Removal of Endocrine Disrupting Chemicals in Wastewater by Nitrifying Sludge (질산화 슬러지에 의한 폐수 중의 내분비계 장애물질 제거)

  • The efficacy of nitrifying sludge existed in biological nutrient removal process was examined for possible removal of endocrine disrupting chemical(EDC) in the effluent of wastewater treatment plant. Some of ammonia oxidizing bacteria causes ammonia oxidation mediated by ammonia monooxygenase(AMO) activity, which has low substrate specificity resulting in cometablic degradation of several chemicals. In this study, the removal of three model EDCs such as bisphenol A(BPA), nonylphenol(NP) and dibutyl phthalate(DBP) was studied in batch cultures using nitrifying sludge, BOD-oxidizing sludge with low nitrifying activity, and sterilized sludge. Nitrifying sludge showed higher initial removal rates in all batches of three EDCs when it was fed with ammonium as an energy source. The acclimation time was required for the removal of EDCs in batches using BOD-oxidizing sludge or nitritefed nitrifying sludge. That retardation seemed to attribute to the slow growth of cells using the EDCs while ammonium-fed nitrifying sludge could degrade EDCs through simultaneous cooxidation with ammonia oxidation. Sterilized sludge was also tested under the same conditions in order to find the contribution of physical adsorption to the removal of EDCs. About 10~20% of initial EDCs dose was removed when using sterilized sludge. Thus the biological activity is likely to play major role for the degradation of BPA, NP, and DBP rather than the physical adsorption from wastewater.

Optimization of Culture Conditions for 1,3-propanediol Production from Glycerol Using Klebsiella pneumoniae (글리세롤로부터 1,3-propanediol 생산을 위한 Klebsiella pneumoniae 배양 조건 최적화)

  • To improve the productivity of 1,3-propanediol(1,3-PD) with K. pneumoniae DSM4799 using pure glycerol and crude glycerol derived from the biodiesel process, optimizing fermentation conditions was performed by changing environmental factors such as anaerobic/aerobic condition, temperature, glycerol concentration, and pH. When anaerobic conditions were maintained, there was an improved 1,3-PD production compared with that from aerobic/anaerobic 2-stage fermentation. From the results with temperature $26{\sim}37^{\circ}C$, the higher 1,3-PD production yield was observed at $30{\sim}33^{\circ}C$. For an initial glycerol concentration higher than 60 g/L, cell growth and 1,3-PD production were inhibited. When crude glycerol was used, the initial 1,3-PD production appeared to be inhibited. After 48 hr of incubation, however, 1,3-PD production with crude glycerol was even higher than that with pure glycerol, demonstrating the feasibility of 1,3-PD production using crude glycerol as a substrate. Fed-batch fermentation was applied for the high concentration of 1,3-PD without substrate inhibition. By regulating pH at 7 during the fed-batch with glycerol lower than 40 g/L, the yield of 1,3-PD was 25% higher than that without pH regulation(0.56 g/g vs. 0.45 g/g). In conclusion, based on our results, anaerobic conditions, temperature at $30^{\circ}C$, pure or crude glycerol lower than 40 g/L, and pH regulation at 7 were the optimized conditions for 1,3-PD production using K. pneumoniae DSM4799, making it more feasible to produce 1,3-PD at higher concentration and a lower price.

Expression and Purification of Recombinant Human Interferon-gamma Produced by Escherichia coli (대장균이 생산한 재조합 인체 감마인터페론의 발현과 정제)

  • For the production of the recombinant human interferon-gamma(rhIFN-${\gamma}$) in Escherichia coli, human glucagon and ferritin heavy chain were used as fusion partners. Even though rhIFN-${\gamma}$ is expressed as an inclusion body form in E. coli because of strong hydrophobicity of itself, over 50% of fused rhIFN-${\gamma}$ was expressed as soluble form in E. coli $Origami^{TM}$(DE3) harboring pT7FH(HE)-IFN-${\gamma}$ which encodes ferritin heavy chain-fused rhIFN-${\gamma}$. In the case of using glucagon-ferritin heavy chain hybrid mutant as a fusion partner, 6X His-tag was additionally introduced to N-terminus of GFHM(HE)-IFN-${\gamma}$ for enhancing purification yields of rhIFN-${\gamma}$. Fusion protein HGFHM(HE)-IFN-${\gamma}$ with two 6X His-tag was more effectively bound to Ni-NTA agarose bead than GFHM(HE)-IFN-${\gamma}$ with a 6X His-tag. rhIFN-${\gamma}$ was completely purified from enterokinase-treated HGFHM(HE)-IFN-${\gamma}$ by Ni-NTA affinity column. For high-level production of rhIFN-${\gamma}$, glucose was used as the sole carbon source with simple exponential feeding rate($2.4{\sim}7.2g/h$) in fed-batch process. The effective lactose concentration for the expression of the rhIFN-${\gamma}$ was $10{\sim}20mM$. Under the fed-batch culture conditions, rhIFN-${\gamma}$ production yield reached 11 g DCW/L for 6 hours after lactose induction.

