The effect of sulfur-containing amino acids on lipid metabolism was studied in rats fed casein as a protein source. Plasma cholesterol, HDL-cholesterol and atherosclerotic index decreased in the cysteine group compared to the methionine group. Plasma triglyceride and phospholipid level were not affected by the supplementation of the sulfur-containing amino acids. The levels of cholesterol and triglyceride in liver decreased by both methionine and cysteine. Cysteine increased the fecal excretion of coprostanol, total neutral steroid and bile acid. The results suggest that plasma cholesterol level is affected by dietary ratio of cysteine/methionine and that the hypocholesterolemic effects of cysteine is, at least in part, through reducing cholesterol absorption from small intestine and through enhancing fecal excretion of bile acids.
대두 펩타이드의 표면소수도가 혈청 콜레스테롤의 농도에 미치는 영향을 조사하기 위하여, 대두 단백질(ISP), 카세인(CNP), 이들 단백질을 펩신으로 가수분해하여 pH에 따른 펩타이드 침전 획분들(SHT, SH8, SH6, SH4, CHT, CH6, CH5, CH4)을 흰쥐에 섭취시키고 혈청 콜레스테롤 농도 및 분변 스테로이드이 배설량을 측정하였다. 그리고 각 펩타이드의 표면소수도를 ANS 형광법 및 SDS 결합법으로 측정하여, 이들의 상관관계를 분석한 결과, 펩타이드의 ANS 표면소수도가 높아질수록 분변으로 배설된 스테로이드량은 증가하였으며(r=0.81), 혈청 콜레스테롤 농도는 낮아졌다.(r=-0.868). 그러나 SDS 표면소수도는 그들과 상관관계가 없었다. 또한 대두 단백질은 가수분해에 의하여 ANS 표면소수도가 증가하였다. 이상의 결과는 흰쥐의 담즙염이, 소화중 생성된 높은 표면소수도의 펩타이드와 결합하여 체외로 배출되므로서 대두단백질의 섭취에 의한 혈청 콜레스테롤 농도가 낮아짐을 시사하였다.
본 연구는 총 4두의 삵에서 발정기와 비발정기, 임신기와 비임신기 동안의 분변에서 steroid metabolic materials를 추출한 후, TR-FIA kit를 이용하여 estradiol과 progesterone의 농도를 측정하였다. 발정기간 (2월) 중 estradiol 농도의 최저는 평균 $4.02{\pm}1.9$ng/g 이었고, 최고는 평균 $86.01{\pm}35.2$ng/g (dry fecal weight) 이었다. 비발정기(11월)의 최저는 평균 $4.42{\pm}1.32$ng/g 이었고, 최고는 평균 $15.62{\pm}6.84$ng/g 이었다. 임신기 (3월) 중 progesterone농도의 최저는 평균 $427{\pm}24.49$ng/g 었고, 최고는 평균 $1490{\pm}265.27$ng/g 이었다. 비임신기 (11월)의 progesterone의 최저는 평균 $71.25{\pm}29.61$ng/g 이었고, 최고는 평균 $291.75{\pm}90.30$ng/g 이었다. 위의 결과에 따라 삵에서 TR-FIA에 의한 분변내 steroid hormone 의 측정은 발정과 임신에 관련된 난소활동을 비 침습적으로 평가하기 위한 적절한 방법으로 판단되었다. 본 연구는 삵과 같이 접근이 어렵고 멸종위기에 처한 야생동물의 사육관리의 효율화와 번식계획의 수립에 도움이 될 것이다.
