a 35% increase in 5-MTHF production (97 ㎍/l). To reduce formation of the by-product 5-formyltetrahydrofolate, overexpression of 5-formyltetrahydrofolate cyclo-ligase converted 5-formyltetrahydrofolate to 5,10-methyltetrahydrofolate, which enhanced the 5-MTHF titer to 132 ㎍/l. Finally, combinatorial addition of folate precursors to the fermentation medium boosted 5-MTHF production, reaching 300 ㎍/l. To the best of our knowledge, this titer is the highest achieved by L. lactis. This study lays the foundation for further engineering of L. lactis for efficient 5-MTHF biosynthesis.
Consumer's interest in sustainable livestock farming methods has grown in response to concerns for the environment and animal welfare. The purpose of this study is to examine the different influences of sustainability product information on sensory characteristics and purchase behaviors. To accomplish this aim, the study used salami, which is an Italian-style sausage processed by fermentation and drying. Three different types of information were provided: salami made from the pork of an antibiotic-free pig (SMAFP), of an animal welfare pig (SMAWP), and of a grazing pig (SMGP). This study was conducted as an off-line experiment with Korean participants (n=140). As a result, there were sensory differences according to the sustainability information. For the SMAFP, it had a significant difference in, sourness (p<0.05). With the SMAWP, there was a difference in gumminess (p<0.10), and the SMGP had significant differences in sourness (p<0.01), sweetness (p<0.01), andmoisture (p<0.05). Moreover, the purchase intention and willingness to pay were significantly higher when the sustainability information was given. Especially, among the three types of salamis, participants were willing to pay the most for the SMAWP. This is one of the first consumer studies to investigate sensory evaluation and purchase behavior for various types of sustainable livestock production. These results contribute by helping sustainable meat producers and marketers become aware of the kind of sustainable information to which consumers are sensitive.
Microorganisms involved in food fermentation not only improve the aroma and taste of the food, but also enhance its preservation. Thus, they are added as starter cultures to boost the final product quality of commercial fermented foods. Although these microorganisms originate from fermented foods and have a long history of consumption, the European Union recently applied the concept of Qualified presumption of Safety (QPS), which is a safety evaluation system for microorganisms used in food or feed in Europe. The QPS system is a species-level safety system and shares results with the European Novel Food System, a strain-level safety evaluation system. In the United States, microorganisms added to fermented foods are considered as food additives or Generally Recognized as Safe substance. In Korea, food microbe lists are presented at the species level. Moreover, the nation has established a strain-oriented evaluation system that applies temporary safety evaluation methods for food raw materials as well as new raw materials. However, when it comes to microorganisms isolated from traditional fermented foods and other fermented food products, there is no definition of the term "species," and there is a lack of an evaluation system at the species level. Therefore, such an evaluation system for microbial species used in Korean fermented foods is necessary.
본 연구에서는 녹용에 함유한 단백질인 케라틴을 Fervidobacterium islandicum AW-1로 고온 혐기 발효를 통하여 케라틴 펩타이드를 생산하고, 인체 모발 관련한 인자를 확인하여, 화장품 원료로서 케라틴 펩타이드의 가능성을 확인하였다. 모유두 세포주에서 녹용 발효 케라틴 펩타이드를 농도에 따라 세포 독성 및 증식을 확인한 결과, 세포 독성은 나타나지 않았고, 세포 증식 효과를 보였다. 인체 모유두 세포에 대하여 녹용 발효 케라틴 펩타이드에 따른 성장인자의 증가를 확인하여 모발에 영향을 준다고 판단하였다. 이러한 녹용 발효 케라틴 펩타이드를 함유한 화장품을 제조하여 피부 안전성 및 탈모 증상 완화 인체 효력 시험을 수행한 결과, 사용 12 주 후에 제품 사용 전 대비 전체 모발 수의 증가 및 대조군과 비교시 전체 모발 개수의 차이가 통계적으로 유의하게 나타났다. 이를 통해 녹용 발효 케라틴 펩타이드는 기능성 화장품 원료뿐만 아니라 건강 기능 식품 소재로의 가능성을 확인하였다.

