An epidemiological survey on Cryptosporidium parvum infection of inhabitants in Chorwon-gun, Kangwon-do

  • The present study was undertaken to know the infection status of Cryptosporidium parvum among the residents of Chorwon-gun, Kangwon-do in 1993. Total 461 fecal samples were collected from the inhabitants residing in Chorwon-gun during the period of August 12 to September 14, 1993. Fecal smears were prepared by formalin-ether sedimentation, and examined after modified acid fast staining. Of the 461 fecal samples,9 (1.9%) were positive for C. parvum oocysts. The positive cases were limited to thirties (4) patients, forties (3), and sixties (2) , and no oocyst was detected in other age groups. The oocyst positive rate for male was 1.4% and that of female was 2.6%.

Fecal and Molecular Survey of Neospora caninum in Farm and Household Dogs in Mashhad Area, Khorasan Province, Iran

  • Neospora caninum is an important cause of abortion in dairy cattle worldwide. Dog is the definitive host for N. caninum and can infect dairy cattle. The aim of this study is to determine the prevalence of Neospora oocysts in feces of dogs from dairy farms. A total of 174 fecal samples was collected from 89 farm dogs and 85 household dogs during 2006 and 2008. Fecal samples of dogs were microscopically examined for detecting Hammondia Neospora-like oocysts (HNLO) by Mini $Parasep^{(R)}SF$ fecal parasite concentrator. HNLO were microscopically detected in 4 fecal samples (2.2%). The fecal samples with HNLO were examined by N. caninum-specific PCR. Two of the samples were positive for N. caninum. The 2 positive fecal samples were selected for inoculation to calves. Two inoculated calves were seronegative by ELISA for 4 months post-infection. This is the first report of finding N. caninum DNA in feces of farm dogs in Mashhad area, Iran.

  • 한국산 개로부터 Isospornohioenis를 분리하기 위하여 형태학적 특성을 관찰하였던 바 다음과 같은 결과를 얻었다. 즉, 전북지방의 개의 분편에서 검출된 Isospora spp. 중 I. ohioensis의 oocyst 하나를 분리하여 강아지에 경구감염시켜 얻은 oocyst를 다시 5두의 개에 감염시킨 후 형태학적 특성을 현미경하에서 관제하였다. 이의 잠복기는 4일이었으며 분편내에 배출된 oocyst의 크기는 $22.9{\;}{\times}{\;}19.8{\;}\mu\textrm{m}$ (R = 1.16)이었고 sporont의 크기는 $17 4{\;}{\times}{\;}16.3{\;}\mu\textrm{m}$ (R = 1.06)이었다. 96시간에 있어서 포자가 완전히 형성되었으며 oocyst 및 sporocyst의 크기는 각각 $22.8{\;}{\times}{\;}20.5{\;}\mu\textrm{m}$ (R = 1.11)과 $15.0{\;}{\times}{\;}10.8{\;}\mu\textrm{m}$ (R = 1.39)이었다. 포자형성 후에 oocyst 및 sporocyst의 크기는 거의 변화가 없었던 점으로 보아 개의 분편검사를 통한 Isosporc속의 감별진단을 위하여 96시간 정도의 포자형성기문이 필요할 것으로 예상되는 바이다.

Effects of Simple and Disposable Chicken Cages for Experimental Eimeria Infections

  • During experimental Eimeria infections in chickens, facilities are often contaminated by fecal oocysts known to be highly resistant to both chemical and enzymatic treatments. Thus, studies using experimental Eimeria infections have been limited due to the difficulty of complete elimination of residual oocysts from both cages and facilities. To overcome this limitation, simple, inexpensive, and disposable cages were constructed from cardboard boxes and tested during experimental Eimeria maxima infections. The cages were used in animal rooms with only a 1.7% evidence of coccidia contamination between adjacent cages. No significant differences in fecal oocyst output and body weight gain were noted between animals housed in disposable cages and animals housed in wire control cages. This cage design is a useful means for preventing oocyst contamination during experimental conditions, suggesting that this disposable cage design could be used for other avian infectious disease studies.