The present study was conducted to evaluate the usefulness of common Korean legumes as a high-fiber supplement in therapeutic diets for diabetic patients. Streptozotocin-induced diabetic rats were used as animal models and four kinds of legumes, black soybean (BS), yellow soybean (YS), green pea (GP) and soybean curd residue (SCR) were tested as high-fiber supplements. Seven groups of normal and streptozotocin-induced diabetic rats were fed isocaloric experimental diets containing 8% dietary fiber from one of four legumes or purified cellulose and pectin for 6 weeks. The effects of the legumes on the glucose and lipid metabolism of diabetic rats was examined and compared with the effects of cellulose and pectin. The legume supplementation did not show any beneficial effect on glucose tolerance, however, it exhibited a plasma cholesterol-lowering effect in diabetic rats. The cholesterol-lowering action was especially strong in BS and the degree of the effect was comparable to that of pectin. The levels of total lipids, cholesterol, and triglyceride in the hepatic tissues of rats fed legume diets were similar to those of the pectin group. All legume supplements induced an increase in fecal steroid excretion. The fecal cholesterol contents were significantly high following the supplementations of YS and SCR (p < 0.05). The excretion of fecal bile acids in the BS and YS groups was significantly higher than it was in the pectin group (p < 0.05). Concentration of lipid peroxidation products in the blood and urine of diabetic animals was lower in the legume groups than in the cellulose group. The levels of hepatic lipid peroxidation products were significantly lower in the BS and YS groups than in the pectin group (p < 0.05). From the results of this study, the plasma cholesterol-lowering effect of BS is possibly due to the significant (p < 0.05) in-crease in fecal steroid excretion, which suggests that BS could be beneficial in improving abnormal lipid metabolism in diabetic rats. The effect of Artemisia princeps var orientalis(mugwort) and Circium japonicum var ussuriense (Unggungqui) on lipid metabolism was examined . Thirty rats of 5 experimental groups were fed with a diet containing 1% cholesterol and 0.25% sodium cholate(control diet) and 5% plant powder or its water soluble extract(experimental diet) for 4 weeks. The activity of enzymes related to liver function, lipid components of liver and principle organs, and fecal steroids were assayed. The activity of enzymes was significantly lower in the unggungqui and mugwort powder diet group than in control. Liver total cholesterol , free cholesterol, and triglyceride levels were significantly lower in Unggungqui powder diet groups that in the control. The concentrations of total cholesterol , triglyceride and phospholipid in heart and kidney were comparatively low in the Unggungqui powder diet groups. In each plant powder diet group, the values of serum bile acid were significantly lowe ran the fecal steroid excretion was higher than in the control and the extract diet groups.
The purpose of the study was to find an effect of Medium Chain Triglycerids (MCT) diet on cholesterol metabolism in rat. Sprague-Dawley rats were fed two different diets containing MCT(trioctanoate) and corn oil respectively. After feeding to each group for four weeks, the levels of serum and liver cholesterol, the excretion rates of fecal and biliary steroids, and also bile acid composition were investigated. The results obtained from the study are as follows : (1) The average body weight gain in MCT group was almost same as that in the corn oil group. (2) The concentration of serum cholesterol in MCT group was lower than that in the corn oil group. Therefore it is confirmed that the cholesterol lowing action of MCT diet was practically high. (3) The concentrations of liver cholesterol and Triglyceride in MCT group were almost same as that in the corn oil group. Therefore it is thought that the level of liver lipids was not influenced by the difference of diet in this study. (4) The excretion rate of fecal neutral steroid in MCT group was significantly lower than that in the corn oil group, while the rate of fecal bile acid excretion was about same in both MCT and corn oil group. (5) The composition rates of fecal bile acid such as cholic acid, chenodeoxycholic acid and deoxycholic acid, a secondary acid of cholic acid, in MCT group were significantly lower than that in the corn oil group. (6) The excretion rates of biliary cholesterol and bile acid in MCT group were significantly higher than that in the corn oil group, while the composition rates of biliary bile acid such as chenoddeoxycholic acid and deoxycholic acid in MCT group were significantly higher than that in the corn oil group.
The present study was conducted to investigate the effect of sea tangle and hypoglycemic agent on lipid metabolism in normal and dabetic rats. Male Sprague-Dawely rats were fed AIN-76 based experimental diets containing 5%(w/w) cellulose or 15%(w/w) sea tangle for 3 weeks, after which diabetic groups were made diabetic by intramuscular injection of streptozotocin(STZ, 45mg/kg BW). Metformin(350mg/kg BW) as a hypoglycemic agent was given once a day using a feeding tube for 5 days. Body weight grains were reduced significantly by STZ treatment, but not influenced by metformin feeding. Blood glucosel levels in sea tangle groups were reduced, compared with those in cellulose groups. Metformin feeding showed the lowering effect of blood glucose. Plasma levels of triglyceride were increased significantly in diabetic rats, but decreased in metformin group by sea tangel feeding. Total cholestero contents showed a similar tendency with triglyceride, but were reduced in diabetic groups without metformin by sea tangle feeding. Plasma levels of HDL-cholesterol were reduced in diabetic rats, compared with those in normal rats. There was a significant increase in fecal weights in diabetic rats fed sea tangle. Fecal contents of cholesterol were lower in diabetic rats than in normal rats. In normal rats, it tended to increase by sea tangle feeding, but not significantly. Fecal excretions of coprostanol and coprostanone were reduced significantly in diabetic rats, compared with those of normal rats. It tended to increase in diabetic rats by simultaneous feeding of sea tangle and metformin, but not significantly. Diabetes reduced fecal excretion of bile acid, but it was increased by sea tangle and metformin feeding.