Yeast extract is a product prepared mainly from waste brewer's yeast, which is rich in nucleotides, proteins, amino acids, sugars and a variety of trace elements, and has the advantages of low production cost and abundant supply of raw material. Consequently, yeast extracts are widely used in various fields as animal feed additives, food flavoring agents and additives, cosmetic supplements, and microbial fermentation media; however, their full potential has not yet been realized. To improve understanding of current research knowledge, this review summarizes the ingredients, production technology, and applications of yeast extracts, and discusses the relationship between their properties and applications. Developmental trends and future prospects of yeast extract are also previewed, with the aim of providing a theoretical basis for the development and expansion of future applications.
지리오갈피(Acanthopanax chilsanensis)를 이용한 지역 특산품 개발 및 소비촉진을 목적으로 지리오갈피 발효주를 개발하여 5~10℃, 60일 동안 숙성시키면서 발효숙성주의 품질특성 등을 검토하였다. 숙성 기간 동안 발효숙성주 중의 환원당 함량은 약간 감소하였으나 총산의 함량은 아주 완만하게 증가하였다. 지리오갈피 뿌리, 줄기 및 열매로 제조한 발효숙성주 중의 유기산 함량은 3종류의 숙성주 모두 lactic acid, formic acid, malic acid, tartaric acid 및 acetic acid의 순으로 그 농도가 높게 검출되었다. 그리고 유리당은 3 종류의 숙성주 모두 sucrose, glucose 및 mannose 등의 유리당이 검출되었으며 이들 중 glucose 함량이 모든 시험구에서 가장 높게 나타났다. Acanthoside-D 함량은 줄기 첨가 발효숙성주에서 64.12 ppm으로 가장 높게 나타났으며 발효주와 발효숙성주의 acanthoside-D 함량을 비교해 보면 발효숙성주에서 약간 높아지기는 했으나 거의 변화가 없는 것으로 나타났다. 발효숙성주 중의 총 유리아미노산 함량은 뿌리, 줄기 및 열매의 발효숙성주에서 각각 1,858.9, 2,855.1 및 2,672.2 mg/l로 나타났다. 숙성주의 색도를 검토한 결과 뿌리와 줄기를 첨가한 숙성주에서는 거의 유사한 L, a, b값을 나타내어 발효숙성주의 색깔도 비슷하게 나타났으나, 열매를 첨가한 숙성주에서는 L값이 낮고, a값이 높아 붉은 색을 많이 띄었다.
1. 5섯가지 젖산균을 대두유에 접종배양 하여 산 생성도를 비교 하였드니 Str. thermophilus 와 Str. diacetilactis가 Str. lactis, Str. cremoris 및 L. bulgaricus보다 높았다. 2. 대두유와 탈지 분유를 7 : 3의 비로 혼합하여 젖산균 발효를 하였을 때에 침전물의 수획량이 가장 많고 또한 그의 단백질 함량이 가장 많으며 수분의 량이 적고 견고한 curd를 만들수 있음을 알았다. 3. 대두유와 탈지분유를 7 : 3의 비로 혼합한 것에 젖산균 starter로 Str. thermophilus를 첨가하고 Pen. caseicolum을 접종하여 제조한 치-즈를 3주간 숙성$(15^{\circ}C,\;RH:\;85{\sim}90%)$ 하였드니 숙성 7일째부터 Pen. caseicolum의 생육이 왕성하게 되며 pH가 상승함과 동시에 proteolytic activity는 14일째에 최고치에 도달하였다, 또 total water soluble nitrogen, water soluble protein nitrogen 및 amino-N도 숙성 7일째부터 증가하기 시작하여 숙성 21일째는 이들의 값이 각각 total nitrogen의 52%, 32% 및 14%에 이르렀다. 그리고 이차원 paper chromatography에 의하여 3주간 숙성한 치-즈에서 17종의 아미노산을 검출 하였다. 또 3주간 숙성한 후 제품의 일반성분 조성은 수분 63.2%, 조단백질 17.5%, 조지방 13.2%, 조회분 2.8% 및 식염 2.5%였다.