  • Although Energy demands of modern society increase rapidly, current energy would be exhausted shortly. Therefore development of bio-ethanol production process from cellulose containing materials was extremly demanded. Therefore development of highly functional cellulase is requisite for this purpose. In this study cellobio-hydrolase (CBH1) gene from Trichorderma reesei was used to increase cellulase activity by directed evolution and highly functional cellobio-hydrolase was obtained and characterized.

Process Optimization of Ginseng Berry Extract Fermentation by Lactobacillus sp. Strain KYH isolated from Fermented Kimchi and Product Analysis (발효 김치로부터 분리한 Lactobacillus sp. Strain KYH를 이용한 진생베리 추출물 최적 발효 공정 확립 및 생성물의 특성 분석)

  • The pharmacological effects of ginseng berry have been known to improve psychological function, immune activities, cardiovascular conditions, and certain cancers. It is also known that fermentation improves the bioavailability of human beneficial natural materials. Accordingly, we investigated the optimal fermentation conditions of ginseng berry extract with strain isolated from conventional foods. We also analyzed the fermentation product and its antioxidant activity. The bacterium isolated from fermented kimchi was identified as Lactobacillus sp. strain KYH. To optimize the process, fermentation was performed in a 5 L fermenter containing 3 L of ginseng berry extract at 200 rpm for 72 hr. Under optimized conditions, batch and fed-batch fermentations were performed. After fermentation, organic acids, amino acids, sugars, ginsenosides, and antioxidant activity were evaluated. The optimum fermentation conditions were determined as pH 7.0 and a temperature of $30^{\circ}C$, respectively. After fermentation, the amounts and compositions of organic acids, amino acids, sugars, ginsenosides, and antioxidant activity were altered. In comparing the distribution of ginsenosides with that before fermentation, the ginsenoside Re was a major product. However, amounts of ginsenosides Rb1, Rc, and Rd were reduced, whereas amounts of ginsenosides Rh1 and Rh2 increased. Total phenol content increased to 43.8%, whereas flavonoid content decreased to 19.8%. The DPPH radical scavenging activity and total antioxidant activity increased to 27.2 and 19.4%, respectively.

Anaerobic Digestion Biochemical Sludge Produced from Municipal Sewage Treatment Process (하수처리시설에서 발생된 약품 잉여슬러지의 혐기성 소화 특성)

  • This study was carried out to get the characteristics of anaerobic digestion for chemical/biological sludge produced from municipal sewage treatment plant for phosphorus. Anaerobic mesophilic batch tests showed that the ultimate biodegradability of waste activated sludge showed 31%, PACl sludge 24%, Alum sludge 26%, respectively. At the S/I 1.0, 75% of total biodegradable volatile solids (TBVS) of waste activated sludge was degraded with an initial rapid decay coefficient, k1 of $0.1129day^{-1}$ and 74% of TBVS of PACl sludge with k1 of $0.0998day^{-1}$, and 76% of TBVS of Alum sludge with k1 of $0.1091day^{-1}$ for 20 days. During the operation of SCFMRs, the 3 reactor (Control, PACl, Alum) pH maintained 6.7~7.0 and the reactor alkalinity maintained 1,800~ 2,200 mg/L as $CaCO_3$. The average biogas production rates of SCFMRs fed with PACl sludge and Alum sludge were 0.089 v/v-d and 0.091 v/v-d, respectively, which was 27~28% lower than that of the control (0.124 v/v-d) at an HRT (hydraulic retention times) of 20 days. And the methane content during the operation ranged 70~76% in 3 reactor. The average TVS removal efficiency of SCFMRs fed with PACl sludge and Alum sludge were 19.6% and 19.9%, respectively, at an HRT of 20 days, which showed 4% lower than that of the control (23.8%). The average BVS removal efficiency of SCFMRs fed with PACl sludge and Alum sludge were 25.8% and 26.9%, respectively, at an HRT of 20 days, which was 8~9% lower than that of the control (34.5%).

Quality Characteristics of High Acidity Apple Vinegar Manufactured Using Two Stage Fermentation (2단계 발효에 의한 고산도 사과식초의 품질특성)

  • This study investigated the manufacturing conditions of apple vinegar with high acetic acid content following fermentation according to alcohol concentration without any nutrients. We compared and analyzed the quality characteristics of high acetic acid fermentation by varying the initial alcohol content (6%, 7%, 8%, and 9%). In the results, it was possible to manufacture high acetic acid vinegar with 12% titratable acidity and an alcohol content of 6% and 7%. Lower initial alcohol content was associated with higher yield due to a shorter lag phase. For quality characteristics of the high acetic acid apple vinegar, pH was 2.91~3.20, titratable acidity was 12.0%, and organic acid consisted of acetic acid, malic acid, critic acid and oxalic acids. Based on the results, high acetic acid apple vinegar was produced using a two stage fermentation process after alcohol fermentation but the further research is needed to reduce the time of fermentation in fed-batch culture for industrial use.

