Characterization of sediment quality is important for the proper management of surface water quality, yet sediment has not been monitored sufficiently. In this study, fecal indicator microorganism concentrations of sediments in the Geum River Basin were monitored. Sampling was carried out at one paddy field, one lakeshore and five monitoring stations in the lower reach of the Geum River Basin. Surface waters and sediments were sampled four times during rainy season. Total coliform concentrations of sediments were 12 times higher in average to those of surface waters while E. coli concentrations of sediments were six times higher. No correlation found between indicator microorganism concentration between surface waters and sediments.
Seafood, if eaten raw, carries the risk of food poisoning. Seafood poisoning is often caused by pathogenic microorganisms originating from fecal contamination, such as Salmonella sp. and norovirus. Fecal coliforms are an important indicator of fecal contamination. Therefore, data on fecal coliforms are very important for evaluating the safety of fisheries in coastal areas. In this study, 2,226 sea water samples were collected from the southeast coast of Korea, and total and fecal coliforms were compared and analyzed. Total coliforms were detected in 76.5% of the samples and 71.4% of the total coliforms were fecal coliforms. At sea water temperatures above $20.0^{\circ}C$, total coliforms were found in 78.8% of the samples and fecal coliforms constituted 72.0% of the total coliforms. In sea water below $19.9^{\circ}C$, the respective values were 74.6% and 70.9%. These results suggest that temperature does not have a significant effect on the detection of fecal coliforms. When the salinity exceeded 30.0%o, total coliforms were found in 72.1% of the samples and fecal coliforms constituted 66.0% of these. At salinities below 29.9%o, the respective values for total and fecal coliforms were 90.4% and 85.2%. These results strongly suggest that the detection of fecal coliforms is proportional to the amount of precipitation.
Objective: This study was performed to detect indicator bacteria in drinking spring water samples in Gwangju City and to identify their genus using the VITEK-II system. Methods: The subjects were ten drinking spring water sites in Gwangju. Samples of spring water were taken every month from September 2019 to August 2020. We analyzed for the indicator bacteria Yersinia and microorganisms isolated from the spring water. Result: According to the research results on indicator bacteria, general bacteria in st1-st7 with sterilization facilities in the spring and summer were investigated in the range of 0-2 CFU/mL and 0-12 CFU/mL. In st9, where a sterilization facility was not installed, the most general bacteria were detected (160 CFU/mL). Total coliform and fecal coliform showed unsatisfied rates of 16.7 and 11.1% in spring and 14.7 and 11.8% in summer, respectively. The unsatisfied rates of total coliform for the designated and non-designated spring water facilities were 3.8 and 47.1%, respectively, and for the fecal coliform group they were 2.5 and 35.3%. The difference was confirmed according to the presence of a sterilization facility. Yersinia spp. was not detected in all drinking spring water. Forty-one strains in 25 species were isolated from ten sites. The results classified as major dominant species are Pseudomonas spp. 14.6%, Pantoea spp. 9.8%, Serratia spp. 9.8%, Acinetobacter spp. 9.8%, Citrobacter spp. 7.3%, Bordetella spp. 7.3%, Delftia spp. 4.9%, and Enterobacter spp. 4.9%. Conclusions: Based on the result that various species derived from fecal pollution and artificial pollutants were detected in the non-specified public spring water facilities that many people use, the facilities need institutional complements such as continuous management or complete shutdowns.
Water quality and the distribution of ammonia oxidizing bacteria were characterized in constructed wetland of Shihwa lake. Both physico-chemical parameters and fecal indicator microorganisms including total coliforms, E.coli, Enterococcus spp. were measured. In addition, denaturant gradient gel electrophoresis (DGGE) was carried out after PCR amplification of amoA gene from input, output, and wetland sites of the Banwol, Donghwa, and Samhwa stream in Shihwa lake area. Physico-chemical parameters were in proper range for typical nitrifying bacteria to grow and perform their biological activities. Average concentrations of fecal indicator microorganisms of wetland samples were lower than those of input sites. These results suggested that microbial water quality improved by the process of constructed wetland. According to phylogenetic information obtained from DGGE from study sites, distribution of nitrifying bacteria from each of input, output, and wetland were generally distinctive one another. In addition, distribution of nitrifying bacteria between Banwol and Donghwa streams showed higher similarity (52.6%) than this of Samhwa stream (15.2%). These results indicated that characteristics of ammonia oxidizing bacteria in Samhwa were unique in comparison with those of Banwol and Donghwa stream.