Anticoccidial Activity of Berberine against Eimeria-Infected Chickens

  • Avian coccidiosis has a major economic impact on the poultry industry, it is caused by 7 species of Eimeria, and has been primarily controlled using chemotherapeutic agents. Due to the emergence of drug-resistant strains, alternative control strategies are needed. We assessed anticoccidial effects of berberine-based diets in broiler chickens following oral infection with 5 Eimeria species (E. acervulina, E. maxima, E. tenella, E. mitis, and E. praecox). When 0.2% berberine, a concentration that does not affect weight gain, was added to the diet, the 4 groups infected with E. acervulina, E. tenella, E. mitis, or E. praecox showed significant reductions in fecal oocyst shedding (P<0.05) compared to their respective infected and untreated controls. In chickens treated 0.5% berberine instead of 0.2% and infected with E. maxima, fecal oocyst production was significantly reduced, but body weight deceased, indicating that berberine treatment was not useful for E. maxima infection. Taken together, these results illustrate the applicability of berberine for prophylactic use to control most Eimeria infections except E. maxima. Further studies on the mechanisms underlying the differences in anticoccidial susceptibility to berberine, particularly E. maxima, are remained.

경기남부지역의 닭에 있어서 Cryptosporidium baileyi의 감염상황 (Prevalence of Cryptosporidium baileyi Infection of Chickens In South Kyeonggi Area)

  • A total of 1050 layer and broiler chickens from 63 flocks of 21 poultry farms in South-Kyeonggi area, aged from 1 to 18 weeks, were investigated for the prevalence of Cryptosporidium baileyi infection from May 1992 to March 1993. The results have shown that 44 chickens(4.19%) were infected with C.(baileyi during the period of investigation) Fecal samples were treated by using Sheather's flotating technique and were examined under the light and phase contrast microscopy and then stained by Kinyoun method. Cryptosporidium oocysts were found in 35 out of 650 broilers(5.4%) and 9 out of 400 layer chickens(2.3%) aged mainly from 2 to 12 weeks. The regional infection rate were 4.7% in Pyeong-taek, 5.1% in An-sung and 2.3% in Yong-in, respectively. The average size of isolated oocyst was about $5.23{\times}4.92{\mu}m$ and the oocysts were orally inoculated into 2-day-old SPF chickens for the histological examination of oocyst in bursa of Fabricius. The study has concluded that C. baileyi infects chickens and oocysts were isolated in South-Kyeonggi area.

한약재의 첨가 급여에 따른 콕시듐 억제 효과 (Inhibitory effects of herbal medicine on coccidiosis)

  • This study examined the efficacy of natural ingredient formulated herbal medicine as feed additives against coccidiosis in broiler chicken. Medicinal herbs used in this study were Artemisia annua, Pulsatilla chinensis, Clematis chinensis and Scutellaria baicalensis. Two week old chicks were administered with 1% ingredient formulated herbal medicine. Observations of the efficacy were confirmed by examining the lesion score, histopathological appearance of cecum, shedding of fecal oocyst and production index in farm. S. baicalensis of the most effective substance were analyzed by UPLC. The results are as follows; among various types of Chinese medicine, the most efficacious ingredient was found to be pure bicalein. The major index substance is bicalein and $13{\mu}g/ml$ per gram was found. Feeding of chickens with S. baicalensis induced the coccidiostat effect and reduced the fecal shedding of E. tenella oocyst by 14%. Experimental results on a farm, feed conversion ratio and weight gain and production rate in the group with natural ingredient formulated additives than in control group increased. The rate of weight gain (%) in each experimental group showed 108% and 126% increase while the mortality rate was 1.4% lower than the control group. The investigation showed that the most efficacious ingredient was found to be S. baicalensis among herbal medicine products for coccidiostats. However, as it was found in the farm experiment that pure S. baicalensis alone is economically unviable. The additives with cocktailed of efficacious ingredient are suggested to be implemented for effective protection against coccidiosis.