Anaerobic Treatment of Leachate Solubilized from Thermal Hydrolysis of Sludge Cake (하수슬러지 케이크 열수분해 탈리액의 혐기성 분해 특성)

  • This study was performed to evaluate the feasibility of anaerobic pretreatment for the leachate solubilized from thermal hydrolysis of sewage sludge cake. Overall process for the treatment of sludge cake consists of thermal hydrolysis, crystallization of magnesium, ammonium, and phosphate (MAP) for the leachate and anaerobic digestion of supernatant from MAP crystallization. The experimental evidence showed that the optimum ratio of Mg : P for the struvite crystallization of leachate solubilized from thermal hydrolysis of sludge cake was 1.5 to 1.0 as weight basis at the pH of 9.5. With this operational condition, the removal efficiencies of ammonia nitrogen and phosphorous achieved 50% and 97%, respectively. The mesophilic batch test showed that the ultimate biodegradability of the supernatant from MAP crystallization reached 63% at S/I ratio of 0.5. The readily biodegradable fraction of 90% ($S_1$) of the MAP supernatant BVS (Biodegradable Volatile Solids, $S_0$) degraded with $k_1$ of $0.207day^{-1}$ for the initial 17 days where as the rest slowly biodegradable fraction ($S_2$) of 10% of BVS degraded with $k_2$ of $0.02day^{-1}$ for the rest of the operational period. Semi-Continuously Fed and Mixed Reactor (SCFMR) was chosen as one of the best candidates to treat the MAP supernatant because of its total solids content over 6%. Maximum average biogas production rates reached 0.45 v/v-d and TVS removal efficiency of 37~41% was achieved at an hydraulic retention time (HRT) of 20 days and its corresponding organic loading rate (OLR) of 1.43 g VS/L-d.

Antimicrobial activities of Burkholderia sp. strains and optimization of culture conditions (Burkholderia sp. OS17의 항균활성 증진을 위한 배양최적화)

  • In this study, we isolated and identified bacteria from freshwater and soil collected from Osang reservoir, to screen antimicrobial bacteria against various pathogenic bacteria. 38 strains were isolated and assigned to the class Proteobacteria (22 strains), Actinobacteria (7 strains), Bacteroidets (6 strains), and Firmicutes (3 strains) based on 16S rRNA gene sequence analysis. Among them, strain OS17 showed a good growth inhibition against 5 methicillin-resistant Staphylococcus aureus subsp. aureus strains and Bacillus cereus, Bacillus subtilis, Filobasidium neoformans. As a result of the 16S rRNA gene sequence analysis, strain OS17 show the high similarity with Burkholderia ambifaria $AMMD^T$, B. diffusa $AM747629^T$, B. tettitorii $LK023503^T$ 99.8%, 99.7%, 99.6%, respectively. We investigated cell growth and antimicrobial activity according to commercial culture medium, temperature, pH for culture optimization of strain OS17. Optimal conditions for growth and antimicrobial activity in strain OS17 were found to be: YPD medium, $35^{\circ}C$ and pH 6.5. When the strain was cultured in LB, NB, TSB, R2A media at $20^{\circ}C$ and $25^{\circ}C$, the antimicrobial activity did not show. Culture filtrate of strain OS17 showed antimicrobial activity against 5 MRSA strains, Bacillus cereus, Bacillus subtilis, and Filobasidium neoformans with inhibition zones from 2 to 8 mm. Optimal reaction time was 48 h in YPD medium, 100 rpm and 0.3 vvm in 2 L-scale fed-batch fermentation process for antimicrobial activity. Culture optimization of strain OS17 can be improved on antimicrobial activity. Therefore, the antimicrobial activity of Burkholderia sp. OS17 had potential as antibiotics for pathogens including MRSA.

Fermentative Water Purification based on Bio-hydrogen (생물학적 수소 발효를 통한 수처리 시스템)

  • Among various techniques for hydrogen production from organic wastewater, a dark fermentation is considered to be the most feasible process due to the rapid hydrogen production rate. However, the main drawback of it is the low hydrogen production yield due to intermediate products such as organic acids. To improve the hydrogen production yield, a co-culture system of dark and photo fermentation bacteria was applied to this research. The maximum specific growth rate of R. sphaeroides was determined to be $2.93h^{-1}$ when acetic acid was used as a carbon source. It was quite high compared to that of using a mixture of volatile fatty acids (VFAs). Acetic acid was the most attractive to the cell growth of R. sphaeroides, however, not less efficient in the hydrogen production. In the co-culture system with glucose, hydrogen could be steadily produced without any lag-phase. There were distinguishable inflection points in the accumulation of hydrogen production graph that resulted from the dynamic production of VFAs or consumption of it by the interaction between the dark and photo fermentation bacteria. Lastly, the hydrogen production rate of a repeated fed-batch run was $15.9mL-H_2/L/h$, which was achievable in the sustainable hydrogen production.
