$\beta$-Sitosterol, campesterol and stigmasterol have been known to the phytosterols the most frequently found in plants. Metabolism of phytosterols was investigated using rat feces and liver microsomes. Feces were collected after phytosterols (a well characterized mixture of $\beta$-sitosterol 40%, campesterol 30% and dihydrobrasicasterol) were administered orally (0.5 ${g/kg$) to rats. Metabolites of phytosterols were identified using GC/MS. Three peaks were eluted at 12.47, 12.65, 12.87 min and had characteristic molecular ions m/z 428, 430, 432, respectively. Three fecal metabolites were identified as androstadienedione, androstenedione, and androstanedione. No metabolites could be detected in the rat liver microsomal reaction mixture. The results suggest that the metabolites of phytosterols in rat feces are formed by oxidation at 3- position, saturation at 5- and 6- position, and 17- side chain cleavage in the rat large intestine.
The cytotoxicity of the ethanol extracts of varietal samples of mulberry leaves and branches and silkworm feces was measured using CT-26 cells originated from murine metastatic colon cancer, using dye uptake assay in order to find potential anticancer agents. Two ethanol extracts (varietal mulberry leaves and mulberry branches) were prepared from 16 varietal mulberries and used as partial extract materials for the activity assay. Among these, the ethanol extracts from Shinkwangppong leaves showed a little anticancer activity, and those from Sugaeppong, Cheongunppong and Gumsulppong branches showed some anticancer activity as well as cytotoxicity. In contrast, ethanol extracts from freeze-dried, the 3rd day of 5th instar feces showed more potent anticancer activity than that of other mulberry leaves, mulberry branches and other 5th silkworm instar larva feces on the basis of high UV absorbance at 665 nm.
Human breast milk is a potential source of bacteria for the development of the intestinal microbiota of infants. Several species within the genera Lactobacillus and Bifidobacterium were demonstrated to shape the gut microbiota of infants. In this study, the bacterial diversity was investigated in the breast milk and feces of a mother-infant pair, and probiotic candidates were identified. Importantly, the novel L. gasseri EJL and B. breve JTL strains were isolated from breast milk and infant feces samples, respectively; their completed genome was resolved using de novo sequencing. In addition, the bacterial composition in the infant's feces at 1 week revealed the prevalence of Bifidobacterium and Streptococcus; a higher diversity was observed after 3 weeks. In particular, the abundance of Akkermansia was sharply increased at 7 weeks, further increasing thereafter, up to 15 weeks. Our results suggest that human breast milk and infant's feces are a source of probiotic candidates.
We have established a SYBR Green-based realtime PCR method using AnyDirect solution, which enhances PCR from whole blood, for direct amplification of the virA gene of Shigella flexneri and the invA gene of Salmonella typhimurium from human feces without prior DNA purification. When we compared the efficiency of conventional or realtime PCR amplification of the virA and invA genes from the supernatant of boiled feces supplemented with S. flexneri and S. typhimurium in the presence or absence of AnyDirect solution, amplification products were detected only in reactions to which AnyDirect solution had been added. The detection limit of real-time PCR was $1{\times}10^4\;CFU/g$ feces for S. flexneri and $2{\times}10^4\;CFU/g$ feces for S. typhimurium; this sensitivity level was comparable to other studies. Our real-time PCR assay with AnyDirect solution is simple, rapid, sensitive, and specific, and allows simultaneous detection of S. flexneri and S. typhimurium directly from fecal samples without prior DNA purification.
A PCR-aided monitoring of verotoxin-producing Escherichia coli (VTEC) was performed over the period of 12 months by using fresh feces collected monthly from 5 dairy cows that had been identified as VTEC carriers. The PCR products were confirmed to be verotoxin genes by Southern hybridization using a gene fragment of verotoxin 2 as a probe. Although seasonal variation of VTEC shedding seemed to depend on each cow, several factors may have influenced the frequency of detection. Shedding of VTEC tended to be reduced during grazing from the middle of May up to the beginning of October. Only one cow was positive for VTEC in August. Dry-off was also suggested to have a depressive effect on VTEC shedding, i.e. 3 of 4 dry cows showed no shedding of VTEC. Contrary to these factors, winter or indoor rearing tended to increase VTEC with only 5/24 samples being negative during the period from November to April. Total VFA concentration was higher (p<0.05) in VTEC-positive feces than in VTEC-negative feces, while fecal pH and VFA proportions were not different. Partial sequences of verotoxin genes from feces of 4 VTEC-positive cows were nearly identical (99-100%), suggesting that gut bacteria sharing the same gene were distributed among the cows. The present results indicate that grazing and dry-off could be factors which reduce VTEC shedding, while winter/indoor rearing may be a factor which increases the shedding, possibly through on-farm interactions.