The aim of this study was to compare crop contamination between two irrigation methods using microbial-contaminated water. The effect of relative humidity on microbial survival of the three indicator microorganisms was also investigated. Escherichia coli ATCC 25922, Clostridium perfringens ATCC 3624, and coliphage PRD1 were applied to irrigation water to grow cantaloupe, lettuce, and bell pepper. Half of the sixteen plots were subsurface drip irrigated (SDI) and the other half were furrow irrigated (FI). Two relative humidity levels were controlled at 15-65 % and 55-80 % for the dry and humid condition experiments, respectively. Samples of produce, surface soil, and subsurface soil at a depth of 10 cm were collected over a two-week period following the application of the study microorganisms. Overall, greater contamination of both produce and soil occurred in the FI plots. For the SDI plots, preferential water paths and resulting water appearance on the seed beds seemed to be responsible for produce contamination. Relative humidity levels did not appear to affect microbial survival in soil. PRD 1 showed lower inactivation rates than 5. coli in both dry and humid conditions. C. perfringens did not experience significant inactivation over the experimental period, suggesting this microorganism can be an effective indicator of fecal contamination.
참굴(Crassostrea gigas) 수하식양식에 있어 상층과 저층 그리고 조석에 따른 해수 및 굴의 위생학적 성상에 관한 변화를 조사하였으며 본 연구는 미국국제연맹처(AID) 연수계획에 의하여 Washington주 Burley Lagoon 뗏목 수하식 양식장에 대하여 실시되었다. 해수의 수온, 염분 및 탁도는 저층이 상층보다 다소 높았고 그 차이는 수온 $0.3^{\circ}C$, 염분 $0.5\%_{\circ}$, 탁도 0.1 JTU 있다. 조석에 따를 수온 및 염분의 변화는 대체로 조석 주기에 일치하는 기복상을 보였다. 해수의 위생지표미생물 함양은 상층이 저층보다 대체로 높았고 일반적으로 Coliform MPN 및 Fecal coliform MPN의 변화는 조고에 역비례하고 있어 이들 미생물군의 출처는 이 해역으로 유입하는 하천임을 알 수 있다. $35^{\circ}C$ Plate count는 조석 변동에 대하며 보다 안정하였다. 굴에 있어 Coliform 및 Fecal coliform MPN의 평균치는 상층굴이 저층보다 일반적으로 높은 값을 시현하였으나 $35^{\circ}C$ Plate count는 이와 같은 경향을 나타내지 않았다. 해수에서와 같이 굴에 있어서도 Coliform 및 Fecal coliform MPN의 평균치는 양자 모두 호고에 역상관관계를 나타내었고 조사기관중 굴의 세균축적비는 Coliform MPN은 $8.6\~19.7$, Fecal coliform MPN은 $16.9\~44.3$이였다. 이상의 결과로서 굴수하식 양식에 있어 패류위생관리상 중요한 것은 창호시에 굴을 채취하는 것이 미생 지표 세균의 함량이 보다 낮다는 것을 들 수 있다.
Photoreactivation of microorganism following UV-disinfection is one of the research topics of interest in assessing the UV-disinfection performance. Apparent photoreactivation was examined under fluorescent lamp and solar radiation as well as in darkness. Total coliform, fecal coliform, and Escherichia coli were used as indicator microorganisms, and their concentration was monitored with time after UV-disinfection. Under the darkness, their initial concentration of 10∼30 MPN/100 mL increased to the level of 100 MPN/100 mL after 24 hours, which implied that part of damaged microorganisms by UV-disinfection might be repairable with time. Under the fluorescent lamp, photoreactivation was more apparent that their concentration increased up to 1,000 MPN/100 mL which might significantly impair the water uses specially in reuse of reclaimed wastewater. However, their concentration further decreased down to below 2 MPN/100 mL under the solar radiation primarily due to additional disinfection by solar radiation rather than photoreactivation. Samples not disinfected by UV-disinfection also demonstrated substantial decrease of their concentration under solar radiation from about 5,000 MPN/100 mL to less than 30 MPN/100 mL in 24 hours. But direct reuse of effluent without disinfection is not recommended because natural decay by solar radiation may take time and be affected by climatic conditions. The result suggests that photoreactivation of pathogenic microorganisms may not be concerned in agricultural reuse of reclaimed wastewater because solar radiation may provide further disinfection after UV-disinfection.
농지-임야유역의 비점원으로부터 발생하는 미생물학적 오염물질 부하량을 추정하기 위하여, 농지와 임야가 혼합된 3개 시험유역에서 동일한 2개 강우사상에 대한 지표미생물 유출 특성을 조사하였으며, 지표미생물항목은 대장균군(total coliform: TC), 분원성 대장균(Fecal coliform: FC), 대장균 (Escherichia coli: EC), 분원성 연쇄상구균(Fecal streptococcus: FS)이었다. 농지-임야 유역의 강우시 유량변화에 따라 토사유실로 인하여 부유물질 농도가 상당히 증가하였다. 지표미생물 농도는 유량변화와 상당히 밀접한 관계를 보였다. 대부분 오염되지 않은 임야로 구성되어 있는 첫번째 유역의 강우유출수 TC EMC(Event Mean Concentration)는 $5.3{\times}10^3CFU/100ml$이었으며, FC EMC는$1.4{\times}10^3CFU/100ml$, EC EMC는 $1.1{\times}10^3CFU/100ml$, FS EMC는 $3.9{\times}10^2CFU/100ml$이었다. 임야유역과 농지유역이 혼합되어 있는 제 2 유역의 지표미생물에 대한 EMC는 TC EMC가 $1.7{\times}10^5CFU/100ml$, FC EMC가 $8.5{\times}10^4CFU/100ml$, EC EMC가 $8.9{\times}10^4CFU/100ml$, FS EMC가 $3.4{\times}10^4CFU/100ml$로 나타났다. 농지와 임야가 혼재되어 있으나, 유역면적이 큰 제 3 시험유역의 지표미생물에 대한 EMC는 TC EMC가 $1.9{\times}10^5CFU/100ml$, FC EMC가 $9.6{\times}10^4CFU/100ml$, EC EMC가 $7.0{\times}10^4CFU/100ml$, FS EMC가 $5.1{\times}10^4CFU/100ml$로 나타났다.
