Acute Intravenous and Oral Toxicity of DWC-751 in Rats and Mice (랫드 및 마우스에서 DWC-751의 급성정맥 및 경구 독성시험)

  • Single intravenous and oral administration to SD rats and ICR mice of both sexes were performed to investigate the acute toxicity of DWC-751, a new parenteral cephalosporin. $LD_50$ values for ICR mice and SD rats administered intravenously with DWC-751 were as follows; 1151.1 mg/kg (male SD rat), 1183.5 mg/kg (female SD rat), 2698.1 mg/kg (male ICR mouse), 2833.0 mg/kg (female ICR mouse). It is suggested that $LD_50$ values in rats and mice of both sexes would be 5000 mg/kg in oral route. Major general symptoms induced by injection intravenously with DWC-751 are decreased motor activity, increased respiratory rate, tremor and convulsion. In oral route, piloerection and soft stool are observed to 4 day after administration. No significant body weight changes were observed at any level in the groups administered with DWC-751. The gross finding of rats administered intravenously was observed cecum distension.

Effect of Sex Hormones on Lipid Peroxidation in Rat Liver

  • The role of sex homones in hepatic lipid peroxidation, and in hepatic adehyde odidase and xanthine oxidase activites were investigated using rat liver homogenates. It was observed that male rt had a significantly greater content of malondialdehyde in liver than female. Among the sex hormones tested, estradiol, one of female hormones, markedly inhibited the formation of lipid peroxides in liver tissues in vitro. Especially, the inhibitory effect of estradiol appeared more remarkably in Fe-induced lipid peroxidation. The hepatic xanthine oxidase activity was decreased about 15% by $10\;^6\;M$ estradiol, wherease, the adehyde oxidase activity was almost completely disappeared at the same concentration of estradiol. It implies that sex differences in lipid peroxidation is attributed to the suppression of radical generating system by estradiol.

  • Kami-honghwa-tang(KH-19) is a prescription for reducing the side effect of radiotherapy. In this study, safety of KH-19 was evaluated by GLP guideline of Korea Food and Drug Administration. In acute oral toxicity study on rat, transient inhibition of weight increase was shown, but change in general symptom was not detected. No dead animal was observed up to 5,000 mg/kg in both male and female animals. In acute oral toxicity study on Beagle dog, transient vomiting, diarrhea, anorexia, and weight reduction was observed. However, no dead animal was observed up to 2,000 mg/kg in both male and female animals.

  • This study investigated changes of mechanical characteristics of the trabecular and cortical bone in the lumbar vertebrae of the ovariectomised (OVX) rat. In previous researches, there were many studies for morphology of osteoporotic bones based on Micro-Computed Tomography (Micro-CT). However, there were few studies for detecting and tracking changes of mechanical characteristics of the trabecular and cortical bone in the lumbar vertebrae of the OVX rat. For this study, the 4th lumbar of the OVX rat (female Sprague-Dawley) was utilized as a specimen. An OVX rat was scanned at week 0 (just before surgery), at week 4, and week 8 after surgery. Micro-finite element (${\mu}-FE$) analysis was used to investigate mechanical characteristics of the trabecular and cortical bone in the lumbar vertebrae for an OVX rat. When the OVX rat (at week 8) was compared with the OVX rat (at week 0), the structural modulus of cortical and trabecualr bone was decreased by 52% and 99%, respectively. This study showed the change of mechanical characteristics of cortical bone as well as trabecular bone of an OVX rat. Detecting and tracking changes of mechanical characteristics could greatly contribute to an experiment test for the trabecular and cortical bone in the lumbar vertebrae of an OVX rat by using In-vivo Micro-CT.