Effects of unprocessed (whole) or steam-flaked corn with or without enzyme additives on in vivo nutrient digestibilities and distribution of corn particles in the feces of Holstein steers were determined in a $4{\times}4$ Latin square experiment using four Holstein steers fed the diets containing 1) whole corn without enzyme additive, 2) whole corn with enzyme additive, 3) flaked corn without enzyme additive, or 4) flaked corn with enzyme additive. With regard to nutrient digestibilities such as DM, CP, CF, NFE, NDF, and ADF, no significant differences were detected among treatments, and also the nutrient digestibilities were not affected by the addition of enzyme additive. When distribution of corn particles in the feces was examined, there were no significant differences in the amount of 2, 8 mm and total corn particles. However, feeding flaked corn resulted in less corn particles (4 mm) in the feces than feeding whole corn (p<0.05). There were no significant differences in amounts of corn particles in the feces due to the addition of enzyme additive.
Presently, viral isolation in the diarrheal feces can be reached by many tools such as fluorescent antibody test(FA), negative contrast electron microscopy(NCEM), virus neutralization test, cell culture, and so on. The purpose of the study was to aimed at the establishment of simplified NCEM technique which can be efficiently applied for diarrheal feces and also the understanding on prevalence of viral-induced diarrhea in calves. One hundred fourty-seven korean indigenous calves with diarrhea were examined to their feces by the modified NCEM. Among them, 98(66.7%) were confirmed to have one or more viruses in feces. The viruses detected were identified as rotavirus(33.3%), coronavirus(16.3% ), togavirus(10.2%) and herpesvirus(0.7%). Ten cases of combined viral infection were consisted of 8 with rotavirus+coronavirus, one with rotavirus+togavlrus and one with rotavirus+herpesvirus. Dirrheal types could classified by yello-wish watery(44.9a ), blood-tinged(19.7% ), white watery(17.7% ) , brownish watery(14.3%), greenish watery(3.4%) diarrhea, respectively. Yellowish watery diarrhea(66cases) was frequently included rotavirus(31.8%), coronavirus(15.2%), and togavirus(13.6%), respectively. Consequently, these results suggest that the modified NCEM is reliable and efficient diagnostic tool for detection of viruses in the diarrheal feces and many calves rearing in Chonnam province have been exposed to some enteric viral agents mainly including rotavirus and coronavirus.
The total global emission of $CO_2$ from soils is recognized as one of the largest fluxes in the global carbon cycle. Especially it is necessary to quantify the amount of $CO_2$ emitted by the organic material decomposition processes of microorganisms in the soil, because it becomes one of a factor for determining the carbon stocks in the soil. This study was conducted to estimate the impact of the Korean water deer(Hydropotes inermis)' feces to the soil organic matter. Also, effects of Korean water deer' feces on $CO_2$ emissions of soil and land use pattern dependent $CO_2$ flux quantification are studied. The organic materials in the Korean water deer' feces significantly changed organic matter content of soil and influenced the activity of soil microorganisms, both changing of respiration of the soil and physical chemical components in soil. In particular, C/N ratio and the $CO_2$ flux of soil of four regions (Rice paddy, Fallow ground, Salix koreensis community, Phragmites australis community) showed a statistically highly significant correlation (P<0.01) with the presence or absence of feces. $CO_2$ flux of soil affected by the feces was 2-20 times higher than the soil unaffected by the feces. This study has great significance to quantify the extent of the material circulation and its impact to the terrestrial ecosystem and soil zone throughout Korean water deer' feces. Feces of wildlife can affect soil and soil material circulation.

In order to find potential anticancer agents, we extracted pheophytin in the silkworm feces from various larval stages by water, chloroform and methanol extraction. The cytotoxicity of the pheophytin extracts of various silkworm feces was measured in the CT-26 cells originated from murine metastatic colon cancer, by dye uptake assay. The cytotoxicity of those pheophytins in 2nd, 3rd and 4th instars was better than remaining larval stages. The in vitro anticoagulant and fibyinolytic activities of ethanol extract from varietal mulberry leaves, mulberry branches and silkworm feces and pheophytin extracts from silkworm feces obtained at various larval stages were evaluated in order to find effective therapeutic drugs for the treatment of myocardial and cerebral thrombosis. The fibrinolytic activity was tested using the activated partial thromboplastin time (APTT) and thrombin time (TT) was measured for blood clotting activity. With regards to the fibrinolytic system, ethanol extracts of silkworm feces were better than varietal mulberry leaves and mulberry branches. The pheophytin extracts from 7th days of 5th instar contained the highest percentage of pheophytin and good fibrinolytic activity.
To investigate the effects of applying animal feces to the mulberry field on the chemical properties and mulberry leaf yield, poultry, pig and cattle feces were applied to the mulberry field making-up the sand loam from 1988 to 1990. The chemical properties of the mulberry field have been improved by the application of the poultry, the pig, and the cattle feces, increasing pH level, organic matter, P2O5 and exchangeable cation, especially Mg and K. Applying the pig feces and poultry feces to the mulberry field increased leaf yield during autumn rearing season, but not increased during spring rearing season. Applying animal feces to the mulberry field increased the content of total-cargbohydrate in the leaf in autumn.