In this study, we evaluated the impacts of land-based pollution sources on seawater and shellfish in the Jindongman area after 20.5 mm and 90.6 mm rainfall events. We analyzed sanitary indicator microorganisms used in survey management, such as total coliform, fecal coliform, Escherichia coli, and male-specific coliphage in Waste water treatment plant (WWTP), major inland pollution source,s seawater, and shellfish for 4 days after rainfall events. Our results showed that the range of coliform group and fecal coliform was 1.8-49 and <1.8-4.5 MPN (most probable number)/100 mL, respectively, after rainfall events in WWTP discharge water. Furthermore, the radius of the calculated impacted area of major inland pollution sources ranged from 5 to 798 m after 20.5 mm of rainfall and 30 to 1,031 m after 90.6 mm of rainfall. The fecal coliform of seawater at 30 stations in the shellfish growing area and areas adjacent to four stations was <1.8-130 and from <1.8-79 MPN/100 mL, respectively. The E. coli level of shellfish at 7 stations in the shellfish growing area was <18-220 MPN/100 g.
이 연구에서는 하수처리수를 농업용수로써 재이용시 발생할 수 있는 위생문제를 검토하기 위해 하천수, biofilter 유출수, 그리고 UV소독수를 관개한 벼재배 실험포트에서 지표미생물의 밀도변화를 조사하였다. 우리나라 하수처리장 유출수 수질기준을 살펴보면, USEPA에서 정하고 있는 농업용수 재이용 수질기준 항목인 $BOD_5$와 SS의 수질기준은 만족시키지만, 대장균의 경우추가적인 처리가 필요한 것으로 나타났다. 벼 식재 직후인 5월과 6월의 경우 관개 후 24시간 동안의 평균밀도가 $10^2{\sim}10^4$의 범위에서 관개용수의 밀도와 상관없이 지표미생물의 밀도변화는 크게 나타나지 않았다. 이 시기에는 기비와 추비로 인해 미생물의 성장에 필요한 영양물질, 교란된 저질의 높은 미생물 밀도 등에 영향을 받고 논의 벼 생육생태가 덜 성숙되었기 때문으로 판단된다. 그러나 7월과 8월은 처리구별로 밀도차이가 컸고 24시간 동안 변화폭도 컸으며, 관개용수 미생물밀도에 영향도 일부 나타났다. 실험기간동안 지표미생물의 밀도를 관개용수별로 분산 분석한 결과, UV소독 후 관개한 경우가 하천수와 biofilter 유출수를 사용한 경우보다 낮은 밀도를 나타내었다. 실제 논농사가 이루어지고 있던 양평군의 하천관개 논과 여주군의 지하수관개 논의시료분석에서 지표미생물 밀도는 FC, E. coli밀도가 TC밀도보다 크게 낮았으며, 시기별로 밀도의 차이가 있으나, TC, FC, 그리고 E. coli의 평균밀도가 약 4,000, 400,300 MPN/100mL범위를 나타내었다. TC의 밀도는 본 실험포트 실험과 유사한 범위이었으며, FC와 E. coli는 실험포트에서 초기 경운시 사용된 biofilter 유출수의 영향으로 실제 논보다 높은 값이었다. 이앙기 때에는 특별히 논 표면수와 신체접촉이 많은 점을 고려할 때 하수처리수만을 관개하여 이앙준비를 하는 것보다 하수처리수를 보조관개용수로 사용하는 것이 보다 안전할 것으로 판단된다. 하수처리수를 농업용수로 재이용할 경우 하수처리수를 관개용수로 직접 이용하기보다는 UV소독으로 보건 위생학적으로 높은 안정성을 확보할 수 있으며 하수처리수재이용 수질기준에도 적합할 수 있을 것이다. 그러나 벼의 생육에 영향을 줄 수 있는 영양물질과다로 인한 도복문제, 하수처리수에 존재할 수 있는 독성물질에 의한 영향 등 하수처리수를 관개강수로 이용하는 데에는 종합적이고 과학적인 고찰이 필요하며, 본연구의 결과는 하수처리수를 농업적으로 이용할 경우 공공의 보건 ·위생적인 안전을 확보하기 위한 기초연구로서의 성격을 갖는다고 할 수 있다.