An animal model using Eimeria live vaccine and to study coccidiosis protozoa pathogenesis

  • Cell culture systems for the protozoan Eimeria are not yet available. The present study was conducted to develop an animal model system by inoculating animals with a live Eimeria vaccine. This study was conducted on 3-day-old chickens (n = 20) pretreated with cyclophosphamide. The chickens were divided into 2 groups: the control group (n = 10) and the inoculated group that received the live Eimeria vaccine (n = 10). During the study period, we compared the clinical signs, changes in body weight, and number of oocysts shed in the feces of the control and inoculated group. This study showed that oocyst shedding was significantly higher in the chickens inoculated with live Eimeria oocysts than in the control chickens. Moreover, body weight gain was lesser in the animals in the inoculated group than in the control animals. Fecal oocyst shedding was observed in the inoculated animals. On the basis of these findings, we suggest that live Eimeria vaccination with cyclophosphamide pretreatment may be used to obtain an effective animal model for studying protozoan infections. This animal study model may eliminate the need for a tedious continuous animal inoculation process every 6 months because the live coccidiosis vaccine contains live oocysts.

한국산 동물로부터 크립토스포리디움의 분리 및 동정 III. 닭으로부터 Cryptosporidium baileyi의 분리 (Isolation and identification of Cyuptosporidium from various animals in Korea)

  • 한국산 닭으로부터 분리한 크립토스포리디움(Cryptosporidium)의 중형 오오시스트를 SPF 병아리에 경구투여하여 그 분변속에의 오오시스트의 크기 및 배출양상과 파브리시우스낭 조직에서 많이 발견되는 여러 발육기의 미세구조를 관찰하였다. 병아리에 있어서 prepateat period는 평균 5.9일간, patent period는 $12.87{\pm}3.4$일간, 오오시스트 배출 정점기는 접종후 $12{\pm}2.78$일째, 그리고 일반적으로 8일째부터 14일째까지 1주일간에 걸적 다수의 오오시스트가 분변으로 배출되었다. 이 원충의 거의 모든 발육기의 미세구조는 C. muris를 제외한 사람을 포함한 포유동물과 조류에서 이미 발견된 것들과 거의 비슷하지만 분변으로 배출되는 오오시스트의 크기는 Kinyoun 항산염색(변법)표본에서 $5.24{\pm}0.44{\times}4.86{\pm}0.37{\mu\textrm{m}}$, 오스윰산 증기고정 Giemsa 염색표본에서 $6.06{\pm}0.23{\times)4.86{\pm}0.34$\mu\textrm{m}$이었다. 이상의 연구결과를 기초로 하여 한국산 닭유래 크립토스포리디움을 C. baileyi라고 동정한다.

국내(國內)소의 Eimeria bukidnonensis와 E. wyomingensis의 감염율조사(感染率調査)와 형태학적(形態學的) 특징(特徵)에 관한 연구(硏究) (Prvalence and Morphological Identification of Eimeria bukidnonensis and E. wyomingensis in Cattle, Korea)

  • To determine the prevalence and the morphological identification of two Eimeria species in fecal examinations, a total of 1092 fecal specimens collected during the period of March to December in 1987 was tested by means of flotation technique. The results obtained were summarized as follow: 1. The prevalence of two species was revealed as 1.83% and 0.92% in E. bukidnonensis and E. wyomingensis, respectively. 2. The oocysts of E. bukidnonensis measured in range of $41.3{\mu}m$ to $49.4{\mu}m(mean\;45.5{\pm}1.7){\times}28.8{\mu}m$ to $35.3{\mu}m(mean\;32.2{\pm}1.2)$, and E. wyomingensis $36.9{\mu}m$ to $44.5{\mu}m(mean\;39.8{\pm}1.7){\times}26.5{\mu}m$ to $29.9{\mu}m(mean\;28.2{\pm}0.9)$. 3. The oocysts of E. bukidnonensis appeared darker and more robust than those of E. wyomingensis. 4. The striations in the oocyst wall of E. bukidnonensis were more peculiar than in E. wyomingensis. 5. Among the morphological characteristic, the widths could be reliably taken as a differential point of view for two Eimeria species.