This study is conducted to evalute the effect of ginseng fraction component (ginseng extract solution, GES; ginseng protein, GP; ginseng saponin, GSA; ginseng residue, GR) upon hyperlipidemia and fatty liver induced by high fat administration. In doing so, the serum, liver and epididymal adpoid tissue have been examined for lipid components level and lipoprotein fraction. Feces bile acid and neutral sterol excretion have been also measured. 1'he results obtained from this study are as follows. 1. Serum, liver, epididymal lipid components of GP and GSA group were significantly lower than the controlgroup. 2. During the feeding experiment, VLDL and LDL increase while HDL decrease in all group. However the degree of VLDL and LDL increase and HDL decrease were signficantly small in GP and GSA group compared with control group. 3. In the excretion of bile acid and neutral sterol, all experiment group showed increased excretion in the comparison of control group.
흰쥐에 cholesterol(0.5%) 및 A. keiskei(5%)를 급여 한 후 혈장 및 간장 지질대사 및 분변 중의 neutral steroids 및 담즙산 배설에 미치는 영향을 관찰하였다. 혈장 cholesterol 농도는 식이중 cholesterol 첨가에 의해 현저히 증가되었으며, A. keiskei 급여시 유의적으로 감소하였다. 식이중 cholesterol 첨가는 혈장 HDL-cholesterol 함량을 유의적으로 감소시키는 반면 LDL-cholesterol을 크게 증가시켜 동맥경화지수를 높였다. A. keiskei 급여는 혈장 LDL-cholesterol의 유의적으로 감소시켰으며, 동맥경화지수도 cholesterol을 급여하지 않은 군에서 0.21, cholesterol 급여 군에서 0.52를 보여 A. keiskei를 급여하지 않은 각각의 대조군에 비하여 유의적 감소를 보여 A. keiskei가 혈장지질 개선효과를 보였다. A. keiskei 급여는 또한 cholesterol 급여와 관계없이 혈장 triglyceride 함량을 유의적으로 감소시키는 결과를 보였으나, 혈장 인지질 함량에는 영향을 미치지 않았다. A. keiskei 급여는 간조직의 total cholesterol, free cholesterol, cholesteryl ester 및 phospholipid 함량에는 영향을 미치지 않았으나, triglyceride 함량은 식이중 cholesterol 첨가와 관계없이 유의적으로 감소시켰다. A. keiskei는 cholestanol, 총 neutral steroid 및 담즙산의 배설을 증가시켰다. 이와 같은 결과로 보아 A. keiskei에 의한 혈장 cholesterol의 감소는 cholesterol 흡수의 감소 및 cholesterol을 cholestanol으로의 전환과 담즙산으로의 배설이 증가됨에 일부 기인하는 것으로 사료된다.
This study was carried out to determine the effect of dietary ${\gamma}$-linolenic acid on decreasing the plasma lipid levels and the thrombotic activity in rats. Sprague-Dawley male rats (B.W 120 g) were fed a experimental diet containing 5% lard (46.05% saturated fatty acids) , corn oil (51.36% linoleic acid) , evening primrose oil (EPO,72.80% linoleic acid and 9.16% ${\gamma}$-linolenic acid) or borage oil (BO,40.29% linoleic acid and 24.25% ${\gamma}$-liolenic acid) for 30 days. Although there were no significant differences in the food intake among the groups, the body weight gain of the BO group was significantly lower than that of the other groups. The bleeding time of the BO group was significantly longer than that of the other groups. There were significantly differences in the whole blood clotting time among the groups except for the EPO and corn oil groups, where the whole blood clotting time of the BO group was the highest among the groups, and that of the lard group was the lowest. The plasma triacyglyceride (TAG) , total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C) concentrations were the lowest in the BO group, but highest in the lard group, and there were significant differences among the groups. The plasma HDL-C concentrations were in the following order: BO, EPO, corn oil and lard groups and there were significant differences among the groups. The excretions of fecal neutial steroids and acidic steroids of the BO group were the highest among the groups, and there were significant differences compared to the other groups. The results suggest that dietary EPO and BO containing ${\gamma}$-linolenic acid has an antithrombotic activity, and inhibits the increasing of plasma TAG, TC and LDL-C concentrations compared to lard, which contains saturated fatty acids, or corn oil, which contains linoleic acid.