Pyometra in the PMSG-treated Immature Female Rats (PMSG를 투여한 미성숙랫드에 발생한 자궁축농증 발생례)

  • Nine immature 30-day-old female rats were injected sc at 0800 hr with pregnant mare serum gonadotrophin(PMSG) to induce ovulation and mating. Fifty-six hours later the animals were placed with mature male rats overnight (one female and one male). Five of 9 immature female rats treated with PMSG were pregnant and allowed to maintain the pregnancy to term. Three of 5 pregnant rats were failed to maintain pregnancy to term. Two of 5 pregnant rats seemed to be developed normally and increased abdominal enlargement as pregnancy progresses, but did not occurred parturition on day of 43 or 48 of pregnancy, respectively. On day 44 or 49, pregnant rats were killed and examined uterus and ovaries. There was no fetus but approximately 50∼60ml. of mucopurulent fluids were accumulated in the uterine cavity and 40 or 42 corpora lutea persisted in the ovaries. Pyometra was developed after coitus in PMSG-treated immature female rat.

Studies on Sexing of Bisected Mouse Embryos by Rat H-Y Antibody (Rat H-Y 항체에 의한 생쥐 분할란의 성 조절에 관한 연구)

  • This experiment was carried out to develop a new technique of identifying XX of XY-bearing bisected embryos prior to implantation by immunological method. H-Y antiserum prepared in inbred Wastar female rats by repeated immunization with spleen cells from males of the same strain. The reactivity of H-Y antibody was confirmed by culturing mouse embryos in the medium containing H-Y antiserum and complement obtained from the guinea pig. The optimal condition for the activity of H-Y antibody was also investigated by culturing embryos under the concentraton or affected H-Y antibody was also investigated by culturing embryos under the concentration or affected H-Y antibody and culture rate. However, production of live young or sex rates of male and female from embryos transferred with psudopregnant. The biological test with the morula stage embryos showed that H-Y antibody was formed in all female rats immunized with spleen cell, but it was formed only in 80% female rats immunized with the antigen. When the bisected mouse embryos were cultured in vitro for 5~6 hours in morula stage, of 457 bisected embryos 81.4% of then were developed to the blastocyst stage. When the concentration rate of complement to H-Y antiserum varied from 1.0~5.0${mu}ell$, the lysis-rate of embryo was 19.5 to 67.3%. The concentration rate of complement did not influence the lysis-rate of embryos(P<0.05). The morphology embryos of bisected, zona-free and intact embryos showed the embryos lysis rate of 58.6, 42.7 and 48.5% respectively(P<0.05). Pregnancy rate were 50.0, 45.5 and 57.1% in psudopregnant recipient transferred with bisected, zona-free and intact blastocyst embryos. However, production of live youngs, sexual rate of male or female was 24(50.0:50.0), 22(45.5:55.5) and 36(58.3:41.7)mice, but affected and non affected half embryos with H-Y antiserum treatment was 23.1 and 26.7%. Also production of live youngs and sexual rate was 14(92.9:7.1) and 17(17.6:82.4)mice in affected and non affected half embryos in H-Y antiserum treatment(P<0.05).

Optimal Condition and Interspecific Cross-Reaction of H-Y Antibody Activity (H-Y항체활성의 최적조건과 종간교차반응)

  • These experiments were carried out to clarify the optimalconditions and interspecific cross reaction of H-Y antibody activity. H-Y antiserum was prepared in inbred SD female rats and Balb/c female mice by repeated immunization of rat newborn testis homogemate, rat and mouse spleen cells obtained from males of same strain. The activity of H-Y antibody in antiserum was tested by ELISA and biological tests. The cross reactivity of H-Y antibody was confirmed by culturing mouse and rabbit embryos in medium containing H-Y antibody and complement obtained from rat and guinea pig, respectively. The optimal condition for the activity of H-Y antibody was also investigated by culturing embryos in medium with different pH and complement concentration. The results obtained in these experiments were summarized as follows: 1. The formation rates of H-Y antibody in rats immunized with newborn testis and spleen cell were 40.0 and 50.0% respectively, and that in mouse immunized with spleen cell was 48.4%. 2. The activity of H-Y antibody was not affected by pH in range of 6.5 to 8.0, and the same was true for the relative concentration of complement to the H-Y antibody. 3. Minimum time needed for the activity of H-Y antibody was confirmed to be 0.5 to 1 hour and 24 to 48 hours respectively for the zona free embryos and intact embryos. 4. When mouse and rabbit embryos were treated with H-Y antibody obtained from rat, 46.4 and 54.8% of embryos were retarded or destroyed. From these results it could be said that H-Y antibody had strong interspecific cross reactivity.
